24-Epibrassinolide Modulate Cellular and Organogenic Response of Explants of Brassica Species, in vitro Culture

  • Brassinosteroids are steroidal plant hormones and are known to modulate physiological and cellular response in a wide range of plant species. Considerable insights has been achieved of the physiological role of brassinosteroid in Brassica species in the past few years, but their effect on direct organogenesis has not been extensively studied. In this sense, under optimal basal media and growth conditions we tested the cellular and organogenic response of 24-epibrassinolide (EBL) in a variable concentration (0.1 to $5.0\;{\mu}M$) and Zeatin (Z) (1.0 to $100\;{\mu}M$) and their synergic effect on hypocotyl explants of cauliflower and broccoli. The isolated EBL accelerated cell elongation and promotes direct organogenesis. One micromolar EBL + $10\;{\mu}M$ of Z was the most efficient combination for cell elongation, cell differentiation as well as for organogenesis. A suppressing effect on root induction was confirmed for all the tested hormone levels. The general results indicate a synergic effect of EBL-Z and EBL potentates Zeatin activity, at least in certain tissues. Besides de genetic factors, we can speculate that the natural hormone concentration in the explants might affect the responses by application of exogenous growth regulators. Experiments with new plant growth regulators, like brassinolide, are important aiming to maximize or accelerate plant regeneration for in vitro multiplication or for genetic transformation.

Effect of plant growth regulators on plant regeneration from the Sedum rotundifolium D. Lee (둥근잎꿩의비름(Sedum rotundifolium D. Lee)의 식물체 재분화에 미치는 식물생장조절제의 영향)

  • To establish the system of In vitro plant regeneration, the floral bud and leaf explants of Sedum rotundifolium were cultured on the MS media supplemented with different concentration of 2,4-D, NAA, and BA. The callus induction was more effective in the floral explants than the leaf explants, and was the best on MS medium containing 1.0 or 2.0 mg/L 2,4-D and 1.0 mg/L BA. The highest numbers of shoots were regenerated when callus were cultured on MS medium containing 2.0 mg/L 2,4-D and 1.0 mg/L BA for 8 weeks. The normal root formation from shoot was effective on the MS medium containing IAA alone. The regenerated plantlets were transferred to the pot and acclimatized successfully.

  • Iris odaesanensis Y. N. Lee. is an important endangered and native plant belonging to the family Iridaceae in Korea. This study describes a method for rapid micropropagation of this species via from leaf, rhizome and root explants derived calli. Leaf, rhizome and root explants were cultured on Murashige and Skoog (MS) medium supplemented with 2,4-dichlorophenoxy acetic acid (2,4-D) for callus induction. Rhizome explants yielded calli at a frequency of 72% when cultured at 1.0 mg/l 2,4-D. Calli were maintained at 1.0 mg/l 2,4-D. These calli were transferred to MS medium supplemented with 0, 0.5, 1.0, and 2.0 mg/l 2,4-D in combination with 0, 0.5, 1.0, and 3.0 mg/l BA for adventitious shoot induction. The highest number of adventitious shoot (228.9 per petri-dish) were formed at 1.0 mg/l 2,4-D and 1.0 mg/l BA. WPM medium was the best to convert calli into plantlets, where up to 98.2% of calli were regenerated into plantlets. This in vitro propagation protocol should be useful for conservation of this endangered plant.

Correlation between in vitro Flowering Frequency and the Structure of Cytokinins in Ginseng (Panax ginseng C.A. Meyer) (인삼 (Panax ginseng C.A. Meyer)의 기내 화아형성 빈도와 cytokinin 구조와의 관계)

  • To elucidate the relationship between in vitro flowering and the structure of cytokinins in ginseng (Panax ginseng C.A. Meyer), zygotic embryos, seedlings, and cotyledonary nodes were cultured on MS medium supplemented with 5 $\mu$M of various cytokinins (BA, kinetin, 2-iP, and zeatin) with or without GA$_3$ (5 $\mu$M). The frequency of in vitro flowering was the highest when explants were cultured on the medium containing BA regardless of the kinds of explants, followed by kinetin, 2-iP, and zeatin. Flowering frequency of cotyledonary node explants was significantly increased by the combined treatment of cytokinin and GA$_3$. Flowering frequency was highly correlated with the logP of cytokinins, indicating that the lipophilicity of each cytokinin may involved in the in vitro flowering of ginseng.

  • Effects of plant growth regulators and condition on shoot induction from internode explants of Actinidia arguta were investigated. Optimal condition for shoot induction was obtained when internode explants were cultured on MS medium containing 3.0 mg/L zeatin. The frequency showed about 62.5% under darks condition at 27$^{\circ}C$ after 6 weeks of culture. Regenerated shoots were rooted on WPM medium supplemented with 0.5 mg/L NAA and the regenerants were acclimated to an artificial soil with mixture (perlite : vermiculite = 1:1, v/v) for further growth and successfully transplanted to the soil in a highly humidified greenhouse. Histological observations revealed that meristemoids were formed from small and isodiametric cells, and then developed to shoot primordia.

  • A propagation for Fatsia japonica using axillary bud explants were established. Cultures were initiated from axillary bud explants on MS medium supplemented with IAA $(1,2,3\;mgl^{-1})$, 2,4-D $(1,2,3\;mgl^{-1})$ or NAA $(1,2,3\;mgl^{-1})$ in combination with BA $(0.5\;mgl^{-1})$. The maximum shoot bud formation was obtained in MS medium supplemented with $0.5\;mgl^{-1}$ BA and $2;mgl^{-1}$ IAA after 4 weeks culture. The microshoot rooted within 4 week in MS medium containing $1.0\;mgl^{-1}$ IBA.

  • In order to investigate the possibility of in vitro mass propagation via somatic embryogenesis from Sicyos angulatus L., effects of plant growth regulators and carbon sources on callus induction and somatic embryogenesis were evaluated. Optimal combinations of plant growth regulator for callus induction from cotyledon and inflorescence explants were 2,4-D 2.0 mg/L + BA 0.1 mg/L and 2,4-D 1.0 mg/L + BA 0.1 mg/L in MS basal medium supplemented with sucrose 30 g/L,, respectively. Somatic embryogenesis was observed from cultured inflorescence explants, but it could not be achieved from leaf or cotyledon explants. The most effective plant growth regulators for somatic embryogenesis from callus was NAA 1.0 mg/L + kinetin 10 mg/L in the half strength of MS basal medium supplemented with 20 g/L sucrose.

Enhanced Development and Germination of Carrot Somatic Embryos on Modified Surface of Medium (당근체세포배의 발생 및 발아에 미치는 배지표면의 물리적변화)

  • Carrot cotyledon explants cultured on MS medium with 1 mg/L 2,4-D were transferred to a hormone-free solid medium overlaid with filter paper in order to elucidate the effect of simple physical treatment on the development and germination of somatic embryos. Transfer of the explants cultured for one week on MS basal medium overlaid with 3 sheets of filter paper on to MS basal medium increased somatic embryo production 2-39 times over the one week culture on medium without filter paper overlay. Maturation and germination of somatic embryos was more prominent on medium overlaid with filter paper than on medium without filter paper. The explants cultured for one week on filter paper overlay added with liquid medium showed prominent decrease in somatic embryo formation compared to filter paper overlay only. It is suggested that the filter paper overlay affected the moisture environment of the somatic embryos developing on it.

  • Callus and shoot formation of leaf explants from in vitro propagated shoots and field grown plants depending on the position of leaf, and four boxthron cultivars were investigated. Callus formation of explants from both in vitro shoot and field grown plants as easily achieved at the cut surfaces of explants but the callus formed from leaf of in vitro shoots was hardened as the duration of culture was proceed. Calli were effectively induced from leaves detached from the middle position of both in vitro and in vitro plants on MS medium containing 0.5 mg/L NAA with 0.2 mg/L BA, and the growth of calli were better in field grown leaves than in vitro grown leaves. Shoot formation were effectively induced from leaves detached from the upper position in vitro plants, and the middle parts of in vitro plants on MS medium containing 0.01 mg/L (NAA with 0.2 mg/ BA. There was difference on the frequency of shoot formation among four different cultivars; 'Jindojaerae' was the best for shoot formation followed by 'Cheonyang', 'Younghagukija' and 'Cheongyangjaerae'.

  • To establish the system of in vitro plant regeneration, the different explants (stem with axillary bud and stem without axillary bud) of Hylotelephium ussuriense were cultured on the Murashige and Skoog's medium containing 6-benzylaminopurine (BA) and indolebutyric acid (IBA). The adventitious shoot induction was more effective in the stem with axillary bud explants than the stem without axillary bud explants, and was the best on MS medium containing 3.0 mg/L BA and 0.01 mg/L IBA. Frequency of plantlet growth was not significantly treated on MS and sucrose. Total chlorophyll contents under ventilation treatment were higher than those in control (non-ventilation). This in vitro propagation protocol will be useful for conservation and mass propagation of this endangered plant.
