Effect of Morphological Factors, Antibiotics and Agrobacterium Co-cultivation in the Efficiency of Somatic Embryogenesis of Eggplant (Solanum melongena L.)

  • Induction of somatic embryogenesis from Brazilian eggplant variety F-100 was studied in response to four auxin types. NAA, at the optimal concentration of 54 $\mu\textrm{m}$, was the only one that resulted in the induction of somatic embryos in either leaf and cotyledon explant and, at murk lower intensity and frequency, in hypocotyl and epicotyl explants. The optimal temperatures for embryo induction were 28 and 35$^{\circ}C$ for cotyledon and leaf explants. Incubation at 22$^{\circ}C$ caused a significant reduction both in the frequency and intensity of induction. This system was used to study the effects of position and orientation of the tissue on the culture medium as well as of antibiotics and explant co-cultivation with Agrobacterium on the efficiency of somatic embryo induction. The intensity of embryo induction was greater in the midsections of cotyledons relative to apical and basal regions, when the abaxial surface was in contact with the culture medium. The presence of antibiotics resulted in approximately 40-60% reduction of embryo induction relative to control explants, which originated 335$\pm$26.6 embryos. Co-cultivation with Agrobacterium before treatment with antibiotics caused a more drastic reduction (80-99%). Ampicilin treatment after cocultivalion with Agrobacterium caused the least inhibitory effect, allowing the production of 60 embryos/explant.

Adventitious Bud Induction and Plant Regeneration from Cotyledon Explants of Camellia japonica L. (동백 자엽절편으로부터 부정아 형성을 통한 식물체의 재분화)

  • Culture conditions for plant regeneration of Camellia japonica were achieved by organogenesis in explants of cotyledon. Seed cotyledons were cultured on MS medium containing various auxin, 2,4-D or NAA and cytokinins BA. The adventitious shoot buds were efficiently formed without embryogenesis on the basal region of cotyledon cultured on MS medium supplemented with $0.1\;{\mu}g/m{\ell}$ 2.4-D and $1\;{\mu}g/m{\ell}$ BA. Seed cotyledons could be used as a source of explants in experiments of genetic transformation of the genotypes evaluated for improving the efficiency of production of transgenic Camellia plants.

Production and Rooting of Shoots in Bioreactor Culture of Scrophularia buergeriana Miquel (생물반응기에서 현삼의 신초 형성과 발근)

  • This study was carried out to know the factors affecting on shoot formation and rooting for stable and routine production of plantlets in bioreactor culture of Scrophularia buergeriana. Multiple shoots were formed effectively when explants were transplanted on the MS media with decreased concentration of $NH_4NO_3$ as 413mg/ l . Three hundred stem explants (0.8-1.0cm) was appeared as proper inoculation size in bioreactor culture. IBA (0.05mg/L) was more effective for rooting of the shoots in liquid as well as solid media. Six weeks long culture of explants in bioreactor gave better shoot shape for rooting on solid half-strength MS media.

  • Multiple shoots were induced from in vitro shoot originated from winter bud of P. yedoensis from Jeju. Most explants grow in similar type among the five different media but affected by supplement of sucrose regardless of media. For mass-propagation various concentrations of BAP or $GA_3$ were treated in the medium respectively. BAP was very effective to produce multiple shoots and 3.5~9.5 shoots were formed on the explant. The shoots induced on the high levels of BAP have short internodes. No shoots were induced on the treatment of $GA_3$ but roots were induced on it. When $GA_3$ was supplemented with the medium containing BAP, multiple shoots were produced from the explants. The medium(WPM) containing with $0.5mg/{\ell}$ BAP and $4.0mg/{\ell}$ $GA_3$ was most effective to produce multiple shoots. When the explants were cultured for 8 weeks, 39.5 shoots were developed in average.
