  • Celiac disease (CD) is an immune-mediated enteropathy of small intestine diagnosed in both childhood and adulthood. CD is caused by gluten, which produces gliadorphin during its digestion. The enzyme dipeptidyl peptidase-4 (DPP4) breaks gliadorphin down nevertheless the last tripeptide remains and eventually inhibits DPP4, thus slowing down its process. Therefore, the idea is to produce an additional DPP4 enzyme which is crucial. Consequently, the functional DPP4 gene was cloned into pCDNA3 intermediate (FLAG+DPP4) vector and finally a recombinant plasmid pSB1C3 (Andersons promoters+FLAG+DPP4) was constructed using synthetic biology. Normally, a DPP4 inhibitor is used as a cure for diabetes. Another important concern was overexpression of DPP4, which might lead to diabetes, accordingly the work was also performed for the regulation of the DPP4 gene expression. In this regard, three types of Anderson promoters (strong, moderate and weak) were utilized to study the control overexpression. This is the first report of idealistic trial for control the exogenous DPP4 gene-expression by molecular biologic tools.

  • Wild rice, Oryza grandiglumis shows hyper-resistance response to pathogen infection. In order to identify genes necessary for defense response in plants, we have carried out a subtractive hybridization coupled with a cDNA macroarray. An acidic PATHOGENESIS-RELATED1 (PR1) gene of the wild rice is highly identical to the acidic PR1 genes of different plant species. The OgPR1a cDNA has an apparent single open reading frame with a predicted molecular mass 40,621 Da and an isoelectic point of 5.14. Both in silico analysis and a transient expression assay in onion epidermal cells revealed that the OgPR1a protein could be localized in intercellular space in plants. The OgPR1a mRNA was strongly transcribed by the exogenous treatment with ethylene and jasmonic acid as well as protein phosphatase inhibitors. Additionally, ectopic expression of the OgPR1a conferred disease resistance on Arabidopsis to the bacterial and fungal infections.

  • Two cultivars Buram-3-ho (susceptible) and CR-Hagwang (moderate resistant) of kimchi cabbage seedlings showed differential defense responses to anthracnose (Colletotrichum higginsianum), black spot (Alternaria brassicicola) and black rot (Xanthomonas campestris pv. campestris, Xcc) diseases in our previous study. Defense-related hormones salicylic acid (SA), jasmonic acid (JA) and ethylene led to different transcriptional regulation of pathogenesis-related (PR) gene expression in both cultivars. In this study, exogenous application of SA suppressed basal defenses to C. higginsianum in the 1st leaves of the susceptible cultivar and cultivar resistance of the 2nd leaves of the resistant cultivar. SA also enhanced susceptibility of the susceptible cultivar to A. brassicicola. By contrast, SA elevated disease resistance to Xcc in the resistant cultivar, but not in the susceptible cultivar. Methyl jasmonate (MJ) treatment did not affect the disease resistance to C. higginsianum and Xcc in either cultivar, but it compromised the disease resistance to A. brassicicola in the resistant cultivar. Treatment with 1-aminocyclopropane-1-carboxylic acid (ACC) ethylene precursor did not change resistance of the either cultivar to C. higginsianum and Xcc. Effect of ACC pretreatment on the resistance to A. brassicicola was not distinguished between susceptible and resistant cultivars, because cultivar resistance of the resistant cultivar was lost by prolonged moist dark conditions. Taken together, exogenously applied SA, JA and ethylene altered defense signaling crosstalk to three diseases of anthracnose, black spot and black rot in a cultivar-dependent manner.

  • In order to investigate the meanings and bases of transforming theories of diseases(病傳論), several relevant theories were collected from medical books since . The outside-to-inside changing process(pyoree jeon) was the fundamental pattern in febrile diseases regardless of slang han and wen bing after Han dynasty. But in case of numb disease the three exogenous pathogenic factors of wind, cold and dampness got into each viscera through five tissues like skin, vessesl, flesh, tendons, bones respectively. It was called corresponding changing process(sanghap jeon) here. The pathogenic changing process(byungsa jeon) had complicated details to explain pathologic processes, but could give useful informations on evaluating relative strength of pathogens and tendency ahead. The changing process of diseases of internal injuries were explained in case of emotional distress which is outbroken abruptly didn't follow regular order of viscera-emotion relations. So it was named random changing process(bulcha jeon) after 's usage. And marasmus and asthenic disease followed top-to-down or down-to-top changing process(sangha jeon) based on . There are many types of changing process of diseases between viscera and viscera or viscera and bowels like generation changing process, restriction changing process, changing process between couple, changing process of mutual transmission based on attributions of each element by the theory of 5 phases(五行論). And changing process to a neighbor doesn't have any special relations within two organs but has anatomical contiguity and physiological continuity between them. The transforming theories of diseases bring forth useful understanding on comings and goings of pathogenic factors and tendency and prognosis of disease, so they are needed to be taught in the course of pathology class.

  • Amyloid precursor protein binding protein-1 (APP-BP1) binds to the carboxyl terminus of amyloid precursor protein and serves as a bipartite activation enzyme for the ubiquitin-like protein, NEDD8. Previously, it has been reported that APP-BP1 rescues the cell cycle S-M checkpoint defect in Ts41 hamster cells, that this rescue is dependent on the interaction of APP-BP1 with hUba3. The exogenous expression of APP-BP1 in neurons has been reported to cause DNA synthesis and apoptosis via a signaling pathway that is dependent on APP-BP1 binding to APP. These results suggest that APP-BP1 overexpression contributes to neurodegeneration. In the present study, we explored whether APP-BP1 expression was altered in the brains of Tg2576 mice, which is an animal model of Alzheimer's disease. APP-BP1 was found to be up-regulated in the hippocampus and cortex of 12 month-old Tg2576 mice compared to age-matched wild-type mice. In addition, APP-BP1 knockdown by siRNA treatment reduced cullin-1 neddylation in fetal neural stem cells, suggesting that APP-BP1 plays a role in cell cycle progression in the cells. Collectively, these results suggest that increased expression of APP-BP1, which has a role in cell cycle progression in neuronal cells, contributes to the pathogenesis of Alzheimer's disease.

  • Serotonin was chemically identified as 5-hydrowytryptamin which occurs in plants, animals, and human beings. The end product of metabolism was excreted as 5-HIAA in urine. Many scientists, specially biochemist and psychiatrist reported that the change of serotonin concentration caused mental disorder and pathological condiions such as schizophrenia and carcinoid. This study was carried out to observe the urinary 5-HIAA excretion rates changed by three recipes treatment (Kamisoyosan, Ondamtang, kalgunhaegitang) according to the classification of endogenous, exogenous and non-exo-endogenous, causes of disease. The urinary 5-HIAA excretion rates before and after three recipes treatment on normal groups divided into three groups on the basis of physical constitutional differences and on patients groups, divided into three groups according to the causal factor were measured. The results were as follows; 1. The urinary 5-HIAA volume of patients groups appeared non-exoendogenous, endogenous, exogenous causes of disease in order of three causative classification. 2. The urinary 5-HIAA volume of Normal groups was $2.50{\pm}088mg/24hrs$(range $0.30{\sim}6.90mg/24hrs$) 3. Thr urinary 5-HIAA volume was $3.70{\pm}0.89mg/24hrs$(range $0.90{\sim}6.50mg/24hrs$) before kamisoyosan treatment and $3.20{\pm}0.72mg/24hrs$ (range $0.80{\sim}6.20mg/24hrs$) after the treatment. 4. The value was $2.60{\pm}0.10mg/24hrs$ (range $0.60{\sim}6.50mg/24hrs$) before Ondamtange treatment and $2.00{\pm}0.12mg/24hrs$(range $0.20{\sim}6.10mg/24hrs$) after the treatment. 5. The value was $4.30{\pm}0.75mg/24hrs$(range $0.92{\sim}6.50mg/24hrs$) before kalgunhaegitang treatment and $3.10{\pm}0.10mg/24hrs$(range $0.80{\sim}5.80mg/24hrs$) after the treatment. Considering the above-mentioned the study on the changes of urinary 5-HIAA volume will make a significant contribution to the diagnosis and the evaluation of therapeutic effect successive research and modified application will be in need.

  • We got some conclusion like below. after comparison and consideration mainly with commentary of "Somun(素問)", "Uihakyimmun(醫學入門)"about pulses, reasons, symptoms, treatments of HaeYok. The name of Haeyok was appeared "Somun(素問)", "Youngchu(靈樞)" for the first time, and from then 'Treatment From Pulse' was promoted into details. About the Pulse of Haeyok, every doctor followed the 'chokmaekwansaek' mentioned in "Somun Pyoungingisanglon(素問 平人氣象論)". But the letter 'wan(緩)' in 'chokmaekwansaek' was translated not pulse but drooped skin in "SomunKumsok(素問今釋)". On the reasons of Haeyok, we can lot out in detail like next; (1) heat of liver and abdomen, lack of blood. (2) declining Gi, little blood, (3) declining and little Gi of spleen, (4) Lack of Gi and Blood, (5) weak kidney energy, (6) nutritional vacancy, (7) no energy circulation from hardened kidney, (8) exogenous disease on void kidney (9) both weak meridian of liver, kidney. And it can be divided broadly into two groups; weak liver and kidney, declining and little Gi of spleen. The symptoms of Haeyok is that patient feels cold, but really that is not cold, feels weak, really not weak, feels vigorous but not. So hardly be named. the members are exhausted, people get lazy, annoying, sick. and have no vitality. Treatments about this, some prescriptions are suggested such as Baekhaptang(白合湯) from "Naekyoungsupyoubanglon(內經拾遺方論)", Yishintang(利腎湯) from "Hwangjesomunsonmyoungbanglon(黃帝素問宣明方論)". In "Dongyanguihakdaesajon", there's some treatment according to some reason; when we are lazy and don't know where is sick, use Haryoungmansudan(遐齡萬壽丹) or Shinsongijedan(神仙旣濟丹), When it is from Liquor, use Galhwahaedokdan(葛花解毒丹), When from humidity use Gamichulbutang(加味朮附湯), from cold use Ganghwalchunghwatang(羌活沖和湯), and when it comes from instable menstruation, it will be good Gamisoyosan(加味逍遙散). And treatment principle from "Uihakyimmun(醫學入門)" is that "Circulate Gi and blood, be thin skin, prescribe exogenous disease over for void organs."

  • This report describes 90 prescriptions related to the use of Platycodi Radix main blended from Dongeuibogam. The following conclusions were reached through investigations on the prescriptions that use Platycodi Radix as a key component. Prescriptions that Platycodi Radix was taken as a monarch drug are utilized for 30 therapeutic purposes, for example, cough disease, throat disease, abscess and pus, and wind disease. In particular, 12.1% of prescriptions appear in the chapter of cough, and 9.9% of those appear in the chapter of throat, and each 8.8% of those appear in the chapter of abscess and of wind disease. Prescriptions that utilize Platycodi Radix as the main ingredient are used in the treatment of cough disease, throat disease and abscess, and they are also used for treating 74 different types of disease. The prescriptions are compounded with Platycodi Radix as a monarch drug are related to exogenous agents such as wind-cold pathogen, wind-heat pathogen, epidemic diseases, and endogenous agents such as seven emotions, and non-endo-exopathogenic factors like excessive labor, deficiency of Qi and blood, phlegm-mass, phlegm-fire, ect. The dosage of Platycodi Radix is 2pun(about 0.75g) to 1nyang(about 37.5g), however 1don (about 3.75g) has been taken the most for clinical application. We can find out that according to herbs or prescriptions blended with it self, Platycodi Radix makes a variety of functions to penetrate lung stagnancy and remove phlegm, relieve throat pain and get rid of pus, and regulate Qi flow. And Gamgiltang is the most useful base prescription which used the Platycodi Radix as the main component.

  • Spinocerebellar ataxias (SCAs) are caused by expansion of (CAG)n triplet repeats. These repeats occur as polymorphic forms in general population; however, beyond a threshold size they become pathogenic. The sizes and distributions of repeats at the SCA1, SCA2, SCA3, SCA7 and DRPLA loci were assessed by molecular analysis of 124 unrelated ataxia patients and 44 controls, and the association of larger normal (LN) alleles with disease prevalence was evaluated. Triplet repeat expansions in the disease range were detected in 8% (10/124) of the cases, with the majority having expansion at the SCA1 locus. Normal allele ranges in the cohort studied were similar to the Caucasian and North Indian populations but differed from the Korean and Japanese populations at various loci. The percentage of individuals with LN alleles at the SCA1 and SCA2 loci was higher than reported in Indians, Japanese and Caucasians. LN alleles showed a good correlation with the incidence of SCA1, indicating that SCA1 is the most prevalent ataxia in our population. The majority of cases with clinical symptoms of SCA could not be diagnosed by established CAG repeat criteria, suggesting that there may be an alternative basis for disease pathogenesis: (i) Repeats lower than the normal range may also result in abnormal phenotypes (ii) LN alleles at different loci in the same individual may contribute to symptoms (iii) Exogenous factors may play a role in triggering disease symptoms in individuals with LN alleles (iv) Triplet repeats may reach the disease range in the brain but not in the blood.

  • Porphyromonas gingivalis, a Gram negative, anaerobic, asaccharolytic rod, is one of the most frequently implicated pathogens in human periodontal disease and has a requirement for hemin for growth. A 30 kDa (heated 24 kDa) hemin-binding protein whose expression is both hemin and iron regulated has recently been purified and characterized in this oral pathogen. This study has identified a hemin-binding P. gingivalis protein by expression of a P. gingivalis genomic library in Escherichia coli, a bacterium which does not require or transport exogenous hemin. A library of genomic DNA fragments from P. gingivalis was constructed in plasmid pUC18, transformed into Escherichia coli strain $DH5{\alpha}$ , and screened for recombinant clones with hemin-binding activity by plating onto hemin-containing agar. Of approximately 10,000 recombinant E. coli colonies screened on LB-amp-hemin agar, 10 exhibited a clearly pigmented phenotype. Each clone contained various insert DNA. The Hind III fragment transferred to the T7 RNA polymerase/promoter expression vector system produced a sligltly smaller (21 kDa) protein, a precursor form, immunoreactive to the antibody against the 24 kDa protein, suggesting that the cloned DNA fragment probably carried an entire gene for the 24 kDa hemin-binding protein.
