Quantification of seleno proteins in Korean blood serum using solid phase extraction and affinity chromatography-inductively coupled plasma/mass spectrometry (고체상 추출과 친화 크로마토 그라피-유도결합 플라즈마 질량분석법을 이용한 한국인 혈청에서의 셀레노 단백질 정량)

  • Interferences were removed using anion exchange solid phase extraction (AE SPE) in quantification of selenoproteins in Korean human blood serum with affinity high performance liquid chromatography (AF HPLC)-inductively coupled plasma/mass spectrometry (ICP/MS). The average selenium level obtained for healthy Koreans was $94.3{\pm}2.3ngg^{-1}$ using isotope dilution method. AE SPE was coupled to AF column to separate 3 selenoproteins, glutathione peroxidase GPx, selenoprotein SelP, and selenoalbumin SeAlb. Post column isotope dilution was employed to quantify the proteins. The certified reference material of human blood serum BCR-637 was analyzed to give total selenoprotein concentration of $85.4{\pm}3.4ngg^{-1}$, which agreed well with the reference value of $81{\pm}7ngg^{-1}$. The pooled concentration of GPx, SelP, and SeAlb from healthy Koreans (n=20) was $12.1{\pm}1.4ngg^{-1}$, $57.2{\pm}2.0ngg^{-1}$, and $20.0{\pm}1.9ngg^{-1}$, respectively. The sum of selenoproteins is $89.3ngg^{-1}$, which is about the same as the total selenium concentration of $94.3ngg^{-1}$. The fact suggests that selenium in blood serum is mostly consisted of selenoproteins. After the removal of interference, GPx showed a significant decrease (more than 50%) from $25.0ngg^{-1}$ to $12.1ngg^{-1}$. It was identified that the interference in blood serum was mostly from GPx and the use of AE SPE was proven to be efficient in eliminating Cl and Br that cause interference to GPx.

  • We report $^{230}Th/^{234}U$ disequilibrium ages of fracture-filling carbonate veins from the Ipsil and Janghangri fault zones, Gyeongju, Korea by multiple collector inductively coupled plasma mass spectrometry. The U and Th fraction was extracted from totally dissolved samples by rapid and convenient coprecipitation and ion exchange chemistry. The recovery was around 80% for Th and 70% for U. The $^{234}U/^{238}U,\;^{230}Th/^{232}Th$ ratios were analysed for this preconcentrated fraction and the U/Th ratio was directly analysed for untreated sample solution. The $^{234}U-^{230}Th$ system is in secular equilibrium for the Ipsil carbonate samples, supporting previously reported ESR ages. The detrital-corrected $^{230}Th/^{234}U$ age of the Janghangri carbonate samples is $48\pm$41 ka, which constrains the minimum age of the fracture zone.

A Design and Implementation of XML DTDs for Integrated Medical Information System (통합의료정보 시스템을 위한 XML DTD 설계 및 구현)

  • The advanced medical information systems usually consist of loosely-coupled interaction of independent systems, such as HIS/RIS and PACS. To support easier information exchange between these systems and between hospitals, and to support new types of medical service such as teleradiology, it becomes essential to integrate separated medical information and allow them to be exchanged and retrieved through internet. This thesis proposes an integrated medical information system using XML. We analyzed HL7 and DICOM standard formats, and designed an integrated XML DTD. We extracted information from HL7 messages and DICOM files and generated XML document instances and XSL stylesheets based on the proposed XML DTD. We implemented the web interface for the integrated medical information system, which supports data sharing, information exchange and retrieval between two different standard formats. The proposed XML-based integrated medical information system will contribute to solve the problems of current medical information systems, by enabling integration of separated medical informations and by allowing data exchange and sharing through internet. The proposed system with XML is more robust than web-based medical information systems developed by using HTML, because XML itself provides more flexibility and extensibility than HTML.

Adsorption Characteristics of Nitrogen monoxide over Dealuminated and Alkali/Alkaline-earth Metal ion Exchanged Y-Zeolites (탈알루미늄 및 알칼리/알칼리토금속 양이온을 교환한 Y형 제올라이트의 NO흡착 특성)

  • The dealuminated and alkali/alkaline-earth metal exchanged Y-zeolites were prepared as a catalyst. Elemental compositions and structures of the prepared catalysts were analyzed by the various spectroscopic techniques such as inductively coupled plasma-atomic emission spectroscopy(ICP-AES), X-ray fluorescence(XRF) and X-ray diffraction(XRD), and the desorption behaviors of adsorbed species on the catalyst surfaces were investigated via NO-TPD experiment. Comparing with the composition of the starting material of NaY zeolite, the magnitudes of Si/Al ratio in catalytic materials were increased after dealumination. The Si/Al ratio of catalytic materials after dealumination followed by Cs and Ba cation exchange were additionally decreased. Dealumination to catalysts induced a destruction of basic frame due to a detachment of aluminum, which results in reducing framework structure, while increasing non-framework structure. This phenomenon becomes more serious with increasing time of steam treatment and even more significant for the cation exchanged catalysts. In NO-TPD experiments, the desorption peaks of NO which indicates an activity point of catalysts shifted to the low temperature region after dealumination and cation exchange. The desorption peaks of the NO-TPD profiles taken after steam treatment also shifted to the low temperature region as the steam treatment time increased. In dealuminated and cation exchanged Y-zeolites, the catalytic activities were more influenced by exchanged cation and the formation of non-framework structure.

  • Spin dynamics of magnetic tunnel junctions with free and fixed reference layers is investigated by ferromagnetic resonance micromagnetic simulations. First, in magnetic tunnel junctions with an exchange biased synthetic ferrimagnetic reference layer, a magnetization direction of each layer and the tunneling magnetoresistance are calculated for a DC magnetic field. To investigate the spin exciting modes in magnetic tunnel junctions, we simulate the ferromagnetic resonance frequency spectra with small RF magnetic fields. Exciting modes of the tunneling magnetoresistance calculated by an included angle between free and reference layers is interpreted from those of each layer. Spin exciting modes are different according to a signs of the DC magnetic field. In a negative magnetic field, FMR frequency spectra of free and reference layers are well elucidated by the modified Kittel's equation. However, in a positive magnetic field, there is no simple analytic solution related to FMR frequency spectra due to the coupled modes. Since ferromagnetic layers in magnetic tunnel junctions are interactive each other, careful considerations of the reference and fixed layer as well as the free layer are required for understanding on the spin dynamics of magnetic tunnel junctions with an exchange biased synthetic ferrimagnetic reference layer.

Corrosion Behaviors of 316L Stainless Steel Bipolar Plate of PEMFC and Measurements of Interfacial Contact Resistance(ICR) between Gas Diffusion Layer(GDL) and Bipolar Plate (고분자 전해질 연료전지 금속분리판 316L 스테인리스강의 부식거동 및 기체확산층(GDL)과의 계면접촉저항 측정)

  • The corrosion behaviors of 316L stainless steel were investigated in simulated anodic and cathodic environments for proton exchange membrane fuel cell (PEMFC) by using electrochemical measurement techniques. Interfacial contact resistance(ICR) between the stainless steel and gas diffusion layer(GDL) was also measured. The possibility of 316L was evaluated as a substitute material for the graphite bipolar plate of PEMFC. The value of ICR decreased with an increase in compaction stress(20 N/$cm^2$~220 N/$cm^2$) showing the higher values than the required value in PEMFC condition. Although 316L was spontaneously passivated in simulated cathodic environment, its passive state was unstable in simulated anodic environment. Potentiostatic and electrochemical impedance spectroscopy (EIS) measurement results showed that the corrosion resistance in cathodic condition was higher and more stable than that in anodic condition. Field emission scanning electron microscopy (FE-SEM), and inductively coupled plasma(ICP) were used to analyze the surface morphology and the metal ion concentration in electrolytes.

Numerical Analysis for the Effect of Ground and Groundwater Conditions on the Performance of Ground Source Heat Pump Systems (토양 및 지하수 조건이 지열공조시스템의 성능에 미치는 영향에 관한 수치 해석적 연구)

  • Recently, ground source heat pump (GSHP) systems have been introduced in many modem buildings which use the annually stable characteristic of underground temperature as one of the renewable energy uses. However, all of GSHP systems cannot achieve high level of energy efficiency and energy-saving, because their performance significantly depends on thermal properties of soil, the condition of groundwater, building loads, etc. In this research, the effect of thermal properties of soil on the performance of GSHP systems has been estimated by a numerical simulation which is coupled with ground heat and water transfer model, ground heat exchanger model and surface heat balance model. The thermal conductivity of soil, the type of soil and the velocity of groundwater flow were used as the calculation parameter in the simulation. A numerical model with a ground heat exchanger was used in the calculation and, their effect on the system performance was estimated through the sensitivity analysis with the developed simulation tool. In the result of simulation, it founds that the faster groundwater flow and the higher heat conductivity the ground has, the more heat exchange rate the system in the site can achieve.

Comparison between Ionospheric and plasmaspheric TECs measured from JASON satellite: plasmaspheric flux

  • The plasmasphere is filled with the ions and electron transported mostly from the mid-latitude ionosphere. In the topside ionosphere where the $O^+$ ions are still major ions, the $O^+$ ions are in chemical equilibrium with the $H^+$ ions and exchange their charges with each other's parent atoms with similar rates in both reactions. During the day, the newly produced $H^+$ ions flow upward to fill the plasmasphere while they flow downward and contribute to the maintenance of the ionospheric density at night under the geomagnetically quiet condition. The ionosphere and plasmasphere are coupled by these plasma fluxes and therefore strongly affect each other. In order to study these coupling we utilized the plasma density measurements from JASON satellite. This satellite measures vertical total electron content (TEC) from the ground to the satellite orbit (about 1336 km) and slant TEC from the satellite orbit to much higher GPS satellites by using the on-board dual-frequency altimeter and GPS receiver, respectively. The former measurement can represent the ionospheric TEC while the latter can represent the plasmaspheric TEC in the equatorial region. We compared these data with different seasons, solar activities and local times, and the results will be presented.

A Sequential Separation Procedure for the Determination of Uranium, Thorium and Their Isotopes in Geological Materials (지질시료 중의 U과 Th 분석 및 자연방사성동위원소 측정을 위한 화학적 분리)

  • Inductively coupled plasma atomic emission spectrometry(ICP/AES) was used for the determination of uranium and thorium in geological materials. Samples were predecomposed by mixed acid digestion technique. The separation of the uranium and thorium was achieved by systematic solvent extraction with TTA(thenoyltrifluoroacetone) and TOA (tri-n-octylamine) and back extraction into HCl. The results for standard rock sample, NIST SRM 278, showed a good agreement with those certified from NIST as well as found values by other non-destructive techniques. Additional purification for extracted portions was carried out by anion exchange chromatography for measurement of several natural radioisotopes of uranium and thorium by alpha spectrometry.

Molecular Characterization of Bile Salt Hydrolase from Bifidobacterium animalis subsp. lactis Bi30

  • The present work describes the identification, purification, and characterization of bile salt hydrolase (BSH) from Bifidobacterium animalis subsp. lactis. The enzyme was purified to electrophoretic homogeneity by hydrophobic chromatography, ion-exchange chromatography and ultrafiltration. SDS-PAGE analysis of putative BSH and gel filtration revealed that the analyzed protein is presumably a tetramer composed of four monomers each of about 35 kDa. The purified enzyme was analyzed by liquid chromatography coupled to LTQ FT ICR mass spectrometry and unambiguously identified as a bile salt hydrolase from B. animalis. The isoelectric point of the studied protein was estimated to be around pH 4.9. The pH optimum of the purified BSH is between 4.7 to 6.5, and the temperature optimum is around 50oC. The BSH of B. animalis could deconjugate all tested bile salts, with clear preference for glycine-conjugated bile salts over taurine-conjugated forms. Genetic analysis of the bsh showed high similarity to the previously sequenced bsh gene from B. animalis and confirmed the usefulness of bile salt hydrolase as a genetic marker for B. animalis identification.
