14-bp Insertion/Deletion Polymorphism of the HLA-G gene in Breast Cancer among Women from North Western Iran

  • Background: The human leukocyte antigen-G (HLA-G) gene is highly expressed in cancer pathologies and is one strategy used by tumor cells to escape immune surveillance. A 14-bp insertion/deletion (InDel) polymorphism of the HLA-G gene has been suggested to be associated with HLA-G mRNA stability and the expression of HLA-G. The aim of present study was to assess any genetic association between this polymorphism and breast cancer among Iranian-Azeri women. Materials and Methods: In this study 227 women affected with breast cancer, in addition to 255 age-sex and ethnically matched healthy individuals as the control group, participated. Genotyping was performed using polymerase chain reaction and electrophoresis assays. The data were compiled according to the genotype and allele frequencies, compared using the Chi-square test. Statistical significance was set at P<0.05. Results: In this case-control study, no significant difference was found between the case and control groups at allelic and genotype levels, although there is a slightly higher allele frequency of HLA-G 14bp deletion in breast cancer affected group. However,when the stage I subgroup was compared with stage II plus stage III subgroup of affected breast cancer, a significant difference was seen with the 14 bp deletion allele frequency. The stage II-III subgroup patients had higher frequency of deletion allele (57.4% vs 45.8%) than stage I cases (${\chi}^2=4.16$, p-value=0.041). Conclusions: Our data support a possible action of HLA-G 14bp InDel polymorphism as a potential genetic risk factor for progression of breast cancer. This finding highlights the necessity of future studies of this gene to establish the exact role of HLA-G in progression steps of breast cancer.

Comparative Assessment of the Diagnostic Value of Transbronchial Lung Biopsy and Bronchoalveolar Lavage Fluid Cytology in Lung Cancer

  • Background: This study was designed to determine the accuracy of bronchoalveolar lavage fluid cytology (BAL) using histopathologic examination of transbronchial biopsy specimens as the gold standard in diagnosis of lung carcinoma at our center. Materials and Methods: A retrospective study was conducted to investigate a total of 388 patients who were suspected of having lung cancer and had undergone fiberoptic bronchoscopy in Shahid Sadoughi hospital from 2006 to 2011. Lung masses were proven to be malignant by histology. Results: Transbronchial lung biopsy (TBLB) identified malignancy in 183 of the 388 cases, including 48 cases (26.2%) with adenocarcinoma, 4(2.1%) with bronchioloalveolar carcinoma, 47(25.6%)with squamous cell carcinoma, 34(18.5%) with well-diffentiated neuroendocrine carcinoma, 35(19.1%) with small cell carcinoma, 14 (7.6%) with non-small cell carcinoma, and 1 (0.54%) with large cell carcinoma. A total of 205 cases were correctly classified as negative. BAL was also performed in 388 patients; 86/103 cases were consistent with the final diagnosis of lung cancer and 188/285 cases were correctly classified as negative. The sensitivity of BAL was 46.9%(CI:41.9%, 51.8%)) and its specificity was 91.6%(CI:88.8%, 94.3%). BAL had a positive predictive value (PPV) of 83.4%(CI:79.7%, 87.1%) and a negative predictive value (NPV) of 65.8%(CI:61%, 70.5%). The overall accuracy of BAL was 70.5% and the exact concordance was 39%. Conclusions: Our findings suggest that BAL cytology is not sensitive but is a specific test for diagnosis of lung carcinoma. If transbronchial lung biopsy is combined with bronchoalveolar lavage, the positive diagnostic rate will be further elevated.

CYP3A4 Expression in Breast Cancer and its Association with Risk Factors in Mexican Women

  • Background: In Mexico, breast cancer (BCa) is the leading type of cancer in women. Cytochrome P450 (CYP450) is a superfamily of major oxidative enzymes that metabolize carcinogens and many antineoplastic drugs. In addition, these enzymes have influence on tumor development and tumor response to therapy. In this report, we analyzed the protein expression in patients with BCa and in healthy women. Links with some clinic-pathological characteristic were also assessed. Materials and Methods: Immunohistochemical analyses were conducted on 48 sets of human breast tumors and normal breast tissues enrolled in Hospital Militar de Especialidades de la Mujer y Neonatologia and Hospital Central Militar, respectively, during the time period from 2010 to 2011. Informed consent was obtained from all participants. Statistical analysis was performed using ${\chi}^2$ or Fisher exact tests to estimate associations and the Mann Whitney U test for comparison of group means. Results: We found a significant CYP3A4 overexpression in BCa stroma and gland regions in comparison with healthy tissue. A significant association between protein expression with smoking, alcoholism and hormonal contraceptives use was also observed. Additionally, we observed estrogen receptor (ER) and progesterone receptor (PR) positive association in BCa. Conclusions: We suggest that CYP3A4 expression promotes BCa development and can be used in the prediction of tumor response to different treatments. One therapeutic approach may thus be to block CYP3A4 function.

Risk of Cancer with Combined Oral Contraceptive Use among Iranian Women

  • Oral contraceptive use is the most common type of contraception. More than 300 million women worldwide take oral contraceptives every day. However, there is a concern about the relationship with the incidence of cancer. This analytical retrospective study aimed to investigate the relationship between the incidence of cervical and breast cancers and oral contraceptive use in 128 Iranian patients with cervical cancer, 235 with breast cancer and equal numbers of controls. Data were collected through interviews with an organized set of questions. Details were also extracted from patient files. Data were analyzed using Student's t-test, chi-square and Fisher's exact tests, and Pearson's correlation analysis. The result revealed correlations between both cervical and breast cancers and history of contraceptive pills use. While cervical cancer significantly correlated with duration of use of pills, breast cancer had significant correlations with the type of oral contraceptive and age at first use. No significant relationships were found between the two types of cancer and age at discontinuation of oral contraceptives, patterns of use, and intervals from the last use. The use of oral contraceptives may triple the incidence of cervical cancer and doubles the incidence of breast cancer. Therefore, performing Pap smears every six months and breast cancer screening are warranted for long-term oral contraceptive users.

Phosphatidylinositol 3-kinase (PI3KCA) Oncogene Mutation Analysis and Gene Expression Profiling in Primary Breast Cancer Patients

  • Background: The phosphatidylinositol 3-kinase (PI3K) pathway plays a significant role in apoptosis, cellular proliferation and motility. The aim of the present study was to analyze mutations and gene expression profiles of the PI3KCA gene to determine any role in breast carcinomas. Materials and Methods: We analyzed 38 breast cancers for mutations in the two PIK3CA hotspots in exons 9 and 20 by direct sequencing of DNA obtained from biopsy samples. We have also analyzed expression of the PI3KCA gene in 38 breast carcinoma tumor and corresponding control tissue samples at the mRNA level by RT-PCR. The Fisher's exact test ($2{\times}2$ only) was performed using MedCalc software for to examine associations with mRNA levels. Results: In the present study a total of 13 cases demonstrated somatic mutations. In 9/13 cases 1633 G>A (E545K) were found in exon 9, whereas in exon 20, 4/13 cases had 3140A>G mutation. Our combined analysis showed PI3KCA mutations present in 34% of human breast cancer patients. In our study, we have also clearly found significantly higher expression in breast cancer tissues in comparison with control tissues (p=0.001). Conclusions: PIK3CA mutation is an emerging tumor marker that, in the future, might be used in the process of choosing a treatment. The detection of PI3KCA mutation might have important clinical implications for diagnosis, progression and therapy.

  • In this paper, we propose a real-time face detection/tracking methodology with Haar wavelets and skin color. The proposed method boosts face detection and face tracking performance by combining skin color and Haar wavelets in an efficient way. The proposed method resolves the problem such as rotation and occlusion due to the characteristic of the condensation algorithm based on sampling despite it uses same features in both detection and tracking. In particular, it can be applied to a variety of applications such as face recognition and facial expression recognition which need an exact position and size of face since it not only keeps track of the position of a face, but also covers the size variation. Our test results show that our method performs well even in a complex background, a scene with varying face orientation and so on.

Route Tracking of Moving Magnetic Sensor Objects and Data Processing Module in a Wireless Sensor Network (무선 센서 네트워크에서의 자기센서기반 이동경로 추적과 데이터 처리 모듈)

  • In sensor network processing environments, current location tracking methods have problems in accuracy on receiving the transmitted data and pinpointing the exact locations depending on the applied methods, and also have limitations on decision making and monitoring the situations because of the lack of considering context-awareness. In order to overcome such limitations, we proposed a method which utilized context-awareness in a data processing module which tracks a location of the magnetic object(Magnetic Line Tracer) and controlled introspection data based on magnetic sensor. Also, in order to prove its effectiveness we have built a wireless sensor network test-bed and conducted various location tracking experiments of line tracer using the data and resulted in processing of context-aware data. Using the new data, we have analyzed the effectiveness of the proposed method for locating the information database entries and for controlling the route of line tracer depending on context-awareness.

  • The use of post installed anchors with adhesive type has lately been increasing when it is necessary to repair, reinforce, or remodel structures. This method provides flexibility and simplicity for construction of structural members that require adhering or fixing. Meanwhile, strength evaluation of anchors with expansion type among post installed anchors systems has nearly reached setting up stage like design code through continual experimental studies for the last ten years, but analyses or experimental studies on anchor system with adhesive type are not yet sufficient. Accordingly, the designers and builders of korea depend on foreign design codes since there are no exact domestic design code they could credit. In this study, the objectives are investigating the effects on adhesive strength of anchors embedded into plain concrete by shear experiments of anchors with variables such as edge distance, anchor interval, and load direction and supplying basic data for enactment of domestic design code.

Analysis of Baseline Accuracy by GPS Relative Positioning (GPS 상대측위에 의한 기선 정확도 분석)

  • As the exact geographical information has been nowadays required for effective developing and using of national land, in the country, there has been interested in using of GPS, and its practical use is expected. Various kinds of fundamental research for practical use of GPS is being accomplished. In this study, a test was carried out over 9 stations with baseline of the range of 1.5 to 210km, and the accuracy of baseline length by GPS relative positioning was variously considered. As result of this study, using a GPS receiving L1 frequency only, baseline accuracy for 2 hour observation was of the order of 0.3ppm for the 10km, and for I hour was below 1ppm. Using a GPS receiving dual frequency(L1/L2), baseline accuracy was of the order of 0.3ppm for the 100km to 200km as 3 hour observation using double difference methods by carrier phase. With basic on the result of this study, when observation and baseline processing are proceeded by the selected optimum observation time and using of baseline processing method, we can expect that geographical information will be acquired effectively.

Computer-Aided Diagnosis for Pulmonary Tuberculosis using Texture Features Analysis in Digital Chest Radiography (질감분석을 이용한 폐결핵의 자동진단)

  • There is no exact standard of detecting pulmonary tuberculosis(TB) in digital image of simple chest radiography. In this study, I experimented on the principal components analysis(PCA) algorithm in the past and suggested six other parameters as identification of TB lesions. The purpose of this study was to develop and test computer aided diagnosis(detection) method for the detection and measurement of pulmonary abnormalities on digital chest radiography. It showed comparatively low recognition diagnosis rate using PCA method, however, six kinds of texture features parameters algorithm showed similar or higher diagnosis rates of pulmonary disease than that of the clinical radiologists. Proposed algorithms using computer-aided of texture analysis can distinguish between areas of abnormality in the chest digital images, differentiate lesions having pulmonary disease. The method could be useful tool for classifying and measuring chest lesions, it would play a major role in radiologist's diagnosis of disease so as to help in pre-reading diagnosis and prevention of pulmonary tuberculosis.
