Previous works have shown the viability of NIRS technology for the prediction of fatty acids in Iberian pig fat, but although the resulting equations showed high precision, in the predictions of new samples important fluctuations were detected, greater with the time passed from calibration development to NIRS analysis. This fact makes the use of NIRS calibrations in routine analysis difficult. Moreover, this problem only appears in products like fat, that show spectrums with very defined absorption peaks at some wavelengths. This circumstance causes a high sensibility to small changes of the instrument, which are not perceived with the normal checks. To avoid these inconveniences, the software WinISI 1.04 has a mathematic algorithm that consist of create a “Repeatability File”. This file is used during calibration development to minimize the variation sources that can affect the NIRS predictions. The objective of the current work is the evaluation of the use of a repeatability file in quantitative NIRS analysis of Iberian pig fat. A total of 188 samples of Iberian pig fat, produced by COVAP, were used. NIR data were recorded using a FOSS NIRSystems 6500 I spectrophotometer equipped with a spinning module. Samples were analysed by folded transmission, using two sample cells of 0.1mm pathlength and gold surface. High accuracy calibration equations were obtained, without and with repeatability file, to determine the content of six fatty acids: miristic (SECV$\sub$without/=0.07% r$^2$$\sub$without/=0.76 and SECV$\sub$with/=0.08% r$^2$$\sub$with/=0.65), Palmitic (SECV$\sub$without/=0.28 r$^2$$\sub$without/=0.97 and SECV$\sub$with/=0.24% r$^2$$\sub$with/=0.98), palmitoleic (SECV$\sub$without/=0.08 r$^2$$\sub$without/=0.94 and SECV$\sub$with/=0.09% r$^2$$\sub$with/=0.92), Stearic (SECV$\sub$without/=0.27 r$^2$$\sub$without/=0.97 and SECV$\sub$with/=0.29% r$^2$$\sub$with/=0.96), oleic (SECV$\sub$without/=0.20 r$^2$$\sub$without/=0.99 and SECV$\sub$with/=0.20% r$^2$$\sub$with/=0.99) and linoleic (SECV$\sub$without/=0.16 r$^2$$\sub$without/=0.98 and SECV$\sub$with/=0.16% r$^2$$\sub$with/=0.98). The use of a repeatability file like a tool to reduce the variation sources that can disturbed the prediction accuracy was very effective. Although in calibration results the differences are negligible, the effect caused by the repeatability file is appreciated mainly when are predicted new samples that are not in the calibration set and whose spectrum were recorded a long time after the equation development. In this case, bias values corresponding to fatty acids predictions were lower when the repeatability file was used: miristic (bias$\sub$without/=-0.05 and bias$\sub$with/=-0.04), Palmitic (bias$\sub$without/=-0.42 and bias$\sub$with/=-0.11), Palmitoleic (bias$\sub$without/=-0.03 and bias$\sub$with/=0.03), Stearic (bias$\sub$without/=0.47 and bias$\sub$with/=0.28), oleic (bias$\sub$without/=0.14 and bias$\sub$with/=-0.04) and linoleic (bias$\sub$without/=0.25 and bias$\sub$with/=-0.20).
Color is one of the quality determining factors for pepper powder. To measure the color of pepper powder, several methods including high-performance liquid chromatography (HPLC), thin layer chromatography (TLC), and ASTA-20 have been used. Among the methods, the ASTA-20 method is most widely used for color measurement of a large number of samples because of its simplicity and accuracy. However it requires time consuming preprocessing steps and generates chemical waste containing acetone. As an alternative, we developed a fast and simple method based on a visible/near infrared (Vis/NIR) hyperspectral method to measure the color of pepper powder. To evaluate correlation between the ASTA-20 and the visible/near infrared (Vis/NIR) hyperspectral methods, we first measured the color of a total of 488 pepper powder samples using the two methods. Then, a partial least squares (PLS) model was postulated using the color values of randomly selected 3 66 samples to predict ASTA values of unknown samples. When the ASTA values predicted by the PLS model were compared with those of the ASTA-20 method for 122 samples not used for model development, there was very high correlation between two methods ($R^2=0.88$) demonstrating reliability of Vis/NIR hyperspectral method. We believe that this simple and fast method is suitable for highthroughput screening of a large number of samples because this method does not require preprocessing steps required for the ASTA-20 method, and takes less than 30 min to measure the color of pepper powder.
Digital Radiography (DR) has rapidly developed in megavoltage X-ray imaging (MVI). Thus, a very simple and general quality assurance (QA) method is required. The purpose of this study was to evaluate the modulation transfer function (MTF), the noise power spectrum (NPS) and the detective quantum efficiency (DQE) for MVI using general QA method and computed radiography (CR) device. We used tungsten edge block with $19{\times}10{\times}1cm^3$ thickness and 6MV energy. For detector, CR-IP (image plate), CR-IP-lead, the CR-IP-back (lanex TM fast back screen), CR-IP-front (lanex TM fast front screen) were used and pre-sampling MTF was calculated. The MTF of CR-IP-front showed the highest value with 1.10 lp/mm although the CR-IP showed the only 0.70 lp/mm. The best NPS was observed in CR-IP front screen. According to the increase in spatial frequency, our results showed that DQE was approximately 1.0 cycles/mm. The present study demonstrates that the QA method with our home-made edge block can be used to evaluate MTF, NPS and DQE for MVI.
In the present study, the performance of culture methods using two selective agars and real-time PCR were compared for selective isolation of Cronobacter in powdered infant formula and dried pumpkin. Two food samples were spiked with the pathogen and then preenriched in distilled water. A small portion of preenrichment (10 mL) was incubated in Enterobacteriaceae enrichment both, followed by inoculation onto Druggan-Forsythe-Iversen agar (DFI agar) and Cronobacter sakazakii chromogenic plating agar (R&F agar). The preenrichment and enrichment (1 mL each) was used in real-time PCR assay. In powdered infant formula (PIF), no statistical difference was observed between both culture methods and real-time PCR with preenrichemt (p > 0.05). However, the number of positives obtained by R&F agar and real-time PCR was much higher than that of culture method using DFI agar in dried pumpkin (p < 0.05). In particular, R&F agar yielded a significantly greater selectivity than DFI agar in dried pumpkin (p < 0.05). Real-time PCR and R&F agar, which are currently recommended by US FDA, could be used as an alternative detection tools for the isolation of Cronobacter in PIF and ingredient of child foods such as dried pumpkin that has high number of competing natural microflora.

Near infrared reflectance spectroscopy (NIRS) has become increasingly used as a rapid, accurate method of evaluating some chemical constituents in cereal and dired animal forages. Analysis of forage quality by NIRS usually involves dry grinding samples. Costs might be reduced if samples could be analyzed without drying or grinding. The objective of this study was to investigate effect of sample preparations on prediction ability of chemical composition and fermentation parameter for Italian ryegrass silages by NIRS. A population of 147 Italian ryegrass silages representing a wide range in chemical parameters were used in this investigation. Samples were scanned at 1nm intervals over the wavelength range 680-2500 nm and the optical data recorded as log 1/Reflectance (log 1/R) and scanned in oven-dried grinding and fresh ungrinding condition. The spectral data were regressed against a range of chemical parameters using partial least squares (PLS) multivariate analysis in conjunction with four spectral math treatments to reduced the effect of extraneous noise. The optimum calibrations were selected on the basis of minimizing the standard error of cross validation (SECV) and maximizing the correlation coefficient of cross validation (${R^2}_{CV}$). The results of this study show that NIRS predicted the chemical parameters with high degree of accuracy in oven-dried grinding treatment except for moisture contents. Prediction accuracy of the moisture contents was better for fresh ungrinding treatment (SECV 1.37%, $R^2$ 0.96) than for oven-dried grinding treatments (SECV 4.31%, $R^2$ 0.68). Although the statistical indexes for accuracy of the prediction were the lower in fresh ungrinding treatment, fresh treatment may be acceptable when processing is costly or when some changes in component due to the processing are expected. Results of this experiment showed the possibility of NIRS method to predict the chemical composition and fermentation parameter of Italian ryegrass silages as routine analysis method in feeding value evaluation and for farmer advice.

Cryopreserving cells which are maintaining their viability are the very complex process. This study has been carried out in order to find the effects of cryopreservation steps and freezing media on the rates of viability of cryopreserved chicken primordial germ cells (PGCs). PGCs obtained from the germinal gonade of 5.5~6 day (stage 28) chick embryos of Korean Ogye (KO) and Commercial breeds (C), using the MACS method were suspended in a freezing medium containing a freezing and protecting agents (e.g. dimethyl sulfoxide (DMSO), ethylene glycol (EG) and propylene glycol (PG)). Gonads were harvested from stage 28 chick embryos and pooled in groups of 5, 10, 15, 20E embryos, contributing gonads to the cell suspension. The gonadal cells, including PGCs, were then frozen in 1 of the following cryoprotectant treatments : 2.5%, 5%, 10%, 15% and 0% cryoprotectant (DMSO, EG, PG) as a control. Effects of exposure to slow freezing and vitrification, with different concentrations of the cryoprotectant solution, were examined. After vitrification and slow freezing, survival rates of the frozen-thawed PGCs from the 10% EG plus FBS treatment were 85.63%, and 66.14% (p<0.05), respectively. The viability of PGCs after freeze-thawing was significantly higher for 10% EG plus FBS treatment than for 10% PG + FBS treatment (p<0.05) (85.63% vs 66.81%) by vitrification. This study established a method for preserving chicken PGCs that enables systematic storage and labeling of cryopreserved PGCs in liquid ($LN_2$) at a germplasm repository and ease of entry into a data base. In the future, the importance for this new technology is that poultry lines can be conserved while work is being conducted on improving the production of germline chimeras.
Recently Korean universities show very rapid increases in both patents and R&D (research and development) expenditures. During the period from 1970 to 2008, university R&D spending has on the average increased 15.3% annually. Along with steady increases in R&D spending, university's research outputs have also continuously increased. In 1990 Korea as a total published 1,613 SCI-level scientific papers and Korean universities applied 27 patents to Korea patent office. In 2008, Korea published more that 35,000 SCI papers and Korean universities applied about 7,300 patents. The growth of scientific articles had begun from the early 1990s whereas the growth of patent has ignited entering the 2000s. The paper tried to investigate university research through the window of patent. Patents lie between invention and innovation and represent the potential value of invention which will be realized at the marketplace. Since Korean patents do not contain citation information, the paper used US patents-NBER patent database-as the main data. The key empirical question is whether Korean university patents granted from USPTO are characteristically different from other Korean patents granted from USPTO. Previous studies on US and Europe show that corporate patents are more stylized in appropriablity of invention, whereas university patents basicness. In case of Korea, the paper confirmed the appropriability characteristic of corporate patents; but the Korean unversity patents are not distinguishable in terms of basicness. The paper estimated the citation frequency function-an empirical model which was firstly developed by Caballero and Jaffe (1993) and later articulated by Jaffe and Trajtenberg (1996, 2002). The model is specified mainly composed of two interacting parts-diffusion effect and obsolescence effect of new ideas or innovations. Estimation results show that differences in forward citations between university and corporate patents are not statistically significant, after controlling self-citation. Since forward citations represent the quality of patents, this estimation result implies that there are no statistically significant quality differences between university and corporate patents. Prior research results, based on the same model of citation frequency function, about US and some European cases show that, in terms of forward citations, university patents are generally superior to corporate patents -for the case of US- or, the former not inferior to the latter-for the case of most of Europe. It is argued that some important and significant policy changes caused the rapid rise of university patents in Korea. Policy changes include the revision of technology transfer act allowing the ownership of publicly-funded research results to researchers and the changes in faculty/professor evaluation which gives more credit to the number of patents. These policy changes have triggered the rapid growth of the number of university patents. The results of the empirical analysis in this paper indicated that Korea now needs to make further efforts to enhance the quality of university patents, not just to produce more numbers of patents.

As part of a research dedicated to the field evaluation of the durability of concrete subjected to freezing-thawing, this study analyzes the relationship between the surface roughness and the relative dynamic elastic modulus through image analysis. Four mix compositions with water-to-binder ratios (W/B) of 40%, 50%, 60% and 70% and without AE agent were considered to provoke early freezing. The basic physical properties of the mixes including the relative dynamic elastic modulus and the compressive strength were first evaluated experimentally according to W/B. Then, tests were performed to measure the surface roughness followed by photographs and SEM image analysis. The measured surface roughness tended to increase with larger number of freezing-thawing cycles regardless of W/B. The relative dynamic elastic modulus appeared to increase gradually with the number of cycles for the relatively denser mixes with W/B of 40% and 50%. Besides, the surface roughness increased only at rupture for the mixes with W/B of 60% and 70%. Moreover, the analysis of the photographs of the surface of the mixes with W/B of 40% and 50% revealed that the degradation progressed gradually from the surface with the freezing-thawing cycles. However, for the mixes with W/B of 60% and 70%, apparent change of the surface remained very insignificant until rupture at which damage like cracking could be observed. Consequently, the analysis of surface photograph or the measurement of the surface roughness presented some limitation in assessing the degree of freezing-thawing-induced degradation in case of relatively porous specimens. On the other hand, the photograph and surface roughness appeared to be sufficient for assessing such degradation for the mixes with W/B of 40% and 50%. Accordingly, the image of the surface and the surface roughness are potentially applicable on site for the assessment of freezing-thawing damages in relatively dense mixes.

Many outbreaks of food-borne illnesses have been associated with the consumption of fresh vegetables and fruits contaminated with food-borne pathogens. Contaminated medium, manure and irrigation water are probable vehicles for the pathogen in many outbreaks. The aim of this study was to determine the potential transfer of Escherichia coli and Bacillus cereus from medium and soil fertilized with contaminated compost or irrigation with contaminated water to the edible parts of lettuce. Moreover, survivals of the two pathogens on lettuce contaminated medium, soil and irrigation water were estimated. Lettuce seeds were planted in medium contaminated with 7.5 log colony forming unit (CFU)/g of E. coli and B. cereus. Seedlings grown in the contaminated medium were transplanted in soil fertilized with contaminated pig manure compost or uncontaminated soil. Contaminated irrigation water with E. coli and B. cereus at 8.0 log CFU/mL was applied only once on the plant by sprinkle irrigation and surface irrigation. Although E. coli and B. cereus in medium and sprouted lettuce after planting seeds were reduced as time passed, these pathogens survived in seedling raising stage for extended periods. The numbers of E. coli and B. cereus in lettuce grown on contaminated soil were detected over 4.0 log CFU/g for 21 days. The numbers of E. coli and B. cereus in lettuce applied by sprinkle irrigation were higher than those of surface irrigation by 5.0 log CFU/g. Our results indicated that contaminated medium, soil and irrigation water can play an important role in the presence of food-borne pathogens on vegetables.

This study was conducted to quantify a carbon budget of major vegetation types established in the campus of the National Institute of Ecology (NIE). Carbon budget was measured for Pinus thunbergii and Castanea crenata stands as the existing vegetation. Net Primary Productivity (NPP) was determined by applying allometric method and soil respiration was measured by EGM-4. Heterotrophic respiration was calculated as 55% of total respiration based on the existing results. Net Ecosystem Production (NEP) was determined by the difference between NPP and heterotrophic respiration (HR). NPPs of P. thunbergii and C. crenata stands were shown in $4.9ton\;C\;ha^{-1}yr^{-1}$ and $5.3ton\;C\;ha^{-1}yr^{-1}$, respectively. Heterotrophic respirations of P. thunbergii and C. crenata stands were shown in $2.4ton\;C\;ha^{-1}yr^{-1}$ and $3.5ton\;C\;ha^{-1}yr^{-1}$, respectively. NEPs of P. thunbergii and C. crenata stands were shown in $2.5ton\;C\;ha^{-1}yr^{-1}$ and $1.8ton\;C\;ha^{-1}yr^{-1}$, respectively. Carbon absorption capacity for the whole set of vegetation types established in the NIE was estimated by applying NEP indices obtained from current study and extrapolating NEP indices from existing studies. The value was shown in $147.6ton\;C\;ha^{-1}yr^{-1}$ and it was calculated as $541.2ton\;CO_2ha^{-1}yr^{-1}$ converted into $CO_2$. This function corresponds to 62% of carbon emission from energy that NIE uses for operation of various facilities including the glass domes known in Ecorium. This carbon offset capacity corresponds to about five times of them of the whole national territory of Korea and the representative rural area, Seocheongun. Considered the fact that ongoing climate change was originated from imbalance of carbon budget at the global level, it is expected that evaluation on carbon budget in the spatial dimension reflected land use pattern could provide us baseline information being required to solve fundamentally climate change problem.
