Quantitative Determination of Eugenol in Eugenia Caryophyllata Thunb by Three Wavelength Spectrophotometry

  • Three wavelength spectrophotometry was used to determine the content of eugenol in Eugenia Caryophyllata Thunb. Using this method could effectively eliminate the deviation of background absorption caused by the change of concentration and the error of quantitative analysis caused by asymmetric peaks, and at the same time the leaning degree of base line was corrected. This method was simple the recovery ratio was 90.05%-116.94% and the coefficient of variation was 3.5%.

정향 열수 및 에탄올 추출물의 항산화 및 항염 효과 (Antioxidant and Anti-Inflammatory Activities of Extracts from Eugenia caryophyllata Thunb.)

  • The objective of this study was to investigate the antioxidant and anti-inflammatory activities of Eugenia caryophyllata Thunb. water and 70% ethanol extracts. The content of total polyphenol was significantly higher in water extract than in 70% ethanol extract. The DPPH radical scavenging activity of water extract was similar to that of Vit. C at a concentration of $1,000{\mu}g/mL$. The ABTS radical scavenging activities of water and 70% ethanol extract were similar to that of Vit. C at a concentration of $1,000{\mu}g/mL$. SOD-like activity of water extract was higher than that of 70% ethanol extract at a concentration of $1,000{\mu}g/mL$ but lower than that of Vit. C. The DPPH radical scavenging activity, ABTS radical scavenging activity, and SOD-like activity increased as concentrations of water and 70% ethanol extracts increased. Cell cytotoxicity was not observed at all concentrations except at $100{\mu}g/mL$ concentration of water extract. Inhibitory activity on NO production effect of water extract was significantly higher than that of 70% ethanol extract. These results show that E. caryophyllata Thunb. has potent biological activities, and their activities were different depending on extraction solvent.

용매별 정향 추출물의 항산화 및 신경세포 보호 효과 (Antioxidant and Neuronal Cell Protective Effects of Eugenia caryophyllata Thunb. by Extraction Solvent)

  • This study examined the antioxidant and neuronal cell protective effects of the water and methanol extracts of Eugenia caryophyllata Thunb. The total polyphenol content was significantly higher in the methanol extract than in the water extract. The DPPH radical scavenging activity in the water extract was similar to Vit. C at a concentration of $100{\sim}200{\mu}g/mL$. The ABTS radical scavenging activity in the water and methanol extract was similar to Vit. C at a concentration of $800{\sim}1,000{\mu}g/mL$. The superoxide dismutase (SOD)-like activity in the methanol extract was similar to Vit. C at a concentration of $800{\sim}1,000{\mu}g/mL$. The DPPH, ABTS radical scavenging and (SOD)-like activity increased with increasing extract concentration. In a cell viability using MTT, the water extract (50 and 100 ppm) and methanol extract (100 ppm) had a protective effect against $H_2O_2$-induced neurotoxicity.The result ssuggest that the extract of E. caryophyllata Thunb. has antioxidant activities and may be useful for treating neurodegenerative disorders.

Anticoagulation and Anticancer Constituents from Eugenia caryophyllata Thunb

  • From the EtOAc fraction of Eugenia caryophyllata, four compounds were isolated through activity-guided silica gel column chromatography, From the result of spectroscopic data including NMR, MS and IR, the chemical structures of the compounds were determined as 1-allyl-4-hydroxy-3-methoxybezene acetate (eugenol acetate, 1), 1-allyl-4-hydroxy-3-methoxybezene (eugenol, 2), $3{\beta}-hydroxyolean-12-en-28-oic$ acid (oleanolic acid, 3) and $2{\alpha}$, $3{\beta}-dihydroxyolean-12-en-28-oic$ acid (maslinic acid, 4). Compounds 3 and 4 were isolated for the first time from this plant. Also, compounds 1, 2 and 3 exhibited relatively high platelet aggregation inhibitory activity with the $IC_{50}$ values of 0.24, 0.09 and 0.07 mM, respectively. Compound 2 significantly prolonged activated partial thromboplastin time (aPTT) with the value of $124{\pm}11.2$ seconds as compared to the control with the value of $37.5{\pm}2.2$ seconds at the concentration of 50 ${\mu}g/ml$. Compounds 1 and 3 revealed inhibitory activity on farnesyl protein transferase (FPTase) with the $IC_{50}$ values of 0.49 and 0.24 mM and compounds 1 and 2 highly inhibited the growth of rat-H-ras cells with the $Gl_{50}$ values of 6.63 and 5.70 ${\mu}M$, respectively.

전통 식재료의 항산화 효과 (Antioxidant Effect of Traditional food Ingredient)

  • 본 연구는 전통 식재료로 많이 사용되는 10종의 약용식물 및 채소류 추출물과 10종 혼합추출물의 항산화 효과를 살펴보기 위해서 디자인되었다. 세포 생존률은 MTS assay로, 항산화효과는 DPPH free radical 소거능, Riboflavin 유래 Superoxide 억제활성(SQA)에 의한 항산화활성을 측정하였다. 그 결과 세포생존률은 10종 약용식물 및 채소류 추출물과 10종 혼합추출물 모두 0.5mg/mL에서 대조군과 유사한 생존율을 보였다. DPPH free radical 소거능은 Cinnamomum cassia Blume, Eugenia caryophyllata Thunb. Arctium lappa, Total extract가 우수하였고, Riboflavin 유래 Superoxide 억제활성(SQA)은 Cinnamomum cassia Blume, Arctium lappa, Prunus mume Sieb. et Zucc., 우수하였으나, Total extract가 가장 우수한 항산화 효과를 보였다. 전통식재료로 사용되는 약용식물 및 채소 추출물과 혼합추출물의 항산화 효과를 비교한 결과, 단독으로 사용하는 것보다 혼합물로 사용시 항산화능이 증가되었다. 이는 항산화 기능성 소재로 활용이 가능한 것으로 사료되며, 향후 항산화 소재 개발 시 가치가 있을 것으로 사료된다.

정향(Eugenia caryophyllata Thunb.) Eugenol 및 그 유도체의 항산화 및 항염증활성 (Antioxidant and Anti-Inflammatory Activities of Eugenol and Its Derivatives from Clove (Eugenia caryophyllata Thunb.))

  • 본 연구는 한방스킨의 원료로 널리 사용되고 있는 8가지 생약의 휘발성증류추출액 중 항산화 및 항염증활성이 가장 강한 정향의 증류추출액으로부터 주된 향기성분을 SDE법으로 추출한 후 GC-MS로 확인하였으며, 주된 향기성분인 eugenol과 그 유도체를 합성한 후 항산화 및 항염증활성을 측정하고 비교하였으며, 아울러 HPLC를 이용하여 정향의 eugenol 및 그 유도체를 정량분석 하였다. 8가지 생약의 휘발성증류추출액 중 정향의 증류추출액이 가장 강한 항산화활성($IC_{50}$=8.85 ${\mu}g/mL$) 및 COX-2 저해활성(10 ${\mu}g/mL$ 농도에서 저해율은 58.15%)을 나타내었으며, 반면 15-LOX 저해 활성(25 ${\mu}g/mL$ 농도에서 저해율은 86.15%)은 당귀 다음으로 가장 높았다. 정향 증류추출액의 휘발성 향기성분을 SDE법으로 추출한 후 GC-MS를 이용하여 분석한 결과, eugenol, trans-caryophyllene 및 acetyl eugenol을 확인하였다. 한편, eugenol 및 그 유도체(methyl eugenol 및 acetyl eugenol)의 항산화 및 항염증 활성을 측정한 결과, eugenol($IC_{50}$=5.99 ${\mu}g/mL$)이 가장 높은 항산화활성을 나타낸 반면, methyl eugenol 및 acetyl eugenol은 거의 활성을 나타내지 않았다. COX-2의 경우 20 ${\mu}g/mL$ 농도에서 eugenol(85.35%)이 가장 강한 저해활성을 나타낸 반면, 15-LOX는 20 ${\mu}g/mL$ 농도에서 methyl eugenol(83.29%)이 가장 높은 저해활성을 나타내었다. 정향 에탄올추출물의 eugenol 및 유도체의 함량을 HPLC로 분석한 결과, eugenol 및 acetyl eugenol이 각각 6.95%, 1.85% 함유되어 있었으며 methyl eugenol은 검출되지 않았다. 이와 같이 정향 유래의 eugenol 및 그 유도체는 안전성이 문제시되고 있는 합성항산화제 및 항염증제를 대체할 수 있는 천연 유래의 항산화 및 항염증물질로서 잠재적 가치가 있어 향후 기능성식품, 화장품 및 의약품 소재로 널리 사용될 수 있을 것으로 생각된다.

죽엽 추출액을 이용한 젤리 품질평가 및 구강세균에 대한 항균효과 (Quality Characteristics and Anti-Oral microbial Activity of Jelly Using Bamboo (Phyllostachys nigra var. henonis) Leaves Extract)

  • This study examined the quality characteristics and anti-oral microbial activity of bamboo leaf jelly prepared with different 5 levels (0, 10, 20, 30, and 40%) of bamboo (Phyllostachys nigra var. henonis) leaf extract. The sugar contents of bamboo leaf jelly were increased significantly by increasing the level of bamboo leaf extract. The luminance and Hunter's a values of the jelly samples increased with increasing bamboo leaf extract, but the 40% bamboo leaf jelly had the lowest Hunter's b values. The hardness, adhesiveness, gumminess, and chewiness increased significantly with increasing bamboo leaf extract. Among the mechanical properties, only the flavor of the jelly with 30 and 40% bamboo leaf extract were increased significantly. The extract of bamboo leaves had strong antimicrobial activity against S. mutans, S. sobriuns, P. gingivalis, and P. intermedia at a concentration of 40%. These results suggest that bamboo leaf extract can be useful in the production of high quality jelly.
