Depolymerization of PET by Ethylene Glycol (에틸렌글리콜을 이용한 PET 해중합 특성)

  • A method for depolymerization of PET by catalyzed glycolysis with an excess ethylene glycol(EG) to recover bis-hydroxyethyl terephthalate(BHET) was investigated. The product was analyzed by high-performance liquid chromatography(HPLC). Effects of operation variables such as reaction temperature, reaction time, EG/PET weight ratio were examined and kinetics of the glycolysis was studied. High temperature increases the rate of depolymerization and the yield of BHET. But, repolymerization rate was also high at too high temperature and the yield at $250^{\circ}C$ was shown to be lower than that at $230^{\circ}C$. First order reaction model was proposed to describe the glycolysis reaction. Activation energies for the reaction were obtained to be 37.8 kJ/mol above $210^{\circ}C$ and 149.6 kJ/mol below $210^{\circ}C$, which shows the glycolysis reaction is a multiple reaction. A maximum yield of BHET of 71% was achieved at a reaction temperature of $230^{\circ}C$ for 6 hr with an EG/PET weight ratio 4.

Selection of Cryoprotectants for Cryopreservation of Pearl Oyster, Pinctada fucata martensii Trochophore (진주조개, Pinctada fucata martensii 담륜자의 냉동보존을 위한 동해방지제의 선택)

  • To find out the desirable cryoprotectant for cryopreservation of bivalve trochophores, four types of cryoprotectant were tested with trochophores of the pearl oyster (pinctada fucata martensii) generally used in the pearl production in Korea. Each cryoprotectant (dimethyl sulfoxide, ethylene glycol, glycerol and 1,2-propanediol) was mixed with 0.2 M sucrose to make final concentrations of 0.01, 0.1, 1.0 and 2.0 M. The trochophores were immersed in each preparation, waiting for 10 minutes to reach equilibration and cryopreserved in the liquid nitrogen (-l96$^{\circ}C$). Survival rate of trochophores thawed after cryopreservation increased as the media concentration increase. However, a few number of the trochophores seemed to be damaged with the efflux of cell inclusions. Our study rsults indicate that desirable cryoprotectants for cryopreservation of pearl oyster trochophores are 1.0~2.0 M dimethyl sulfoxide and ethylene glycol : 82.8~97.4% of the trochophores cryopreserved with these media survived after thawing.

  • The volume phase transition of poly(N-isopropylacrylamide) (PNIPAAm) and poly (N-isopropylacrylamide-co-sodium methacrylate) (P (NIPAAm-co-SMA)) hydrogels crosslinked with poly (ethylene glycol) diacrylate (PEGDA) was investigated in consideration of water content and surface area. The volume phase transition temperature of hydrogel was not affected by the concentration of crosslinking agent, which increased over 40$\^{C}$ by incorporating a small amount of SMA. Higher volume phase transition temperature was obtained when PEGAD was used as a crosslinking agent, suggesting that the chain length of crosslinking agent had a significant effect on the volume phase transition temperature. The surface area of PNIPAAm and P (NIPAAm-co-SMA) gels fell off around the volume phase transition temperature, resulting from the fact that the size of pores reduced remarkably in the course of the volume phase transition. Hence, the surface area and the pore size were considered to be important factors indicating the volume phase transition.

  • This study was carried out with the view of fundamental investigating to improve the tactile and the hygroscopicity of Poly(ethylene Terephthalate) (PET)fibers. Mono-sodium ethylene glycolate in ethylene glycol (MSEG-EG) solution was prepared and PET films were treated with it. The following conclusions were obtained. When PET films were decomposed in MSEG-EG solution, decomposition rate constant showed an exponential relationship with treating temperature; activition energy was 23.30 Kcal/mol, activation enthalpy was 22.52~22.60 Kcal/mol and activation entropy was -29.20~ -29.41 e.u. On the basis of the results obtained above and structure identification of decomposition products, it was found that the decomposition reaction proceeded through ester interchange reaction.

  • This experiment was carried out to investigate the effects of alcohols and carbohyrates upon heat resistance of the amylase produced by Bacillus subtilis var. M-181. The results obtained are as follows: 1) Alcohols showed the effects of increasing heat resistance in the order of glycerin, propylene glycol, ethylene glycol, ethanol and methanol, while higher monohydroxy alcohol and dipropylene glycol affected reversely. 2) Inositol affected on heat resistance of the amlase better than sugar alcohols which effects were in the order of mannitol, dulcitol and sorbitol. 3) Monosaccharides affected on heat resistance of the amylase in the order of mannose, galactose, xylose, glucose, arabinose, rhamnose and fructose. 4) Among oligosaccharides, the effects of increasing heat resistance were recognized in the order of melibiose, lactose, maltose and sucrose. 5) Polysaccharides showed the effects of increasing heat resistance in the order of glycogen, soluble starch and dextrin, while the effects of inulin were not recognized.

  • The resin matrix in the dental composite is generally composed of 2,2-bis[4-(2-hydroxy-3-methacryloyloxy propoxy) phenyl] propane (Bis-GMA) as a base resin and triethylene glycol dimethacrylate (TEGDMA) as a diluent for the reduction of viscosity. The applications of dental composite were often limited in dentistry due to the relatively large amounts of volumetric shrinkage during polymerization and water uptake caused by the addition of TEGDMA to the resin matrix. In this study, in order to solve problems stemmed from the TEGDMA by reducing amount of diluent added to resin matrix, diethylene glycol dimethacrylate (DEGDMA) and ethylene glycol dimethacrylate (EGDMA) were explored as new diluents. A decrease in the volumetric shrinkage and an increase in the mechanical strength were observed by replacing TEGDMA in the dental composite to DEGDMA (or EGDMA). Reduction in the mechanical strength of the dental composite containing DEGDMA (or EGDMA), was not serious in comparison with that of the dental composite containing TEGDMA after water uptake.

Quantitative Evaluation of Free CaO in Electric Arc Furnace Reduction Slag using the Ethylene Glycol Method (에틸렌 글리콜법을 이용한 전기로 환원슬래그의 Free CaO 정량 평가에 관한 연구)

  • Blast furnace slag has been actively used as a substitute for cement in the construction field with high value-added through resource recycling research. However, most of the slag cannot find a clear recycling purpose. This is because some slags contain unstable materials and are used for road-use asphalt and embankment, which are low value-added materials. Electric arc furnace reduction slag(ERS) has been reported to contain a large amount of unstable free CaO due to deoxidation and component adjustment. In this study, free CaO of ERS which is generated in Korean steelmakers is quantitatively evaluated by using ethylene glycol method. As a result of free CaO quantitative evaluation of ERS, it was confirmed that there is a big difference according to the location of each field. In addition, ERS generally existed in powder form as undifferentiated characteristics, but it was confirmed that free CaO content was different due to hydration product in aggregate form due to water treatment. In addition, free CaO is an amorphous material and its crystallization characteristics are different due to the influence of temperature when it is cooled. ERS requires a long-term aging period as it contains a lot of free CaO.

Effects of Exposure to Vitrification Solution on Maturation, Fertilization and Development of Immature Porcine Oocytes In Vitro (유리화 동결액 노출이 돼지 미성숙 난포란의 성숙율, 수정율 및 배발달율에 미치는 영향)

  • This study was conducted to investigate the toxi-cological effects of different vitrification solution on development of immature porcine oocytes in vitro. Oocytes were exposed to EFS solution [40% ethylene glycol (EG) + 18% Ficoll + 0.3M sucrose], ES solution (5.5M EG + 1.0M sucrose) or GE solution [10% glycol (G) + 20% EG], and these oocytes were transferred to sucrose solution directly. Maturation rates were significantly (P<0.05) higher in the ES solution (44.5%) and control (57.6%) than in the EFS solution (38.8%) and GE solution (22.4%). No differences among three solution were found in fertilization rates. Cleavage rates was significantly (P<0.05) higher in the ES solution (47.1%) and control (65.9%) than in the EFS solution (21.9%) and GE solution (19.0%), but no difference among three solutions was found in the blastocyst formation rates. These results indicate that combination of EG and sucrose solutions had effects on development of immature porcine oocytes.

Effect of Ethylene Glycol (EG) on the Viability of Mammalian Embryo during Cryopreservation (포유류 초기 배아의 동결 시 생존율에 미치는 Ethylene Glycol(EG)의 영향)

  • Ethylene glycol (EG) has been successfully used as a cryoprotectant for vitrification of mammalian embryos (including human embryos) due to its low formula weight and high permeation into cells compared with other cryoprotectants, including propylene glycol (PROH). Cryopreservation is able to store the surplus pre-embryos for freezing and furthermore thawing and transfer in a subsequent cycle. This study was carried out to evaluate the effects of embryonic stage, cryoprotectant, and freezing-thawing method on the rates of survival and development of the cryopreserved mouse early embryo and finally to establish the cryopreservation method of surplus embryos obtained during assisted reproductive technology (ART). Female ICR mice (6~8 weeks old) were induced to superovulate by sequential intraperitoneal injection of 5 IU PMSG and 5 IU hCG 48 h apart. Mouse embryos were collected according to its developmental stage after the injection of hCG. Embryos were cryopreserved not only during cryoprotectant step (1~4 step) but also in a variety of media (HTF, IVF medium, D-PBS) and cell stage. The results were as follows : There is no clear advantage in these freezing media of rapid method, but 4 cell and 8 cell of slow method (2, 3 and 4 step) have advantage in D-PBS. The development of embryos according to cell stage become greater in 8 cell stage. In the treatment steps of cryopreservation, the development of embryo to blastocyst was similar among rapid method, but the development of 4 cell and 8 cell embryos to blastocyst according to slow method was better than rapid method.
