Clinical, Morphological and Immunohistochemical Characteristics of Canine Lipid-Rich Sertoli Cell Tumor

  • Some canine typical Sertoli cell tumors (SCT) induce signs of hyperestrinism. However, whether variant lipid-rich SCTs have signs of hyperestrinism remains largely unknown. In the present study, an 11-year-old male Pekingese dog showed significantly elevated serum estrogen and characteristic signs of hyperestrinism such as gynecomastia and alopecia. Cytological, radiological and ultrasound examinations found testicular mass, prostatitis with squamous metaplasia, and cystitis. Pathologically, the tumor lesions consisted mainly of lipid-rich tumor cells with signet-ring appearance, which were immunohistochemically positive for vimentin and anti-$M{\ddot{u}}llerian$ hormone. Based on the findings, a diagnosis of lipid-rich Sertoli cell tumor was established. In conclusion, a canine lipid-rich SCT induced signs of hyperestrinism and caused prostatitis via squamous metaplasia due to its excessive secretion of estrogen.

Effect of Natural Compounds on Catechol Estrogen-Induced Carcinogenesis

  • The hydroxylation of estradiol results in the formation of catechol estrogens such as 2-hydroxyestradiol ($2-OHE_2$) and 4-hydroxyestradiol ($4-OHE_2$). These catechol estrogens are further oxidized to quinone metabolites by peroxidases or cytochrome P450 (CYP450) enzymes. Catechol estrogens contribute to hormone-induced carcinogenesis by generating DNA adducts or reactive oxygen species (ROS). Interestingly, many of the natural products found in living organisms have been reported to show protective effects against carcinogenesis induced by catechol estrogens. Although some compounds have been reported to increase the activity of catechol estrogens via oxidation to quinone metabolites, many natural products decreased the activity of catechol estrogens by inhibiting DNA adduct formation, ROS production, or oxidative cell damage. Here we focus specifically on the chemopreventive effects of these natural compounds against carcinogenesis induced by catechol estrogens.

Effect of Growth Hormone and Androgen on Vitellogenin and Estrogen Receptor Gene Expression in the Japanese eel, Anguilla japonica (뱀장어 Vitellogenin과 Estrogen 수용체 유전자 발현에 대한 성장호르몬 및 웅성호르몬의 영향)

  • Vitellogenin(Vg) is a sex specific serum protein present in sexually maturing female blood of oviparous vertebrates. Estrogen($E_2$) is a main inducer of hepatic Vg synthesis. We investigated the effects of androgen and growth hormone(GH) on regulation of Vg and estrogen receptor(ER) genes in Japanese eel. Immature eels($200{\sim}250\;g$) were given a single injection of $E_2(5{\sim}5,000\;{\mu}g/kg\;bw)$ alone, or in combination with eel recombinant GH(eGH, $1{\sim}10\;{\mu}g/kg$) or methyltestosterone(MT, $1{\sim}5\;mg/kg$) and sacrificed 10 days after the hormone treatments. Expression levels of ER and Vg genes from the liver were determined by means of reverse transcription and polymerase chain reaction(RT-PCR). Administration of $E_2$ stimulated Vg gene expression in a dose dependent manner. Levels of Vg mRNA after the injection of $E_2(500\;{\mu}g/kg)$ with MT(5mg/kg) or eGH($10\;{\mu}g/kg$) were much higher than in that of $E_2$ alone($500\;{\mu}g/kg$). Whereas, injection of either vehicle, eGH ($10\;{\mu}g/kg$) or MT(5mg/kg) alone did not induce the expression of Vg gene in the liver. ER mRNA was detected from the fish treated with vehicle alone. $E_2$ injection($5{\sim}500\;{\mu}g/kg\;bw$) increased this ER expression but dose dependent response was not clear. Addition of MT(5mg/kg) or eGH($10\;{\mu}g/kg$) did not affect $E_2-stimulated$ ER mRNA expression. This study confirms the necessity of $E_2$ as the primary factor for Vg gene expression and requirement of additional hormones such as MT or GH for the full expression of Vg mRNA, and suggests that the additive effect of MT or GH on Vg gene expression would be mediated by some unknown factors other than ER.

The Effects of regular physical activities on female senior adults aging related hormone levels and a risk factor in cardiovascular disease (규칙적인 신체활동 참여가 여성노인의 노화과련 호르몬 농도 및 심혈관질환 위험요소에 미치는 영향)

  • A purpose of this study is to investigate the effects of 12-Weeks regular physical activities on female senior adults aging related hormone(HGH, DHEA-S, estrogen) levels and a risk factor in cardiovascular disease(adiponectin, hs-CRP). Twenty female senior adults participated in this study and divided intoexercise group(n=10) and control group(n=10). The Korean traditional dance program was conducted three times a week(60min) for 12-weeks. Paired t-test was used to examone the differences within experimental groups and independent t-test was used to changes the differences between groups. The followings are the results of this study. Estrogen(p=0.025), HGH(p=0.009), DHEA-S(p=0.009), adiponectin(p=0.014) was significantly increased after 12-weeks program for exercise group, And hs-CRP(p=0.010) was significantly decreased for exercise group. Estrogen(p=0.006), DHEA-S(p<.0.002), adiponectin(p=0.008), hs-CRP(p=0.004) was significantly changed between exercise group and control group.

Concentration Effect of Estrogen Receptor-${\alpha}$ Selective Agonist on the Morphology of Reproductive Organs of the Male Mice (수컷 생쥐 생식기관의 형태에 미치는 에스트로겐 수용체 알파의 선택적 촉진제의 농도별 영향)

  • Known as a female hormone, estrogen, has an effect on the male reproductive organs. The estrogen has to combine with the estrogen receptor to communicate a signal. Propyl pyrazole triol (PPT) is an estrogen receptor alpha selective agonist with a 410-, or 1,000-fold relative binding affinity for estrogen receptor alpha versus estrogen receptor beta. In this study, adult male mice were treated weekly with subcutaneously injections of PPT (0.01 mg, 0.1 mg, 1mg and 4 mg) suspended in castor oil (as control) for 8 weeks and observed histologically changes in testis, efferent ductule and epididymis. In the high concentrations of PPT 4 mg treatment group, a remarkable reduction was observed in the weight of the body, testis and epididymis. Microscopic examination revealed a reduction in seminiferous tubular diameter of the testis, and epithelial cell height of the epididymis in treated group during the experiment. In addition, as the diameter of the efferent ductule increased gradually, the height of epithelial cells was decreased. PPT 4 mg treatment group caused inhibition of spermatogenesis due to atrophied germinal epithelium in the testis, and decrease of adipocyte size attached to the epididymis. Sperm was not observed in the caudal epididymis of PPT 4 mg treated group. In conclusion, the injection of high concentrations of PPT into adult male mice induced physiological changes, such as an inhibition of spermatogenesis, and also histological changes within the reproductive organs.

Detection of Tissue-specific Expression of Porcine Cytochrome P450 Aromatase Genes by Use of Denaturing High Performance Liquid Chromatography(DHPLC) Technique (DHPLC 기술을 이용한 돼지 Cytochrome P450 Aromatase 유전자의 조직 - 특이적 발현양상 관찰)

  • Cytochrome P450 aromatase is the enzyme responsible for biosynthesis of female sex hormone(estrogen) and 19-nortestosterone(nandrolone), a unique steroid hormone endogenously synthesized in the pig. By use of RT-PCR coupled with DHPLC technique (WAVE analysis), expression pattern of isoforms of porcine cytochrome P450 aromatase gene was investigated. Relatively higher expression of aromatase mRNA was observed in testis than in ovary and this result accounted for the previous findings of higher blood estrogen level in male compared with female in this species. The result from the DHPLC demonstrated that PCR amplified DNA fragments of ovary and testis tissues. using unique PCR primers for all three types of aromatase genes, were different from those of type II and ill genes. Further nucleotide sequence analyses of the plasmid clones containing the PCR products revealed that nucleotide sequences of all clones were identical to type I aromatase gene(ovary type). Thus, the result from the present study indicates that the ovary and testis express the same type of aromatase gene. Therefore, the efficacy of DHPLC techniques used for this study helped us to analyze tissue-specific expression of isoform of genes containing the nucleotide sequences with high homology.

Effects of Soy and Isoflavones on Bone Metabolism in Growing Female Rats (성장기 암컷 쥐에서 콩 단백질과 Isoflavones가 골 대사에 미치는 영향)

  • The purpose of this study was to determine which differences in the source of protein (soy vs casein) and isoflavones in soy protein are responsible for the differential effects of bone marks and hormones in growing female rats. Forty-two 21-day-old Sprague-Dawley female rats were randomly assigned to one of three groups, consuming casein (control group), soy protein isolate (57 mg isoflavones/100 g diet), or soy protein concentrate (about 1.2 mg isoflavones/100 g diet). All rats were fed on experimental diet and deionized water ad libitum for 9 weeks. Bone formation was measured by serum osteocalcin and alkaline phosphatase (ALP) concentrations. And bone resorption rate was measured by deoxypyridinoline (DPD) crosslinks immunoassay and corrected for creatinine. Serum osteocalcin, growth hormone, estrogen and calcitonin were analyzed using radioimmunoassay kits. Diet did not affect weight gain and mean food intake. Food efficiency ratio was lower in the soy protein groups. The soy isolate group had a higher ALP and osteocalcin concentration and lower crosslinks value than the casein group. Therefore, the soy isolate groups had a higher bone formation/resorption ratio than the casein group. And, the soy group had significantly higher growth hormone than the casein group. The findings of this study suggest that soy protein and isoflavones are beneficial for bone formation in growing female rats. Therefore, exposure to these soy protein and isoflavones early in life may have benefits for osteoporosis prevention.

Influence of Occupational Exposure to Bisphenol A on the Sex Hormones of Male Epoxy Resin Painters

  • Epidemiological studies demonstrate an adverse effect of several environmental and occupational exposures on male sex hormone. Bisphenol A (BPA) is a weak estrogen and a widely used industrial chemical. Epoxy resin painters may be continuously exposed to BPA at high concentrations. The effect of occupational exposure of BPA on male reproduction was examined by measuring the urinary BPA, testosterone and gonadotropic hormones of epoxy resin painters in the shipyard. The painters had significantly higher concentrations of urinary BPA (2.61${\pm}$1.08 ${\mu}g$/g creatinine) than controls (1.38${\pm}$0.5 9 ${\mu}g$/g creatinine). In serum, the testosterone level of painters was significantly decreased but the luteinizing hormone (LH) and follicle-stimulating hormone (FSH) levels of painters were significantly higher than controls. Occupational exposure to BPA influences testosterone and gonadotropic hormones in male workers.
