  • 여성의 breast cancer의 1/3 의 경우가 hormone-dependent에 의한 것이다. 따라서 그 치료의 접근 방법으로써 estrogen receptor에 작용하거나, receptor-mediated gene trenscription을 저해하는 antiestrogen을 사용하는 것이 있다. Estrogen은 microsomal cytochrome P-450 enzyme complex system에 의해 androgen으로부터 생합성되는 hormone이며, estrogen product는 steroid product의 biosynthetic sequence의 마지막 단계에서 생산되고, aromatase는 이에 관여하는 enzyme으로써, aromatase를 선택적으로 저해하는 경우에 다른 steroid의 생산에 영향을 미치지 않고 estrogen 생산을 감소 시킬 수 있다. 이러한 관점에서, aromatase를 선택적으로 저해하는 것은 hormone-dependent breast cancer를 완화시킬 수 있는 가능성을 가진 cancer chernopreventive agents의 새로운 방법이라 할 수 있다.

  • Estrogen is the most important endocrine hormone that has reproduction and physiological process in a number of tissues. However, an excess of estrogen can promotes the growth of hormone-dependent breast cancer. Thus the regulation of estrogen level is important a prevention of estrogen-related cancer. It has been reported that some of flavonoids could inhibit estrogen-dependent cancer. And these compounds are expected as chemopreventive agents on estrogen related disease. (omitted)

  • Hormone이 암발생과 밀접한 관계가 있음은 이미 1935년대에 밝혀진 바 있다. 즉 lacassagne은 estrogen이 토끼 자궁벽의 fibromyomatous transformation에 영향을 준다고 하였다. Nelson는 guinea-pig를 시험동물로 해서 estrogen을 장기투여한 결과 uterine fibroids가 수발됨을 확인한 이래, 유발됨을 확인한 이래, 이러한 hormone이 암발생의 장기요인이 됨으리 밝히게 되었다. 이러한 업적은 Lipschutz에 의해 더욱 수식되었다. 이러한 업적의 결과가 여러편의 종설로 발표되었다. 즉 Lacassagne, Zuckerman, Burrows등, Gardner는 각기 종설을 발표하여 이를 확증하기에 이르렀다. 한편 각개 hormone, 그중에서도 sex-hormone의 opposite action은 Steinach등 및 Sand에 의해 밝혀졌고 거세백서에 estrogen을 투여했을때 유기되는 vaginal mucosa의 epidermization이 testosterone의 투여에 의해 급속히 소진됨을 Currier 일파가 증명하였고 estrogen에 의해 유발된 웅백서 부속성선의 조직변성이 androgen의 동시투여의 소실됨이 homone에 의해 유발된 neoplastic에 대해 유효하리하는 것은 백명한 일이고 이러한 사실은 현재 실시로 허다히 이용되고 있는 것이다.

Estrogen, Androgen, and Retinoic Acid Hormone Activity of Ginseng Total Saponin (인삼 총 사포닌의 에스트로젠, 안드로젠, 레티노익산 호르몬 수용체에 대한 활성)

  • Alternative or complementary medicine plays an important role in health care system. Ginseng, being one of the most popular oriental herbs, is believed to contain various steroid hormone activity. Ginseng has been demonstrated pharmacological effect in the cardiovascular, endocrine, central nervous, and immune system. Our objective was to study that total saponin might mediate some of their actions by binding to the steroid hormone receptor, as they share many of the actions of steroid hormone in various physiological system. Using total saponin from Panax Ginseng, we have studied the possibility of total saponin being a potential estrogen receptor, androgen receptor, and retinoic acid receptor ligand. Total saponin activated the transcription of both the estrogen and androgen responsive luciferase reporter plasmids at a concentration of 100$\mu\textrm{g}$/ml in COS cells transiently transfected with the corresponding receptor and hormone responsive receptor plasmids. And total saponin caused a concentration-dependent stimulation of estrogen receptor. Total saponin increased the expression of estrogen responsive c-fos proto-oncogene at the protein level in MCF7 cells at 24 h treatment as examined by Western analysis. The c-fos induction was used as a specific marker of estrogen responsiveness. This activation was inhibited by the specific estrogen receptor antagonist, ICI 182,780. However, total saponin failed to activate the retinoic acid receptor in COS cells transiently transfected with the corresponding receptor and retinoic acid responsive reporter plasmids. These results show that total saponin is capable of activating estrogen and androgen receptors.

  • 여성호르몬으로 잘 알려진 estrogen은 난소의 granulosa 세포와 정소의 Sertoli 세포에서 주로 생성되는 것으로 알려져 있으며 최근엔 사람의 지방, 근육, 뇌, 뼈세포 등에서의 합성 가능성과 각 조직에서 생성되는 estrogen 의 생리학적인 기능에 관해 많은 관심이 모여져 왔다. 다른 steroid처럼 지방친화적인 (lipophilic) estrogen은 세포막과 핵막을 쉽게 통과하여 목표세포 (target cell)의 핵에 존재하는 estrogen receptor (ER)에 결합하여 특정 유전자를 발현에 관여하는 것으로 알려져 있다. Cytochrome P450 Aromatase (간략히, aromatase)는 steroid hormone을 합성하는 마지막 단계에서 androgens을 estrogens으로 전환시키는데 관여하는 효소이다. Aromatase의 substrate (기질)로 사용되는 androgen에는 androstenedione과 testosterone 등이 있으며, 최종 산물로써 estrone(E1)이나 17$\beta$-estradiol(E2) 등의 estrogen이 생성된다. Aromatase는 steroidogenic tissues의 세포내 골지체에 존재하는 것으로 알려져 있으며, NADPH-cytochrome P450 reductase와 함께 복합체로써 존재한다. NADPH-cytochrome P450 reductase는 다른 steroid hormone에 관여하는 효소들과도 복합체를 형성하여 다른 steroid hormone을 합성할 수 있으므로, 어떤 조직에서 여성호르몬이 만들어질 수 있느냐 하는 것은 그 조직에서 aromatase 단백질이 만들어지느냐에 따라서 결정된다.

  • In situ and circulating estrogen is the most important endocrine hormone that promotes the growth of hormone-dependent breast cancer. Consequently. decrease of estrogen on in situ and circulation can inhibit breast cancer. Estrogen is mainly produced by the ovary in premenopausal women and by peripheral tissues such as adipose tissues in postmenopausal women. Effect of Calcium on Estrogen and Follicle Stimulating Hormone Secretion in Rabbits

  • The effect of calcium gluconate on estrogen (estradiol) serum level as well as follicle stimulating hormone level was studied. Our results revealed that oral administration of calcium gluconate (100 mg/kg body wight) to adult non-pregnant female rabbits caused a significant increase of serum levels of estradiol and follicle stimulating hormone. On the other hand, oral contraceptive (Norminest tablets) decreased significantly follicle stimulating hormone serum level, while combined administration of calcium gluconate and oral contraceptive caused significant increase of serum level of follicle stimulating hormone compared with control values. Also, concurrent administration of calcium gluconate and Norminest tablet increased significantly the rate of conception compared with group recieved Norminest tablets only. These results indicated that combined administration of calcium and oral contraceptives must be cautiously.

  • The present study aims to discover the effects of Dance sports program upon the hormone reaction of middle aged women. For this aim, 18 women in their 40 s were randomly selected and assigned to the experiment group and control group, evenly. The experiment group underwent a 12-week lesson of Dance sports program. Before training, and 4, 8 and 12 weeks after the onset of the lesson, the effects of hormone reaction of the group were investigated and the outcomes of the analysis could be concluded as follows: Both estrogen and growth hormone in the experiment group 12 weeks later result compared with the control group 0 week that practiced Dance Sports programs increased significantly (p<.001). In conclusion, it may be said that the 12-week session of Dance sports program exercised a positive effect to training seems to suggest the possibility of the dance lesson to have effects on the increase of estrogen and growth hormone. The study indicates a number of further are as into understanding that is worked out in consideration of practice period and proper weight for more efficient results, instead of a program that is just composed of an aerobic exercise only.

  • The authors observed the effect of estrogen and progesterone on the submaxillary glands of female albino rabbits ovariectomized bilaterally. The animals were injected estrogen 25 ㎍ /kg, day intramuscullarly for 1 week, Progesterone was also injected intramuscullarly 0.5 ㎍/kg, day for five days after estrogen injection. After hormone injection, salivary glands were removed under ether anesthesia and the sections were stained with H-E stain for microscopic examination. In the estrogen injected-ovariectomized rabbits, glandular epithelial cells and a part of striated duct cells have high reactivity for hematoxylin and observed the proliferation or hypertrophy of interlobular connective tissues. In the estrogen-progesterone injected group, lesser significant changes than estrogen injected group were observed.

  • After infection of male mice with the plerocercoids (spargana) of Spirometra mansoni, serum levels of estrogen and testicular weight were analyzed by enzyme-linked immunosorbent assay (ELISA) and weighing machine, respectively. The serum level of estrogen increased progressively in infected mice compared with normal controls, whereas the testicular weight of infected mice decreased significantly (P < 0.05). These results suggest that certain substances from spargana change the steroid hormone metabolisms in the host by unknown pathways, and chronic infection may contribute to change of the function of steroid hormone target organ, i.e., testis, in male mice.
