Essential oil yields and chemical compositions from 5 populations of Chamaecyparis obtusa with several environmental factors were investigated through essential oil extracted distillation apparatus and metabolite profiling by GC-MS analysis. Among the populations, content of essential oil at Gokseong was significantly higher than other populations. To compare the several environmental factors affecting on chemical composition and essential oil yields from C. obtuse at Gokseong, the environmental factors (soil condition, temperature, humidity, and moisture content) were measured during 1 year. The essential oils at Goksung based on humidity on March, July, and November was significantly different from other months. The essential oils at Goksung based on temperature on July and August was significantly different from other months. The essential oils at Goksung based on the moisture content on September were significantly different from other months. The percentage of T-N, OM, and yield of oil at Gokseong were significantly different on from other populations. The main constituents of C. obtusa at all populations were ${\alpha}$-pinene, ${\beta}$-pinene, ${\alpha}$-terpinene, ${\gamma}$-terpinene, terpinene-4-ol, isobonyl acetate, terpinyl acetate, and cedar acetate. Specially, Essential oil compositions (%) of ${\alpha}$-terpinene and cedar acetate were higher at Gokseong than at other populations. The chemical compositions of essential oils were variable depend on populations and environmental conditions. Therefore, this study might be used as fundamental research on study for selection of high productive terpenoids and for understanding about biosynthesis of essential oils in C. obtusa.
This study was conducted to establish standard fertilization rates for the cultivation of Korean valerian(Valeriana fauriei var. dasycarpa Hara) which is useful for medicinal and aromatic resources. Nitrogen, phosphate and potassium were treated with four application levels, 0, 6, 9, 12kg per 10a, and the growth characteristics, root yield and essential oil compositions of valerian were measured. The growth and root yield of valerian were the highest in the rate of 9kg per 10a for nitrogen, phosphate and potassium, respectively. Nitrogen had the most powerful effective followed by potassium and phosphate. The contents and compositions of essential oil also varied with the amount of fertilization. The contents of the essential oil was the highest in the application level of 9kg per 10a for nitrogen, phosphate and potassium, respectively. The essential oil compositions were affected by the fertilization rate.
The chemical compositions and antibacterial and antifungal activities of essential oils extracted from Abies holophylla and A. koreana were investigated. GC-MS analysis revealed that 38 compounds comprised 95.88% of the A. holophylla essential oil, with the main components being bicyclo[2.2.1]heptan-2-ol(28.05%), ${\delta}3$-carene(13.85%), $\alpha$-pinene(11.68%), camphene(10.41%), dl-limonene(7.61%), $\beta$-myrcene(7.11%), trans-caryophyllene(5.36%), and $\alpha$-bisabolol(3.67%). In the essential oil from A. koreana, 36 compounds comprised 98.67% of the oil, and the main compounds were bornyl ester(41.79%), camphene(15.31%), $\alpha$-pinene(11.19%), dl-limonene(8.58%), fenchyl acetate(5.55%), and $\alpha$-terpinene(2.29%). Both essential oils showed great potential of antibacterial activity against several bacteria tested, in the range of 2.2-$8.8{\mu}g$ per disc by the agar disc diffusion method, and minimal inhibitory concentration(MIC) values of 5.5-21.8 mg/ml by the microdilution method. Both oils showed very effective antifungal activities toward all pathogenic strains tested, including Candida glabrata, with MIC values in the range of 0.5-2.2 mg/ml. As a whole, A. koreana oil showed better antibacterial and antifungal properties than A. holophylla oil.
The purpose of the present research was to evaluate the antimicrobial activity of the essential oil extracted from Pinus koraiensis seed against pathogens related to acne. The essential oil was extracted by steam distillation method. The chemical compositions of essential oil were analyzed by GC-MS. Alpha-pinene (29.87%), D-limonene (19.26%), betapinene (11.19%), beta-myrcene (3.84%), n-hexadecanoi acid (3.2%), beta-caryphyllene (2.72%), and cyclohexene (2.17%) were main components. This essential oil had antimicrobial activities against Malasseizia furfur, Propionibacterium acnes, and Staphylococcus epidermidis.
Callus was derived from the seedlings of Agastache rugosa(Labiatae). The growth rate of callus and the production of essential oil were studied with the variation of culturing conditions. 2, 4-D 2ppm in the medium was more effective for the production of essential oil than NAA 2ppm. The growth rate of callus and the production of essential oil were inhibited by the illumination of the light. The essential oils from Agastache rugosa and the callus cultivated on the medium containing 2, 4-D 2 ppm and kinetin 0.2 ppm were analysed by TLC, gas chromatography and mass spectrometry. These two oils showed different compositions. The main component of the plant oil, methyl chavicol was not contained in the callus oil.
The chemical compositions, and antibacterial and antifungal effects of essential oils extracted from three coniferous species, Pinus densiflora, Cryptomeria japonica, and Chamaecyparis obtusa, were investigated. Gas chromatography mass analysis of the essential oils revealed that the major components and the percentage of each essential oil were 16.66% $\beta$-phellandrene and 14.85% $\alpha$-pinene in P. densiflora; 31.45% kaur-16-ene and 11.06% sabinene in C. japonica; and 18.75% bicyclo [2,2,1] heptan-2-ol and 17.41% 2-carene in Ch. obtusa. The antimicrobial assay by agar disc diffusion method showed that $2.2{\mu}g$ of Ch. obtusa oil inhibited most effectively the growth of Escherichia coli ATCC 33312 and Klebsiella oxytoca ATCC 10031, whereas the C. japonica oil gave weak antimicrobial activity. The minimal inhibitory concentration(MIC) values for bacterial strains were in the range of 5.45-21.8 mg/ml depending on essential oils, but most Gram-negative bacteria were resistant even at 21.8 mg oil/ml. P. densiflora oil showed the most effective antifungal activity and the MIC values for Cryptococcus neoformans B42419 and Candida glabrata YFCC 062CCM 11658 were as low as 0.545 and 2.18 mg/ml, respectively. Cryp. neoformans B42419 was the most sensitive to all essential oils in the range of 0.545-2.18 mg/ml. Our data clearly showed that the essential oils from the three conifers had effective antimicrobial activity, especially against fungi.
Angelica acutiloba is one of the most intensively cultivated medicinal plants in Korea. The roots of this plant have been used as an important herbal drug, especially for the treatment of various female disorders, as the traditional therapy in Korea and other Asian countries. Consumption of its fresh leaves as a healthy vegetable has recently increased. In this study, essential oil fractions were extracted from the roots and leaves of this plant by steam distillation. Compositions of the two oils were compared by gas chromatography-mass spectrometry (GC-MS). The antibacterial activities of the essential oil were determined against three strains of Escherichia coli. DPPH radical scavenging and reducing power tests were performed to evaluateits antioxidant activities. The cytotoxic activities of the essential oil against a human breast and a uterine cancer cell line were estimated by MTT tests. Additionally, the morphological changes after treatment of the oil fraction were observed under a microscope. The essential oil fraction and its main components, Z-ligustilide and butylidene phthalide, inhibited the growth of three E. coli strains examined, with minimum inhibiting concentrations (MICs) ranging from 1.0 mg/ml to 8.0 mg/ml. Additionally, the essential oil fraction of A. acutiloba exhibited significant DPPH free radical scavenging activity and reducing power. Significant cytotoxic activities of the A. acutiloba essential oil were observed for human uterine (Hela) and breast (MCF-7) cancer cell lines.
The essential oils from the roots of Codonopsis lanceolata and the cultivated callus were analysed and compared by gaschromatography-mass spectrometry. In the experimental study of cell culture, it appeared that 2,4-dichlorophenoxy acetic acid in the culture medium induced higher production of essential oils in the callus than indole acetic acid. The growth of callus was inhibited by illumination of the light. The production of essential oil in cultured cells was increased by the addition of biosynthetic precursors. The essential oils from the roots of Codonopsis lanceolata and the cultured callus showed different compositions. Tetradecanoic acid, 1,1,-dimethoxyl 4-methoxy phenol, 9,12-octadecanoic acid and hexadecanoic acid were identified as main components of the cultured callus oil.
The role of fat is important from nutritional standpoint. The physiological functions of fat are energy yielding as well as the carrier of fat soluble vitamins, with special activities of essential fatty acids. It is fortunate that Korean families prefer to use vegetable oils rather than those from animal origin. But the problems are focused on better qaulity of food oils with attempt to exploit the available resources. This study was undertaken to inevestigate the fatty acid compositions of Korean origin both from conventional and unconventional resources of gas-liquid chromatography. The results obtained are as follows. 1. The total lipid contents of seeds of red pepper, Korean squash, sesame, perilla (var Japonica), and Korean pine nuts and walnuts were 24.3%, 56.6%, 56.4%, 46.9%, 69.8%, and 67.2%, respectively. 2. The saponification numbers of samples ranged from 190 to 200. It showed that the mean molecular weights of fatty acids composing the lipids were very much closed each other. 3. Iodine numbers of varing seed oils ranged from 96.5 of Korean squash seed oil to 124.6 of walnut oil. Oils obtained from squash seeds and sesame seeds showed significantly lower value, while others ranged from 122 to 125. 4. In the fatty acid compositions, squash seed oil was composed mainly of unsaturated fatty acids, 74.9% of which was oleic acid, whereas red pepper seed oil, pine nut oil, and sesame oil contained linoleic acid as a major component showing 64.4%, 56%, 48.8%, and 47.8%, respectively. In perilla seed oil, the amounts of linoleic and linolenic acids were 14.1% and 58%, respectively which meant nearly three-fourths of the total fatty acidsbeing the unsaturated essential fatty acids. This study will encourage the use of these conventional and unconventional vegetable oils which have highr ratios of polyunsaturated to saturated fatty acids.
This study investigates the chemical compositions of Aster koraiensis Nakai essential oils and the quantitative changes of major terpene compounds according to various harvesting times. The essential oils obtained by hydrodistillation from the aerial parts of Aster koraiensis Nakai are being analyzed by GC and GC-MS. The essential oil compositions of Aster koraiensis Nakai are characterized byhigher contents of sesquiterpene compounds. Ninety-seven volatile flavor compounds are being identified in the essential oils from Aster koraiensis Nakai harvested in 2010, and caryophyllene oxide (8.38%), aristolene (7.08%), epiglobulol (5.57%), and ethyl furanone (4.73%) are the most abundant compounds. Ninety-five compounds are identified in the essential oils from the plants harvested in 2011, and aristolene (11.56%), calarene (9.33%), phytol (8.28%), ethyl furanone (7.63%), and epiglobulol (7.18%) are the most abundant compounds. Ninety-five compounds are being identified in the essential oils from the plants harvested in 2012, and calarene (15.3%), aristolene (14.24%), ethyl furanone (7.21%), phytol (6.98%) are the major compounds. The contents of aristolene, ${\alpha}$-caryophyllene, ${\alpha}$-muurolene, ${\alpha}$-calacorene, aromadendrene oxide, and calarene increase significantly from 2010 to 2012. The contents of ${\alpha}$-cubebene, isocaryophyllene, and diepi-${\alpha}$-cedrene epoxide decrease significantly from 2010 to 2012. The quantitative changes of aristolene and calarene according to harvesting time can be served as a quality index of the Aster koraiensis Nakai essential oils.
