Pharmacophore Modeling, Virtual Screening and Molecular Docking Studies for Identification of New Inverse Agonists of Human Histamine H1 Receptor

  • Human histamine H1 receptor (HHR1) is a G protein-coupled receptor and a primary target for antiallergic therapy. Here, the ligand-based three-dimensional pharmacophore models were built from a set of known HHR1 inverse agonists using HypoGen module of CATALYST software. All ten generated pharmacophore models consist of five essential features: hydrogen bond acceptor, ring aromatic, positive ionizable and two hydrophobic functions. Best model had a correlation coefficient of 0.854 for training set compounds and it was validated with an external test set with a high correlation value of 0.925. Using this model Maybridge database containing 60,000 compounds was screened for potential leads. A rigorous screening for drug-like compounds unveiled RH01692 and SPB00834, two novel molecules for HHR1 with good CATALYST fit and estimated activity values. The new lead molecules were docked into the active site of constructed HHR1 homology model based on recently crystallized squid rhodopsin as template. Both the hit compounds were found to have critical interactions with Glu177, Phe432 and other important amino acids. The interpretations of this study may effectively be deployed in designing of novel HHR1 inverse agonists.

Effects of Stabilized Rice Bran on Obesity and Antioxidative Enzyme Activity in High Fat Diet-induced Obese C57BL/6 Mice (안정화 미강이 고지방 식이로 유도된 C57BL/6 Mice의 비만과 항산화 효소 활성에 미치는 영향)

  • Rice bran (RB), a by-product obtained during polishing of un-milled rice, contains a large quantity of essential nutrients such as minerals, vitamins, fiber, amino acids, and antioxidants. In this study, the anti-obesity effects of stabilized RB addition as a food material in a diet-induced obese animal model were investigated. For the analysis, a total of 32 mice were randomly divided into four groups: normal diet group (ND, n=8), high-fat diet group (HFD, n=8), 20% RB added high fat diet group (HFRB20, n=8), and 40% RB added high fat diet group (HFRB40, n=8). The animals were sacrificed after 7 weeks. Body weight gain, various adipose tissue weights, blood glucose concentration, and serum leptin level of the HFRB40 group were found to be significantly lower than those of the HFD group. Moreover, antioxidant enzyme activity of the HFRB40 group was significantly higher than that of the HFD group. However, the HFRB20 group did not show significantly different values. The results of this study show that RB (40%) addition reduces body weight gain and improves obesity-related parameters. These results suggest RB could be considered as a functional additive material for the manufacture of anti-obesity products, and 40% RB addition would be the physiologically effective level.

Characterization of acetic acid fermentation of detoxified Rhus verniciflua vinegar produced by various yeast strains (효모 종류에 따른 무독화 옻식초의 초산발효 특성)

  • This study was conducted to investigate and compare the characteristics of acetic acid fermentation in detoxified Rhus verniciflua vinegar (DRV) produced by different yeast strains. The DRVs were prepared by static acetic acid fermentation using six different yeast strains (Saccharomyces cerevisiae Fermivin, Pichia kudriavzerii N77-4, Hanseniaspora pountiae HP1-2, Candida tropicalis Y447, Wickerhamomyces anomalus N43-8, and Pichia kluyveri Frootzen). Alcohol content of the S. cerevisiae Fermivin fermented DRV was highest 16.07%. Among the yeast strain DRVs, there were significant differences in alcohol content, but all alcohol levels were 11%. Moreover, there were differences in pH and titratable acidity of the DRVs. The organic acid content of the DRVs ranged from 35.88 to 55.49 mg/mL and there were significant differences among the yeast strain DRVs. Essential free amino acids, particularly glutamic acid, alanine, leucine and valine, were detected in each of the 6 DRVs. Electronic nose analysis revealed that three different volatile chemical patterns were present in the 6 DRVs. The results indicate that yeast strains with different characteristics can produce vinegars with different characteristics.

Physicochemical Characteristics of Agastache rugosa O. Kuntze Extracts by Extraction Conditions (추출조건에 따른 배초향 추출물들의 이화학적 특성)

  • Fifty-seven extracts from flower, leaf and stem of Agastache rugosa O. Kuntze were prepared by varing extraction conditions: solvents (hot water, ethanol and NaCl solution); temp.$(60,\;80\;and\;100^{\circ}C)$ and solvent ratios (10 to 35 times per material). Lipid content of leaf was 9.54% and protein content of the flower where the essential amino acids were most abundant among all parts was 16.8%. Among six minerals (Na. Ca, Fe, P, K and Mg) detected from all parts of the plant, the content of K was the highest. Extraction yield rated higher in odor of water, ethanol and NaCl solution, respectively and the extractions reached eqilibrium in about 15 to 20 min, regardless of extraction conditions. pH values of all extracted solvents maintained neutral upon extraction and a few free sugars in forms of glucose and fructose were detected from ethanol extracts. Colors and organoleptic characteristics of the extracts which determine the properties of final products varied with extraction conditions.

Conditions for Processing of Meaty Textured Fish Protein Concentration from Filefish and Sardine (축육(畜肉) 조직(組織)과 유사(類似)한 말쥐치 및 정어리의 조직(組織) 단백질(蛋白質) 농축물(濃縮物)의 가공(加工) 조건(條件)에 관한 연구(硏究))

  • For the effective utilization of the fish resource in coastal regions, an investigation on optimum processing conditions and meat quality textured fish protein concentrate (FPC) was carried out with the fish meat of filefish and sardine. Optimum pH and sodium chloride content of fish meat were 7.5 and 1.0 %, respectively. The most effective soaking conditions were as follows ; soaking time, 30 min ; temperature of ethanol, 5 to $20^{\circ}C$ ; amount of added ethanol, 3 times the weight of the fishmeat paste ; repeated number of soaking in ethanol for filefish and sardine, 2 and 4, respectively. The ethanol remaining is meaty textured FPC could be removed effectively by forced-air drying. Yields of the product to the minced meat weight and the contents of protein lipid in meaty textured from filefish were 21.1, 77.6 and 0.2 % and those from sardine were 24.3, 75.8 and 3.6 %, respectively. Contents of essential amino acids in meaty textured FPC of filefish and sardine were not inferior to those of beef, textured soybean protein and FAO pattern. Beef meat could be substituted with the meaty textured FPC up to 50 % in the processing of typical meat balls and hamburger without any significant loss in its taste, odor and texture.

Effect of Sea Urchin Shell on Egg Quality (성게껍질이 계란의 품질에 미치는 영향)

  • We investigated the quality characteristics of eggs produced from laying hens fed with non-supplemented diets (A) and diets supplemented with 3% (B) and 5% (C) of sea urchin shell powder for efficient applications of sea urchin shell. There was no significant difference in the proximate composition. Ca and Fe contents of (B) and (C) groups were higher than those of (A) group. Contents of phosphorus and magnesium, however, showed no significant differences among the groups. (B) and (C) groups had higher in essential amino acid contents than (A) group except tryptophan. Taurine was detected in all groups. Analysis of fatty acid showed that (B) and (C) groups contained more unsaturated fatty acids. The DHA contents of (A), (B) and (C) groups were 0.56%, 0.68% and 0.89%, respectively. These results show that sea urchin shell possesses the potential as supplement of laying hens diets to produce functional eggs.

In Vitro Isolation and Proliferation of Mouse Male Germ-Line Stem Cells (생쥐 생식줄기세포의 체외 분리 및 증식)

  • Sperrnatogenesis, the process by which the male germ-line stem cells(GSCs; type A spermatogonia) divide and differentiate to produce the mature spermatozoa, occurs in the seminiferous tubules of the testis. The GSCs proliferate actively to produce two types of cells: other GSCs and differentiating spermatogonia. GSCs have unipotentcy, devoted solely to the generation of sperm. The function of GSCs has broad implications for development, disease, and evolution. Spermatogenesis is fundamental for propagation of species and the defects of this system can result in infertility or disease. The ability to identify, isolate, culture, and alter GSCs will allow powerful new approaches in animal transgenesis and human gene therapy relating to infertility. Until recently, research on stem cells in the testis has been limited because of technical difficulties in isolating and identifying these cell populations. Here, we were trying to find out optimal conditions for in vitro culture of GSCs for identifying and isolating GSCs. We collected mouse GSCs from 3-days old mouse by two-step enzyme digestion method. GSCs were plated and grown on mouse embryonic fibroblasts in Dulbecco's modified Eagle's medium (DMEM) containing 15% fatal bovine serum, 10 mM 2-mercaptoethanol, 1% non-essential amino acids, 1 ng/$m\ell$ bFGF, 10 $\mu$M forskolin, 1500 U/$m\ell$ human recombinant leukemia inhibitory factor (LIF). Over a period 3∼5 days, GSCs gave rise to large multicellular colonies resembling those of mouse pluripotent stem cells. After 5th passages, cells within the colonies continued to be alkaline phosphatase and Oct-4 positive and tested positive against a panel of two immunological markers(Integrin $\alpha$ 6 and Integrin $\beta$ 1) that have been recognized generally to characterize GSCs. SSEA-1, SSEA-3, and SSEA-4 also showed positive signals. Based on our data, these GSCs-derived cultures meet the criteria for GSCs itself and even other pluripotent stem cells. We reported here the establishment of in vitro cultures from mouse male GSCs.

Effects of Beverage Including Extracts of Artemisia capillaris on Fatigue-Recovery Materials, Heart Rate and Serum Lipids in University Male Athletes (인진쑥 추출물을 함유한 음료가 대학 운동선수의 피로회복 물질과 심박수 및 혈청지질에 미치는 영향)

  • The purpose of this study was to investigate the possibility of medicinal plants application as an edible functional food resource. This research was planned to apply Artesmisia capillaris, which is one of the most widely used oriental medicinal resource into beverages, and to evaluate the validity and the nutrients of the developed beverage with Artemisia capillaris. Following this plan, we analyzed contents of the general nutritional composition, mineral and amino acid contents. And to evaluate the improvement of blood circulation and the ability to recover from fatigue, we had a group of athletes to regularly take the beverage before and after exercise and checked the heart rates, and blood components before exercise and after exercise. Brix, pH and titratable acidity of develoued drink were 9.2%, 4.4 and 0.04%, respectively. The approximate nutritional composition of beverage was carbohydrate, 2.70%, crude protein, 6.00%, crude ash, 0.20%, and crude fat, 0.10%. And developed beverage contained essential amono acids and minerals (p, Ca, K, Na, Fe, Mn, Zn). Heart rates, lactic acid, glucose, creatinine, LDH, GOT, GPT and lipid concentratios in blood were decreased after taking the beverage with Artemisia capillaris for 6 weeks, both in stable condition and after exercise. Since the athletes who participated In our research were doing aerobic exercise regularly, we judged that this result was formed not only by aerobic exercise, but also by the function of developed beverage with Artemisia capillaris the athletes took for 6 weeks. We believe that taking the beverage regularly will help the improvement of the public health and the athleticism of the athletes. We hope that this result will be used as the basic resource in developing of the beverage with Artemisia capillaris in the near future.

Screening of the Single Nucleotide Polymorphisms in the Protamine 1 and 2 Genes of Korean Infertile Men (한국 남성 불임환자에서 Protamine 1과 2 유전자의 Single Nucleotide Polymorphism에 관한 연구)

  • Objective: Although several genetic factors have been associated with defects in human spermatogenesis, the unambiguous causative genes have not been elucidated. The male infertility by haploinsufficiency of PRM1 or PRM2 has been reported in mouse model. The aim of this study was to identify the single nucleotide polymorphisms (SNPs) of PRM1 and PRM2, related to the genotype of Korean infertile men. Methods: Genomic DNAs were extracted from peripheral bloods of infertile men with oligozoospermia or azoospermia, and analyzed using polymerase chain reaction (PCR) and direct sequencing. We carried out the direct sequencing analysis of amplified fragments in PRM1 (557 nucleotides from -42 to 515) and PRM2 (599 nucleotides from 49 to 648) genes, respectively. Results: Three SNPs of coding region in the PRM1 gene was found in the analysis of 130 infertile men. However, the SNPs at a133g (aa 96.9%, ag 3.1% and gg 0.0%), c160a (cc 99.2%, ca 0.8% and aa 0.0%) and c321a (cc 56.9%, ca 35.4% and aa 7.7%) coded the same amino acids, in terms of silence phenotypes. On the other hand, as results of the PRM2 gene sequencing in 164 infertile men, only two SNPs, g398c (gg 62.2%, gc 31.1% and ga 6.7%) and a473c (aa 63.4%, ac 29.9% and cc 6.7%), were identified in the intron of the PRM2 gene. Conclusions: There was no mutation and significant SNPs on PRM1 and PRM2 gene in Korean infertile men. These results suggest that the PRM1 and PRM2 genes are highly conserved and essential for normal fertility of men.

Proximate and nutritional compositions of freeze-dried silkworm powder as edible insect resources (동결건조 누에분말의 일반 성분 및 영양 성분)

  • Freeze-dried silkworm powder has been approved as a functional ingredients for blood glucose control since 2009. Proximate and nutrient compositions of silkworm powder was summerized as bellow. Silkworm powder contained 4.7% moisture, 57.7% crude protein, 9.5% crude fat, 5.4% crude fiber, and 9.3% ash. The larval protein contained 18 known amino acid including glutamic acid and aspartic acid. Fat is composed of 3:7 ratio of saturated fatty acid to unsaturated fatty acid including various essential fatty acids. It contained 34.7 linolenic acid, 24.4% oleic acid, 20.2% palmitic acid, 9.2% stearic acid, and 7.7% linoleic acid. Five minerals, K, P, Ca, Mg, S were abundant in the silkworm powder with other minerals such as Cu, Mn, Fe, and Zn.
