  • Tenecin fragments are antimicrobial and antifungal peptide from Tenebrio molitor with highly positive charged amino acid residues. To elucidate their membrane selectivity and molecular mechanism, various forms of tenecin fragments were synthesized, and their interaction with acidic phospholipid, Gram (+), fungal and human erythrocyte membrane were investigated by ANTS/DPX leakage, membrane binding and fusion assay. The Effect of Woohwangcheongsim-won on Circulatory Disturbance in Diabetes (우황청심원이 당뇨병 Rat의 혈액순환장애에 미치는 영향)

  • Object: Death rate due to hypertension, atherosclerosis, ischemic heart disease and cerebral infarction induced by Westernized diet and increased average life span is on the rise. Decrease in blood circulation, activation of thrombus generation and intravascular lipid accumulation, cited as the principal causes of the above mentioned diseases in recent studies, result in circulatory disturbance and blood vessel obstruction leading to ischemic cell death of heart, brain and peripheral vessels. Method: We investigated the biochemical changes in microvascular permeability, aggregation of platelet and the intravascular lipid accumulation in induced-diabetic rat using Streptozotocin. We also studied the effects of Woohwangcheongsirn-won after oral administration on blood circulation, platelet function and lipid metabolism. The results are as follows: I. Woohwangcheongsim-won increased blood circulation in microvessels. 2. Woohwangcheongsim-won increased the reduced erythrocyte deformability in diabetes. 3. Woohwangcheongsim-won induced the reduction of contents of 2, 3-DPG, but failed to affect the reduced contents of ATP in erythrocyte in diabetes. 4. Woohwangcheongsim-won reduced the activity of Ca/sup 2+/-ATPase in the membrane of erythrocyte. 5. Woohwangcheongsim-won reduced the platelet aggregation evoked by platelet agglutinin factor. 6. Woohwangcheongsim-won reduced the production of platelet-derived granules. 7. Woohwangcheongsim-won reduced the production of metabolites of arachidonic acid in diabetes, and also reduced the production of increased thromboxane B2. 8. Woohwangcheongsim-won reduced the synthesis of oxidized LDL-cholesterol. In conclusion, Woohwangcheongsim-won enhanced blood circulation in microvesseles, erythrocyte deformability and inhibited the increased platelet aggregation and the synthesis of oxidized LDL-cholesterol in diabetes. Therefore Woohwangcheongsim-won is believed to positively affect blood circulation (J Korean Oriental Med 2002;23(2):164-179)

  • The effect of captafol on the hematological value, erythrocyte membrane and plasma biochemical value was investigated using blood of SPF Sprague-Dawley male rats in vitro. For the anticoagulations, we used 0.5mg of heparin per $10m{\ell}$ of blood from vena cava. Three con-centrations($0.1{\times}10^{-4}M$, $1{\times}10^{-3}M$ and $1{\times}10^{-2}M$) captafol in ethanol were added to the each $2.5m{\ell}$ blood so that the linal concentration of ethanol was 1%. The blood contained with each concentration of captafol was incubated at $37^{\circ}C$ C for 2 hours under 5% $CO_2$ gas The whole blood and plasma were examined for hematological vaJues, erythrocyte membrane damage and biochemical values, respectively. The results obtained were summarized as follows ; 1. The number of RBC in $1{\times}10^{-2}M$ group and the concentration of MCHC in $1{\times}10^{-3}M$ group were significantly (p<0.05) decreased and increased from that of control values, respectively. The percentage of Hct was significantly (p<0.05) decreased with dose-response. 2. Erythrocyte fragility rate of $1{\times}10^{-3}M$ and $1{\times}10^{-2}M$ group were significantly (p<0.01) increased from that control with dose response. 3. Potassium ion level of $1{\times}10^{-2}M$ was significantly (p<0.05) increased from that of control. 4. The concentration of total bilirubin in the $1{\times}10^{-3}M$ and $1{\times}10^{-2}M$ groups were significantly increased from that of control. The enzyme level of creatine kinase in $1{\times}10^{-2}M$ group was significanlty (p<0.05) decreased from control value.

  • Atg5 and Atg7 have long been considered as essential molecules for autophagy. However, we found that cells lacking these molecules still form autophagic vacuoles and perform autophagic protein degradation when subjected to certain stressors. During this unconventional autophagy pathway, autophagosomes appeared to be generated in a Rab9-dependent manner by the fusion of vesicles derived from the trans-Golgi and late endosomes. Therefore, mammalian autophagy can occur via at least two different pathways; the Atg5/Atg7-dependent conventional pathway and an Atg5/Atg7-independent alternative pathway.

Studies on the Hemolytic Mechanism of Antitumor Platinum Complex (항암성 백금화합물의 용혈독성기전에 관한 연구)

  • This study was designed to determine the hemolytic mechanism of antitumor agent tetraphosphine platinum (II) complex (RC-1), which was synthesized recently. Erythrocytes treated with RC-1 showed concentration and time dependent lipid peroxydation, methemoglobin synthesis and hemolysis. And also treatment of radical scavengers showed the inhibitory effect of hemolysis and the decrease of malondialdehyde levels in RC-1 treated erythrocytes. So, the mechanism of hemolysis was considered to be the generation of free radicals, methemoglobin synthesis and the lipid peroxidation of phospholipid which composed of erythrocyte membrane.

Anti-lipid Peroxidative Effect of Extracts and its Fractions Polygonatum odoratum (둥굴레 추출물 및 분획의 과산화지질 생성 저해효과)

  • The rhizoma of Polygonatum odoratum (Liliaceae) promotes the production of body fluid and relives dryness symptoms and has hypoglycemic effect. In order to evaluate the prevention of lipid peroxidative efficacy of P. odoratum, its extracts (Et$_2$O and MeOH ex.) and its fractions ($H_2O$, 20% MeOH, 40% MeOH, 60% MeOH and 100% MeOH fr.) were measured by TBARS assay on rat liver S9 and human erythrocyte ghost membrane. The order of anti-lipid peroxidative effect was as follows: Et$_2$O ex. >40% MeOH fr. >60% MeOH fr.>100% MeOH fr. Based on these results, we conclude that membrane lipid peroxidation is inhibited in vitro by addition of P. odoratum ether extract and its gradient MeOH fractions excepts $H_2O$ fr., and which have significant hepatoprotective activity in $CCl_4$-treated rats.

Studies on hemolytic reaction of normal chicken serum and sheep erythrocytes (닭혈청(血淸)에 의한 면양적혈구(緬羊赤血球)의 용혈(溶血)에 관한 연구(硏究))

  • Hemolytic reaction of normal fresh chicken serum on sheep erythrocytes was studied and the following experimental results were obtained and summarized. 1. Chicken sera, 258 (78%) out of 344 samples showed hemolytic activity on sheep erythrocytes. 2. Distribution of a different hemolytic titer of chicken sera was not dependent to sex and age difference of test chicken. 3. Hemolytic activity of serum component obtained from normal fresh chicken was heat inactivated at $56^{\circ}C$. 30 minutes heating. 4. The most enhanced hemolytic activity of chicken serum on sheep erythrocytes was observed at the incubation temperature of $46^{\circ}C$. 5. The most effective pH for the hemolytic reaction of chicken serum on sheep erythrocytes was observed at 7.0, and pH 6.0 or 8.5 resulted less or no hemolysis. 6. Hemolytic reaction of chicken serum and sheep erythrocytes required Mg⧻ and Ca⧻ ions as, co-factor, and the former was required more compared to the latter. 7. Hemolytic activity of chicken serum was observed in ChC 2, 4 fraction but not in ChC 1, 3, ChC 3, 4, ChC 1, 2, 4 and ChC 1, 2, 3 fractions. 8. In electron micrography, morphological changes of sheep erythrocyte membrane by normal chicken serum was similar to that of immune hemolysis: that was, the hemolytic hole was circular and it was surrounded with a white ring. 9. Electron micrography of morphological changes on sheep erythrocyte membrane indicated that the size of hemolytic hole and white ring were functional to the chicken serum concentration used and reaction time.

Influence of Heating, Cooling and Acidity on the Permeability of the Rabbit Erythrocyte Membrane (고온, 한냉 및 산도가 토끼 적혈구막 투과성에 미치는 영향)

  • Outward movement of hemoglobin and $K^+$ ion across rabbit erythrocyte membrane after heating, cooling and in acid medium was studied. One milliliter of rabbit blood was centrifuged and packed red cells were obtained. Packed red cells were resuspended by addition of 4 ml of 0.9% NaCl solution and were subjected to heating $(57^{\circ}C\;for\;5\;minutes)$ or cooling $(-4^{\circ}C{\sim}-8^{\circ}C\;of\;-10^{\circ}C{\sim}-11^{\circ}C\;for\;10\;minutes) $. For acid medium experiment packed ref cells were resuspended by addition of 4 ml of acid medium of PH 4.5 consisting of 0.01% glacial acetic acid in 0.85% NaCl solution and kept standing for 10 minutes. All red cell suspensions were centrifuged again and packed red cells were separated. This packed red cells were again suspended in 4 ml of NaCl solution of 0.8%, 0.7%, 0.6%, and 0.5% concentration respectively and kept standing for 20 minutes. The concentration of hemoglobin and $K^+$ in the supernatant of the above red cell suspensions were measured and the following results were obtained. 1. Outward movement of hemoglobin and $K^+$ was greatest in red cells subjected to heating. The movement paralled to the osmolal concentration gradient between extra- and intra-cellular phase of red cells. 2. In acid medium the outflux of hemoglobin and $K^+$ increased as compared to the control. 3. In red cells subjected to the cold of $-10^{\circ}C{\sim}-11^{\circ}C$ the outflux of hemoglobin and $K^+$ increased. Whereas in the environment of $-4^{\circ}C{\sim}-8^{\circ}C$ there was no change in the outflux of $K^+$. The he-moglobin outflux showed rather a decreased as compared to tile control.

Effect on Fatty Acid Composition of red Blood Cell Membranes of supplement of Korean Pinenut Oil, rich in 5-Olefinic acids, in Normocholesterolemic and Atherosclerotic New Zealand White Rabbits (한국산 잣기름이 정상토끼 및 동맥경화증 유발 토기의 적혈구막 지방산조성에 미치는 영향)

  • The present study was carried out in normal and atherosclerotic New Zealand white(NZW) rabbits, to evaluate the effect of dietary supplementation with Korean pinenut oil, on the total fatty acid composition of red blood cell membranes. Erythrocyte fatty acids of chain lengths ranging fro 12 : 0 to 22 : 6$\omega$3 were identified. In study I, palmitic acid(16 : 0) was found to be the main erythrocyte membranes fatty acid, followed in turn by stearic(18 : 0), oleic(18 : 1), and linoleic acids(18 : 2$\omega$6). There was a large increase in the proportion of 20 : 4$\omega$6 in 10% pinenut oil group on days 40 and 80(compared with control), although the increase was not statistically significant. Most erythrocyte fatty acids were less affected by the types or levels of oils supplemented for periods fo 40 and 80days. In study II,at the end of the dietary treatment, the levels of 12 : 0, 14 : 0, 15 : 0 and 18 : 0 were significantly decreased in the 5% soybean oil/cholesterol group, whereas those of 12 : 0, 14 : 0 and 18 : 0 were significantly decreased, but that of 18 : 2$\omega$6 was significantly increased in the 10% soybean oil/cholesterol group, compared to the cholesterol group. No significant differences in fatty acid composition were seen between the pinenut oil/cholesterol group, whereas the 10% soybean oil/cholesterol group had significantly elevated 18 : 2$\omega$6, compared to the cholesterol group. The proportion of 18 : 4$\omega$6 was significantly raised in the 5% soybean oil/cholesterol and 5% pinenut oil/cholesterol group, and the content of 16 : 1$\omega$7 was significantly lowered in the 5% and 10% soybean/cholesterol groups, compared to the cholesterol group at 10 weeks supplementation. In studies I,II and III, the total proportions of saturated fatty acids were not significantly affected by the types of levels of oils supplemented throughout the study period.

  • Phospholipids were examined for their capacity to protect human erythrocytes against hemolysis induced by hypotonic solution, p-hydroxymercuribenzoate or hematin. The following results were obtained. 1. Phosphatidyl choline, lysophosphatidyl choline and phophatidyl ethanoleamine as well as chlorpromazine prevented the osmotic hemolysis of human erythrocytes which occurred due to water influx into erythrocytes from medium, but showed no effect on hematin-induced hemolysis which occurred without the volume change of erythrocytes. 2. Human erythrocytes were found to be most sensitive to the antihemolytic action of phospholipids among mammalian erythrocytes from sheep, rabbit, rat and mouse. 3. Phospholipids at the concentrations showing their strong antihemolytic effect on human erythrocytes against osmotic hemolysis had no influence on methylene blue uptake and volume change of erythrocytes in hypotonic solution. 4. Phospholipids increased erythrocyte deformability 2 to 3 times over control group and there was aclose relationship between their antihemolytic action and increase of deformability as a function of their concentrations. 5. The phospholipids increased the resistance to osmotic hemolysis of human erythrocytes by increasing membrane elasticity through their incorporation into lipid bilayer without altering glucose metabolism and water influx to erythrocytes.
