Establishment of Acceptable Daily Intakes (ADIs) and Risk Assessment for Ephedrine, Menichlopholan, Anacolin, and Etisazole Hydrochloride

  • BACKGROUND: Prior to implementing a positive list system (PLS), there is a need to establish acceptable daily intake (ADI) and maximum residue limit (MRL) for veterinary drugs that have been approved a few decades ago in South Korea. On top of that, chronic dietary exposure assessment of veterinary drug residues should be performed to determine whether the use of these veterinary drugs would cause health concerns or not. METHODS AND RESULTS: To establish the ADI, the relevant toxicological data were collected from evaluation reports issued by international organizations. A slightly modified global estimate of chronic dietary exposure (GECDE) model was employed in the exposure assessment owing to the limited residual data. Therefore, only the ADI of ephedrine was established due to insufficient data for the other veterinary drugs. Thus, instead of ADI, the threshold of toxicological concern (TTC) value was used for the other drugs. Lastly, the hazard index (HI) was calculated, except for etizazole hydrochloride, due to the potential of mutagenicity. CONCLUSION(S): The HI values of ephedrine, menichlopholan, and anacolin were found to be as high as 6.4%, suggesting that chronic dietary exposure to the residues from these uses was unlikely to be a public health concern. Further research for exposure assessment of veterinary drug residues should be performed using up-todate Korean national health and nutrition examination survey (KNHANES) food consumption data. In addition, all relevant available data sources should be utilized for identifying the potentials of toxicity.

HPLC Chromatographic Methods for Simultaneous Determination of Pholcodine and Ephedrine HCI with Other Active Ingredients in Antitussive-Antihistamine Oral Liquid Formulations

  • A description of simple, isocratic and precise reversed phase HPLC methods is given for simultaneous quantification of pholcodine and ephedrine hydrochloride together with either carbinoxamine maleate or terfenadine in antitussive-antihistaminic oral pharmaceutical formulations. Separations were carried out on X-Terra and symmetry shield C18 column $(250\;{\times}\;4.6\;mm,\;5\;{\mu}m)$. The used isocratic elution systems were either $0.02\;M\;KH_2PO_4-acetonitrile$ in the ratio of 75 : 25 and pH adjusted to 7.70 with orthophosphoric acid or sodium hydroxide, for syrup (method A), or 0.02 octanesulphonic acid sodium salt solution-acetonitrile-acetic acid in the ratio of 75 : 25 : 0.5 for suspension (method B). The elution of both mixtures was achieved with a flow rate of 1 ml/min. Detection was carried out by UV absorbance at wavelengths of 220 and 250 nm for syrup and suspension, respectively. The quantification of the components in synthetic mixtures and actual syrup and suspension were calculated using the internal standard technique with metoclopramide HCl and codeine phosphate as internal standards (IS), respectively. The methods, for both mixtures, were validated and met all the requirements for the quality control analysis recommended by FDA and ICH.

  • DF formula is comprised of three traditional herbs, Ephedra intermedia, Rheum palmatum and Lithospermum erythrorhizon, and locally used for treating of the metabolic diseases, such as obesity and diabetes in Korea. We tried to optimize the extraction conditions of two major components, (-)-ephedrine and (+)-pseudoephedrine, in DF formula using response surface methodology with Box-Behnken design (BBD). The experimental conditions with 70% for EtOH concentrations, 4.8 hour for extraction hours and 8.7 times for the solvent to material ratio were suggested for the optimized extraction of DF formula with the highest amounts of (-)-ephedrine and (+)-pseudoephedrine in the designed model.

  • A new fluorescent derivatizing reagent was developed to be used in HPLC for the trace determination of primary and secondary amines. This new reagent, 1-(N,N-dimethylamino)pyrene-6-sulfonyl chloride, was synthesized by the chlorination of sodium 1-(N,N-dimethylamino)pyrene-6-sulfonate which was obtained from 1-(N,N-dimethylamino)pyrene after sulfonation. Ephedrine and norephedrine were derivatized quantitatively by this reagent. The optimum conditions for the derivatization such as pH, reagent concentration, reaction time and reaction temperature ware examined. The structures of derivatives were identified by IR, $^{1}$H-NMR and MS methods. The fluorescence properties and the stability of the derivatives were examined. The derivatives were separated on silica column with an isocratic elution using the mixture of n-hexane and ethylacetate and monitored by fluorescene detector. Linear calibration curves were obtained and detection limits in a 10 $\mu$l injection volume were 5 picomole for ephedrine and norephedrine.

  • Background : Mahuang (Ephedra sinica) has been used widely to treat respiratory disease in traditional medicine for over thousands years. Mahuang preparations contain approximately $0.75{\sim}1%$ ephedrine alkaloids. Recently in North America, the Eephedra alkaloids has aroused a controversy due to its adverse effects in those using whole extracts as "dietary supplements" for weight loss or athletic performance enhancement. Objective : To discuss the need for clinical practice guidelines on the use of Mahuang. Results and Conclusion : It is widely assumed that Mahuang is one of the useful medicines and is safe for human body. Most of scientific literatures support that the safety of Mahuang prescription in common dosage. However, it has been claimed that there were some warnings and apprehensions about the harmfulness of Mahuang as dietary supplements. This article does not negate the fact that it is necessary to pay more attention for use of Mahuang in clinical situations. We assert that the establishment of clinical practice guideline on the use of Mahuang, which is based on scientific materials and consensus, is required in traditional medicine.

Impurity Profiling Analysis of Illicit Methamphetamine Seized in Korea (우리나라에서 불법 유통되는 메스암페타민의 불순물 프로화일 분석)

  • Impurity profiling analysis of methamphetamine seized in Korea was investigated for the evidential and intelligent purpose. Samples were extracted with ethylacetate which contai ns internal standard of dioctylsebacate under basic condition and extracts were analyzed by GC-FID. Ephedrine, chloroephedrine & 1,2-dimethyl-3-phenylaziridine were identified impurities in illicit methamphetamine by GC-MS. These impurities revealed that most of abused methamphetamine in Korea were synthesized from ephedrine as a starting material. For the classification of samples. firstly, 24 impurity peaks were selected after inspection of every peak in 50 samples as the specific markers of impurities. Secondly, corresponding peak retention time and area ratio to the internal standard were calculated and database was created with values of 24 peaks by in-house program. Finally, cluster analysis was attempted with the resultant profiles using the STAR plot, which was based on the Euclidian distance for evaluating similarity among samples. A total of 76 samples were divided into 8 different groups within 90% statistical similarity and inter-batch samples showed similar impurity patterns by this procedure. In conclusion, the analysis of impurities is a suitable index for estimation the common or different origin of methamphetamine sample.

Neuroprotective Activity of So Cheong Ryong Tang with Steaming Process (증숙 과정을 통한 소청룡탕의 뇌신경세포 보호 활성 변화에 관한 연구)

  • So Cheong Ryong Tang (SCRT) has been used as a traditional herbal medicine for the treatment of the bronchial asthma. In this study, the effect of steaming process on changes in 11 major compounds (homogentisic acid, ephedrine HCl, paeoniflorin, cinnamic acid, cinnam aldehyde, glycyrrhizin, 6-gingerol, schizandrin, methyleugenol, gomisin A and gomisin N) content and neuroprotective activity of SCRT were evaluated. Major compound content was slightly different with steaming produce. The contents of paeoniflorin, cinnamic acid, cinnam aldehyde, 6-gingerol and methyleugenol were decreased and homogentisic acid, ephedrine HCl, glycyrrhizin, schizandrin and gomisin A were increased by steaming process. The neuroprotective activity of steaming SCRT was determined in mouse hippocampal HT22 cells by MTT assay. As a result, neuroprotective activity of a steaming SCRT was higher than that of a non steaming SCRT. This study demonstrated that neuroprotective activity of SCRT can be improved through steaming process.

  • The herbal formula, DF-02, consisting of Ephedra intermedia and Rheum palmatum are used for the treatment of the metabolic diseases such as obesity and liver fibrosis in Korean local clinics. We aimed to develop the simultaneous analytical conditions for four standards, (+)-pseudoephedrine (PSEP) and (-)-ephedrine (EP) for E. intermedia, and aloe-emodin (AE) and chrysophanol (CP) for R. palmatum using HPLC-UV techniques. The validated conditions yielded the high precision (relative standard deviation (RSD) < 3.65%) and the recoveries (94 - 106%) using the calibration curves with high linearity ($R^2$ > 0.9994). As a result, four standards of DF-02 were simultaneously determined under the developed method, which will be utilized for the quality control or evaluation of DF-02 and many herbal preparations containing E. intermedia and R. palmatum.
