Two Cases of Mycotic Pneumonia in Dogs (개에서 곰팡이성 폐렴 2례)

  • A two-year-old male Laika weighing 19 kg and a two-year-old female Rottweiler dog weighing 33 kg were presented to the Veterinary Medical Center, Chungbuk National University. Chest radiographs showed miliary interstitial lung pattern in the overall lung fields and the ill-defined mass in the perihilar area, which is consistent with lymphadenopathy in both cases. In the tracheal wash fluid smears in two cases, 90% of cells were eosinophils. Also, Aspergillus fumigatus was isolated from the tracheal wash fluid in Czapek's agar in one case. Ketoconazol (10 mg/kg, PO, ql2hr) alleviated the clinical signs in two dogs. Follow up radiographs showed marked reduction of lung lesions in one case.

  • Objective : The purpose of this research is to examine the effects of Gleditsiae Spina (GS) extract on immune cells and cytokines in ovalbumin (OVA)-induced asthmatic mice. Methods : In vivo, C57BL/6 mice were sensitized and handicapped by OVA for 12 weeks. During this experiment, the one group was then treated with GS extract for the later 8 weeks (3 times per week) and analyzed by ELISA, flow cytometer and RT-PCR. Results : In vivo, there were significant decreases in eosinophils, IL-4, IL-5, IL-13, IgE in BALF (bronchoalveolar lavage fluid). However, $IFN-{\gamma}$ in BALF of GS group increased significantly, compared with that of control group. Additionally, the population of $CD3e^-/CCR3^+,\;CD69^+/CD3e^+,\;IgE^+/B220^+,\;CD11b^+/Gr-l^+$ cells in the GS group decreased. Conclusion : The results of this study support a role for GS as an effective treatment for asthma in its experimental success in significantly decreasing inflammation and asthma reactions, and in increasing $IFN-{\gamma}$, which helps prevent such reactions.

  • Chronic eosinophilic pneumonia is a very rare disorder of unknown etiology characterized by striking systemic and pulmonary manifestations such as fever, weight loss, dyspnea, blood eosinophilia, and fluffy peripheral opacities on chest radiograph. A number of these patients developed asthma before or with the onset of illness. The roentgenographic lesion rapidly resoluted with corticosteroid and recurrence was occasionally occured in the same location. Histopathologic features of chronic eosinophilic pneumonia include dense aggregates of eosinophils, histiocytes, and multinucleated giant cells within alveolar spaces, interstitium, and bronchioles associated with scattered lymphocytes and plasma cells. We report a case of chronic eosinophilic pneumonia diagnosed by clinical, radiographic, and histologic findings with review of the literature.

Effects of Saposhnikoviae Radix on allergic responses in OVA-induced Allergic rhinitis mice (방풍(防風)의 난알부민 유도 알레르기 비염 마우스에서의 항알레르기 효능 연구)

  • Objectives : In this study, we investigated the effect of Saposhnikoviae Radix on allergic responses in ovalbumin(OVA)-induced Allergic rhinitis(AR) mice. Methods : BALB/c mice were orally administrated with Saposhnikoviae Radix water extract (SRW, 50 mg/kg) or anti-histamine drug, Ketotifen (10 mg/kg) as a reference drug, followed by sensitization and challenge of OVA. Mice were measured clinical symptoms and the serum levels of histamine, IgE, IL-4, and IFN-${\gamma}$, and observed histopathological changes of nasal mucosa H&E staining. Results : SRW significantly decreased rubs and the serum levels of histamine, IgE, and IL-4, and then increased the serum levels of IFN-${\gamma}$ in OVA-induced AR mice, and inhibited histopathological changes of nasal mucosa with inflammation and the eosinophils infilteration. Conclusions : These data suggest that SRW has anti-allergic effect through the inhibitory property of Saposhnikoviae Radix against allergic responses in allergic rhinitis.

Effects of NR on Production Levels of Cytokines and Distribution of BAL Cells in Allergic Asthma (강활추출물이 알레르기 천식 모델 생쥐에 기관지폐포세척액의 면역세포에 미치는 영향)

  • Objectives : This study was designed to investigate therapeutic effects fo NR on allergic asthma in terms of measurement of cytokine profiles, distribution of BAL and expression levels of asthma induced by OVA. Methods : C57BL/6 mouse was divided into normal, NRI, control, and NRII, and Notopterygii Rhizoma Extract was orally administered to the C57BL/6 mice in the normal group and the normal mouse in NRI group while it was orally administered to the control group after inducing asthma by OVA and NRII. The study measured them by means of ELISA after dividing BALF of each group, and analyzed RT-PCR by using lung tissues. Results : In our results, lung weight and the number of total cells in the lung and of Leukocyte and Eosinophils in BALF were significantly decreased. In addition, IL-4, IL-5, IL-13, and IgE in BALF were significantly decreased compared to the control group. Conclusions : These results demonstrate that NR Extract be a desirable altemative therapy for allergic asthma by inhibiting the expressin of immune cells, the activation of inflammatory mediator.

A case of eosinophilic cystitis in a dog (개에서 호산구성 방광염 1례)

  • Eosinophilic cystitis (EC) is a rare inflammatory disorder in dogs. Although EC has been associated with various etiological factors, the precise cause of this disease remains unclear in human and animals. A 7-year-old female Yorkshire terrier was presented with 3-week-history of hematuria, dysuria, and suprapubic pain. A bladder mass in ventral surface and urinary calculi were noted on ultrasonography and gross findings. Biopsy sample was taken from the protruded mass of bladder for histopathologic examination. Hyperplastic transitional epithelium with focal ulceration covered the bladder mass. A large nodule of fibrous tissue that contained fibrocytes, fibroblasts, Iymphocytes, plasma cells, macrophages, numerous eosinophils, and abundant blood vessels was occupied in lamina propria and submucosa of bladder mass. Based on the clinical, gross, and histopathologic examinations, this case was diagnosed as EC in a dog. In our knowledge, this is the first report of an eosinophilic cystitis in dog in Korea.

Anti-inflammatory Effects and its Mechanisms of Hesperidin in an Asthmatic Mouse Model Induced by Ovalbumin

  • Hesperidin, a member of the flavanone group of flavonoids, can be isolated in large amounts from the rinds of some citrus species [e.g., Citrus aurantium L. (bitter orange), Citrus sinensis L. (sweet orange) and Citrus unshiu Marcov. (satsuma mandarin)], and has been reported to have anticarcinogenic, antihypotensive and antimicrobial properties. Despite the efficacy of these polyphenolic compounds as immune modulators, the effects of the flavonoids are poorly understood about allergic effect. In this study, we investigated whether hesperidin could influence on Th1 and Th2 balance. Allergic reactions included an increase in the number of eosinophils in bronchoalveolar lavage (BAL) fluid, an increase in inflammatory cell infiltration into the lung tissue around blood vessels and airways, airway luminal narrowing, the development of airway hyper-responsiveness (AHR). The administration of hesperidin before the last airway OVA challenge resulted in a significant inhibition of all asthmatic reactions. Accordingly, this study may provide evidence that hesperidin plays a critical role in the amelioration of the pathogenetic process of asthma in mice. These findings provide new insight into the immunopharmacological role of hesperidin in terms of its effects in a murine model of asthma, and also broaden current perspectives in our understanding of the immunopharmacological functions of hesperidin.

Crosstalk between Adipocytes and Immune Cells in Adipose Tissue Inflammation and Metabolic Dysregulation in Obesity

  • Recent findings, notably on adipokines and adipose tissue inflammation, have revised the concept of adipose tissues being a mere storage depot for body energy. Instead, adipose tissues are emerging as endocrine and immunologically active organs with multiple effects on the regulation of systemic energy homeostasis. Notably, compared with other metabolic organs such as liver and muscle, various inflammatory responses are dynamically regulated in adipose tissues and most of the immune cells in adipose tissues are involved in obesity-mediated metabolic complications, including insulin resistance. Here, we summarize recent findings on the key roles of innate (neutrophils, macrophages, mast cells, eosinophils) and adaptive (regulatory T cells, type 1 helper T cells, CD8 T cells, B cells) immune cells in adipose tissue inflammation and metabolic dysregulation in obesity. In particular, the roles of natural killer T cells, one type of innate lymphocyte, in adipose tissue inflammation will be discussed. Finally, a new role of adipocytes as antigen presenting cells to modulate T cell activity and subsequent adipose tissue inflammation will be proposed.

Studies on the Coliform Mastitis I. Histopathological Observation on Experimentally Induced Escherichia coli Mastitis in Rabbits (Coliform 유방염(乳房炎)에 관한 연구(硏究) 1. Escherichia coli에 의(依)한 유방염(乳房炎)의 실험병리학적(實驗病理學的) 관찰(觀察))

  • In order to clarify the morphological changes in the mammary glands of cows affected with coliform (Escherichia coli) mastitis, histopathological observations were undertaken on the mastitis of the lactating rabbits which was experimentally induced with E. coli or its endotoxin isolated from cases of acute and chronic matitis in dairy cattle. In the bacterial suspension-infused groups the affected quarters of udder showed cloudy swelling, hyperemia and hemorrhage to local necrosis and firmness. The microscopic findings of early stage of the mastitis were appearance of large numbers of heterophils in the glandular lumina and ducts accompanied by degeneration, necrosis and desquamation of epithelial cells, and also infiltration of heterophils, hemorrhage and edema in the interstitial tissue, and destruction of alveoli. Later, proliferation of firoblasts, plasma cells, lymphocytes, eosinophils and histiocytes appeared in the glandular tissue and necrotic foci of glandular tissue were surrounded by highly proliferated connective tissue. Granuloma-like inflammatory changes could be observed in the glandular tissue on the 7th days after infusion. The inflammatory response in the group infused with E. coli strain isolated from the natural case of acute mastitis was rapid and severe as compared with that of chronic mastitis. In the endotoxin-infused group the morphological changes were similiar to those of the bacterial suspension-infused groups.
