분쇄마찰매체 효소반응계에서 생전분 효소당화를 위한 Glucoamylase와 Alpha-Amylase의 보완작용 (Synergistic Effect of Glucoamylase and $\alpha$-Amylase in Enzymatic Hydrolysis of Raw Corn Starch in an Agitated Bead Reaction System)

  • 분쇄마찰매체효소반응계에서 glucoamylase와 Alpha-amylase의 보완작용에 의한 옥수수 생전분의 효소당화 mechanism을 규명코자, 분리.정제된 상기 효소의 혼합 비율에 따른 당화양상 및 생성당 조성, 전분입자 구조, 생전분입자의 particle size 분포, X-ray 회절양샹의 변화 등을 관찰하였다. 분쇄마찰매체 효소반응계에서도 효소를 분리하여 사용하였을 때보다 glucoamylase와 $\alpha$-amylase를 혼합사용할 경우 당화율이 크게 향상되었으며, 고순도의 glucose 생산에 적합한 두 효소의 혼합비율은 약 1:1-1:2(unit) 로 판명되었다.

  • 1회의 회분실험 데이터로부터 효소반응 속도식의 매개 변수를 쉽게 추정할 수 있는 방법을 제시하였다. 여러 가지 반응패턴을 갖는 모델식에 있어서 효소반응 데이터를 적용시킨 결과 정확하고 간편하게 매개변수값을 추정할 수 있었다. 가역반응의 경우에는 3개의 매개변수를 갖는 모델식의 형태로 평형 기질농도 및 매개변수를 추정할 수 있었다. 또한 반응특성이 잘 알려져 있지 않은 효소 반응 시스템에 있어서는 효소반응이 기질이나 생산물의 저해작용을 받는지 여부와 그 저해패턴을 확인할 수 있었다.

Effect of [EMIM]Ac Recycling on Salix gracilistyla Miq. Pretreatment for Enzymatic Saccharification

  • Recycling of ionic liquid (1-ethyl-3-methylimidazolium acetate, [EMIM]Ac) after the pretreatment of Salix gracilistyla Miq. was conducted and the effect of the recycling number on the enzymatic saccharification yield was investigated. Enzymatic saccharification was performed using an enzyme cocktail (Acremonium cellulase and Optimash BG) at 50 ℃ for 72 h. All recycled [EMIM]Ac samples showed a lower amount of water soluble fraction than pure [EMIM]Ac. On increasing the recycling number from 1 to 4, the amount of water soluble fraction decreased from 18% to 15%. The X-ray diffraction pattern of the products pretreated with recycled [EMIM]Ac showed cellulose I crystalline polymorph. The crystallinity of the product pretreated with recycled [EMIM]Ac was 47-49%, which was lower than 33% of that with pure [EMIM]Ac. The yields of glucose and xylose decreased in the pretreatment with recycled [EMIM]Ac compared to that with pure [EMIM]Ac.

각종 섬유질 효소당화 반응조내의 현탁액의 혼합교반양상이 효소당화에 미치는 영향 (Effect of Mixing Pattern of Different Types of Bioreactor on Enzymatic Hydrolysis of Cellulose)

  • 각종 섬유질 효소당화반응조내의 현탁액 혼합교반 양상이 효소당화에 미치는 영향을 검토하기 위하여 3종의 대표적인 bioreactor;vertical impeller type bioreactor-(VITB), horizonatl paddle type bioreactor(TDTB)를 제작하여 그 효용성을 비교 검토하였다. 최적 교반강도는 impel-ler나 paddle로 현탁핵을 교반하는 VITB와 HPTB가 각각 100rpm, 그리고 통자체를 회전시켜 교반하는 TDTB가 200rpm이었으며 TDTB에 부착된 baffle의 형태 및 크기는 큰 영향을 주지 않았다. 또한 VITB나 TDTB의 경우 과다한 교반은 효소의 실활을 유발시켜 오히려 섬유소당화를 저해하였다. 3종의 bioreactor의 최적 교반조건에서의 당화효율을 비교한 결과 VITB가 가장 높았으며 특히 고농도 섬유소 당화시 가장 높은 당화효율을 보여, 가장 효율적인 교반양상을 나타내는 것으로 보인다. 이로 미루어 impeller나 paddle의 물리적 충격에 의한 현탁액의 cellulose network를 파괴시키는 효소반응기가 단순히 교반혼합하는 반응기에 비하여 우수한 결과를 줌을 알 수 있었다. 반면 주어진 양의 당을 생산하는데 소요되는 동력소모를 검토한 결과 TDTB가 VITB에 비하여 다소 낮았으나 고농도의 경우는 급속히 증가되었다. 적절한 bioreactor의 선정에는 기질의 가격, 당화수율, 고농도 당화, 그리고 반응기의 scale-up의 용이성등이 종합적으로 검토되어져야 할 것이다.

Comparison of Three Substrates (Casein, Fibrin, and Gelatin) in Zymographic Gel

  • Three zymographic techniques using casein, fibrin, and gelatin as substrates in SDS-PAGE were compared based on three aspects: (1) The proteolytic pattern of extracellular enzymes from the three bacterial strains, Bacillus sp. DJ-1, DJ-2, and DJ-3. (2) The enzymatic sensitivity of their activity on zymogram gels. (3) The stability of stained zymogram gels with Coomassie brilliant blue in the destaining solution. There was no significant difference on the pattern of extracellular enzymes from the three strains. The bands in the fibrin gel were clearer and more distinct from the extensive destaining process. It was also shown that the gelatin gel revealed the highest enzymatic sensitivity among the three gels, based on the densitometric analysis. In the casein gel, a trace that could be mistaken as a proteolytic band appeared around 40-50 kDa.

Comparison of the Cell Surface Barrier and Enzymatic Modification System in Brevibacterium flavum and B. Lactofermentum

  • To investigate impediments to plasmid transformation in Brevibacterium flavum BF4 and B. lactofermentum BL1, cell surface barriers were determined by measuring growth inhibition whilst enzymatic barriers were determined by comparing DNA methylation properties. B. lactofermentum was more sensitive to growth inhibition by glycine than B. flavum. Release of cellular proteins during sonication was more rapid for B. lactofermentum than for B. flavum. Plasmid DNA (pCSL 17) isolated from B. flavum transformed recipient $McrBC^+$ strains of Escherichia coli with lower efficiency than $McrBC^-$. McrBC digestion of this DNA confirmed that B. flavum contain methylated cytidines in the target sequence of McrBc sequences but B. lactofermentum contained a different methylation pattern. DNA derived from the B. lactofermentum transformed recipient $EcoKR^+$ strains of E. coli with lower efficiency than $EcoKR^-$, indicating the presence of methylated adenosines in the target sequence of EcoK sequences. The present data describe the differences in the physical and enzymatic barriers between two species of corynebacteria and also provide some insight into the successful foreign gene expression in corynebacteria.

  • 장내 세균의 생리적 연구를 목적으로 한국인의 장내 상재균을 분리하여 효소 pattern을 관찰하였다. 분리된 Bifidobacterium sp. Int-57은 다른 장내 균종에 비하여 $\alpha$-glucosidase, $\beta$-glucosidase, $\alpha$-galactosidase, $\beta$-galactosidase, $\beta$-xylosidase, $\alpha$-arabinofuranosidase역가가 높았다. Bifidobacterium sp. Int-57의 각 효소 생산에 미치는 탄소원의 영향을 조사하였다. $\alpha$-glucosidase는 maltose, $\beta$-glucosidase는 cellobiose, $\alpha$-galactosidase는 raffinose, 는 lactose, $\beta$-xylosidase와 $\alpha$-arabinofuranosidase는 xyloserk 각각 최적의 탄소원이었다. 또한 각 효소들의 최적 조건과 pH 안정성을 조사 하였다. $\alpha$-glucosidase는 pH 6.0 $40^{\circ}C$에서 $\beta$-glucosidasessm pH 7.0 50에서, $\beta$-galactosidase는 pH 7.0 50에서, $\beta$-xylosidase는 pH 6.0 $40^{\circ}C$에서, $\alpha$-arabinofurnaosidase는 pH 5.0 $50^{\circ}C$에서 각각 최적이었다. $\alpha$-glucosidase는 pH 4.0~9.0 $\beta$-glucosidase는 pH 4.0~7.0 $\beta$-galactosidase는 pH 4.0~9.0, $\beta$-xylosidase는 pH 4.0~6.0, $\alpha$-arabinofuranosidase는 pH 7.0~9.0에서 각각 안정하였다.

  • Antioxidative and cytoprotective effects of tectorigenin and glycitein isolated from the pueraria thunbergiana and its derivative, genistein, were determined. Among these three compounds, tectorigenin and glycitein bearing 6-methoxyl groups in both isoflavones showed significant free radical scavenging activities on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and xanthine/xanthine oxidase (XOD) generating superoxide anion radical. Tectorigenin only showed a slight inhibitory effect on XOD. We further studied the inhibitory effects of these isoflavones on the lipid peroxidation in rat liver microsomes induced by enzymatic and non-enzymatic methods. Each of them exhibited inhibitory effect on both ascorbic $acid/Fe^{2+}-{\;}and{\;}ADP/NADPH/Fe^{+3}-induced$ lipid peroxidation. Moreover, tectorigenin exhibited the highest protection of hydrogen peroxide damage on HepG2 and Vero cells among the three isoflavones, in the cytoprotective assay. It was suggested that the pattern of antioxidative and cytoprotective effect of isoflavones could be crucially by the aromatic substitution of oxygen-containing groups.

  • Today's computers are built up from a minimal set of standard pattern recognition operations. Logic gates, such as NAND, are common examples. Biomolecular materials offer an alternative approach, both in terms of variety and context sensitivity. Enzymes, the basic switching elements in biological cells, are notable for their ability to discriminate specific molecules in a complex background and to do so in a manner that is sensitive to particular milieu features and indifferent to others, The enzyme, in effect, is a powerful context sensitivity pattern processor that in a rough way can be analogized to a neuron whose input-output behavior is controlled by enzymatic dynamics.

녹화중 보리유식물에서 Phosphorylcholine대사의 변화 (Changes of Phosphorylcholine Metabolism in Barley Seedlings during Greening)

  • We investigated the activities of choline kinase, CTP: phosphorylcholine cytidyltransferase, and phosphatase during the greening of etiolated barley seedlings. Activities of choline kinase in leaves increased until 6 hours after illumination and decreased considerably after 6 hours, while activities of CTP: phosphorylcholine cytidyltransferase increased after illumination. On the contrary, changes of these two enzymatic activities showed reverse pattern in roots. The activities of phosphatase which hydrolyze phosphorylcholine decreased in leaves but changed little in roots during greening. The concentration of phosphorylcholine increased in xylem exudate and in roots during greening, while decreased in leaves. These results suggested that more phosphorylcholine arrive in leaves from roots as greening of etiolated barley seedlings.
