Multi-step Reactions on Microchip Platform Using Nitrocellulose Membrane Reactor

  • A straightforward and effective method is presented for immobilizing enzymes on a microchip platform without chemically modifying a micro-channel or technically microfabricating a column reactor and fluid channel network. The proposed method consists of three steps: the reconstitution of a nitrocellulose (NC) membrane on a plane substrate without a channel network, enzyme immobilization on the NC membrane, and the assembly of another substrate with a fabricated channel network. As a result, enzymes can be stably and efficiently immobilized on a microchip. To evaluate the proposed method, two kinds of enzymatic reaction are applied: a sequential two-step reaction by one enzyme, alkaline phosphatase, and a coupled reaction by two enzymes, glucose oxidase and peroxidase, for a glucose assay.

Angiotensin- I Converting Enzyme Inhibitory Properties of Bovine Casein Hydrolysates in Different Enzymatic hydrolysis Conditions (효소가수분해 조건에 따른 우유 케이신의 Angiotensin-I 전환효소 저해효과)

  • Angiotensiri-I converting enzyme(ACE) catalyst the removal of the C-terminal dipeptide from the angiotensin-I to give the angiotensin-II, a potent peptide that causes constriction of regulation of blood pressure. Recently, ACE inhibitor peptides have been isolated from enzymatic digests of food protein. The aim of this study was to identify bovine casein hydrolysates with ACE inhibitory properties in different enzymatic hydrolysis conditions. The casein were hydrolyzed neutrase, alcalase, protamax, flavourzyme, premed 192, sumizyme MP, sumizyme LP and pescalase alone and with an enzyme combination. Premed 192 produced ACE inhibitory peptides most efficiently. In order to ACE inhibitory peptide produced enzymatic hydrolysis condition were premed 192 added to casein ratio of 1:100(w/w), and incubated at 47$\^{C}$ for 12hrs. Casein hydrolysate gave 50% inhibition(IC$\_$50/ value) of ACE activity at concentration with 248ug/ml(general method) and 265ug/ml(pretreatment method) respectively.

  • Choline is important an organic compound for normal membrane function, acetylcholine synthesis, lipid transport, and methyl metabolism. In biological tissues and foods, there are multiple choline compounds that contribute to choline content. There are so many analytical methods for choline determination, such as radioisotopic, high-performance liquid chromatography, and gas chromatography/mass spectrometry. However, these existing methods are expensive, unmanageable, and time-consuming. In this study, we modified enzymatic method, which is applicable for the determination of choline in milk and infant formulas, and applied to bovine serum and muscle. The calibration curves were linear with higher correlation coefficients than 0.994. Recoveries obtained by calibration curves from the spiked bovine serum and muscle samples varied between 70.6 and 85.2%. The method may be suitable for use as a routine method in the determination of choline for biological tissue and food samples.

Study on the Pretreatment of Rice Hull to Enhance Enzymatic Saccharification Efficiency (효소 당화효율 증진을 위한 왕겨의 전처리 방법 연구)

  • The objective of this study was to investigate the efficient pretreatment method for bioethanol production from rice hull. Ammonia and sodium hydroxide as an alkaline solution and dilute sulfuric acid as an acidic solution were used in a batch reactor under high-temperature and high-pressure conditions. The highest enzymatic saccharification efficiency of 82.8% and ash removal rate of 94.7% were obtained in the dilute sulfuric acid treated sample after the sodium hydroxide solution treatment. The enzymatic saccharification efficiencies and ash removals of pretreated rice hull samples have very similar variation tendency. This means that the maximum obstructive factor for the enzymatic saccharification of rice hull is the ash (silicate) content in biomass. The findings suggest that the combined sodium hydroxide-dilute sulfuric acid treatment system under high-temperature and high-pressure conditions is a promising pretreatment method to enhance the enzymatic saccharification of the silica-rich biomass.

A Study of Enzymatic and Water Degumming Using Crude Canola Oil (Crude Canola Oil의 효소 탈검과 수용성 탈검에 관한 연구)

  • In this study, degumming process was carried out for reducing to less than 10 ppm of phosphorus contents and primary properties of crude canola oil including 0.64 mgKOH/g of acid value, 0.09% of water contents, 0.13% of insoluble impurities, and 40 ppm of phosphorus contents. Efficiency of water degumming and enzymatic degumming was compared for the selection of suitable process obtaining feedstock of biodiesel. Degumming method was determined for preparation of raw material of biodiesel, and reaction conditions were also established. The most effective conditions for water degumming were 2% distilled water (w/w oil), $30^{\circ}C$ of reaction temperature, 900 rpm of agitation speed, and 30 min of reaction time, respectively. In case of enzymatic degumming, optimal conditions were found to be 90 ppm of phospholipase A2 (w/w oil), $50^{\circ}C$ of reaction temperature at pH 5, respectively. When comparing water degumming with enzymatic degumming, efficiency of enzymatic degumming was better than water degumming. However, water degumming method was much more suitable for the production of biodiesel feedstock considering reaction time and process feasibility.

Enzymatic Isolation of Husk from Barley Kernel (효소적 방법에 의한 보리피층의 분리)

  • The amounts of husk materials from barley kernel were determined by an enzymatic method and compared with the values determined by conventional methods involving acid or alkaline treatments. The enzymatic method consists of boiling in distilled water and pressing to help squeeze out the gelatinized starch from the husk matrix, and enzymatic removal of starch by ${\alpha}-amylase$ and weighing the residual husk materials after washing 3 times with hot water and then drying at $95^{\circ}C$. Husk materials amounted about 15 of the covered barley (Gangbori and Olbori) and 10-12% of naked variety (Backdong and Sedohadaga) and the values were always somewhat higher than those obtained by the conventional methods. The husk materials prepared by the enzymatic procedure contained protein 4-8%, lipid 5-10%, ash 0.2-0.6% and crude fiber 20-40%. Although it took longer time, the enzymatic procedures can provide more intack husk materials for further characterization of the materials.

Antioxidant and angiotensin I-converting enzyme inhibitory activities of northern shrimp (Pandalus borealis) by-products hydrolysate by enzymatic hydrolysis

  • In the present study, we investigated to the antioxidant and angiotensin I-converting enzyme (ACE) inhibitory activities of the northern shrimp (Pandalus borealis) by-products (PBB) hydrolysates prepared by enzymatic hydrolysis. The antioxidant and ACE inhibitory activities of five enzymatic hydrolysates (alcalase, protamex, flavourzyme, papain, and trypsin) of PBB were evaluated by the 2, 2'-azino-bis [3-ethylbenzothiazoline-6-sulfonic acid] ($ABTS^+$) radical scavenging and superoxide dismutase (SOD)-like activities, reducing power and Li's method for ACE inhibitory activity. Of these PBB hydrolysates, the protamex hydrolysate exhibited the most potent ACE inhibitory activity with $IC_{50}$ value of $0.08{\pm}0.00mg/mL$. The PBB protamex hydrolysate was fractionated by two ultrafiltration membranes with 3 and 10 kDa (below 3 kDa, between 3 and 10 kDa, and above 10 kDa). These three fractions were evaluated for the total amino acids composition, antioxidant, and ACE inhibitory activities. Among these fractions, the < 3 kDa and 3-10 kDa fractions showed more potent $ABTS^+$ radical scavenging activity than that of > 10 kDa fraction, while the > 10 kDa fraction exhibited the significant reducing power than others. In addition, 3-10 kDa and > 10 kDa fractions showed the significant ACE inhibitory activity. These results suggested that the high molecular weight enzymatic hydrolysate derived from PBB could be used for control oxidative stress and prevent hypertension.

Comparison of Differential Scanning Calorimetry with Enzymatic Method for the Determination of Gelatinization Degree of Corn Starch (DSC에 의한 전분의 Endothermic peak와 효소분석법에 의한 호화도 비교)

  • Gelatinization degrees of torn and waxy corn starches in the low-moisture environment were determined by DSC thermogram and enzymatic analysis, the results were compared each other As the moisture content increased from 20% to 70%, the enthalpy of endothermic peak of starch increased linearly in DSC thermograms. When the moisture content exceeded above 70%, the DSC enthalpy of starch remained constant in DSC thermogram. The enthalpies for gelatinization of corn and waxy corn starches were 3.23 cal/g and 4.2 cal/g, respectively. When gelatinization degrees of starches were measured by enzymatic analysis, the gelatinization degree increased linearly as the moisture content increased from 20% to 80%. A linear correlation between DSC and enzymatic analysis was obtained only when the moisture content was under 70%.

Development of Combined Method for Extraction of Sea Tangle (다시마의 효과적 추출을 위한 종합적인 추출방법의 개발)

  • Development for extraction method of sea tangle was investigated with addition of sucrose, NaCl, EDTA-2Na, SHMP, and hydrolysis with commercial polysaccharide hydrolyzing enzymes. The total solids and protein yields were significantly increased by enzymatic hydrolysis or addition of NaCl and EDTA-2Na during boiling. The extract prepared by enzymatic hydrolysis followed by 2 hrs of boiling in 1.5% NaCl solution was significantly improved in solids and protein yield from 24.1% and 13.9% of 2 hrs of water boiling to 44.6% and 32.2%, respectively. The combined method increased the amino-nitrogen content but reduced the turbidity and viscosity. Extracts with a high intensity of total and seaweed-like taste as revealed by sensory evalution were obtained by combined method.

A Technique for Increasing Cellulose Hydrolysis (섬유소의 가수분해에 대한 기술 개발 모색(I))

  • A novel pretreatment of rice straw has been developed to increase the reactivity of cellulose, in particular to increase the rate and extent of cellulose enzymatic hydrolysis. This technique is called ammonia-freeze-explosion method and relies on treatment of the lignocellulosic material with a volatile liquid under pressure followed by pressure release to evaporate the liquid and reduce the temperature by Bme E. Dale of Texas A & M University. Volatile liquids which also chemically explosion and swell lignocellulosic materials are particularly effective when used in this technique. Above four times of conversion of cellulose to glucose has been achived by enzymatic hydrolysis of rice straw with this method.
