Anti-oxidative Activities of 3'-Hydroxygenkwanin from the Flower Buds of Daphne genkwa in Caenorhabditis elegans

  • In the course of screening for antioxidant compounds from natural plants in Korea by measuring the radical scavenging effect, a methanol extract of the flower buds of Daphne genkwa S. et Z. (Thymelaeaceae) was found to show a potent antioxidant activity. Subsequent activity-guided fractionation of methanol extract of D. genkwa led to the isolation of four compounds from the ethyl acetate soluble fraction. The chemical structures were elucidated as genkwanin (1), 3'-hydroxygenkwanin (2), apigenin (3), and tiliroside (4) by spectroscopic techniques. Among them, compound 2 showed the significant anti-oxidative effect on DPPH. And compound 2 showed the significant riboflavin-and xanthine-originated superoxide quenching activities. To verify the antioxidant enzymatic activities of compound 2, the SOD enzymatic activity was measured spectrophtometrically using prepared Caenorhabditis elegans homogenates. The results showed that compound 2 was able to elevate SOD activity of C. elegans in a dose dependent manner. Moreover, compound 2 decreased the intracellular ROS accumulation of worms.

On-line Conversion Estimation for Solvent-free Enzymatic Esterification System with Water Activity Control

  • On-line conversion estimation of enzymatic esterification reactions in solvent-free media was investigated. In principle, conversion to ester can be determined from the amount of water produced by the reaction, because water is formed as a by-product in a stoichiometric manner. In this study, we estimated the water production rate only from some measurements of relative humidity and water balances without using any analytical methods. In order to test the performance of the on-line conversion estimation, the lipase-catalyzed esterification of n-capric acid and n-decal alcohol in solvent-free media was performed whilst controlling water activity at various values. The reaction conversions estimated on-line were similar to those determined by offline gas chromatographic analysis. However, when the water activity was controlled at higher values, discrepancies between the estimated conversion values and the measured values became significant. The deviation was found to be due to the inaccurate measurement of the water content in the reaction medium during the initial stages of the reaction. Using a digital filter, we were able to improve the accuracy of the on-line conversion estimation method considerably. Despite the simplicity of this method, the on-line estimated conversions were in good agreement with the off-line measured values.

In vitro Anti-obesity Effect of 4-hydroxybenzyl Alcohol from Cudrania tricuspidata

  • The present study was investigated on in vitro anti-obesity effect of 4-hydroxybenzyl alcohol from Cudrania tricuspidata. We isolated various compounds from Cudrania tricuspidata. Among these compounds, anti-obesity effects of 4-hydroxybenzyl alcohol was examined by lipase activity assay, cyclic adenosine monophosphate (cAMP)-specific phosphodiesterase type IV (PDE4) activity assay, and citrate synthase activity assay. 4-hydroxybenzyl alcohol and Cudrania tricuspidata extracts inhibited the enzymatic activities of lipase, PDE4, and citrate synthase. Lipase is known to mediate the hydrolysis of triacylglycerol in adipose tissue and cholesterol esters in other tissue or cells. Also, PDE4 hydrolyses cAMP, a crucial secondary messenger for in metabolic pathways including glucose and lipid metabolism, lipolysis, and thermogenic function. 4-hydroxybenzyl alcohol and Cudrania tricuspidata extracts induced the inhibitory effect against each enzymatic activity on several specific substrates as observed by detection at 405 or 412 nm. These findings might be attributable to the inhibition of adipogenesis, and partial prevention of obesity. In conclusion, these results show that 4-hydroxybenzyl alcohol and Cudrania tricuspidata may be a critical candidate as a natural anti-obesity source.

해조류의 Angiotensin-I 전환효소 저해작용 (Angiotensin-I Converting Enzyme Inhibitory Activity of Algae)

  • 해조자원의 기능 특성을 밝혀 그 이용도를 증진시키기 위한 연구의 일환으로 일반 해조류의 단백질 함량 및 물추출물과 해조 가수분해물의 ACE 저해효과를 조사한 결과는 다음과 같다. 1 단백질 함량은 김이 $39.6\%$로 가장 높았으며, 파래 $22.1\%$, 미역 $21.1\%$, 우뭇가사리 $18.3\%$, 청각 $15.7\%$, 톳 $12.4\%$, 다시마 $8.2\%$의 순으로 나타났다. 2. 해조 물추출물의 ACE 저해효과는 $50^{\circ}C,\;70^{\circ}C,\;98^{\circ}C$의 세 조건에서 각각 추출한 결과 $70^{\circ}C$에서 추출시 시료 모두에서 가장 높게 나타났으며, ACE 저해율이 가장 높은 $70^{\circ}C$를 기준으로 비교했을 때 우뭇가사리 $10.9\%$, 김 $9.3\%$, 파래 $8.9\%$, 미역 $8.2\%$, 톳 $7.5\%$, 다시마 $7.1\%$, 청각 $7.0\%$로 나타났다. 3. maxazyme과 papain에 의한 해조 가수분해물의 ACE 저해효과는 김이 파래, 미역, 우뭇가사리에 비해 월등히 높았고 pep-tide-nitrogen함량 역시 매우 높았다. 김의 경우 가수분해 8시간에서 ACE 저해효과가 maxazyme $31.3\%$, papain $27.9\%$로 가장 높았고 peptide-nitrogen함량도 이 시간에서 가장 높게 나타났다. 파래의 ACE 저해효과 역시 8시간에서 maxazyme $9.6\%$, papain $8.5\%$로 다른 시간대에 비해 대체로 높았으며 peptide-nitrogen함량은 16시간까지 완만히 증가하였다.

Hydrogen Peroxide produced by Two Amino Acid Oxidases Mediates Antibacterial Actions

  • The antibacterial actions of two amino acid oxidases, a D-amino acid oxidase from hog kidney and a L-amino acid oxidase from the venom of Agkistrodon halys, were investigated, demonstrating that both enzymes were able to inhibit the growth of both Gram-positive and Gram-negative bacteria, and that hydrogen peroxide, a product of their enzymatic reactions, was the antibacterial factor. However, hydrogen peroxide generated in the enzymatic reactions was not sufficient to explain the degree to which bacterial growth was inhibited. A fluorescence labeling assay showed that both of these two enzymes could bind to the surfaces of bacteria. To the best of our knowledge, this is the first report regarding the antibacterial activity of the D-amino acid oxidases.

Functionalization of Isoflavones with Enzymes

  • Considerable progress has been made in functionalization of the soy isoflavones through enzymatic modification of daidzin, genistin, and glycitin. After hydrolysis of $\beta$-glucosides into their corresponding aglycones, these compounds were structurally modified via biotransformations such as regioselective hydroxylation, enantioselective reduction, regioselective methylation, and polymerization. These reactions often resulted in an increase of the biological activities (e.g., anti oxidative activity, antiproliferative activity) and/or improvement of the physico-chemcial properties (e.g., water solubility, bioavailability). This review briefly summarizes on-going research activities on the biofunctionalization of the soy isoflavones.

Cloning and Characterization of a Bifunctional Cellulase-Chitosanase Gene from Bacillus lichenformis NBL420

  • A 1,3 kb cellulase gene encoding novel bifunctional cellulase-chitosanase activity was cloned from biopolymer-producing alkali-tolerant B. lichenformis NBL420 in E. coli. A recombinant cellulase-chitosanase, named CelA, was expressed and purified to homogeneity. The activity staining and the enzymatic characterization of the purified CeIA revealed bifunctional activities on carboxymethyl cellulose (CMC) and glycol-chitosan. The similar characteristics of the enzymatic activities at the optimum pH, optimum temperature, and thermostability Indicated that CelA used a common catalytic domain with relaxed substrate specificity. A comparison of the deduced amino acids in the N-terminal region revealed that the mature CelA had a high homology with the previously identified bifunctional cellulase-chitosanase of Myxobacter sp. AL- 1.

Cyclodextrin Glucanotransferase 고생산 호알칼리성 세균의 탐색과 분비 효소의 특성 (Screening of Alkalophilic Bacillus sp. for Overproduction of Cyclodextrin Glucanotransferase and Its Enzymatic Properties)

  • An alkalophilic microorganism for overproduction of cyclodextrin glucanotransferase (CGTase) was newly isolated from hot-water spring soil, and identified as Bacillus firmus var. alkalophilus H609. The strain maintained stability during preservation and cultivation for the enzyme production, and produced significant amount of CGTase corresponding to the volumetric activity of 75 units/mL at 37C, initial pH of 11.2, and after 40 hours. The strain excreted several different proteins showing CGTase activity that catalyzed the formation of mainly beta-and Gamma-type cyclodextrin (ratio of 7:1) from soluble starch without accumulation of alpha-type. Other enzymatic properties were also investigated.

고초균에서 폴리페놀로 유도된 DNA 손상에 대한 폴리페놀산화효소의 억제효과 (The Inhibitory Effect of Polyphenol Oxidase on Polyphenol-Induced DNA Damage of Bacillus subtilis)

  • Antimutagenic activity of the enzymatic browning reaction products (EBRPs) was investigated by using the spore rec-assay with Bacillus subtilis strains H17 $(rec^+)\;and\;M45 (rec^-)$. The EBRPs tested were prepared from the reactions of five different kinds of polyphenols with polyphenol oxidase isolated from the leaves Perilla frutescens. In the spore rec-assay, most of the polyphenolic compounds tested showed positive, whereas only their tested compound showed negative respectively. In addition of polyphenol oxidase inhibitors such as cysteine, glutathione and ascorbic acid to the reaction mixtures consisted with the polyphenol oxidase and polyphenols, the mutagenic effects were increased in the spore recassay. These results show that the activity of polyphenol oxidase may play an important role in the reduction of mutagenicity of polyphenols.

Bacillus sonorensis KCTC13918로부터 새로운 laccase유전자 (soncotA)의 클로닝과 대장균에서의 발현 (Cloning and expression of new laccase gene (soncotA) from Bacillus sonorensis KCTC13918 in E. coli)

  • A new putative laccase gene (soncotA) which show 78% homology with that from Bacillus licheniformis (liccotA) was isolated from draft genome sequence of Bacillus sonorensis KCTC 13918. A 1,545 bp of PCR product corresponding 514 amino acids was cloned into NdeI-NotI site of pET21c and expressed as soluble form in E. coli. About 59 kDa size of recombinant laccase was purified into homogenity by Ni-NTA column and laccase activity was confirmed by zymography. The enzymatic properties of recombinant laccase were characterized. The specific activity of B. sonorensis laccase was 0.033 fold lower than that of Bacillus licheniformis laccase. The finding of new laccase gene broadened the enzymatic diversity of Bacillus species laccases.
