Objectives: The objective of this study is to identify toxicants causing acute toxicity in effluents from the aluminum rolling industry that violate the discharge limits in Korea. Methods: Whole effluent toxicity tests (WET) were conducted on effluent discharged from the aluminum rolling industry following the US EPA WET test methods. We collected effluent samples three times and evaluated acute toxicity by using Daphnia magna. We employed toxicity identification evaluation (TIE) procedures to identify toxicants causing toxicity in the effluent. Results: No specific chemical groups were identified in the seven different manipulations applied to the of wastewater effluent samples showing 1.3 toxic units (TU) according to the TIE phase I procedures. Water quality parameters for water hardness, electric conductivity and heavy metals (Mn) were 4,322 mg/l as $CaCO_3$, 11.39 mS/cm, and $5,551{\mu}g/l$, respectively. Considering water hardness and reference toxicity, high concentrations of Mn can be disqualified from the causative toxicants. Consequently, high ionic concentrations of $Na^+$(1,648 mg/l), $Ca^{2+}$(1,048 mg/l), $Mg^{2+}$(1,428 mg/l) and $SO_4{^{2-}}$(7,472 mg/l) were identified to be causative toxicants. Water hardness and electric conductivity exceed the $EC_{50}$ value obtained by biological toxicity tests using Daphnia magna. Conclusion: According to TIE procedures, high salt concentration is determined to be a major toxicant in the effluent of agro-industrial wastewater treatment plants receiving wastewater from the aluminum rolling industry.
The heavy metal cadmium is a xenobiotic toxicant of environmental and occupational concern and it has been classified as a human carcinogen. Inhalation of cadmium has been implicated in the development of emphysema and pulmonary fibrosis, but, the detailed mechanism by which cadmium induces adverse biological effects is not yet known. Therefore, we undertook the investigation of genes that are induced after cadmium exposure to illustrate the mechanism of cadmium toxicity For this purpose, we employed the polymerase chain reaction-based suppression subtractive hybridization technique. We identified 29 different cadmium-inducible genes in human peripheral mononuclear cells, such as macrophage migration inhibitory factor, lysophosphatidic acid acyltransferase-${\alpha}$, enolase-1${\alpha}$, VEGF, Bax, neuron-derived orphan receptor-1, and Nur77, which are known to be associated with inflammation, cell survival, and apoptosis. Induction of these genes by cadmium treatment was further confirmed by semi-quantitative reverse-transcription polymerase chain reaction. Further, we found that these genes were also induced after cadmium exposure in normal human lung fibroblast cell line, WI-38, suggesting potential use of this induction profile to monitor cadmium toxicity in the lung. Next, Nur77, one of cadmium-inducible genes, was further studied since the products of Nur77 are known to be involved in the apoptotic process of lung cells. Following cadmium treatment, Nur77 gene expression was increased at protein-level in A549 cells. Consistently, the reporter containing Nur77 binding sequence was activated by 2.5-fold after exposure to cadmium in reporter gene analysis by transient transfection experiments. When the plasmid encoding dominant negative Nur77 that represses the transcriptional function of wild-type Nur77 was transfected into A549 cells, the expression of Bax was significantly reduced, suggesting that induction of Nur77 was an important process in cadmium-induced apoptosis in the cells. Cadmium induced the expression of Nur77 in vivo, confirming the relevance of the data obtained in viro. Together our results suggest that Nur77 gene expression in exposure to cadmium leads apoptosis of lung cells which may cause pathological changes in lung.
지난 반세기 이상 작물보호제의 수단으로 사용되어 왔던 화학농약은 사용 후 식물과 환경구성 요소 중에 남게 되는 잔류성 문제로 일반사회의 지대한 관심거리로 부상하고 있다. 세계 각 국이 국가차원의 규제로써 등록시 안전성 요구자료를 강화하고 있고 일단 등록된 농약이라 할지라도 주기적으로 안전성은 다시 평가하는 재등록제를 도입하고 있다. 특히 최근 지구환경보호 차원에서 국제기구를 중심으로 잔류성 유기 오염물질 (POPs), 내분비계 장애물질 (EDs)에 대한 관심이 집중됨에 따라 국가차원에서 우리 먹거리의 안전성을 확보하고 환경보전을 위하여 다양한 조사사업과 정책 등이 수립 시행되고 있다. 이들 조사결과에 대하여 과학적인 평가와 진단 그리고 대책들은 소비자의 알 권리를 충족하기 위하여 일반 대중에게 공개되어야 할 것이다.

The relationship between the layered structure of granules in UASB reactors and microbial resistance to toxicity was investigated using disintegrated granules. When no toxic materials were added to the media, the intact and disintegrated granules exhibited almost the same ability to decrease COD and to produce methane. However, when metal ions and organic toxic chemicals were added to a synthetic wastewater, he intact granules were found to be more resistant to toxicity than the disintegrated granules, as determined by the methane production. The difference in resistance between the intact and disintegrated granules was maximal, with toxicant concentrations ranging from 0.5 mM to 2 mM for trichloroethylene with toluene and 5 mM to 20 mM for metal ions (copper, nickel, zinc. chromium, and cadmium ions). The augmented COD removal rate by granulation compared to disintegrated granules was also measured in the treatment of synthetic and real wastewaters; synthetic wastewater, $-2.6\%$; municipal wastewater, $2.8\%$; swine wastewater, $6.4\%$; food wastewater, $25.0\%$; dye works wastewater, $42.9\%$; and landfill leachate, $61.8\%$. Continuous reactor operation also demonstrated that the granules in the UASB reactor were helpful in treating toxic wastewater, such as landfill leachate.
Background: Research on the status of many chemicals used in the semiconductor industry is needed. The purpose of this study was to describe the overall status of chemical use in the semiconductor industry in Korea and to examine it from a health perspective. Methods: Data on the status of chemical use and safety data sheets at 11 of 12 major semiconductor workplaces in Korea were collected. The number of chemical products and chemical constituents, quantities of chemicals, and trade secret ingredients used, as well as the health hazards were examined. Results: On average, 210 chemical products and 135 chemical constituents were used at the surveyed workplaces. Among all chemical products, 33% (range: 16-56%) contained at least one trade secret ingredient. Most of the trade secret ingredients were used in the photolithography process. Several carcinogens, including sulfuric acid, chromic acid, ethylene oxide, crystalline silica, potassium dichromate, and formaldehyde were also used. Only 29% (39 of 135) of the chemical constituents had occupational exposure limits, and more than 60% had no National Fire Protection Association health, safety, and reactivity ratings. Based on the aforementioned results, this study revealed the following. First, many chemical products and constituents are being used in the semiconductor industry and many products contained trade secret ingredients. Second, many products contained significant amounts of carcinogenic, mutagenic, and reproductive toxicant materials. Conclusion: We conclude that protecting workers in the semiconductor industry against harm from chemical substances will be difficult, due to widespread use of trade secret ingredients and a lack of hazard information. The findings of the status of chemical use and the health and safety risks in semiconductor industry will contribute to epidemiological studies, safe workplace, and worker health protection.
Recent studies reveal that the endocrine disrupter nonylphenol can also influence the growth of planktonic organisms. To clarify the effect of nonylphenol on the whole planktonic community, we monitored planktonic abundances after addition of nonylphenol using small-scale microcosms in a laboratory. Nonylphenol was added at final concentrations of 1.25 and $2.5{\mu}g\;L^{-1}$, close to the EC50 for the growth of the rotifer, Brachionus calyciflorus. Chlorophyll $\alpha$ concentration increased significantly between 2 to 5 days after nonylphenol treatment compared to the control. The abundance of the predominant phytoplankton, Stephanodiscus hantzschii, followed the same pattern as chlorophyll a concentration. While there was no negative effect on the abundance of ciliates and rotifers, crustacean zooplankton abundance was higher in nonylphenol treatments. Although the relationship did not reach significance, the growth rate of rotifers tended to decline with increasing nonylphenol dosing. It is likely that the decreased rotifer grazing on S. hantzschii caused significant increase in their abundance. This study emphasizes the importance of considering indirect effects of environmental pollutants when predicting the response of biological community to toxicant exposure.
Toxicogenomics combines transcriptome, proteome and metabolome profiling with conventional toxicology to investigate the interaction between biological molecules and toxicant or environmental stress in disease caution. Toxicogenomics faces the problems of comparison and integration across different sources of data. Cause of unusual characteristics of toxicogenomic data, researcher should be assisted by data analysis and annotation for getting meaningful information. There are already existing repositories which claim to stand for toxicogenomics database. However, those just contain limited abilities for toxicogenomic research. For supporting toxicologist who comes up against toxicogenomic data flood, now we propose novel toxicogenomics knowledgebase system, XPERANTO-TOX. XPERANTO-TOX is an integrated system for toxicogenomic data management and analysis. It is composed of three distinct but closely connected parts. Firstly, Data Storage System is for reposit many kinds of '-omics' data and conventional toxicology data. Secondly, Data Analysis System consists of analytical modules for integrated toxicogenomics data. At last, Data Annotation System is for giving extensive insight of data to researcher.
Upflow anaerobic sludge blanket (UASB) system employs granular sludge to treat various wastewaters including landfill leachate. CH$_4$ production of the granules determines overall performance of a UASB reactor. Sludge granules are developed by self-granulation of microorganisms and dynamic balance between granule growth and decay results in coexistence of granules with different sizes in the reactor. In this study, granules taken from a laboratory-scale UASB reactor were classified into 4 groups based on their diameters and their Physicochemical characteristics we were investigated. Each group was analyzed for settling ability, specific methanogenic activity (SMA), and elemental content. Settling ability was proportional to granule diameter. suggesting effective detainment of larger granules in the reactor. When acetate or glucose was used as a substrate, all groups showed relatively slight difference in SMA. However SMA with a volatile fatty acid mixture showed significant increase with granule diameter, suggesting better establishment of syntrophic relationship in larger granules. Larger granules showed higher value of SMA upon environmental changes (i.e., PH, temperature, or toxicant concentration). Comparative analysis of elemental contents showed that content (dry weight %) of most tested elements (iron, calcium, phosphorus, zinc, nickel. and manganese) deceased with granule diameter, suggesting importance of these elements for initial granulation. Taken together, this study verified experimentally that Physicochemical Properties of granules are related to granule size distributions. Overall results of physicochemical characterization supports that larger.
폐수의 복합독성과 다수의 미지 화합물의 영향을 평가하는 것은 많은 시간이 요구되고 분석상 어려움이 존재하는 경우가 많다. 그러므로 폐수중 독성을 나타내는 원인물질의 탐색과 처리효과를 평가하기 위한 방법을 확립하는 것이 절실히 요구된다. 본 연구에서는 질산화 저해제를 이용하여 혼합미생물중 질산화 미생물과 종속영양미생물을 구분하여 암모니아 저해도 실험을 수행하였으며, 이때 저해도는 dehydrogenase activity(DHA)를 이용한 미생물활성도의 차이로서 평가하였다. DHA에 의한 암모니아 저해 연구는 총 암모니아의 영향, 유리 암모니아의 영향, 질산화 미생물과 종속영양미생물의 암모니아 저해도에 대해 각각 평가되었다. 암모니아 독성은 총 암모니아 농도와 pH가 증가할수록 크게 나타나 유리 암모니아의 역할이 중요하게 작용함이 밝혀졌으나 미생물의 DHA는 총 암모니아와 유리 암모니아가 함께 고려되어야 정확히 평가될 수 있었다. 3.000mg/L 이상의 고농도의 암모니아에서는 질산화 미생물이 종속영양미생물 보다 심한 저해를 받는 것으로 나타났다.
농약의 만성어독성을 평가하기 위한 노력의 일환으로 제초제인 butachlor에 대하여 송사리 (Oryzias latipes)을 대상으로 하여 90일간 아급성 어독성 실험을 실시하였고 또한 만성독성 평가를 위한 생물학적 검정어류로써 송사리의 타당성을 제고한 바 그 결과는 다음과 같다. 1) 송사리는 우리나라에서 고르게 분포하고 있는 소형 민물고기로써 시험기간중 대조군 등에서 산란 및 부화가 이루어져 풍부한 생물학적 자료와 함께 농약에 대한 만성 어독성 평가에 적합하다고 사료된다. 2) Butachlor의 0.16 ppm 처리군은 약간의 길이 성장감소와 산란된 수정란의 부화율이 감소되는 경향을 나타내었고, 0.04ppm과 0.01 ppm의 butachlor 처리군에서는 대조군과 비교할 때 별 다른 영향을 받지 않았음을 관찰할 수 있었다. 고로, butachlor는 송사리에서 0.04${\sim}$0. 16ppm의 MATC 값을 갖는 것으로 사료되며 또한 실험기간중 DO와 pH및 온도를 매일 측정한 결과 송사리에 영향을 주지 않았음이 입증되었다.
