  • The single cell gel electrophoressis(SCGE) assay, also known as the comet assay, is a rapid, simple, visual and sensitive technique for measuring and analysing DNA breakage in mammalian cells. The SCGE or comet assay is a promising test for the detection of DNA damage and repair in individnal cells. It has widespread potential applications in DNA damage and repair studies, genotoxicity testing and biomonitoring. In this microgel electrophoresis technique, cells are embedded in agarose gel on microscope slides, iysed and electrophoresed under alkaline conditions. Cells with increased DNA damage display increased migration of DNA from the nucleus towards the anode. The length of DNA migration indicates the amount of DNA breakage in the cell. The comet assay is also capable of identifying apoptotic cells which contain highly fragmented DNA. Here we review the development of the SCGE assay, existing protocols for the detection and analysis of comets, the relevant underlying principles determining the behaviour of DNA and the potential applications of the technique.

Implementation of Context-Aware Services Platform Supporting Mobile Agents

  • A context-aware services platform supporting mobile agents consists of sensor nodes and a sensor coordinator. Sensor nodes collect environmental information and transmit the collected information to the sensor coordinator through wireless sensor networks. The sensor coordinator passes the information to the context-aware service module, and the mobile agent. The context-aware service module or the mobile agent performs services suitable for a user's situation based on the environmental information and a service actuation message is delivered to an actuation node through the sensor coordinator. In this paper, we present a context-aware services platform structure employed in our project, and describe context-aware services platform interfaces with a context-aware service module and mobile agents.

  • This paper described the overall operations and applications of the water environment management and evaluation systems (WEMES) for environmental management, conservation, and evaluation for a small watershed. WEMES was to be applied with field data from small watersheds ranging from 0.4 to 29.79km2 in size which have steep topography and complex landuses. WEMES was a computer aided decision support system, that was comprised of the database subsystem, the simulation model subsystem, the environmental evaluation model subsystem, and the graphic user interface subsystem that visualizes the simulated results. The water environment management and evaluation with WEMES was successfully performed.

  • Review of the global change in various energy usages and resulting environmental impacts were made in terms of population increase, economic development and energy consumption. Greater use of fossil fuels in past couple of centuries give rise to acid rain and gradual climate changes mainly due to Green House inducing gases emissions from fossil fuel combustion. In view of the forthcoming Kyoto conference in December, various alternative options were assessed. To cope with rapidly developing robust Korean economy, the alternative energy options for the sustainable development in 21st Century would be the wider use of Nuclear Energy in parallel with constrained use of fossil fuel and renewable energy development. However there are many hurdles to overcome. One of the most important element is to improve public acceptability of those alternatives. Since public acceptance depend heavily upon individual perception on specific energy technology applications, the basic energy technologies education from primary up to high school education and the related curriculum organization is important. The suggested improvement in education for Nuclear Energy Application was made on the basis of advanced industrial countries with substantial Nuclear Energy Application programs.

Mechanical and Durability Performance of Roller-Compacted Concrete with Fly Ash for Dam Applications

  • This study investigated the mechanical and durability performance of roller-compacted concrete (RCC) with fly ash for dam applications. A test program studied the effects on the properties of fresh and hardened RCC with fly ash replacement ratio, as well as the long-term durability of the resulting mixture. Fly ash replaced 20, 30, 40, and 50% by mass of the cement. Laboratory tests of the compressive strength, splitting tensile strength, shear strength, chloride ion permeability, abrasion, and drying shrinkage were conducted. The test results demonstrated that 30% fly ash replacement is an optimum level, and that this mixture has excellent mechanical and durability properties.

Modeling and Analysis of a Multi Bossed Beam Membrane Sensor for Environmental Applications

  • This paper presents a unique pressure sensor design for environmental applications. The design uses a new geometry for a multi bossed beam-membrane structure with a SOI (silicon-on-insulator) substrate and a mechanical transducer. The Intellisuite MEMS CAD design tool was used to build and analyze the structure with FEM (finite element modeling). The working principle of the multi bossed beam structure is explained. FEM calculations show that a sensing diaphragm with Mises stress can provide superior linear response compared to a stress-free diaphragm. These simulation results are validated by comparing the estimated deflection response. The results show that, the sensitivity is enhanced by using both the novel geometry and the SOI substrate.

Environmental Effects Analysis by the Fertilizer Change with Wastewater Reuse in Paddy Fields (하수처리수의 농업용수 재이용시 시비량 변화에 따른 환경영향 분석)

  • The objective of this paper is to analyze the environmental effects by the fertilizer change with wastewater reuse for agriculture. For this research, Lysimeter tests are being implemented to cultivate rice with different levels of fertilizer applications with wastewater irrigation., and to analyze the nutrient loading by wastwater reuse in paddy fields was examined the CREAMS-PADDY model. CREAMS-PADDY model is modified from CREAMS model for considering the hydrologic cycles in paddy field. As a result, in the lysimeter treated by irrigation with wastewater and chemical fertilizer with half of the conventional amount showed generally similar tendency to the control plot. This may require the modifications of standard cultural practices for rice in terms of fertilizer and pesticide applications. However, high concentration of sodium in wastewater might cause damage to physico-chemical properties of paddy soil. And the wastewater reuse effects on nutrient loads were quantitatively analyzed and this results provide the reasonable management for agricultural reuse.

  • Anisotropy of magnetic susceptibility (AMS), directional variation of susceptibility in a sample, indicates the relative magnitude and orientation of preferentially aligned magnetic grains in a rock sample and represents important aspects of rock fabrics. This observation leads to various studies which applied the AMS technique to numerous geological problems, especially sedimentology, petrology, structural geology, and tectonics. Since AMS is, relative to conventional methods of petrofabric measurement, quickly measured, easily interpretated, and providing quantitative data, AMS technique has evolved into a powerful tool for detection of rock fabrics and continues to draw the interest of non-specialists as well as magnetists. This paper represents the theory, techniques, and parameters of the relatively new AMS method for estimating petrofabrics and their various applications.

  • The behavior of rocks and microcrack development due to fatigue stresses are investigated using cyclic loading tests and ultrasonic velocity measurements. Twenty six medium-grained granite samples from the Pochon area are selected for measurements. Ultrasonic velocities are measured for samples before fatigue test to characterize the pre-existing microcracks. Then, thirteen different cycles of loadings with 70% and 80% dynamic strength are applied to the samples. The ultrasonic velocities are measured again to compare velocities after applications of fatigue stress with those before applications of fatigue stress. The results show that most microcracks are developed along the direction parallel to the axis of loading and that the amount of microcracks increases, as loading levels and numbers of cycle increase.

  • This paper describes the 40kV solid state pulsed power supply for environmental applications. The output specifications of the pulsed power supply are 40kV, 300A, 3kHz, and average output power of 13kW. In order to generate a high voltage, a series stacking cell structure is used which is charged in parallel and discharged in series. Due to this structure, there is no dynamic voltage balancing problem as well as static voltage balancing problem for switches used in high voltage pulse power supplies. To verify this pulse power supply design, PSpice modeling was performed. Finally, experimental results with non-inductive resistive load and gas treatment reactor proved the reliability of the solid state pulsed power supply.
