Photochemical Index Analysis on the Influence of LED Illumination Color Temperature on Donarium Cherry (도시야간조명의 LED 색온도별 겹벚나무의 광화학적 생리지표 해석)

  • In this study, the efficiency of photosynthetic electron transfer according to LED color temperature was verified in order to find a way to efficiently grow trees under night illumination. The experiment was carried out with White treatment, Warmwhite treatment, and non-treatment with donarium cherry. The study uses to a method for analyzing and evaluating the color temperature of an LED light source by photochemical analysis. We found that all treatments 115 DAT of maximum fluorescence amount(P) had the lowest. In the treatment using white light and the Warm-white light, the T amount of florescence of the late stage during the transition of the J-I level was increased, and the photosystem I electron transfer efficiency was decreased. Therefore, the electron transport efficiency of $RE1_O/CS$ and RE1o/RC were reduced. Especially, compared to Warmwhite, the light intensity increased greatly in the white-light treatment, The $PI_{TOTALABS}$ of 7 DAT was the highest value, but it was decreased to the lowest value on 115 DAT. This study has shown that the white treatment was low in electron transfer efficiency and soundness. Warmwhite-light treatments showed lower stress.

Biochemical Characterization of $\small{L}$-Asparaginase in NaCl-Tolerant Staphylococcus sp. OJ82 Isolated from Fermented Seafood

  • $\small{L}$-Asparaginase from gram-positive bacteria has been poorly explored. We conducted recombinant overexpression and purification of $\small{L}$-asparaginase from Staphylococcus sp. OJ82 (SoAsn) isolated from Korean fermented seafood to evaluate its biotechnological potential as an antileukemic agent. SoAsn was expressed in Escherichia coli BL21 (DE3) with an estimated molecular mass of 37.5 kDa, determined using sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Consistent with asparaginases in gram-negative bacteria, size-exclusion chromatography determined SoAsn as a homodimer. Interestingly, the optimal temperature of SoAsn was $37^{\circ}C$ and over 90% of activity was retained between $37^{\circ}C$ and $50^{\circ}C$, and its thermal stability range was narrower than that of commercial E. coli $\small{L}$-asparaginase (EcAsn). Both SoAsn and EcAsn were active between pH 9 and 10, although their overall pH-dependent enzyme activities were slightly different. The $K_m$ value of SoAsn was 2.2 mM, which is higher than that of EcAsn. Among eight metals tested for enzyme activity, cobalt and magnesium greatly enhanced the SoAsn and EcAsn activity, respectively. Interestingly, SoAsn retained more than 60% of its activity under 2 M NaCl condition, but the activity of EcAsn was reduced to 48%. Overall, the biochemical characteristics of SoAsn were similar to those of EcAsn, but its kinetics, cofactor requirements, and NaCl tolerance differed from those of EcAsn.

Study on the distribution law and influencing factors of pressure field distribution before exploitation in heavy oilfield

  • A calculation model of reservoir pressure field distribution around multiple production wells in a heavy oil reservoir is established, which can overcome the unreasonable uniform-pressure value calculated by the traditional mathematical model in the multiwell mining areas. A calculating program is developed based on the deduced equations by using Visual Basic computer language. Based on the proposed mathematical model, the effects of drainage rate and formation permeability on the distribution of reservoir pressure are studied. Results show that the reservoir pressure drops most at the wellbore. The farther the distance away from the borehole, the sparser the isobaric lines distribute. Increasing drainage rate results in decreasing reservoir pressure and bottom-hole pressure, especially the latter. The permeability has a significant effect on bottom hole pressure. The study provides a reference basis for studying the dynamic pressure field distribution before thermal recovery technology in heavy oilfield and optimizing construction parameters.

An Experimental Study on the MR Characteristics of Subbase Materials (보조기층재료의 MR 특성에 관한 실험적 연구)

  • The resilient behavior of subbase materials, locally used in Korea. were evaluated in this study by performing laboratory MR tests. The variations of the MR value according to LVDT's location in experiments and moisture conditions, respectively, were evaluated. And, in order to determine the relevant MR characteristics of the prototype materials, laboratory model tests containing smaller particles than those of the prototype were conducted. Based on above results, the relationship of the MR and the bulk stress (${\theta}$) was suggested. The case using internal LVDT. gave much larger $M_R$ value than that using external LVDT, since the external LVDT somewhat overestimates the resilient strain. The measured $M_R$ in damp conditions ($S_r$=40%) was larger than that in wet conditions ($S_r$=70%) by about 10%. The relationship between the $M_R$ and the void ratio was linear according to particle size effect. The relationship of the $M_R$ and the bulk stress (${\theta}$) on subbase materials in damp conditions to be used in Korea could be recommended as $M_R=3960{\cdot}{\theta}^{0.60}$ psi.

  • A direct shear test is classified roughly by one side simple shear test of confining horizontal displacement type and torsional shear test of non-confining one. Direct shear test that has been widely used so far has some problems with test apparatus, testing and the analysis, and in particular that its strength value is everestimated in sandy soils. Also, progressive failure of shearing process happens from shear apparatus restriction and because the shear strain and shear stress are erratic in specimen, we can not define the shear strain value. In the meantime, a simple shear test having advantage of direct shear test is an ideal test method that can get stress-strain relation on shear because it can deliver constant shearing deformation to specimen. However, simple shear test cannot be used practically, because its structure makes tester manufacturing difficult. This paper described a on outline of test apparatus, improvement of test method, and constant pressure test results based on the obtained from improved direct shear apparatus and the standardization of JGS soil testing method.

A Change of Porewater Pressure under Particle Crushing of Carbonate Sand of Sabkha Layer (Sabkha층 탄산질 모래의 입자파쇄에 따른 간극수압 변화)

  • Carbonate sand of Sabkha layer in the middle east was made of deposition of shell fragments and it consisted of porous particles containing inner void. Generally, at yield stress the soil structure begins to break down, so the porewater pressure and the settlement are increased rapidly. In carbonate sand, unlike quartz sand if particle crushing happens, the inner voids are exposed and porewater pressure can be decreased under yield stress. Porewater pressure can be determined as the sum of excess porewater pressure due to increase of relative density, inner void expose of particle under particle crushing stress and rearrangement of crushed particle fragments. The porewater pressure can be negative value in case of greater amount of inner void expose, so if particle crushing is bigger, the porewater pressure value is smaller. The negative value zone of porewater pressure from triaxial test result means particle crushing effect is bigger than outer void decrease effect and the particle crushing effect dominant zone size was 1.50∼3.46% from triaxial test result of Sabkha layer.

  • If cement could be manufactured with industrial byproducts such as granulated blast furnace slag, phosphogypsum, and waste lime rather than clinker, there would be many advantages, including the maximization of the use of these industrial byproducts for high value-added resources, the conservation of natural resources and energy by omitting the use of clinker, the minimization of environmental pollution problems caused by $CO_2$ discharge, and the reduction of the production cost. For this reason, in this study, mechanical behavior tests of non-sintered cement concrete were performed, and elasticity modulus and stress-strain relationship of non-sintered cement concrete were proposed. Nine test members were manufactured and tested according to reinforcement ratio and concrete compressive strength. According to the test results, there was no difference between general cement concrete and non-sintered cement concrete in terms of flexure and shear behavior.

  • As a considerable, experimental approach, an Auto-carriage type of $CO_2$ welding machine and a MIG(Metal Inert Gas) welding robot under inert gas atmosphere were utilized in order to realize Al 5083 welding applied to hull and relevant components of green Al leisure ships. This study aims at investigating the effect of welding conditions(current, voltage, welding speed, etc) on thermal deformation that occurs as welding operation and tensile characteristics after welding, by using Al 5083, non-ferrous material, applied to manufacturing of co-environmental Al leisure ships. With respect to welding condition to minimize the thermal deformation, 150A and 16V at the wire-feed rate of 6mm/sec were acquired in the process of welding Al 5083 through an auto carriage type of $CO_2$ welding feeder. As to tensile characteristics of Al 5083 welding through a MIG welding robot, most of tensile specimens showed the fracture behavior on HAZ(Heat Affected Zone) located at the area joined with weld metal, except for some cases. Especially, for the case of the Al specimen with 5mm thickness, 284.62MPa of tensile strength and 11.41% of elongation were obtained as an actual allowable tensile stress-strain value. Mostly, after acquiring the optimum welding condition, the relevant welding data and technical requirements might be provided for actual welding operation site and welding procedure specification(WPS).

Layer Interface and Approximated Nonlinear Analysis Method for Consolidation Prediction (압밀현상 예측을 위한 경계면 및 근사 비선형 해석기법)

  • The interface layer having different consolidation properties and nonlinear material function with permeability needs to be considered to predict consolidation behavior. In this study, interface equation between different layers has been derived and then applied to existing finite difference scheme for conducting consolidation analysis. These results have been compared with those by conventional method in which different layers are converted to single layer having conversion value of properties. Also, although the conventional consoilidation analysis is used to consider non-linearity of the permeability with effective stress, an approximated nonlinear method as a function of consoilidation coefficient with effective stress have been developed and applied to the consoilidation analysis for various cases.

Shear Strength Estimation of Clean Sands via Shear Wave Velocity (전단파 속도를 통한 모래의 전단강도 예측)

  • We perform a series of experimental tests to evaluate whether the shear strength of clean sands can be reliably predicted from shear wave velocity. Isotropic drained triaxial tests on clean sands reconstituted at different relative densities are performed to measure the shear strength and bender elements are used to measure the shear wave velocity. Laboratory tests reveal that a correlation between shear wave velocity, void ratio, and confining pressure can be made. The correlation can be used to determine the void ratio from measured shear wave velocity, from which the shear strength is predicted. We also show that a unique relationship exists between maximum shear modulus and effective axial stress at failure. The accuracy of the equation can be enhanced by including the normalized confining pressure in the equation. Comparisons between measured and predicted effective friction angle demonstrate that the proposed equation can accurately predict the internal friction angle of granular soils, accounting for the effect of the relative density, from shear wave velocity.
