Isolation and Characterization of PERV-C env from Domestic Pig in Korea

  • Clone PERV-C (A3) env was isolated from the genomic DNA of domestic pig (Sus scrofa domesticus) in Korea to investigate the molecular properties of PERV-C. The nucleic acid homologies between the PERV-MSL (type C) reference and the PERV-C(A3) clone was 99% for env, but a single base pair deletion was found in the transmembrane (TM) region of the env open reading frame. To examine the functional characteristics of truncated PERV-C env, we constructed a replication-incompetent retroviral vector by replacing the env gene of the pCL-Eco retrovirus vector with PERV-C env. A retroviral vector bearing PERV-C/A chimeric envelopes was also created to complement the TM defect. Our results indicated that truncated PERV-C env was not infectious in human cells as expected. Interestingly, however, the vector with the PERV-C/A envelope was able to infect 293 cells. This observation suggests that recombination within PERV-C TM could render PERV-C infectious in humans. To further characterize PERV-C/A envelopes, we constructed an infectious molecular clone by using a PCR-based technique. This infectious molecular clone will be useful to examine more specific regions that are critical for human cell tropism.

부탄 이용 미생물에 의한 MTBE(Methyl tert-Butyl Ether) 분해 특성

  • In this study, we have examined potential degradation of MTBE (methy1 tert-butyl ether) by pure culture ENV425 and mixed culture isolated from gasoline contaminated soil using n-butane as the sources of carbon and energy. The results described in this study suggest that MTBE is degraded cometabolically by ENV425 and mixed culture grown n-butane, and the disappearance of TBA after complete degradation of MTBE suggest the further degradation of TBA. Butane and MTBE degradation was completely inhibited by acetylene, which indicated that both substrates were degraded by butane-utilizing bacteria. MTBE was degraded ENV425 and mixed culture grown n-butane, and TBA (tert-butyl alcohol) was produced as product of MTBE oxidation. TBA production was accounted 54.7% and 58.6% for MTBE oxidation by ENV425 and mixed culture, respectively. The observed maximal transformation yield (T$_{y}$) were 44.7 and 34.0 (nmol MTRE degraded/$\mu$mol n-butane Utilized) by ENV425 and mixed culture, respectively.y.

Characterization of BLV env gene in Korean Holstein dairy cattle (한국형 홀스타인종 젖소의 BLV env 유전자의 특성분석)

  • This study was performed to investigate the characterization of infectious BLV env gene isolated form Korean Holstein Cattle and to determine its incoming origin. Gp51 region of BLV env gene known as having important role in immunological function was characterized using PCR-RFLP sequencing and phylogenetic analysis. BLV env gene was grouped into PCR-RFLP patterns with three restriction endonucleases including Pvu II, BamHI and Hae III, and we identified two new RFLP patterns from nucleotide sequences of each group. Phylogenetic analysis showed that 80% of the Korean Holstein was included in the USA and Japanese group. These results here can provide a valuable information about the character of the BLV env gene and research on infection route of BLV.

EFFECT OF CIGARETTE PAPER POROSITY ON THE VENTILATION OF CIGARETTES (궐련지 기공도가 담배연기 희석에 미치는 영향)

  • From the combination of cigarette papers, tipping papers and plug wrapping papers, whose physical properties and effect of cigarette smoke dilution were studied. The result obtained from this study are as follows. Envelop ventilation (Env-V'R) cigarettes used high porous cigarette papers showed less changes of unencapsulated pressure drop VPD) compared to tip ventilation (Tip-VR) cigarettes. Higher cigarette paper porosity increased, more decreased the WD of cigarette by Tip-VR. With cigarette length shortening, the UPD or VR changed a little in Tip-VR cigarette, while changed very much in Env-VR cigarettes. The ratio of nicotine/tar was higher in Env-VB cigarettes than in Tip-VR cigarettes, and CO/nicotine value of the former was lower than those of the later. The effect of Tip-VR cigarettes on the delivery reduction of tar and nicotine was more effective compared to Env-VR cigarettes and the reduction prefer Env-VR cigarettes to Tip-VR cigarettes in CO delivery. From the results investigated above it was noted that each defect appeared in the Tip-VR and the Env-VR cigarettes could be complemented by the combination of them.

Use of Waste Woods for Developing Environment-friendly Shock-absorbing Materials

  • Environment-friendly shock-absorbing materials were made using a vacuum forming method from waste wood collected from local mountains in Korea. The waste wood was pulped by thermomechanical pulping. The TMP cushions showed superior shock-absorbing properties with lower elastic moduli compared to EPS and pulp mold. Even though the TMP cushions made using different suction times had many free voids in their inner fiber structure, their apparent densities were a little higher than EPS and much lower than pulp mold. The addition of cationic starch improved the elastic modulus of the TMP cushions without increasing the apparent density, which was different from surface sizing with starch. The porosity of the TMP cushions was a little greater than EPS and much less than pulp mold. Finally, the TMP cushions have great potential to endure external impacts occurring during goods distribution.

  • Thermomechanical pulping process uses large amounts of energy, mostly electricity to run electrical facilities. Thermomechanical pulp (TMP) made from Pinus densiflora also has a big drawback that refining consumes 90 per cent of the total energy used in TMP process. This study explored to draw up a way to save refining energy through different thermal treatment at the stages of presteaming and refining. Presteaming temperature was $80^{\circ}C$, $100^{\circ}C$, and $120^{\circ}C$. After presteaming at each temperature, refining was carried out at $100^{\circ}C$, $120^{\circ}C$, and $140^{\circ}C$ respectively. In a presteaming stage, steaming temperature over $120^{\circ}C$ greatly contributed to the decrease of refining energy leading to earlier attainment of a target freeness, irrespective of refining temperature. In addition, NaOH treatment with presteaming enhanced better development of fiber properties during refining than presteaming without NaOH. High temperature refining at $140^{\circ}C$ produced a high strength paper, and wood chips treated by alkali responded better to refining than at over $120^{\circ}C$. Improved softening effect on wood chips led to the decrease in shives contents but it gave no effect on pitch contents of TMP.

Defective Interfering HIV-1 Pseudotypes Carrying Chimeric CD4 Protein

  • Chimeric CD4 proteins were assembled. They contained the entire CD4 ectodomain that is linked to different membrane anchors. Membrane anchors consisted of either glucosyl phosphatidyl inositol (gpi), the transmembrane and cytoplasmic regions of HIV-1 Env protein, or the vesicular stomatitis virus G glycoprotein, respectively. The HIV-1 co-receptor CXCR4 and CD4 were independently inserted into viral envelopes. We compared the insertion of six different CD4/CXCR4 constructs into HIV-1 envelopes, as well as their functionality in targeting and specific infection of cells that constitutively express the HIV-1 Env protein. All of the six different HIV-1 (CD4/CXCR4) pseudotypes were able to transduce Env (+) cells at similar efficiency. In addition, stable transduction of the Env (+) recipient cells demonstrated that all chimeric proteins were functional as receptors for Env when inserted into HIV-1 envelopes. In fact, these results demonstrate for the first time a stable transduction by a targeted HIV-1 pseudotype virus.
