전자코를 이용한 헥사날과 싸이클로덱스트린의 결합 분석 (Analysis for Cyclodextrins to Entrap with Hexanal using Electronic Nose)

  • 이취성분의 하나인 헥사날이 CD 종류, CD 농도, 저장기간에 따라 결합하는 정도를 전자코를 이용하여 분석하였다. 여러 종류의 CD 중에서 ${\alpha}-CD$가 헥사날과 가장 많은 양이 반응하면서 CD복합체를 이루었다. CD의 농도가 증가할수록 잔존하는 헥사날의 양이 감소하였는데 5% ${\beta}-CD$ 첨가시 86%의 헥사날이 감소하여 헥사날-CD복합체를 이루고 있음을 보여 주었다. CD복합체를 이룬 후 저장초기 상태에서는 헥사날의 양 변화에 별다른 차이가 없었지만 저장일이 지남에 따라 헥사날의 양이 점차 감소하였다. 또한 저장기간이 지남에 따라 자연 휘발되는 양보다는 CD와의 결합에 의해 헥사날의 양의 감소효과가 더 큰 것을 알 수 있었으며 이런 정도를 전자코로 분석할 수 있었다.

  • The effect of entrapping isothiocyanates extracted from horseradish root by microencapsulation on the shelf life of Myeongran jeotgal was evaluated. The total viable cell count of Myeongran jeotgal reached $10^7CFU/g$ (initial decomposition level) on days 43 and 45 of treatment with 1.0% and 2.0% microcapsules (4.0 mg/mL), respectively, compared with day 21 of storage at $5^{\circ}C$ as the control treatment. The proteolytic bacterial counts of Myeongran jeotgal treated with 1.0% and 2.0% microcapsules were $2.0{\times}10^5$ and $9.5{\times}10^4CFU/g$, respectively, with 2 and 3 log reductions compared with the control count ($1.1{\times}10^7CFU/g$) on day 33 of storage at $5^{\circ}C$. The total volatile basic nitrogen (TVB-N) level reached 30.0 mg% (initial decomposition level) on days 47 and 48 of treatment with 1.0% and 2.0% microcapsules, respectively, compared with day 33 of storage at $5^{\circ}C$ as the control. Based on the sensory evaluation, the freshness of Myeongran jeotgal treated with 1.0% microcapsules was best compared with the other treatments. Based on the total viable cell count, TVB-N and sensory evaluation, the shelf life of Myeongran jeotgal treated with 1.0% microcapsules was extended by 22, 16 and 15 days, respectively, compared with the control at $5^{\circ}C$.

근병증을 동반한 콩팥성전신섬유화증 (Nephrogenic Systemic Fibrosis with Myopathy)

  • Nephrogenic systemic fibrosis (NSF) is a systemic disease that affects the skin and other tissues in patients with renal insufficiency and exposure to gadolinium-containing contrast. A 55-year-old woman with end-stage renal disease on hemodialysis was consulted for progressive general weakness. After she had undergone multiple MRIs with gadolinium-containing contrast media, muscle weakness and skin lesions were developed. Her skin and muscle biopsy specimens showed CD34+ fibroblast entrapping collagen bundles. There are few reports of NSF with myopathy.

  • The hydrolyzed-lactose milk for the lactase-deficient subject is sweeter than whole milk, and some subjects dislike its taste. To overcome this shortcoming the dried liposomes containing ${\beta}$-galactosidase to digest lactose in milk after drinking were prepared and examined the possible application of this dried liposomes to the lactase-deficient subjects. To improve the stability of conventional liposome suspension, the dried liposomes in the presence of trehalose were prepared by the dehydration-rehydration vesicles method. Small unilamellar vesicles, prepared with egg phosphatidyl cholesterol, and cholesterol, were mixed with ${\beta}$-galactosidase solution and then ;up[jo;ozed. The freeze-dried liposome was rehydrated and centrifuged. The resultant multilamellar vesicles were mixed with trehalose(4g/g lipid) and then lyophilized to produce final dried liposome. Trehalose increased the entrapping efficiency of liposomes by 3 fo1d compared to the liposomes without trehalose (13% vs. 46%).

Convenient Preparation of Tumor-specific Immunoliposomes Containing Doxorubicin

  • Two innovative methods to prepare target-sensitive immunoliposomes containing doxorubicin by coupling monoclonal antibodies (mAb DH2, SH1) specific to cancer cell surface antigens ($G_{M3}$, $Le^X$) have been developed and are described here. Firstly, liposomes containing N-glutaryl phosphatidylethanolamine (NGPE) were prepared, followed by the encapsulation of doxorubicin, DH2 or SH1 antibodies were conjugated to NGPE in the liposomes (direct coupling). Secondly, liposomes were prepared with NGPE/mAb conjugates by the detergent dialysis method (conjugate insertion), and then doxorubicin was encapsulated by proton gradient. The immunoliposomes prepared by both methods were able to specifically bind to the surface of the tumor cells - B16BL6 mouse melanoma cells. The efficiencies of doxorubicin-entrapping into liposomes prepared by direct coupling and conjugate insertion was about 98% and 25%, respectively. These types of liposomal formulation are sensitive to target cells, which can be useful for various clinical applications.

Micellar Effects on Intramolecular Charge Transfer Emission from Biphenylcarboxylic Acids

  • The intramolecular charge transfer (ICT) phenomena of the photoexcited 2-biphenylcarboxylic acid (2BPCA) and 4-biphenylcarboxylic acid (4BPCA) have been investigated in some surfactant micellar solutions. The ICT emission of 4BPCA and 2BPCA in aqueous solution at sufficiently low pH (1-3) has been observed to be markedly quenched and blue-shifted upon addition of a cationic surfactant, cetyltrimethylammonium chloride (CTAC) in contrast to little change in anionic sodium dodecyl sulfate (SDS) and neutral Brij 35. An anionic emission band has been observed to be enhanced at expense of the ICT emission as a function of the concentration of CTAC. These results with the micellar effects on the fluorescence decay kinetics of 4BPCA suggest that formation of the ICT state of the excited acids is inhibited by CTAC-induced proton transfer as well as the decrease in the polarity and/or hydrogen-bonding ability of the micellar microenvironment entrapping the acids.

The Preparation of Poly(N-methylpyrrole) Bilayers with Entrapped Anthraquinone-2-sulfonate

  • Anthraquinone-2-sulfonate (AQS) release from poly(N-methylpyrrole anthraquinone-2-sulfonate) (PNMP-AQS) was investigated at open circuit and compared with electrochemically stimulated release during potential cycling. It was found that the fast AQS release from PNMP-AQS single layers is substantially retarded and the amounts of spontaneously and electrochemically releasable AQS can be reduced by constructing bilayers, consisting of PNMP-AQS inner layers and PNMP outer layers. PNMP-Cl outer layers exhibited higher effectiveness for entrapping AQS within inner layers than PNMP/poly(styrene slfonate). The effects of outer layer thicknesses on AQS release were also examined with PNMP-AQS:PMP-Cl. The electroactivity enhancement of PNMP-AQS:PNMP-Cl bilayers due to entrapped AQS was confirmed by chronocoulometry.

파클리탁셀을 함유한 지질나노입자의 제조와 인공 소화액에서의 안정성 평가 (Preparation of Lipid Nanoparticles Containing Paclitaxel and their in vitro Gastrointestinal Stability)

  • Peroral administration is the most convenient one for the administration of pharmaceutically active compounds. Most of poorly water-soluble drugs administered via the oral route, however, remain poorly available due to their precipitation in the gastrointestinal (GI) tract and low permeability through intestinal mucosa. In this study, one of drug delivery carriers, lipid nanoparticles (LNPs) were designed in order to reduce side effects and improve solubility and stability in GI tract of the poorly water soluble drugs. However, plain LNPs are generally unstable in the GI tract and susceptible to the action of acids, bile salts and enzymes. Accordingly, the surface of LNPs was modified with polyethylene glycol (PEG) for the purpose of improving solubility and GI stability of paclitaxel (PTX) in vitro. PEG-modified LNPs containing PTX was prepared by spontaneous emulsification and solvent evaporation (SESE) method and characterized for mean particle diameter, entrapping efficiency, zeta potential value and in vitro GI stability. Mean particle diameter and zeta potential value of PEG-modified LNP containing PTX showed approximately 86.9 nm and -22.9 mV, respectively. PTX entrapping efficiency was about 70.5% determined by UV/VIS spectrophotometer. Futhermore, change of particle diameter of PTX-loaded PEG-LNPs in simulated GI fluids and bile fluid was evaluated as a criteria of GI stability. Particle diameter of PTX-loaded PEG-LNPs were preserved under 200 nm for 6 hrs in simulated GI fluids and bile fluid at $37^{\circ}C$ when DSPE-mPEG2000 was added to formulation of LNPs above 4 mole ratio. As a result, PEG-modified LNPs improved stability of plain LNPs that would aggregate in simulated GI fluids and bile solution. These results indicate that LNPs modified with biocompatible and nontoxic polymer such as PEG might be useful for enhancement of GI stability of poorly water-soluble drugs and they might affect PTX absorption affirmatively in gastrointestinal mucosa.

hydroquinone의 혼합기체 포집 거동 특성분석 (Study on entrapping behavior of gas mixtures into hydroquinone clathrate)

  • clathrate compound란 호스트 분자가 수소 결합에 의하여 3차원 골격구조를 만들고, 이 격자 내부의 동공으로 저분자량의 기체 게스트 분자가 포집되며 형성되는 고체 결정 화합물이다. 현재까지 다양한 호스트 분자가 clathrate 화합물을 형성하는 것으로 보고되어 있으며, 이 중 유기물인 hydroquinone 역시 clathrate compound를 형성할 수 있는 것으로 알려져 있다. clathrate compound는 작은 고체 부피 내부에 막대한 양의 기체 분자를 저장할 수 있는 특성을 지니고 있기 때문에, 에너지 가스의 저장/수송이나 혼합 가스의 선택적 분리와 같은 다양한 응용을 위한 연구가 활발히 진행되고 있다. 본 연구에서는 clathrate compound를 형성하는 유기 호스트 분자인 hydroquinone을 이용하여 다양한 기체분자들에 대한 포집 거동을 파악하였다. 순수 기체로는 $N_2$, $H_2$, $CO_2$, $CH_4$의 4종류를 가지고 고압 반응기에서 50bar의 압력, 상온에서의 반응 조건으로 반응을 시켰다. 이렇게 형성된 반응 샘플들은 clathrate 형성 여부(기체의 포집 여부)를 확인하기 위하여 x-ray 회절을 통한 고체 결정 구조 분석을 수행하였다. 또한 순수 기체 이외에 다양한 비율(20%, 40%, 60%, 80%)의 조성을 갖는 $CO_2+N_2$ 혼합가스를 이용하여 clathrate compound의 형성과 조성 분석을 수행하였는데, x-ray 회절 분석과 13C 고체 NMR 분석을 통해 미세 구조 분석 연구를 수행하였고, Raman분석을 통하여 그 조성을 확인하였다. 본 연구에서 얻어진 결과는 기체의 저장/수송이나 혼합 가스의 선택적 분리와 같은 응용 분야에서 중요한 정보를 제공할 수 있을 것으로 기대된다.

건조 고정화 Zymomonas mobilis에 의한 sorbitol 생산 (Production of Sorbitol Using Dried and Immobilized Zymomonas mobilis)

  • 투과성이 증가된 Z. mobilis를 K-carrageenan으로 고정화한 건조beads를 이용하여 sorbitol연속생성공정의 향상에 관한 실험을 하였다. 투과성을 증가시킨 균체를 K-carrageenan으로 고정화 하여서 연속공정을 실시한 결과 효소의 안정성이 30일 이상 지속되었다. K-carrageenan으로 고정화한 균체를 건조시 bead의 견고성과 저장성이 향상되었다. 건조 bead를 이용한 72시간의 반회분식 공정에서 효소 활성도의 감소는 8%였으며, 건조 bead에서의 Vmax값은 $39^{\circ}C$와 pH6.2에서 free cell의 거의 절반값을 나타내었다. 연속공정에서 희석속도 $0.1h^{-1}$일 때 wet bead와 건조bead에서의 sorbitol생산성은 각각 3.4, 2.88g/l-h를 나타내었다.
