Characterization of Drug-Resistant Salmonella enterica Serotype Typhimurium by Antibiograms, Plasmids, Integrons, Resistance Genes, and PFGE

  • Forty-seven Salmonella Typhimurium (33 zoonotic, 14 clinical) strains were tested for antimicrobial resistance using the standard disk diffusion method. The presence of relevant resistance genes and class 1 integrons were investigated by using PCR. Pulsed-field gel electrophoresis (PFGE) and plasmid profiling were carried out to determine the genomic diversity of Salmonella Typhimurium. Approximately 57.4% of the S. Typhimurium strains were multidrug resistant (MDR) and showed high resistance rates to tetracycline (70.2%), sulfonamides (57.4%), streptomycin (53.1%), ampicillin (29.7%), nalidixic acid (27.6%), kanamycin (23.4%), chloramphenicol (21.2%), and trimethoprim (19.1%). Resistance towards cephalosporins was noted for cephalothin (27.6%), cephradine (21.2%), amoxicillin clavulanic acid (17.0%), and cephalexin (17.0%). Resistance genes, $bla_{TEM}$, strA, aadA, sul1, sul2, tetA, tetB, and tetC, were detected among the drug-resistant strains. Thirtythree strains (70.2%) carried class 1 integrons, which were grouped in 9 different profiles. DNA sequencing identified sat, aadA, pse-1, and dfrA genes in variable regions on class 1 integrons. Thirty-five strains (74.4%) were subtyped to 22 different plasmid profiles, each with 1-6 plasmids (2.0 to 95 kb). PFGE subtyped the 47 strains into 39 profiles. In conclusion, high rates of multidrug resistance were found among the Malaysian Salmonella Typhimurium strains. The emergence of multidrug-resistant Salmonella Typhimurium to cephalosporin antibiotics was also observed. The strains were very diverse and no persistent clone was observed. The emergence of MDR Salmonella Typhimurium is a worldwide problem, and this report provides information for the better understanding of the prevalence and epidemiology of MDR S. Typhimurium in Malaysia.

Solar Flare Occurrence Rate and Probability in Terms of the Sunspot Classification Supplemented with Sunspot Area and Its Changes

  • We investigate the solar flare occurrence rate and daily flare probability in terms of the sunspot classification supplemented with sunspot area and its changes. For this we use the NOAA active region data and GOES solar flare data for 15 years (from January 1996 to December 2010). We consider the most flare-productive eleven sunspot classes in the McIntosh sunspot group classification. Sunspot area and its changes can be a proxy of magnetic flux and its emergence/cancellation, respectively. We classify each sunspot group into two sub-groups by its area: "Large" and "Small". In addition, for each group, we classify it into three sub-groups according to sunspot area changes: "Decrease", "Steady", and "Increase". As a result, in the case of compact groups, their flare occurrence rates and daily flare probabilities noticeably increase with sunspot group area. We also find that the flare occurrence rates and daily flare probabilities for the "Increase" sub-groups are noticeably higher than those for the other sub-groups. In case of the (M+X)-class flares in the 'Dkc' group, the flare occurrence rate of the "Increase" sub-group is three times higher than that of the "Steady" sub-group. Our results statistically demonstrate that magnetic flux and its emergence enhance the occurrence of major solar flares.

Physio-ecological Characteristics of Sagittaria trifolia L., a Perennial Weed in Paddy Field 1. Effect of Environmental Factors on Emergence of Tuber of S. trifolia (논 다년생잡초(多年生雜草) 벗풀의 생리생태적(生理生態的) 특성(特性)에 관한 연구(硏究) - 1. 벗풀 지하경(地下莖)의 출아특성(出芽特性))

  • Arrowhead (Sagittaria trifolia L.), a perennial weed dominant in paddy field, is considerably difficult to control by herbicides. The investigation on the effects of environmental factors on emergence of arrowhead tuber can aid for controlling this weed. The results exmined were as follows : From the results of emergence characteristics at different levels of temperature, arrowhead tuber could germinated at 15$^{\circ}C$-40$^{\circ}C$. The optimal temperature for basal emergence was ranged from 25 to 30$^{\circ}C$. Basal emergence at 15$^{\circ}C$ and 20$^{\circ}C$ began at 56 and 12 days after tuber plantation, respectively and were later than those at the optimal temperature. At the different levels of shading, all the tested tubers could emerged. The days required for basal emergence at 25% and 50% shading plots were shorter than those at 70%, 80% and non-shading plots. The basal emergence was more effective under blue and clear films than any other phyto-selective materials. Some of arrowhead tubers could not emerged under green, red and yellow films. On the soil pH, the basal emergence was best at soil pH 6.0 and 7.0. At soil pH 4.0, 5.0 and 8.0, the required days for emergence were longer than those at soil pH 6.0 and 7.0 and some of tubers could not emerged. Arrowhead tubers more then 0.5 g showed the shorter shooting days than those less than 0.5 g. Tubers were able to emerge at water depth ranging from 0 to 20 cm. At 3 cm and 5 cm water depth, the required days for basal shooting were shorter than those at any other levels of water depth. The emergence of basals was best at molding depth of 0 cm, and the rates at 3 to 5 cm of plantation depth were decreased. At 10 cm plantation depth, the shooting rate was significantly decreased.

Effect of Altitude and Tuber Weight on the Growth and Yield of Pinellia ternata (Thunb.) Breit (해발고도별 반하 종구 무게가 생육 및 수량에 미치는 영향)

  • The plant growth and yield of Pinellia ternata (Thunb.) Breit. were studied by altitude and tuber weight. The emergence rates in low land area were not different by tuber weights, but it showed earlier emergence date in heavier weight of seed-tuber and low land area. The higher aerial growth such as plant height and number of leaves per plant was the heavier tuber weight in a planting year, but the growth was not different by the weight of tuber at second year after planting. The distribution pattern of tuber size per $m^2$ was not influenced by different seed-tuber weight. The number of harvested tuber was highest at more than 1 g of tuber weight, and followed 1~2 g and less than 2 g. The distribution pattern of fresh tuber yield was not influenced by different altitude and seed-tuber weight. The marketable tuber, 2 g or more, tends to be produced with more than 0.6 g seed-tuber. As the results above-mentioned, it was thought that the high yield was supposed to use seed-tuber over 0.6 g in the fertile soil.

Effects of Environment Friendly Agricultural Materials to Insect Parasitoids in the Laboratory (실내조건에서 친환경농자재가 기생성 천적곤충에 미치는 영향)

  • For the biological control of the greenhouse whitefly, aphids, American leaf-miners, Encarsia formosa, Aphidius coiemani, Diglyphus isaea, and Dacnusa sibirica were used as biological control factors. Otherwise, many kinds of environment friendly agricultural materials were also used in the kindly environment friendly farming. For testing the toxicity of environment friendly agricultural materials against to insect parasitoids as biological control factors, 61 environment friendly agricultural materials were selected by using methods and periods with insect parasitoids in the greenhouses. Environment friendly agricultural materials were sprayed to mummies and adults of E. formosa and A. colemani, and adults of Diglyphus isaea and Dacnusa sibirica in laboratory condition. Emergence rates were checked during 12 days after spray to mummies of E. formosa and A. colemani, and survival rates were at 24 and 48 hours after viral test for adult parasitoids, with 10% sugar solution. Emergence rates of E. formosa mummies were inhibited by sprayed the fungicidal environment friendly agricultural material (FEFAM) E and the environment friendly agricultural materials contained molybdenum (EFAMMo) G as 0.4 and 2.7%, respectively. E. formosa adults were not survived in vial for 48hours after sprayed and dried with the environment friendly agricultural materials contained plant extract (EFAMPE) J and the EFAMMo C. The environment friendly agricultural materials contained microelements (EFAMMEs) B and EFAMPE H, environment friendly agricultural materials contained useful soil microorganisms (EFAMSMs) H, FEFAM A and FEFAM D were recorded low survival rates of E. formosa adults with 6.7, 13.3, 13.3, 20.0 and 20.0, respectively. In case of the spraying environment friendly agricultural materials to mummies of A. colemani there were low emergence rate from mummies to adults as around 20s % with water spraying as control. A. colemani adults were not survived in vial with EFAMMo C. EFAMPE J, EFAMPE F, EFAMPE H, EFAMSM H were recorded low survival rates under 20% as 6.7, 13.3, 13.3 and 13.3, respectively, D. isaea and D. sibirica adults were not survived in vial with EFAMPE J. EFAMMo C was 53.3% of survival rates in 48 hours vial tests. D. isaea and D. sibirica were not affected by environment friendly agricultural materials compared with E. formosa and A. colemani.

Effect of Organic Soil Amendments on Establishment Vigor, Seedling Emergence, and Top Growth in Kentucky Bluegrass

  • Due to limited supplies and expensive importing costs, it is a goal to replace overseas peat with local soil amendments in turf industry of Korea. The study was initiated to compare the performances of five domestic and imported organic soil amendments (OSAs) on establishment characteristics and to provide basic information for root zone composition on sports turf design and construction. The study was conducted in Kentucky bluegrass (Poa pratensis L., KB) under greenhouse conditions from March to June in 2008. A total of 25 treatments of OSA + sand were prepared. These amendments were Berger Peat (OMA), Eco-Peat (OMB), G1-Soil (OMC), Premier Peat (OMD), and Supersoil I (OME). Significant differences were observed in establishment vigor, seedling emergence, and top growth. Results varied depending upon the type of OSAs and their rates in rootzone mixtures. OMA reached over 70% in establishment vigor in 5 WAS (weeks after seeding). OMC produced a maximum vigor of approximately 60% in 6 WAS. The OME amendment, however, showed poor performance lower than 30% in establishing KB turf until 8 WAS. There were considerable variations of top growth, being 3.8 to 14.5 cm. Greater differences in top growth resulted from OME mixtures. Shoot growth orientation in KB is also influenced by OSAs. In general, optimum mixing rate was considered as 10 to 20% for establishment vigor and 20 to 40% for top growth. Considering overall responses to establishment vigor, seedling emergence, and shoot growth, both local OMC and overseas OMD are considered as the useful soil amendments applicable for sports turfs. Domestic OME amendment would be applied for a low maintenance turfs such as rough and utility areas due to greater shoot growth. Information on these amendments would be of practical use for sports turf design and construction. Repeated experiments and field performance test are required to evaluate these OSA effect on other major turfgrass species and also to determine local OSA as imported peat substitute.

Collection and Evaluation of Korean Red Rices V. Germination Characteristics on Different Water and Soil Depth. (한국 재래 적미 수집 및 특성 검정 V. 수심 및 토심에 따른 발아 특성)

  • This experiment was carried out to evaluate the germination characteristics of Korean red rice seeds. Ten lines of the long grain and ten lines of the short grain Korean red rices were tested for dormancy, shoot emergence rate at different water and soil depth. Both the long and short grain type seeds of Korean red rice germinated over than 99% immediately after harvest, so that no dormancy was observed in the Korean red rices. Shoot emergence rate of the long grain red rices at 7cm of water depth and that of the short grain red rices at 11cm were 83.3% and 83.5% respectively, while those of the indica check Samgangbyeo and japonica check Seomjinbyeo were 0%. Shoot emergence rates of the long and short grain red rices at 7cm of soil depth were 71.1% and 73.9% respectively, while those of the check varieties Samgangbyeo and Seomjinbyeo were 40.0% and 28.0% respectively. Thus, the Korean red rices were expected to be useful germplasm source for direct seeding.

Seed Germination and Effect of Water Depths on Seedling Establishment of Iron-coated Rice Seeds (벼 철분코팅종자의 발아 특성 및 담수깊이에 따른 초기입모 영향)

  • Germination characteristics, seedling emergence, and early seedling growth of iron-coated rice seeds, cultivars Daebo and Samdeokbyeo, under different water depths were compared with those of non-coated seeds (control) and the results evaluated to obtain basic information for establishing stable seedlings in direct water seeding. The total germination percentage of the two seed treatments was similar, but iron-coated seeds had slightly faster germination and shorter mean germination time than non-coated seeds. Water absorption rates of iron-coated seeds were lower than that of non-coated seeds during seed germination. The germination percentage of the two iron-coated rice seed cultivars showed a significant decline of 15-22% after one year of storage under natural conditions. The seedling emergence percentage and uniformity of the two rice cultivars were significantly higher in the iron-coated seeds at 1-13 cm water depths but the percentage of floating seedlings was lower in iron-coated seeds than in non-coated seeds. The iron-coated seeds had a high seedling emergence percentage of 91.3-93.3% at all flooding depths whereas the non-coated seeds had a significantly low seedling emergence percentage of 57.7-71.7% at a water depth of 13 cm. Moreover, the shoot dry weight and seedling health score of iron-coated seeds were significantly higher than those of non-coated seeds, while root dry weights were similar in iron-coated and non-coated seeds, regardless of water depth. These results suggest that iron-coated seeds are more appropriate for stable seedling establishment in direct water seeding than are non-coated seeds.

Effects of Polymer Coating on Seed Vigour in Rice (벼 종자의 Polymer 피복처리가 종자세에 미치는 영향)

  • These experiments were conducted to evaluate the effects of seed coating with ten environmentally acceptable polymers, on germination percentage, water uptake, respiration, emergence and seedling growth characteristics. The water absorption of polymer-coated seeds in saturation condition was highest in klucel and lowest in polyvinyl pyrrolidone(PVP), and that in water was highest in klucel and lowest in maltrin. Respiration rates of polymer-coated seeds in Hwayoungbyeo and Ilpumbyeo were lower than those of none-coated seeds, and those in Daecheongbyeo and Jinmibyeo were higher than that of none-coated seeds, and those of sepiret coated seeds were higher than PVP coated seeds. The germination polimer-coated seed was reduced by one percentage by coating with seed coating machine. Germination percentage was not affected by any of polymer coating in high quality seed, but there were significant effects in low quality seed coating with waterlock, surelease 46 and sepiret significantly reduced germination some cultivars. Germination percentage after accelated ageing treatment were slightly higher most of polymer-coated seeds than in none-coated seeds, but those of sepiret-and klucel-coated seeds were lower significantly. Germination percentage of seeds coated with daran 8600, rnaltrin, sacrust and opadry were enhanced slightly under cold test other polimers reduced germination. The seedling height of polymer-coated seeds were longer than those of none-coated seeds, but those of waterlock, PVP and maltrin coating seeds were shorter, seedling hight was shortened by polimer coating under cold test. Polymer-coated seeds showed higher emergence percentage, shorter emergence time in field condition. The highest emergence percentage and the shortest emergence time was shown at 90% soil moisture content.

Seedling - Emergence of Rice, Weedy Rice, and Echinochloa species Sown before Wintering and in the Early Spring (월동전(越冬前) 초춘(初春)에 파종(播種)한 재배(栽培)벼, 잡초성(雜草性)벼 및 피의 출현특성(出現特性))

  • In recent years dry direct-seeding of rice has been encouraged by the government and increasingly practiced by farmers in Korea. This has been bringing up an increased occurrence of weedy rites. Some farmers in the southern region dare to sow the rice before winter after harvest, while most farmers wish to sow as early as possible in the spring to secure the growing period, and to disperse the intensive labor in early May. The purpose of this study was to determine the feasibility of moving the sowing of rice to an earlier date under dry direct-seeding, and to elucidate the nature of emergence of an old strain of rice, weedy rites, and barnyardgrasses tinder this farming practice and their adaptive competence over present cultivate. The presently recommended rice cultivar, Dongjinbyo and an old rice strain, Dadajo which prevailed in early 1900s, almost could not emerge from soil deeper than 6cm and could emerge to only 5.3% at best from 1cm deep loamy soil field when the seeds were sown on Nov. 28. However, two strains of weedy rites being weedy for over 200 years emerged by 17.0 to 63.0% from the loamy and sandy clay loam field 1 to 6cm deep. Emergence of the weedy rites was greater in the loamy soil and at a shallow depth, and negligible from the soil depth of 9cm. Barnyardgrasses sown on Nov. 28 emerged by 13.4 to 51 % from the 1 to 3cm deep loamy soil, and 8.6 to 46.7% from the 1cm deep sandy clay loam. Echinochloa crus-galli var. crus-galli emerged more than var. praticola, and var. oryzicola least. Most of the non-emerged barnyardgrasses seem to have entered secondary dormancy. Seeding rice a month earlier than the season lowered the emergence of Dongjinbyo by ca. 10, 18, and 26%, respectively at 1, 3, and 6cm soil depths, indicating that moving the seeding date a month earlier is impractical. The old strain, Dadajo sown in the soil at a depth of 6cm responded similarly. However, the strain has shown a significantly higher ability in emergence from 9cm deep soil. Weedy rices sown a month earlier A month earlier sown weedy rices have shown very similar emergence rates at various soil depths to those sown on May 1. Barnyardgrasses have also shown similar emergence rates when sown between April 3 and May 1. Like barnyardgrasses, the old strain and weedy rices apparantly posessed a greater adaptability to emerge under lower temperatures, and from deeper soil ; Dongjinbyo${\leq}$ Echinochloa species in that order. However, emergence- speed under lower temperature(sown on April 3) was faster in the order of weedy rice
