The ICR pregnant mice were irradiated at 1.5Gy in every 6 hours in the period of organogenesis in order to classify the stage specificity of the embryonic effects of radiation and the stage of development differentiation of the primordium of each major organ. Intrauterine death, fetal body weight and external malformation in live fetuses were observed on day 18 of gestation. There was no statistically significant difference in the intrauterine mortality at any stage organogenesis. The fetal body weight of the mice irradiated in the intermediate stage of organogenesis showed significantly lower. There were specific highly sensitive stages in the incidences of each external malformation, that is exencephalia, open eyelid, cleft palate, anomalies of extremities and anomalies of the tail. At these stage, the primordia of the major organs are established in ICR mice.
Embryotoxicity tests were performed in Daphnia magna to assess aquatic ecotoxicity of bisphenol A, a well known industrial compound showing estrogen-like activity in vivo, and to examine their effectiveness in the toxicological assessment. The whole embryonic developmental period was classified into 6 stages and developmental abnormality was checked to evaluate the embryotoxicity. In the present study, bisphenol A showed the ability to interfere with embryonic development, suggesting its antiecdysteroidal activity. The rates of mortality, delayed development, deformity and immobility all showed good concentration-response relationship, demonstrating their possibility as useful toxicological indices in daphnid embryotoxicity tests that have been rarely performed so far. It seemed favorable to the test sensitivity that embryos are removed from maternal daphnids around 7 hr after deposition from the ovaries to the brood chamber. These results suggest that daphnid embryotoxicity tests can be one of useful tools available for the assessment of ecotoxicity of various chemicals in the aquatic environment.
This study investigated the effects of breed, laying age, and egg storage period on hatching performance. Eggs were collected from early laying period (27 to 29 weeks old) and late laying period (50 to 52 weeks old) in Hwanggalsaek Jaeraejong, Korean Rhode Island Red, and Korean White Leghorn chicken breeds. Eggs laid for each period were stored for 3, 7 and 14 days and then hatched. Fertility, hatchability, and embryonic mortality were observed to determine the hatching performance. The results showed that hatching performance was affected by breed, laying age, and egg storage period with independently or interactively. The Korean Rhode Island Red breed had the lowest hatching performance. Hatchability and early embryonic mortality rate increased in early laying period eggs compared to the late laying period, but the late embryonic mortality rate increased in late laying period eggs. Additionally, hatching performance decreased as the egg storage period increased. Using breed-specific hatching management techniques, avoiding late laying period eggs, and shortening the egg storage period to seven days after laying may improve the hatching performance.
The effect of adding transparent fluid (TF) to fowl semen on fertilizing capacity of fowl spermatozoa and on hatchability was studied. Diluted semen and semen containing 15% TF were stored for 24 h at $3-5^{\circ}C$ and inseminated at weekly basis for 5 consecutive weeks. No significant differences were observed in fertility, hatchability and embryonic mortality among the treatments. The results suggest that TF is not necessarily detrimental to fowl spermatozoa even when mixed with semen and stored outside the body.
Objectives: The present study investigated the potential adverse effects of multi-wall carbon nanotubes (MWCNTs) on pregnant dams and embryonic development following maternal exposure in rats. Methods: MWCNTs were orally administered to pregnant rats from gestational day (GD) 6 through 19 at dose levels of 0, 8, 40, 200, and 1000 mg/kg/day. During the test period, clinical signs, mortality, body weights, food consumption, serum biochemistry, oxidant-antioxidant status, gross findings, organ weights, and Caesarean section findings were examined. Results: All animals survived to the end of the study. A decrease in thymus weight was observed in the highest dose group. However, maternal body weight, food consumption, serum biochemical parameters, and oxidant-antioxidant balance in the kidneys were not affected by treatment with MWCNTs. No treatment-related differences in gestational index, embryo-fetal mortality, or fetal and placental weights were observed between treated and control groups. Conclusions: The results show that 14-day repeated oral dosing of MWCNTs during pregnancy induces minimal maternal toxicity at 1000 mg/kg/day in rats. Under these experimental conditions, the no-observed-adverse-effect level of MWCNTs is considered to be 200 mg/kg/day for dams and 1000 mg/kg/day for embryonic development.
Objectives: Advancements in nanotechnology have led to nanoparticle (NP) use in various fields of medicine. Although the potential of NPs is promising, the lack of documented evidence on the toxicological effects of NPs is concerning. A few studies have documented that homeopathy uses NPs. Unfortunately, very few sound scientific studies have explored the toxic effects of homeopathic drugs. Citing this lack of high-quality scientific evidence, regulatory agencies have been reluctant to endorse homeopathic treatment as an alternative or adjunct treatment. This study aimed to enhance our insight into the impact of commercially-available homeopathic drugs, to study the presence of NPs in those drugs and any deleterious effects they might have, and to determine the distribution pattern of NPs in zebrafish embryos (Danio rerio). Methods: Homeopathic dilutions were studied using high-resolution transmission electron microscopy with selected area electron diffraction (SAED). For the toxicity assessment on Zebrafish, embryos were exposed to a test solution from 4 - 6 hours post-fertilization, and embryos/larvae were assessed up to 5 days post-fertilization (dpf ) for viability and morphology. Toxicity was recorded in terms of mortality, hatching delay, phenotypic defects and metal accumulation. Around 5 dpf was found to be the optimum developmental stage for evaluation. Results: The present study aimed to conclusively prove the presence of NPs in all high dilutions of homeopathic drugs. Embryonic zebrafish were exposed to three homeopathic drugs with two potencies (30CH, 200CH) during early embryogenesis. The resulting morphological and cellular responses were observed. Exposure to these potencies produced no visibly significant malformations, pericardial edema, and mortality and no necrotic and apoptotic cellular death. Conclusion: Our findings clearly demonstrate that no toxic effects were observed for these three homeopathic drugs at the potencies and exposure times used in this study. The embryonic zebrafish model is recommended as a well-established method for rapidly assessing the toxicity of homeopathic drugs.
In this experiment, we investigated the toxicity of tebuconazole (fungicide) using domestic frog embryos, along the FETAX (Frog Embryo Teratogenesis Assay-Xenopus) protocol. Bufo gargarizans, Hyla japonica, and Pelophylax nigromaculatus embryos were incubated, and investigation of the tebuconazole effect was performed by the probit analysis. As a result, depending on the concentrations of tebuconazole, the mortality and malformation rates were increased and larval body length was decreased. The teratogenic concentrations (EC50) of tebuconazole were 34.4mg L-1, 10.6mg L-1, and 14.9mg L-1, respectively, and the embryo lethal concentrations(LC50) of tebuconazole were 74.7 mg L-1, 38.5 mg L-1, and 39.1 mg L-1, respectively. The teratogenic index (TI) valuesof tebuconazole were 2.19, 3.58, and 2.65; thus, it showed teratogenicity in embryonic development of these three frogs. These results revealed that in this experiment, tebuconazole suppressed the development of embryos at a relatively low concentration. In addition, mortality, malformation ratios, malformation patterns, and growth rates were similar to the results from the other assay systems. Therefore, tebuconazole was thought to have an effect on the embryo development of domestic frogs. In future, it will be necessary to identify species specificity in order to the clarify the causes of differences in mortality, malformation rate, and malformation patterns depending on the species.
Effects on nonylphenol (NP) treatment on early development of fertilized eggs and survival of larvae in olive flounder, Paralichthys olivaceus were investigated. Fertilized eggs and hatched larvae were exposed to aqueous solutions of NP at nominal concentrations of 50, 100, 150 and 200 $\mu\textrm{g}$/L. In the control I and II (methanol-carrier control) groups, the hatchability of fertilized eggs were 90.7$\pm$7.02 and 90.0$\pm$5.29% (P <0.05), respectively. However, treatments of NP concentrations at 50, 100, 150 and 200 ug/L were reduced to 78.7$\pm$4.16, 46.0$\pm$9.17, 48.0$\pm$3.46 and 33.3$\pm$11.02% (P<0.05), respectively. The time to hatching of fertilized eggs was delayed in high-dose NP treatment groups rather than control groups. Fertilized eggs of NP immersed group could not hatch normally, and also vertebra of the larvae observed as abnormal shape. Cumulative mortality of hatched larvae after 26hr NP treatment was 38.3, 78.3, 88.3 and 100% in NP 50, 100, 150 and 200 ug/L treatment groups, respectively. Whereas, control I and II were 11.7, 16.7% (P<0.05). Hatched larvae showed most death at the embryonic development stage with abnormalities. These results suggest that NP treatment in fertilized eggs and larvae was inhibited as normally development for fertilized egg and growing of larvae.
The combined effect of radiation and ultrasound has been studied in mouse embryos. Radiation and/or ultrasound were adminstered to ICR mice on day 8 of gestation. Intrauterine death, gross malformation, and fetal body weight were selected as indicators of effects. Does of whole-body ${\gamma}-irradiation$ were 0.5 to 2.5 Gy and those of ultrasound were $0.5\;W/cm^2$ to $3\;W/cm^2$. Intrautrine mortality increased with increasing radiation dose ; this trend was more remarkable in combination with ultrasound. Gross malformations such as exencephaly and anophthalmia/microphthalmia appeared frequently in the fetuses treated with both radiation and ultrasound. Decreased fetal weight was observed even in mice treated with 1.5 Gy of radiation or $1\;W/cm^2$ of ultrasound. There was a linear relationship between dose and reduction of fetal weight. The fetal weight was sensitive, precise and easy-to-handle indicator for the effects of growth retardation. Intrauterine mortality and frequencies of exencephaly and anophthalmia/microphthalmia were higher than the sum of those induced by radiation and by ultrasound. The results indicatied that the combined action of radiation and ultrasound on intrauterine death and malformations was synergistic.
As a result of intensive breeding, litter size has considerably increased in pig production over the last three decades. This has resulted in an increase in farrowing complications. Prolonged farrowing will shorten the window for suckling colostrum and reduce the chances for high-quality colostrum intake. Studies also agree that increasing litter sizes concomitantly resulted in decreased piglet birth weight and increased within-litter birth weight variations. Birth weight, however, is one of the critical factors affecting the prognosis of colostrum intake, and piglet growth, welfare, and survival. Litters of uneven birth weight distribution will suffer and lead to increased piglet mortality before weaning. The proper management is key to handle the situation. Feeding strategies before farrowing, management routines during parturition (e.g., drying and moving piglets to the udder and cross-fostering) and feeding an energy source to piglets after birth may be beneficial management tools with large litters. Insulin-like growth factor 1 (IGF-1)-driven recovery from energy losses during lactation appears critical for supporting follicle development, the viability of oocytes and embryos, and, eventually, litter uniformity. This paper explores certain management routines for neonatal piglets that can lead to the optimization of their colostrum intake and thereby their survival in large litters. In addition, this paper reviews the evidence concerning nutritional factors, particularly lactation feeding that may reduce the loss of sow body reserves, affecting the growth of the next oocyte generation. In conclusion, decreasing birth weight and compromised immunity are subjects warranting investigation in the search for novel management tools. Furthermore, to increase litter uniformity, more focus should be placed on nutritional factors that affect IGF-1-driven follicle development before ovulation.
