Successful Superovulation and Recovery of Embryos in Repeat Breeding Crossbred Cows

  • The present study was undertaken to find out possibilities for superovulation and recovery of freezable/transferable embryo from repeat breeding crossbred cows. For this study a total of 10 Holstein crossbred cows having history of failure of conception even after more than 4~5inseminations were taken and superovulated using Folltropin-V at 100 to 140% dose schedule. The results indicated that out of 10 donors, 8 responded to superovulatory treatment and yielding a total of 94 embryos, out of which 45 were of transferable/freezable quality. The mean ovulations and mean transferable embryos were 11.5$\pm$2.91 and 4.5$\pm$1.66, respectively. These observations suggests that the repeat breeding cows having conception failure after several artificial inseminations can be used as donors for production of embryos and calves through embryo transfer technology.

  • Over the years, the embryo transfer industry has grown from the simple collection & transfer of embryos into an advanced field of embryo biotechnology. Currently a large demand exists for bovine oocytes and early embryos in both research and commercial settings. Bovine embryos can now be produced in-vitro. Primary oocytes collected from antral follicles of abattoir - obtained ovaries can be induced to undergo the maturation process. In-vitor maturation system, however must ensure that the resulting oocyte is capable of undergoing normal fertilization and yields a zygote competent of developing to term after embryo transfer. Sperm preparation for IVF has improved with the use of heparine. The use of co-culture system has proved beneficial in circumventing the developmental block in IVM/IVF bovine embryos.

  • Background: The collection of ovaries from slaughterhouses is an important source of oocytes for in vitro embryo production. On the other hand, the physiological stage of slaughtered females varies and influences embryo production. Objectives: The study examined the in vitro efficiency of embryos and demi-embryos from young, non-pregnant adult, and pregnant adult ewes from a local slaughterhouse. Methods: One thousand three hundred ovaries were collected from August to October 2020. The recovered oocytes were matured, fertilized, and cultured at 5% CO2, 38.5℃, and 100% humidity. Embryo bisection was performed in 96 blastocysts (n = 32 per treatment). The demiembryo pairs were incubated for their reconstitution for 12 h. SAS was used for data analysis. Results: The number of oocytes collected from the experimental group of non-pregnant adult ewes was higher (p ≤ 0.007) than those collected from the group of pregnant adult ewes (2.67 ± 0.19 vs. 2.18 ± 0.15 oocytes/group, respectively). The blastocyst rate was higher (p ≤ 0.0001) in the non-pregnant adult group (36.39%) than in the young (17.96%). The ratio of demi-embryos that recovered the blastocoelic cavity was higher (p < 0.05) in the young group (81.25%) than in the pregnant adult group (59.38%). The diameter of the demi-embryos was higher (p < 0.05) in the non-pregnant adult group (186.54 ± 8.70 ㎛) than those in the young and pregnant adult groups. Conclusions: In conclusion, the in vitro embryo production efficiency was highest when using oocytes from non-pregnant adult ewes under the conditions of this study.

Effect of Follicle Stimulating Hormone Dosage on the Production and Pregnancy of Embryos Derived from Korean Brindle Cows (난포자극호르몬(Follicle Stimulating Hormone) 투여 농도가 칡소 수정란의 생산과 이식에 미치는 영향)

  • Korean native cattles (Han woo) are divided yellow, tiger(blinder), and black by coat color. Chikso has tiger coat color, which cultured about 1,700 heads within Korea. However there were a few researches reported about preservation and production. This study was conducted to investigated that the effect of follicle stimulating hormone (FSH) dosage(240 mg and 280 mg) on the embryo production, pregnancy and calf birth weights. There were no difference total embryo and transferable embryo between FSH 240 mg or 280 mg treatment, but pregnancy rate was increased 46% in embryos derived from 280 mg group than 240 mg group's 29.6%. The birth weight of Chikso calves was similar between artificial insemination(25.6 kg) and embryo transfer(24.8 kg) groups.

Antioxidants as alleviating agents of in-vitro embryo production oxidative stress

  • Despite numerous advances in in-vitro embryo production (IVP), many documented factors have been shown to influence the development of mammalian preimplantation embryos and the success of IVP. In this sense, elevated levels of reactive oxygen species (ROS) correlate with poor outcomes in assisted reproductive technologies (ART) due to oxidative stress (OS), which results from an imbalance between ROS production and neutralization. Indeed, excessive production of ROS compromises the structural and functional integrity of gametes and embryos both in vivo and in vitro. In particular, OS damages proteins, lipids, and DNA and accelerates cell apoptosis. Several in-vivo and in-vitro studies report an improvement in qualityrelevant parameters after the use of various antioxidants. In this review, we focus on OS and the source of free radicals and their effects on oocytes, sperm, and the embryo during IVP. In addition, antioxidants and their important role in IVP, supplementation during oocyte in vitro maturation (IVM), in vitro culture (IVC), and semen extenders were discussed. Nevertheless, various methods for determining the level of ROS in germ cells have been briefly described. Still, it is crucial to develop standardized antioxidant supplement systems to improve overall IVP success. Further studies should explore the safety, efficacy, mechanism of action, and combination of different antioxidants to improve IVP outcomes.

Efficiency of In Vivo Embryo Production following Superovulation with Sex-soted Semen in Hanwoo (Korean Native Cattle) (과배란 처리에 있어 성감별 정액을 이용한 한우 체내 수정란의 생산 효율)

  • Sexed semen is commonly used for the production of calves of the desired gender. Gender selection is important in animal production industries. For example, female cattle are required for the dairy industry while males are preferred in the beef cattle industry. The present study was to assess the in vivo embryo production efficiency using the semen separated according to sex during superovulation in Hanwoo. Seventy Hanwoo donor cows were flushed on day 7 of estrus cycle with same FSH and artificial insemination by the same technicians. Embryos were recovered on 7 days after the third insemination by flushing the uterus with embryo collection medium. KPN semen straws used artificial insemination contained 20 million sperm (total number 60 million per donor). Sex-sorted semen straws contained 4 million sperm (total number 12 million per donor). The results obtained were as follows: No differences were observed in the efficiency of superovulation rates on KPN semen 87%, and sexed semen 100%, respectively. The mean numbers of total embryos are each $12.58{\pm}8.31$ and $13.25{\pm}7.86$. The mean numbers of transferable embryos, sexed semen were significantly lower than KPN semen ($3.75{\pm}1.98$ vs. $8.23{\pm}6.07$, P<0.05). The rates of unfertilized embryos from superovulation using sexed semen were significantly higher than KPN semen (50% vs. 15%, P<0.05). The rate of degenerated 2-cell embryos from sexed and KPN semen was 60.87% and 11.11%, respectively (p<0.05). In conclusion, these results indicate that superovulation using sexed semen was useful, but efficient embryo production was important to reducing the damage caused by the Flowcytometer-based sperm sorting procedure.

Effect of Gonadotropin Treatments on Ovarian Response, Ovulation and Embryo Production in Rabbits (토끼에서 선성자극호르몬 처리법에 따른 난소 반응, 배란 및 수정란 생산효율에 관한 연구)

  • This experiment was carried out to investigate the factors affecting superovulation in rabbits and to determine the effect of pFSH and PMSG on ovarian superovulatory responses and embryo production, and the effect of superovulation treatment with a single injection of pFSH dissolved in polyvinylpyrrolidone on the ovarian responses and the embryo quality. The results obtained were suonmerized as follows: Superovulatory response resulted in significantly (P<0.05) higher ovulation rates and more embryos in spring or autumn, compared with summer or winter. Repeated superovulatory treatments with PMSG leaded to a significantly(P<0.05) decreased number of total follicles and recovered ova. Superovulation with pFSH resulted in the higher number of ovulated follicles and recovered ova than with PMSG. A single subcutaneous injection of pFSH dissolved in 25% PVP resulted in the more ovulation points(33.2) and recovered embryos(30.2), which were comparable to the multiple injections of pFSH(44.8 vs 37.7).These results indicated that the treatment with a single injection of FSH dissolved in PVP was an efficient and simple alternative method to the conventional multiple FSH injections for superovulation in rabbits.

MIGRATION OF THE PRIMORDIAL GERM CELLS AND GONAD FORMATION IN THE EARLY CHICKEN EMBRYO

  • In this study, characteristics of chick primordial germ cells (PGCs), which is the founder cell of the germline, and gonadal development of the chick embryo between 12hrs and 6 day of incubation were investigated by transverse serial sections of chick embryos under the light microscopic observation. In embryo stage 20 (3 day of incubation), there are a lot of PGCs at the mesenchym, which were moving to the thickened epithelium (gonadal ridge). The PGCs arrive at both right and left gonad primordial in equal number prior to stage 24 (4 day of incubation), but in the following stages, the distribution of the PGCs became asymmetrical. More PGCs colonized the left than the right gonad, but the reason for the unequal distribution of PGCs is uncertain. The PGCs have mostly settled in the gonadal ridge (GR) at 6 day embryo. This study was conducted to investigate characteristics of the PGC migration and gonadal formation and observe the best condition for PGC isolation, culture and to attempt the possibility of the production for transgenic germline chimeras with manipulated PGCs.

Factors Affecting on Production of Dairy Cattle Embryos (젖소 수정란의 생산에 영향을 미치는 요인에 관한 연구)

  • The present study was carried out to investigate effects of gonadotropin, age of donor, day of estrus cycle gonadotropin injection started and season on embryo production after superovulation in dairy cattles. Embryo collection records were obtained from 177 embryo donor collections from 98 Holstein cows aged from 3 to 9 years during 4 years(1993~1996) at National Livestock Re-search Institute. Superovulation was induced by injections of 3 gonadotropins(FSH-P, FOLLTROPHIN-v* or SUPER-OV*) beginning on days 9 to 14 of the estrus cycle. Em-bryos were collected from donors using a nonsurgical technique on days 6 to 8 after insemi-nation. The results were as follows ;Number of total and freezable embryos per donor cow was affected by gonadotropin(P<0.01). The more number of total and freezable embryos wereobtained by use of FOLLTROPHIN-V (13.2, 7.4) or FSH-P (11.0, 5.7) than SUPER-OV* (5.0, 2.4). Age of donor, the day that gonadotropin was started or season didn't affect total or freezable embryos(P >0.05).

Abnormal Seedlings Emerged during Embryo Rescue and Its Remedy for Seedless Grape Breeding

  • The abnormal seedlings, a common physiological anomalies, emerged during embryo rescue severely restricted grape breeding. To enhance the efficiency of the seedless grape breeding by reducing the production of abnormal seedlings in the course of embryo rescue, we investigated the effects of genotype, media type, embryo style, pre-chilling on the deformity rate of the abnormal seedlings during embryo rescue. The abnormal seedlings were firstly classified into seven categories based on their morphology. Our results indicated that the emergence of abnormal seedlings was highly dependent on the female parent genotype. Polyembryony was advantageous to diminish the number of abnormal plantlets and the germination rate of embryo was 100%. We also found that pre-chilling treatment could reduce the number of abnormal plantlets and promote the embryo germination. The abnormal plantlets were reduced significantly by the addition of $ZnSO_4$ $10{\mu}mol{\cdot}L^{-1}$ or mashed-banana $500mg{\cdot}L^{-1}$ to either embryo development or germination media. Transferring the abnormal seedlings onto the suitable fresh media in 4 weeks after embryo germination provided an effective way to transform them into normal seedlings.
