New antitumor active polymers, poly(methacryloyl-2-oxy-1,2,3-propanetricarboxylic acid-co-exo-3,6-epoxy-l,2,3,6-tetrahydrophthalic acid) [poly(MTCA-co-ETAc)], poly(methacryloyl-2-oxy-l,2,3-propanetricarboxylic acid-co-hydrogen ethyl-exo-3,6-epoxy-l,2,3,6-tetrahydrophthalate) [poly(MTCA-co-HEET)], and poly(methacryloyl-2-oxy-l,2,3-propanetricarboxylic acid-co-a-ethoxy-exo-3,6-epoxy-1,2,3,6-tetrahydrophthaloyl-5-fluorouracil) [poly(MTCA-co-EETFU)] were synthesized and characterized. Their antitumor activity, inhibition of DNA replication and antiangiogenesis were examined. The structures of the polymers were identified by FT-IR, $^1H$ and $^{13}C$-NMR spectroscopy. The number average molecular weights of the fractionated polymers determined by GPC ranged from 9,400 to 14,900, and polydispersity indices were less than 1.7. The in vitro cytotoxicity of these polymers was determined and their antitumor activity was evaluated. The $IC_{50}$ values (the drug concentration at inhibition of 50% tumor growth) indicated that the synthesized polymers were much better inhibitors of cancer cells and showed lower cytotoxicity than the free 5-FU. The in vivo antitumor activity of the conjugates was examined using mice bearing the sarcoma 180 tumor cell line. The life spans (TIC) of the mice treated with the conjugates were higher than those treated with the free 5-FU. In addition, the synthesized conjugates showed excellent antiangiogenic activity based on an embryo chorioallantoic membrane assay.
The hormonal regulation of organ differentiation was investigated in the tissue culture of sweet potato. Embryogenic callus was induced from shoot tips cultured on MS medium supplemented with 1 mg/L 2,4-D. When embryogenic callus was transferred to medium containing 0.1 mg/L GA$_4$, it proliferation was stimulated. The callus gave rise to plantlets when cultured on medium containing 0.1 mg/L BA. Addition of 0.1 mg/L jasmonic acid or 0.01 mg/L brassinolide to medium was effective for the development of healthy normal plantlets.

Plants have evolved physiological, biochemical and metabolic mechanisms to increase their survival under the adverse conditions. This present study has been performed to select salt-tolerant rice mutant lines through in vivo and in vitro mutagenesis with gamma-rays. For the selection of the salt-tolerant rice mutants, we conducted three times of selection procedure using 1,500 gamma ray mutant lines resulted from an embryo culture of the original rice cv. Dongan (wild-type, WT): first, selection in the a nutrient solution with 171 mM NaCl; second, selection under in vitro condition with 171 mM NaCl; and third, selection in a reclaimed saline land. Based on a growth comparison of the entries, out of the mutant lines, two putative 2 salt tolerant (ST) rice mutant lines, ST-87 and ST-301, were finally selected. The survival rate of the WT, ST-87 and ST-301 were 36.6%, 60% and 66.3% after 7 days in 171 mM NaCl treatment, respectively. The WT and two salt tolerant mutant lines were used to analyze their genetic variations. A total of 21 EcoRI and Msel primer combinations were used to analyze the genetic relationship of among the two salt-tolerant lines and the WT using the ABI3130 capillary electrophoresis system. In the AFLP analysis, a total of 1469 bands were produced by the 21 primer combinations, and 700 (47.6%) of them were identified as having polymorphism. The genetic similarity coefficients were ranged from 0.52 between the ST-87 and WT to 0.24 between the ST-301 and the WT. These rice mutant lines will be used as a control plot for physiological analysis and genetic research on salt tolerance.
This study investigates the mechanism of MnS precipitation on $Al_2O_3-SiO_2$ inclusions during the solidification of non-oriented silicon steel, especially the influence of the phase structures and sizes of the oxides on the MnS precipitation, by scanning electron microscopy and transmission electron microscopy coupled with energy dispersive spectrometry. The investigation results show that MnS tends to nucleate on submicron-sized $Al_2O_3-SiO_2$ inclusions formed by interdendritic segregation and that it covers the oxides completely. In addition, MnS can precipitate on micron-sized oxides and its precipitation behavior is governed by the phase structure of the oxides. The MnS embryo formed in a MnO-containing oxide can act as a substrate for MnS precipitation, thus permitting further growth via diffusion of solute atoms from the matrix. MnS also precipitates in a MnO-free oxide by the heterogeneous nucleation mechanism. Furthermore, MnS is less prone to precipitation in the $Al_2O_3$-rich regions of the $Al_2O_3-SiO_2$ inclusions; this can be explained by the high lattice disregistry between MnS and $Al_2O_3$.

Objectives: This study was performed to investigate the effects of pregnancy-related four herbal medicines (Samul-tang, Onkyung-tang, Chokyungjongok-tang and Moutan Radicis Cortex) on the endometrial and placental cells. Methods: In this study, we examined viability and decidualization of telomerase immortalized human endometrial stromal cell lines (T-HESCs) and viability and invasion ability of human first trimester trophoblast cell lines Sw.71 by four herbal medicines (Samul-tang, Onkyung-tang, Chokyungjongok-tang and Moutan Radicis Cortex) Results: In the study, we showed that Samul-tang, Onkyung-tang, Chokyungjongok-tang increased decidualization marker prolactin (PRL) and insulin-like growth factor-binding protein 1 (IGFBP1) in T-HESCs. Moutan Radicis Cortex decreased the mRNA level of PRL and IGFBP1, and the protein level of PRL and IGFBP1 had no significant effect. Moreover, four herbal medicines reduced invasion ability of Sw.71 cells. Conclusions: These results suggest that Samul-tang, Onkyung-tang, and Chokyungjongok-tang have beneficial effects on successful embryo implantation and pregnancy maintenance by increasing decidualization markers such as PRL and IGFBP1. Moutan Radicis Cortex reduces the mRNA levels of PRL and IGFBP1, which may adversely affect pregnancy. Further investigations are needed to elucidate the significance of the decreased invasive ability of Sw.71 cells induced by four herbal medications.

The 1,081 soybean genotypes including 972 black soybeans and 109 other colored soybeans were collected in 197 locations from January to April, 1991. Seed chemical components of soybean collections in relation to growth characters were evaluated to survey germplasms for black soybean breeding for seed quality. Seeds of 1,081 genotypes collected were analyzed for crude protein, crude lipid, and total sugar contents. The crude protein content was averaged to 39.8% and ranged from 34.1% to 48.0%. The average crude lipid and total sugar contents were 20.1% and 10.1%, and the ranges of those were 14.1% to 23.8% and 8.3% to 12.1%, respectively. Higher crude protein content was shown in early soybean maturity group, whereas higher total sugar content was in late group. Black soybean collections with green seed embryo tended to be higher in total sugar content than those with yellow one. Black soybean oil is generally composed of about 22% oleic, 55% linoleic, 7% linolenic acid, and 16% the others.

Pectin methylesterase (PME) plays an important role in vegetative and reproductive development and biotic/abiotic stress responses by regulating the degree of methyl-esterification of pectic polysaccharides in the plant cell wall. PMEs are encoded by a large multigene family in higher land plant genomes. In general, the expression of plant PME genes shows tissue- or cell-specific patterns and is induced by endogenous and exogenous stimuli. In this study, we identified PME multigene family members (CsPMEs) from the sweet orange genome and report detailed molecular characterization and expression profiling in different citrus tissues and two fruit developmental stages. We also discussed the possible functional roles of some CsPME genes by comparing them with the known functions of PMEs from other plant species. We identified 48 CsPME genes from the citrus genome. A phylogenetic tree analysis revealed that the identified CsPMEs were divided into two groups/types. Some CsPMEs showed very close phylogenetic relationships with the PMEs whose functions were formerly addressed in Arabidopsis, tomato, and maize. Expression profiling showed that some CsPME genes are highly or specifically expressed in the leaf, root, flower, or fruit. Based on the phylogenetic relationships and gene expression profiling results, we suggest that some CsPMEs could play functional roles in pollen development, pollen tube growth, cross incompatibility, root development, embryo/seed development, stomata movement, and biotic/abiotic stress responses. Our results shed light on the biological roles of individual CsPME isoforms and contribute to the search for genetic variations in citrus genetic resources.
Objective: This study aimed to determine the protective efficacy of Buddha's Temple (BT) extract against tert-butyl hydroperoxide (t-BHP)-induced oxidative stress in Gallus gallus chicken embryo fibroblast cell line (DF-1) and its effects on the cell lipid metabolism. Methods: In this experimental study, Gallus gallus DF-1 fibroblast cells were pretreated with BT 10-7 for 24 hours, followed by their six-hour exposure to t-BHP (100 μM). Water-soluble tetrazolium salt-8 (WST-8) assays were performed, and the growth curve was computed. The intracellular gene expression changes caused by BT extract were confirmed through quantitative polymerase chain reaction (qPCR). Flow cytometry, oil red O staining experiment, and thin-layer chromatography were performed for the detection of intracellular metabolic mechanism changes. Results: The WST-8 assay results showed that the BT pretreatment of Gallus gallus DF-1 fibroblast cell increased their cell survival rate by 1.08%±0.04%, decreased the reactive oxygen species (ROS) level by 0.93%±0.12% even after exposure to oxidants, and stabilized mitochondrial activity by 1.37%±0.36%. In addition, qPCR results confirmed that the gene expression levels of tumor necrosis factor α (TNFα), TIR domain-containing adapter inducing IFN-beta (TICAM1), and glucose-regulated protein 78 (GRP78) were regulated, which contributed to cell stabilization. Thin-layer chromatography and oil red O analyses showed a clear decrease in the contents of lipid metabolites such as triacylglycerol and free fatty acids. Conclusion: In this study, we confirmed that the examined BT extract exerted selective protective effects on Gallus gallus DF-1 fibroblast cells against cell damage caused by t-BHP, which is a strong oxidative inducer. Furthermore, we established that this extract significantly reduced the intracellular ROS accumulation due to oxidative stress, which contributes to an increase in poultry production and higher incomes.
Seed germination test done in laboratory does not coincide with field emergence in general. The experiments were carried out to examine the effect of priming and $GA_3$, treatment to seeds of Platycodon grandiflorum; Codonopsis lanceolata and C. pilosula on lapsed time to first seedling emergence, seedling emergence, morphological characters and growth and the cause of poor emergence of C. pilosula. No-treatment as Control (water), priming or $GA_3$ treatment was done with only distilled water for 2 days, $CA(NO_3)_2$ 150 mM for 2 days or $GA_3$ 0.1 mM for 3 days, respectively. Seedling emergence rate was counted every 2 days but morphological characters and dry weight of shoot and root were measured on 38 days after sowing. Their internal seed structures were examined with Scanning Electron Microscope. C. pilosula had poorer seedling emergence rate than P grandiflorum and C. lanceolata showing nearly same rate: Compared to the other treatment (s) P. grandiflorum displayed higher rate in priming and $GA_3$, treatments but C. lanceolata or C. pilosula did the greatest rate in only $GA_3$ or priming treatment, respectively. $GA_3$ treatment to seeds of P. grandiflorum and C. lanceolata shortened the lapsed time to seedling emergence in comparison with Control, 2-days water imbibition before sowing. In all the species plant height and number of leaves per seedling became shorter and less in priming treatment than the other treatments except plant height of C. Pilosula while their hypocotyl length was nearly same in all treatments. Although priming treatment had nearly similar effect to morphological characters, $GA_3$ treatment forced greater shoot, root and aftermath total dry weight per seedling. Poor seedling emergence of C. pilosula was caused by its seed defect like cleavage or lack of embryo, poor development of embryo and endosperm or their separation.
Induction of triploid cherry salmon, Oncorhynchus ma.sou was performed by the heat shock procedure. The triploids were induced by 15 and 20 min heat shock treatments after 10 min fertilization at $28^{\circ}C$. Incidences of the triploidy were $91.4\%\;and\;89.8\%$, respectively. Nucleus and erythrocyte from the induced triploids larger than those of the diploids in major axis, minor axis, major axis/minor axis, surface area and volume. The chromosome number of diploid was 2n= 66 (17 pairs of metacentrics or submetacentrics, 16 pairs of acrocentrics or telocentrics), and that of triploid was 3n=99. Satellites were observed in the short arm of the largest telocentrics, and these may be useful marker to determine the polyploidization. Diploid have outgrown to triploid, and female have also outgrown to male in the triploid group during the 22 months of observation period after hatching. During the spawning season, 22 to 26 months after hatching, diploid were observed the growth retardation because of their sexual maturation, however the triploid showed continuous growth.
