Elsholtzia splendens Nakai has been used in North-East Asia as an ingredient of folk medicines for treating cough, headache and inflammation. The present investigation was carried out to establish its in vivo anti-inflammatory activity using several animal models of inflammation and pain. The 75% ethanol extract of the aerial part of E. splendens significantly inhibited mouse croton oil-induced, as well as arachidonic acid-induced, ear edema by oral administration (44.6% inhibition of croton oil-induced edema at 400 mg/kg). This plant material also showed significant inhibitory activity against the mouse ear edema induced by multiple treatment of phorbol ester for 3 days, which is an animal model of subchronic inflammation. In addition, E. splendens exhibited significant analgesic activity against mouse acetic acid-induced writhing (50% inhibition at 400 mg/kg), while indomethacin (5 mg/kg) demonstrated 95% inhibition. E. splendens ($5-100{\;}{\mu}g/mL$) significantly inhibited $PGE_2$ production by pre-induced cyclooxygenase-2 of lipopolysaccharide-treated RAW 264.7 cells, suggesting that cyclooxygenase-2 inhibition might be one of the cellular mechanisms of anti-inflammation.
The antioxidative properties of an Elsholtzia splendens ethanol extract (ESE) were examined in vivo. Oral administration of 10 or 50 mg ESE/kg BW in mice for 50 days resulted in a dose-dependent decrease in several biomarkers of oxidative stress including thiobarbituric acid reactive substance (TBARS), protein carbonyls, and serum 8-hydroxy-2'-deoxy guanosine (8-OH-dG). Moreover, the level of activity and mRNA expression of catalase and superoxide dismutase (SOD) were significantly increased by ESE treatment. Taken together, these results indicate that ESE may be beneficial to human health via its antioxidative properties.
최근 화장품 소재로서 환경친화적인 식물성 유래 천연물질이 주목받고 있으며, 천연물의 다양한 생리활성에 대한 연구가 활발히 이루어지고 있다. 본 연구에서는 다양한 생리활성이 있다고 알려져 있는 약용식물 원지 (P olygala tenuifolia), 백지 (Angelica dahurica), 꽃향유 (Elsholtzia splendens) 3종 추출물의 배합 비율을 달리하여 항산화, 항염증, 보습 및 항균 효능을 조사하였다. 배합 비율은 7가지 조건 (M1, 1:1:1; M2, 0.5:1.5:1; M3, 1.5:0.5:1; M4, 0.1:0.95:0.95; M5, 0.5:0.5:2; M6, 0.95:1.95:0.1; M7, 1.45:0.1:1.45)으로 설정하여 추출하였고, 화장품 소재로 활용하기위해 최적의 혼합 비율을 확인하였다. DPPH 및 ABTS radical 소거 활성은 M6 1,000 ㎍/mL에서 각각 75.37% 및99.19%의 소거능을 나타냈다. M6 200 ㎍/mL 농도에서는 염증반응을 유도한 lipopolysaccharide 처리군 대비 50% 의 Nitric oxide 생성 억제 효과를 보였다. M3와 M6은 50 ㎍/mL 농도에서 대조군에 비해 hyaluronic acid 생성량이 각각 1.47배, 1.49배 높은 것을 확인하였다. Disc diffiusion 테스트에서는 M6 8 ㎍/mL에서 clear zone이 9.75 mm로 나타나 Staplylococcus aureus 균주의 생장 억제를 확인하였다. 이상의 결과로 원지, 백지, 꽃향유 혼합 추출물은 화장품의 기능성 천연소재로 사용 가능할 것이라 판단된다.
전북 변산에서 향유속의 신종인 변산향유(Elsholtzia byeonsanensis M. Kim)를 새로이 발견하여 기재 하였다. 분자생물학적인 결과는 변산향유가 근연종들과 구별되는 신종임을 지지해 주었다. 변산향유는 꽃이 한쪽 방향으로 밀생하는 수상화서이며 난형인 잎과 화관길이가 길어 꽃향유와 유사하다. 그러나 변산향유는 식물체는 작고 줄기는 자주색이며 잎은 혁질이고 윤기가 나며 엽병과 엽신 표면 및 화서의 포 표면에 털이 없고 바닷가 바위틈에 자라는 반면에, 꽃향유는 식물체는 크고 줄기는 녹색이며 잎은 초질이고 윤기가 없으며 엽병과 엽신 표면 및 화서의 포 표면에 털이 있고 숲 가장자리에서 자라는 점에서 두 종이 뚜렷하게 차이가 난다.
이 연구는 국내 8개 지역에서 수집한 꽃향유의 정유성분을 분석하여 화학형을 분류하고, 이들 화학형 성분이 항산화 작용과 식품부패균을 억제하는 효과가 있는지를 알고자 하였다. 정유성분 분석결과 3개의 화학형으로 구분할 수 있었다. 화학형 1의 주성분은 dihydrotaketone (DT type)이었다. 개화기를 기준하여 꽃향유의 개체에 따라 naginatketone (NK type)이 60% 이상을 함유한 것과 elsholtziaketone (EK type)을 60% 이상 함유한 개체로 분류할 수 있었고, 또한 이들 두 성분은 종자세대 후에도 EK type은 elsholtziaketone이 주성분으로 나타났고 NK type은 naginataketone이 주성분인 것이 확인되었다. 따라서 NK type은 제2 화학형으로 그리고 EK type은 제3 화학형으로 구별하였다. 항산화 효과는 $6\;mg/{\ell}$ 농도에서 나타났으며, 같은 농도에서 DT>NK>EK 순으로 항산화능을 보였다. NK type은 Streptomyces와 Bicillus에서, EK type은 Streptomyces와 Pseudomonas에서 높은 항균능력을 보였다.
꽃향유로부터 정유물을 추출하여 천연향료로 개발하기 위하여 수증기증류법으로 추출하고 추출공정의 최적 조건을 반응표면 분석법으로 얻고자 하였다. 수증기증류법의 추출조건에 따른 꽃향유 추출물의 품질특성으로 수율과 총페놀화합물, 전자공여능과 주요 향기성분으로 estragole 과 thymol, beta-caryophyllene의 함량을 분석하였다. 추출온도가 추출시간보다 더 큰 영향을 주는 것으로 나타났으며, 각 품질 특성에 대하여 수립된 예측모델식이 모두 유의성이 높아 각 변수의 예측이 가능함을 보였다. 실험구간내에서 능선분석을 통하여 최대점과 추출조건을 예측하였으며 품질특성 값을 최대로 하는 제한 조건으로 얻어진 최적 추출조건은 $108^{\circ}C$와 2.1시간이었다.
The present study was conducted to investigate the effects of flowers ethanol extract of Elsholtzia splendens (ESE) on the antioxidant defence system in mice with 7,12-dimethylbenz(a)anthracene (DMBA)-induced oxidative stress. The ESE was pre-administered orally to 2 groups of mice at 10 and 50mg/kg body weight (BW) for 5 weeks. Subsequently, mice with pretreatment of ESE received DMBA intragastrically at a dose of 34 mg/kg BW twice a week for 2 weeks. In DMBA alone group, biomarkers of oxidative stress (TBARS value, carbonyl content, and serum 8-OH-dG) were significantly increased. Also, the antioxidant enzymes were down-regulated. ESE significantly restored the TBARS value and carbonyl content at both doses, while a decrease in the elevated serum 8-OH-dG content was observed only at the higher dose. The DMBA-induced decreases in catalase and superoxide dismutase (SOD) activities were restored to nearly control levels by ESE. Glutathione peroxidase (GSH-px) and glutathione reductase (GR) activities, as well as the reduced glutathione (GSH)/oxidized GSH (GSSG) ratio, were significantly affected by ESE at the higher dose. These results suggest that ESE possesses antioxidant activity, which plays a protective role against DMBA-induced oxidative stress.
This study was to evaluate the biological activity of flavor components extracted from E. ciliata and E. splendens in order to survey the possibility applicable to tobacco and food industry. Flavor components were extracted with dividing into three parts; essential oil, absolute, oleoresin. In the nonenzymatic lipid peroxidation system, the inhibition rate($\%$) of essential oil were $67.3\;\pm\;20.7\%,\;58.1\;\pm\;19.3\%$ at the concentration of 50 ${\mu}g/mL$ of E. ciliata and E. splendens, respectively. The inhibition rate($\%$) of the oleoresin in E. ciliata was higher than one in E. splendens. In the enzymatic lipid peroxidation system, the inhibition rate($\%$) of essential oil and oleoresin was$14.28\;\pm\;2.38\%,\;and\;65.93\;\pm\;0.01\%,\;and\;was\;22.58\;\pm\;2.84\%\;and\;40.73\;pm\;6.04\%$. The oleoresin of two species were showed above $90\%$ of the inhibition rate($90\%$) against autooxidative lipid peroxidation system. $EC_{50}$ values in neutral red uptake assays 24 h of exposure times were $23.3\;{\mu}g/mL,\;341.0\;{\mu}g/mL\;and\;17.2\;{\mu}g/mL$ in essential oil, absolute and oleoresin from E. ciliata respectively, and were $46.4\;{\mu}g/mL,\;681.7\;{\mu}g/mL\;17.6\;{\mu}g/mL$ in three extractions of E. splendens. Oleoresin of two species showed high rate in the cytotoxic effect by neutral red uptake assay. Absolute and oleoresin did not show antibiotic and mutagenic activity. On the contrary, essential oil with over 500 ug/plate showed antibiotic and mutagenic activity in Ames test. Essential oil and oleoresin have a prolongating effect the ciliostasis of rat trachea. This results indicate that flavor components extracted from E. ciliata and E. splendens can be considered to be toxicological safe and to be the possibility applicable the cigarette, food and drug industry as a flavor for expectoration.
본 연구에서는 독특한 방향을 지닌 식용 및 약용식물인 꽃향유의 휘발성 향기성분을 SDE 및 HS-SPME법에 의해 추출한 후, GC-MSD를 이용하여 분석 동정하였으며, GC-O에 의하여 꽃향유의 aroma-active compounds를 확인하였다. SDE법에 의해 꽃에서는 aldehydes 3종, alcohols 7종, hydro-carbons 11종, ketones 5종 및 기타 3종의 총 29종이 확인되었고, 잎에서는 aldehydes 3종, alcohols 6종, hydrocarbons 11종, ketones 6종 및 기타 4종의 총 30종이 확인되었다. PDMS fiber를 사용한 HS-SPUE법에 의해 꽃과 잎에서는 aldehyde가 검출 되지 않았으며 alcohols 3종, hydrocarbons 18종, ketones 3종 및 기타 2종의 총 26종이 확인되었고, 잎에서는 alcohols 7종, hydrocarbons 15종, ketones 7종 및 기타 2종의 총 31종이 확인되었다. 꽃향유의 주요 향기성분으로 알려진 elsholtziaketone과 naginataketone은 SDE법과 HS-SPME법 모두에서 확인되었으며 꽃과 잎 부위에서 상대적으로 많은 양이 나타났다. 꽃향유 향기분석에서 SDE 및 PDMS fiber를 이용한 HS-SPME방법의 주요한 차이는 비극성인 SPME fiber에서 비교적 극성 물질인 aldehyde류가 확인되지 않았다는 점이다. 또한 GC-O 분석을 통하여 elsholtziaketone과 naginataketone이 강렬한 운향(rue-like)을 지니며, 톡 쏘는 듯한 향신료의 향을 갖는 것으로 확인되었다. 따라서 elsholtziaketone과 naginataketone이 꽃향유 특유의 향 발현에 기여하는 것으로 판단된다.
