Antioxidant Properties of Tannic Acid and its Inhibitory Effects on Paraquat-Induced Oxidative Stress in Mice

  • The tannins represent a highly heterogeneous group of water-soluble plant polyphenols that may play an important role in antimutagenic and antioxidant properties. We investigated the antioxidant function of tannic acid in comparison to other phenolic compounds including catechin, chlorogenic acid, cinnamic acid, ellagic acid, and gallic acid for their ability to scavenge several stable radicals and reactive oxygen species (ROS) such as ${\bullet}DPPH^+$, ${\bullet}ABTS^+$, hydrogen peroxide, hydroxyl radical, and superoxide radical. The ability of tannic acid to decrease paraquat-induced lipid oxidation in mouse liver and lung through its antioxidant properties was also assessed. The results showed that almost all the tested compounds have stable radical scavenging activity except cinnamic acid. Tannic acid, gallic acid, and ellagic acid demonstrated remarkable ROS scavenging properties toward $H_2O_2$, ${\bullet}OH^-$, ${\bullet}O_2^-$ and especially only tannic acid could inhibit paraquat-induced lipid peroxidation effectively in mouse liver and lung. Based on these results, it appears that increased number of galloyl and ortho-hydroxyl groups enhances the antioxidant activity of phenolic compounds and tannic acid is evaluated as the most effective antioxidant among all the tested compounds. These results suggest that the tannins, especially tannic acid, can be used as therapeutic agent for various diseases caused by ROS.

Rubus coreanus Extract Attenuates Acetaminophen Induced Hepatotoxicity; Involvement of Cytochrome P450 3A4

  • Foods of plant origin, especially fruits and vegetables, have attracted attention because of their potential benefits to human health. In this report, Rubi Fructus (RF), the dried unripe fruit of Rubus coreanus Miq (Rosaceae) and ellagic acid (EA) purified from RF were used to test their potential hepatoprotective effect against acetaminophen (AAP)-induced hepatotoxicity in rats. RF extract (RFext) and EA reduced the elevated levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST) in serum and the content of lipid peroxide in liver by AAP administration, while the increment of the cellular glutathione (GSH) content and the induction of glutathione S-transferase (GST) and glutathione peroxidase (GSH-PX) which were decreased by AAP administration. RFext and EA from RFext did not affect the two major form of cytochrome P450s, cytochrome P450 2E1 (CYP2E1) and cytochrome P450 1A2 (CYP1A2), but downregulated the cytochrome P450 3A4 (CYP3A4) related to the conversion of AAP to N-acetyl-P-benzoquinone imine (NAPQI). These results suggest that RFext and EA from RF exhibit a hepatoprotective effect not only by increasing antioxidant activities but also by down-regulating CYP3A4 in the AAP-intoxicated rat.

율피, 솔잎, 호프의 아세톤 추출물의 항산화작용 (The Antioxidant Activities of Acetone Extracts of Chestnut Inner Shell, Pine Needle and Hop)

  • 율피, 솔잎, 호프로부터 70% 아세톤 추출물을 획득하여 대두유, 돈지, 대두유 유탁액, 돈지 유탁액에 대한 항산화력을 토코페롤과 비교하였다. Rancimat의 유도기간에 따른 항산화력 비교에서는, 대두유에서는 솔잎>율피>호프>토코페롤>무첨가구, 돈지에서는 토코페롤>율피>솔잎>호프>무첨가구로서 항산화력이 있었다. Peroxide value의 측정을 통한 항산화력 비교에서는, 대두유나 대두유 유탁액, 돈지나 돈지 유탁액에서 이들 세 추출물은 같거나 우수한 항산화력을 나타내었으며, 율피가 가장 좋은 것으로 판단되었다. 폴리페놀화합물의 분석결과, 율피 추출물에는 ellagic acid가 솔잎 추출물에는 flavanol이 항산화력에 큰 영향을 미친 것으로 보인다.

Beta-carotene prevents the spermatogenic disorders induced by exogenous scrotal hyperthermia through modulations of oxidative stress, apoptosis, and androgen biosynthesis in mice

  • We investigated whether ${\beta}$-carotene (${\beta}-CA$) or ellagic acid (EA), originating from various fruits and vegetables, has a preventive effect against male infertility induced by exogenous scrotal hyperthermia. ICR adult mice were intraperitoneally treated with 10 mg/kg of ${\beta}-CA$ or EA daily for 13 days consecutively. During this time, mice were subjected to transient scrotal heat stress in a water bath at $43^{\circ}C$ for 20 min on day 7, and their testes and blood were obtained on day 14 for histopathologic and biochemical analyses. Heat stress induced significant testicular weight reduction, germ cell loss and degeneration, as well as abnormal localization of phospholipid hydroperoxide glutathione peroxidase (PHGPx) and manganese superoxide dismutase (MnSOD) in spermatogenic and Leydig cells. Heat stress also altered the levels of oxidative stress (lipid peroxidation, SOD activity, and PHGPx, MnSOD, and $HIF-1{\alpha}$ mRNAs), apoptosis (Bax, Bcl-xL, caspase 3, $NF-{\kappa}B$, and $TGF-{\beta}1$ mRNAs), and androgen biosynthesis (serological testosterone concentration and $3{\beta}$-hydroxysteroid dehydrogenase mRNA) in testes. These changes were all improved significantly by ${\beta}-CA$ treatment, but only slightly improved by EA treatment. These findings indicate that ${\beta}-CA$, through modulations of oxidative stress, apoptosis, and androgen biosynthesis, is a potent preventive agent against testicular injuries induced by scrotal hyperthermia.

밤송이 이용(利用)에 관(關)한 연구(硏究) (1) - 밤송이 성분(成分) - (Studies on the Utilization of Chestnut Bur(1) - Components of Chestnut Bur -)

  • As the production of chestnuts from chestnut trees has been increased annually in the past several years in this country, developing the utilities of chestnut bur has been considered as one of main research problems. Therefore. in this paper, the chemical components were analysed to find a way of utilizing the chestnut bur. It was proved that chestnut bur is abundant in extractives (hot water 16.0%, 1%-NaOH 40.6%) and tannin content (16.5%). Of the tannin, the soluble tannin (3.2%) content is higher than condensed tannin (2.2%), and gallic acid content(3.3-3.9%) is slightly higher than ellagic acid(2.8-3.0%), For more effectively utilizing as a tannin resources, the chestnut bur should be collected immediately after yield of chestnuts, maybe in October every year.

  • This study aimed to search for separate and identify of Quercus acutissima CARRUTHERS tannins. Tannins were extracted with methanol and ethylacetate from acorn powder and identified TLC, UV spectrum, HPLC, IR, GC/MS, and $^{1}H$ NMR. Three spots($R_{f}$ 0.94, 0.84 and 0.29) detected on TLC. These spots gave dark blue color fairly on spraying with 0.3% potassium ferricyanid${\cdot}$0.3% ferricchloride reagent, and these tannins identified as gallic acid, caffeic acid and ellagic acid by UV spectrum, HPLC, IR, GC/ MS, and $^{1}H$ NMR.

Antioxidant Effect of Juglans mandshurica Bark Gallotannins

  • The bark of Juglans mandshurica Maxim. was collected, extracted with acetone-$H_2O$ (7:3, v/v), fractioned with n-hexane, $CH_2Cl_2$ and EtOAc, then each fraction was freeze-dried to give some powders. A portion of the EtOAc (28.4 g) fraction was chromatographed on a Sephadex LH-20 column eluting with a series of MeOH-$H_2O$ and EtOH-hexane mixture. Four gallotannins, gallic acid (1), ellagic acid (2), 1,2,4,6-tetra-O-galloyl-${\beta}$-D-glucose (3) and 1,2,3,4,6-penta-O-galloyl-${\beta}$-D-glucose (4), have been isolated from the EtOAc fraction Their structures were elucidated on the basis of chemical evidence and spectrometric analysis such as NMR and MS. The antioxidant activities on each fraction and the isolated gallotannins were evaluated by DPPH radical scavenging test.

  • Post-transcriptional regulation of mRNA stability by Hu proteins is an important mechanism for tumorigenesis. We focused on the molecular interactions between the HuC protein and AU-rich elements (AREs) to find chemical inhibitors of RNA-protein interactions using RNA electrophoretic mobility shift assay with non-radioactive probes. Screening of 52 natural compounds identified 14 candidate compounds that displayed potent inhibitory activity. Six (quercetin, myricetin, (-)-epigallocatechin gallate, ellagic acid, (-)-epicatechin gallate, and rhamnetin) were categorized as phytochemicals, and their $IC_{50}$ values were low ($0.2-1.8\;{\mu}M$).

  • The present study aimed to investigate the effects of low temperature on the growth, yield, quality, and biologically active compounds of strawberry and obtain basic information for developing a technology for stable growth of strawberry in greenhouses. Growth of strawberry, including leaf number, area, and length, plant height, and dry weight was better at the optimum growth temperature of $20^{\circ}C$ than at a lower temperature of $15^{\circ}C$. At the low temperature of $15^{\circ}C$, the cultivar 'Maehyang' was more tolerant and displayed better growth rate than 'Seolhyang'. At $15^{\circ}C$, the fruit production per week and fruit weight was lower than that at $20^{\circ}C$. In contrast, fruit length and diameter were not significantly different between the two growth temperatures. Growth temperature also did not affect the fruit color index, Hunter L, a, b value, or fruit firmness. However, the sugar content of strawberries grown at $15^{\circ}C$ was higher by 0.8 and 1.5 Brix for 'Seolhyang' and 'Maehyang', respectively, than of those grown at $20^{\circ}C$. There was no difference in the content of fisetin, a biologically active compound, for 'Seolhyang' at both growth temperatures, however, the fisetin content of 'Maehyang' was higher at $20^{\circ}C$ than at $15^{\circ}C$. Cinchonine and ellagic acid content of 'Seolhyang' was higher at $20^{\circ}C$ than at $15^{\circ}C$, whereas that of 'Maehyang' was higher at $15^{\circ}C$ than at $20^{\circ}C$. Quercetin content showed no significant differences with respect to growth temperature, however, it tended to increase at $20^{\circ}C$. The cinnamic acid content of 'Seolhyang' was higher at $15^{\circ}C$ than at $20^{\circ}C$, whereas that of 'Maehyang' increased at $20^{\circ}C$. Collectively, the biologically active compounds of strawberry were affected by growth temperature. Moreover, the content of these compounds tended to increase at $20^{\circ}C$, the optimum growth temperature, rather than at the sub-optimal growth temperature of $15^{\circ}C$.
