Anti-hyperlipidemic Effect of Phragmites communis and Its Active Principles (갈대의 고지혈증 개선효과와 그 활성성분)

  • Administration of an MeOH extract of Pharagmites communis on hyperlipidemic rats produced a significant decrease in blood triglyceride. The fatty acid mixtures, $\beta$ -sitosterol and its glucoside, p-coumaric acid, vanilic acid, ferulic acid, p-hydroxybenzaldehyde and sucrose were isolated. Sterol and p-coumaric acid were shown to be effective in reducing the elevated level of triglyceride. The wet weight and the level of triglyceride in epididymal adipose tissue increase following administration of the methanol extract in rats fed the purified diets at the 1% level for 4 weeks. This results suggest that the methanol extract may accelerate the accumulation of lipid in adipose tissue.

  • Effect of ethanol on the absorption rate, blood level and bioavailability of sulfamethazine (SM) in rats was determined. Absorption rate of SM was determined both by the in vitro and in situ experiment. In vitro, absorption rate of SM in rat small intestine was increased by 0.3, 1.0 and 3.0% ethanol. In situ, absorption rate of SM was increased by 0.3 and 1.0% ethanol but not by 3.0% ethanol. After oral administration, blood level of SM was elevated and relative bioavailability was significantly increased to 114.8% at the dose of 0.6g/kg ethanol but not significantly at the dose of 3.0g/kg ethanol. The time for attainment of peak blood level was changed from 2.5 to 1.5hr. Ethanol enhanced absorption rate constant of SM significantly and reduced elimination rate constant of SM administered orally at the dose of 0.6g/kg ethanol.

Effect of oral guava leaf extract administration on antioxidant and vasculoprotective activity in ovariectomized rats (구아바잎 추출물이 난소절제 흰쥐에 미치는 항산화 및 혈관보호 효과)

  • Purpose: The aim of this study was to assess the effects of guava leaf extract (GLE) supplementation on antioxidant enzyme activity and expression of endothelial nitric oxide synthase (eNOS) mRNA in ovariectomized rats. Methods: All animals were randomly assigned to four groups (n = 7 for each group): non-ovariectomized control (Sham), the ovariectomized control (OVX), ovariectomy + 150 mg/kg b.w. of GLE ($OVX{\cdot}GL$), and ovariectomy + 300 mg/kg b.w. of GLE ($OVX{\cdot}GH$). Treatment groups were administered GLE for 8 weeks every day. Results: Body weight gain was significantly reduced in the $OVX{\cdot}GL$ group compared with the OVX group (p < 0.05). The level of serum $17{\beta}$-estradiol (E2) was significantly lower in the OVX groups than the Sham group (p < 0.05). Serum triglyceride (TG) and HDL-cholesterol levels were not significantly different between all groups. However, serum total cholesterol (TC) level was significantly reduced in the $OVX{\cdot}GH$ group compared with the OVX group (p < 0.05). Serum free fatty acid (FFA) level and liver TG level were significantly reduced in both $OVX{\cdot}GL$ and $OVX{\cdot}GH$ groups compared with the OVX group (p < 0.05). Furthermore, serum glutathione peroxidase (GPx) activity was significantly elevated in the GLE groups (p < 0.05). The mRNA expression level of GPx was not affected by ovariectomy. However, administration of GLE resulted in significantly increased nuclear factor erythroid 2-related factor (Nrf2) and catalase (CAT) mRNA expression levels in the liver (p < 0.05). In addition, liver nitric oxide (NO) level was significantly reduced in the $OVX{\cdot}GH$ group compared with the OVX group (p < 0.05). Expression level of endothelial nitric oxide synthase (eNOS) was significantly elevated in the $OVX{\cdot}GH$ group compared with the OVX group (p < 0.05). Conclusion: These results suggest that GLE could have protective effects in OVX rats by stimulating eNOS expression and improving the antioxidant defense system.

Effects of Dietary Vitamin E Level and Caffeine on Lipid Peroxidation in Rat Liver (흰쥐 간에 있어 지질과산화에 미치는 식이성 비타민 E의 수준 및 카페인의 영향)

  • This study was designed to evaluate the effects of dietary vitamin E and caffeine on the activities of lipid peroxidation related enzymes in rat liver . Male Sprague-Dawley rats were fed on diets containing three level of vitamin E (37.5, 750 or 1,5oomg/kg diet) and with or without 0.3% caffeine. The rats were sacrificed after 5 and 10 weeks of feeding. Results obtained from this study were as follows ; The content of cytochrome P450 tended to increase as dietary vitamin E level was raised. The activity of xanthine oxidase increased in the caffeine groups, but it decreased by the increasing level of vitamin E. Superoxide dismutase and catalase activity were slightly elevated by dietary supplementation of vitamin E. And there was a tendency of higher these enzyme activity of caffeine groups. The activity of glutathione perxidase tended to decrease as dietary vitamin E level increased. But it was raised by caffeine supplementation . Liver glutathione content was not affected by dietary supplementation of vitamin E, but it showed a decreasing tendency in caffeine groups. There was a tendency of more lipid peroxide content of caffeine groups than that of the only vitamin E supplemented group. But the degree of increment of this decreased as dietary vitamin E level increased.

The Effects of Music Therapy on Vital Sign Changes of operational Patients (음악요법이 수술환자의 활력징후 변화에 미치는 영향)

  • The purpose of this study was to determine the effects of music therapy on changes in the vital signs of patients about to undergo an operation. The patients listened to the music at a time when they were feeling preoperative anxiety up until the preanesthesia was given in the operating room. The subjects for this study were selected from sixty patients to undergo operations, who were hospitalized at Dong Eui hospital in Pusan city. They were assigned to two groups, thirty to the experimental group and thirty to the control group. The subjects were from 20 years old to 69 years old, and had no other problem except the one requiring the operation, and no premedication. The data were collected during the period from July 1 to September 30, 1993. The method used in this study was to measure state-anxiety on the ward in the morning of the operation, and vital signs immediately before leaving for the operating room. Vital signs were measured immediately before the anesthesia was given and after the experimental group had listened to the music during the ten minutes needed to prepare the operation setting. The control group just waited during ten mimutes. Vital signs were check again before the anesthesia was given. The data were analyzed by descriptive statistics, mean±SD, p-value, and t-test using the SPSS progrom. The results of this study are : 1. Systolic blood pressure taken in the operating room was elevated, over the level measured on the ward, by 5.00 ± 15.26㎜Hg in experimental group and 18.67±14.56㎜Hg in control group. (t=-3.5496, p=.0008) 2. Diastolic blood pressure was elevated by 6.67±12.95㎜Hg in experimental group and 18.67±12. 79㎜Hg in control group. (t=-3.6100, p=.0006) 3. Pulse was elevated by 2.931±9.44 / min in experimental group and 8.03±8.37 /min in control group. (t=-2.2144, p=.0307) 4. Respiration was elevated by 0.60±1.35 /min in experimental group and 1.57±1.48 /min in control group. (t=-2.6409, p=.0106) 5. Body temperature was down by 0.13±1.91'c in experimental group and elevated by 1.13±1.11'c in control group. (t=-3.1471, p=.0026) Thus, in this study there was a statistically significant difference in the change in the vital signs between the experimental group treated with music therapy and the control group which received no treatment. Because music therapy is valuable to decrease the anxiety of patients facing operations, the result of this study support its effect in relieving anxiety as a valuable nursing intervention. From this study, the following recommendations can be made : First, it is necessary to further study music therapy to develope a better system and determine optimal time. Second, it is necessary that more detailed re-search on measurement of changes in vital signs be done to determine changes over time intervals.

Differential Effects of Fumonisin $B_1$ on Cell Death in Cultured Cells: the Significance of the Elevated Sphinganine

  • Fumonisins are specific inhibitors of ceramide synthase in sphingolipid metabolism. An alteration in sphingolipid metabolism as a result of fumonisin exposure is related to cell death (Yoo et al., 1992). The objective of this study was to investigate whether elevated free sphinganine levels are related to the sensitivity of cultured cells to fumonisin exposure. Fumonisin $B_1$ elevated the intracellular free sphinganine concentraions in both LLC-$PK_1$ and Chinese hamster ovary (CHO) cells. However, CHO cells are resistant to fumonisin cytotoxicity at 50${u}m$, while LLC-$PK_1$ cells are sensitive at concentrations greater than 357M. The intracellular concentration of free sphinganine in LLC-$PK_1$ cells treated at 50${u}m$ fumonisin $B_1$ for 72 h was approximately 1450 pmol/mg protein relative to the 37 pmol observed in the control culture. Under the same conditions, the population of apoptotic cells in the 50${u}m$ fumonisin $B_1$-treated culture was approximately 37% of the total compared to 12% in the control. The caspase III-like activity after 72 h in the 50${\mu}$M fumonisin $B_1$-exposed culture Increased to approximately 50 $pmol/mg$ protein/hr compared to 6 $pmol/mg$ protein/hr in the control. L-cycloserine, a serine palmitoyltransferase inhibitory reduced the fumonisin $B_1$-stimulated caspase III-like activity down to the control level. Under the same culture conditions, the intracellular concentration of free sphinganine after-cycloserine plus fumonisin $B_1$ treatment was 140 pmol/mg protein compared to 1450 $pmol/mg$ protein in fumonisin $B_1$ alone. The intracellular concentration of free sphinganine in CHO cells treated with 50${u}m$ fumonisin $B_1$ for 72 h was al)proximately 460 pmol/mg protein, indicating that the mass amount of elevated free sphinganine in the CHO cells was about 32% of that in LLC-$PK_1$ cells. Adding exogenous sphinganine to the CHO cells along with 50${u}m$ fumonisin $B_1$ treatment for 72 h caused both necrosis and apoptosis. In conclusion, the elevated endogenous sphinganine acts as a contributing factor to the fumonisin-induced cell death.

Effect of Lead Exposure During Lactational Period on Anxiety in Rat Using Elevated Plus Maze Test (수유기동안 납 투여가 성숙 쥐의 불안감 관련 행동양상에 미치는 영향)

  • Lead (Pb) exposure during development can produce neurological deficits. In this study, the effect of Pb exposure during neonatal development via lactation on anxiety of brain function was investigated. Long-Evans strain rats were raised through two generations. At the birth of the second generation, the dams were subdivided into two groups and supplied drinking water containing either $0.2\%$ Pb (Pb-treated group) or sodium (Na, Control group) acetate until weaning. Rats were sacrificed at 3 (weaning) and 11 weeks (maturity) for brain Pb and fatty acid analysis. Motor activity and elevated plus maze tests were initiated at 9 weeks. The brains in the Pb-treated group at weaning and maturity contained 1486$\pm$98 and $270{\pm}46$ ng Pb/g, respectively The control group showed the background level of Pb ($3.7{\pm}1.0_{ng}$ Pb/g) in both ages. The alterations in brain fatty acid composition induced by Pb exposure were more evident in 3 wks old than 11 wks old. For example, in 3 wks old, the percentages of $18:2_{n-6}$, $20:2_{n-6}$ and $18:2_{n-6}$ were decreased in the Pb-treated group with an increase in $20:4_{n-6}$ In motor activity test, there was a tendency of hyperactivity in the Pb-treated group compared with the control group but the difference was not significant. In elevated plus maze test, the Pb-treated group showed fewer numbers of visits to the open arms (P < 0.05), indicating that Pb exposure may lead to anxiogenic effect.

  • The drug interaction between probenecid and lithium carbonate was studied pharmacokinetically in rabbits. The blood level and the area under the concentration curve (AUC) of lithium carbonate administered orally were elevated by coadministration of probenecid. Probenecid inhibited the urinary excretion of lithium carbonate in rabbits. Biological half-life and $t_{max}$ of lithium carbonate were prolonged by coadministration of probenecid. From these results, dosage regimen of lithium carbonate is considered to be adjusted for effective and safe therapy in the coadministration of probenecid.
