Carbon paper에 직접적으로 생산한 CNT를 polyol 방법으로 Pt deposition하여 PEMFC cathode 개발 (The development of PEMFC cathode using polyol method with directly grown CNT on carbon paper)

  • Since the discovery of the carbon nanotube(CNTs), they have attracted much attention because of unique properties that may impact many fields of science and technology. The considerable properties of CNTs include high surface area, outstanding thermal, electrical conductivity and mechanical stability. However, uniform deposition of Pt nanoparticles on carbon surface remains inaccessible territory because of the inert carbon surface. In this study, we prepared directly oriented CNTs on carbon paper as a catalyst support in cathode electrode. carbon surface was functionalized using aryl diazonium salt for increasing adhesion of Ni particles which is precursor for growing CNTs. For fabricate electrode, CNTs on carbon paper were grown by chemical vapor deposition using Ni catalyst and Pt nanoparticles were deposited on CNTs oriented carbon paper by polyol method. The performance was measured using Proton electrolyte Membrane Fuel Cell(PEMFC). The structure and morphology of the Pt nanoparticles on CNTs were characterized by Scanning electron Microscopy(SEM) and Transmission electron Microscopy (TEM). The average diameter of Pt nanoparticles was 3nm.

  • Electron microscopy (EM) can provide a valuable contribution to light microscopy (LM) In the Interpretation of fine needle aspiration cytology (FNAC) specimen, especially in the diagnosis of the tumor. However, considerable care in processing the specimen is mandatory to recover the cells and avoid altering the fine structures. We experienced a case of malignant melanoma in 33-yrs-old female, diagnosed by EM study of FNAC specimen from the axillary mass, who was initially thought as disseminated carcinomatosis on LM study. The technique of EM study on FNAC specimen consisted of washing the needle and syringe in 2.5% glutaraldehyde after a rapid stain (Diff-Quik), which was used to obtain a preliminary diagnostic impression and to assure the adequacy of the EM specimen. After centrifugation in the steps of fixation and dehydration, the sediment was made into an epon block and examined. The whole processing time of EM study can be shortened within 7 or 8 hours, and results can be available within 48 to 72 hours. Our experience suggests the EM study on FNAC can be a useful diagnostic method in the diagnosis of difficult FNAC cases.

  • The aim of this research was to discover the action mechanism of various antituberculosis agents (isoniazid, paraaminosalicylic acid and streptomycin) which act on Mycobacteria tuberculosis hominis $H_{37}R_v$ and also to study the relationship of ultrastructural changes and the growth pattern in Mycobacterium tuberculosis hominis. The ultrastructural change was observed with an electron microscope while the growth pattern was studied through in vitro culture. The results are summarized as follows: 1. The ultrastructural changes found in the group treated only with isoniazid were the loss of nuclear materials and the appearence of electron dense granules. 2. In the group treated with paraaminosalicylic acid, thickening of nuclear filaments and meso some arrangement disorders were observed. 3. In the group treated with streptomycin, the ribosome particles appeared indistinct and the cytoplasm was denaturalized. 4. In the group cross treated with all three agents, all the ultrastructural changes mentioned above could be observed in the cell just as they appeared in the single treated groups. 5. In all of the single and in the crossly treated group, there were no significant changes note in the cell wall or cytoplasmic membranes of any of the cells observed. 6. In the cultural data in vitro, through the crossly treated group and single treated group. growth was observed in 3-5 weeks of culture.

장대끈끈이주걱 분비모의 미세구조와 peroxidase 활성 (Ultrastructure and Activity Pattern of Peroxidase in Secretory Trichomes of Drosera capensis)

  • Glandular trichomes present on the leaf surface of Drosera capensis were examined using transmission electron microscopy. A large number of stalked glands exist on the adaxial surfaces of the leaf blade. The secretory head is composed of two layers of secretory cells, one layer of middle cells, and the inner tracheids. The secretory cells contain rough endoplasmic reticulum, mitochondria, plastids, Golgi apparatus, and vacuoles. The secretory cells show prominent cell wall ingrowth, and thick cuticle restricted on the subcuticular wall. Frequently, the cuticle has some pores, canal-like structures, showing electron -dense granules being penetrated through them. Ultrastructural localization using diaminobenzidine showed the electron-dense deposits in the vacuole. No peroxidase activity was seen in the cell wall and cytolasm. The activity of peroxidase (POX) isozymes in Drosera which isoelectric point (pI) is 3.6 and some anionic POX isozymes which pIs are laid between 3.6 and 4.6 were especially increased according to the development and the formation of glandular trichomes. Also, the activity of some POX isozymes which isoelectric points are laid between 4.6 and 5.1 were increased in the regions of leaves which has trichomes.

Scanning Electron Microscopic Study on the Head Lice Eggs Detected in Korean Children

  • Fine structural characteristics of eggs (nits) found on the scalp hairs of Korean children were observed with scanning electron microscopy. An egg is structurally composed of four parts: the cementum, nit body, operculum, and aerophyle. The total length of an egg is about 1.5 mm. The glue secreted from the female louse completely surrounds the 1 mm hair shaft and forms the cementum. The thickness of the cementum at the end of the nit body was found to be more than 5 times the thickness at the end toward the scalp. The nit body is shaped like a goblet with a very smooth surface. The operculum and aerophyles are located at the apiculus area. In the circular operculum, there are 10 aerophyles concentrated on the side of the hair shaft surface that are shaped like a dome. Three aerophyles in the center are surrounded by 7 aerophyles. Each aerophyle is dome shaped with a diameter of $65{\mu}m$ and a respiratory pathway with a $15{\sim}25{\mu}m$ opening at the center. On the cut surface where the operculum is separated as the egg hatches, long grooves about $1{\mu}m$ thick are uniformly formed transversely. These long grooves facilitate the separation of the operculum through body expansion at the time of hatching.

Morphology of Nanocelluloses and Micro-sized Cellulose Fibers Isolated by Acid Hydrolysis Method

  • As a part of utilizing the nanocellulose (NC) from lignocellulosic components of wood biomass, this paper reports preliminary results on the products of sulfuric acid hydrolysis. The purpose of this study was to investigate the morphology of both NC and micro-sized cellulose fiber (MCF) isolated by acid hydrolysis from commercial microcrystalline cellulose (MCC). Field emission.scanning electron microscopy (FE-SEM) and transmission electron microscopy (TEM) were employed to observe the acid hydrolysis suspension, NC, and MCF. The electron microscopy observations showed that the acid hydrolysis suspension, before separation into NC and MCF by centrifugation, was composed of nano-sized NCs and micro-sized MCFs. The morphology of isolated NCs was a whisker form of rod-like NCs. Measurements of individual NCs using TEM indicated dimensions of 6.96$\pm$0.87 nm wide by 178$\pm$55 nm long. Observations of the MCFs showed that most of the MCC particles had de-fibered into relatively long fibers with a diameter of 3-9 ${\mu}m$, depending on the degree of acid hydrolysis. These results suggest that proper technologies are required to effectively realize the potentials of both NCs and MCFs.

집속이온빔 연마에 의한 패턴의 형태에 관한 연구 (A Study on the Shape of the Pattern Milled Using FIB)

  • For the measurements of surface shape milled using FIB (focused ion beam), the silicon bulk, $Si_3N_4/Si$, and Al/Si samples are used and observed the shapes milled from different sputtering rates, incident angles of $Ga^+$ ions bombardment, beam current, and target material. These conditions also can be influenced the sputtering rate, raster image, and milled shape. The fundamental ion-solid interactions of FIB milling are discussed and explained using TRIM programs (SRIM, TC, and T-dyn). The damaged layers caused by bombarding of $Ga^+$ ions were observed on the surface of target materials. The simulated results were shown a little bit deviation with the experimental data due to relatively small sputtering rate on the sample surface. The simulation results showed about 10.6% tolerance from the measured data at 200 pA. On the other hand, the improved analytical model of damaged layer was matched well with experimental XTEM (cross-sectional transmission electron microscopy) data.

In Vitro Activity of Taurine-5-Bromosalicylaldehyde Schiff Base Against Planktonic and Biofilm Cultures of Methicillin-Resistant Staphylococcus aureus

  • Staphylococcus aureus is a major human pathogen, implicated in both community and hospital acquired infections. The therapy for methicillin-resistant Staphylococcus aureus (MRSA) infections is becoming more difficult because of multidrug resistance and strong biofilm-forming properties. Schiff bases have attracted attention as promising antibacterial agents. In this study, we investigated the in vitro activity of taurine-5-bromosalicylaldehyde Schiff base (TBSSB) against MRSA. The minimum inhibitory concentrations (MIC) and minimum bactericidal concentration (MBC) were determined using a microtiter broth dilution method. TBSSB effectively inhibited planktonic MRSA, with an MIC of $32{\mu}g/ml$. The time-kill curve confirmed that TBSSB exhibited bactericidal activity against MRSA. TBSSB was also found to significantly inhibit MRSA biofilm formation at 24 h, especially at $1{\times}MIC$ and sub-MIC levels. Furthermore, scanning electron microscopy and transmission electron microscopy showed remarkable morphological and ultrastructural changes on the MRSA cell surface, due to exposure to TBSSB. This study indicated that TBSSB may be an effective bactericidal agent against MRSA.

Microscopic Patterns of Decay caused by Tyromyces palustris and Gloeophyllum trabeum in Korean Red Pine and Radiata Pine Woods

  • The objective of this study was to elucidate the microscopic patterns of decay caused by brown-rot fungi of Tyromyces palustri and Gloeophyllum trabeum in Korean red pine (Pinus densiflora) and radiata pine (Pinus radiata) woods through light and electron microscopies. The ultrastructural changes of cell walls attacked by the two brown-rot fungi were compared in this respect. Macroscopically, radiata pine showed more ring and radial checks than Korean red pine. Microscopically, with the progress of decay, spiral checks associated with cross-field pits and bore holes in the cell wall were more remarkably numerous in the radiata pine than in the Korean red pine. In the radiata pine, G. trabeum produced more spiral checks in the cell wall than T. palustris. In the advanced stages of decay by G. trabeum, the erosions of ray cell walls were identified both in the Korean red pine and radiata pine but $S_3$ layers of tracheid walls were eroded only in the Korean red pine.

Light and Electron Microscopic Characterization of Husk from Korean Rice

  • Microscopic techniques were used to observe the microstructure of rice husk. Microscopic examination showed that two main components of husk, lemma and palea consisted of outer epidermis, layers of fibers, vascular bundles, parenchyma cells, and inner epidermis, in sequence from the outer to the inner surface. The outer epidermal walls were extremely thick, highly convoluted and lignified. The underlying fibers were also thick-walled and lignified. Parenchyma cells were thin-walled and unlignified. Inner epidermal cells were also unlignified. The outer surface of both lemma and palea were conspicuously ridged, but the lower surface had a flat appearance. As part of a detailed study to characterize rice husk using microscopic and micro-analytical techniques, distribution of silica was also examined, and is presented elsewhere. Rice husk can potentially be used as a raw material for making composite products and the observations presented here form valuable background information for our future work related to product development.
