Spawning and Growth of eightspine stickleback, Pungitius sinensis kaibarae (Tanaka) (Gasterosteidae, Pisces), in the Chavans Stream, a tributary of Kumho River, Korea (잔가시고기 Pungitius sinensis koibaroe (Tanaka) (큰가시고기과, 어강)의 산란과 성장)

  • The spawning and growth of eightspine stickleback,Pungitius sinensis kaiborae (Tanaka, 19151, were investigated in the Chavang Stream from March 1990 to february 1991 as a part of study on its life history. Nest building, spavming and parental behaviors were similar to those known on the sticklebacks of Europe, North America and Japan. Bht f sinensis kaibarae in the Chayang Stream, peculiarly, showed only superficial gluing behavior in nest building phase and made two nurseries iust behind the nest in Parental Phase. The individuals with fully mature eggs were found from late February to late June. The fecundity of an adult female was about 21 to 110 (mean 47.07) eggs and the diameter of kllv matured ovarian eggs ranged 1.3 to 1.5 mm. The smallest female with fully mature eggs was 25.8 mm in body length and had 21 eggs. The relationship between the number of mature eggs in ovary and body length was No. of eggs = 2.857BL -55.134 (r = 0.890). The number of deposited eggs in a nest was 27 to 637 and thew were composed of several clusters of various developmental stages. The distribution pattern of gonadsomatic index (GSl) against the body length shown that in females 1+ year-old fishes mainly participate in spawning but in males 11 and 2+ year-old fishes equally participate. From the fluctuat지n pattern of GSI and the number of mature eggs in ovary, it was assumed that the spawning season was from February to June and the peak of spawning was early March to late April. The newly hatched vouns was found at late April and their body length was about 10 mm. The vouns (O+) grew rapidly until late August. Then their srowth was slowed down and finallY ceased in midautumn. The young reached about 35 mm BL in their first year of life. The halting of growth was lasted to next Sune when the spawning season would be nearly closed. They (1+) began to grow asain from late june, grew rapidly until tate september and reached about 415 mm BL. Then there was no more grouvth until to die as 2+ yearold fish. Therefore it was assumed that the life span of P. sinensis kaibarae in the Chavang Stream would be about ko years. The relationship between the body length and the body weight was logBW = 2.9541ogBL -4.802 (r = 0.998).

Effect of the supplement of the earthworm cast, earthworm and B. bassiana on the improvement of both the productivity of hens and nutritional constituents of eggs (지렁이, 분변토 및 백강균 첨가에 의한 산란계의 생산성 및 계란의 영양적 조성 향상)

  • Effects of the supplement of earthworm cast produced from the feeding of organic wastes and earthworm on the productivity and nutritional constituents of functional eggs were investigated. Compared with control experiments, the case supplemented with earthworm cast showed high ratios in egg production, selection and the reserved feed. According to the experiment with earthworm, both the number of jumbo eggs and the quantity of reserved feed were increased. Therefore, the nutritional effect of earthworm in the feed was positive. The optimum percentage of earthworm cast in the feed was 10%: the average laying increased to 96.8, which was a 5% increase; the ratio of the large eggs increased by 5% although the ratio of jumbo eggs and of extra large eggs decreased by 5% and 1.1%, respectively; the average reserved feed was 662.5g. Also, Beauveria bassiana was inoculated into the feed as valuable microorganisms to prevent the growth of pathogen and to obtain essential amino acid. With the inoculatio of B. bassiana KACC 40039, the average laying was 0.82/hen and with B. bassiana HYB, it was 0.77/hen. Those numbers were three to eight percentage over the control. As for the effect of inoculation of B. bassiana in the feed on the production of broken eggs, B. bassiana KACC 40039 produced no broken eggs. Analysis of nutritional contents of eggs showed the increase in protein content and decrease in lipid content when compared with the control. According to these results, increase in the income of farmers can be expected.

A Study on the Method of Culture for Paecilomyces japonica Using an Egg (계란을 이용한 눈꽃동충하초 재배방법에 관한 연구)

  • This study was conducted to established method of culture for Paecilomyces japonica using an egg. Mycelia grew favorably at the temperature of 22~26$^{\circ}C$ on eggs. 5.1g of dry matter basis(average 7.2cm of longer and 199.6 of numbers) of artificial fruiting bodies were harvested at 60 days after inoculation from one of egg. Commercial fruiting bodies of Paecilomyces japonica from silkworms was used for comparative nutriental contents. Cordycepin contents of fruit bodies of Paecilomyces japonica cultivated on eggs and silkworms were not significantly different. Crude fat contents of fruiting bodies of Paeilomyces japonica cultivated from eggs was significantly higher than from silkworms(P<0.05). Mn and Cu contents of fruiting bodies of Paecilomyces japonica cultivated from silkworms were significantly higher than from eggs(P<0.05), but Na, Mg, Fe and Zn contents were significantly higher from eggs(P<0.05). Glycine, Arginine and Proline contents in the fruiting bodies of Paecilomyces japonica cultivated from silkworms were tend to higher than from eggs, but Serine, cystein. methionine, isoleucine and phenylalanin were tend to higher from eggs. These results were made possible that possible mass production of artificial fruiting bodies of Paecilomyces Japonica cultivated on eggs.

A study on the residues of fluoroquinolones in hen's eggs consumed in Incheon Metropolitan city (인천광역시에서 유통되는 계란의 플루오로퀴놀론계 항균물질 잔류 실태 조사)

  • This study was carried out to investigate the residue level of fluoroquinolones in hen's general eggs and specific eggs by microbiological assay method and high performance liquid chromatography (HPLC) method. HPLC separation was carried out by reversed phase chromatography on a Symmetry $C_{18}$ (250${\times}$4.6 mm, $5{\mu}m$ particle size) with a phase composed of distilled water (containing 0.4% triethylamine and phosphoric acid) : Methanol (780 : 220, v/v), pumped isocratically at a flow rate of 1.0ml/min. A fluorescence detector was utilized with an excitation wavelength of 278nm and an emission wavelength of 456nm. The calibration curves were linear $({\gamma}^2{\geq}0.999)$ over a concentration range of $0.025{\sim}0.4{\mu}g/ml$. Average recoveries of the five fluoroquinolones in whole eggs at fortified levels of $0.05{\sim}0.2{\mu}g/g$ were ranged mean $78.1{\sim}91.7%$ and low coefficient of variation was less than 10% for all analysed samples. The limits of detection and limits of quantification for whole eggs were $1.2{\sim}6.0ng/g$ and $2.3{\sim}9.1ng/g$, respectively. Only one hen's general eggfrom chicken farm in Incheon was detected with the residual fluoroquinolones (Microbiological assay method; 1 of 47 general eggs) ; the range of residual concentration enrofloxacin was 0.12ppm. Those in food stores were detected with the residual fluoroquinolones (Microbiological assay method; 4 of 88 general eggs) ; the ranges of residual concentration enrofloxacin were $0.15{\sim}2.2 ppm$, ciprofloxacin $0.01{\sim}0.06ppm$, and hen's specific eggs (40) in food stores were not detected. For the microbiological assay method of fluoroquinolones in hen's eggs, as the results of comparative analysis, the disc diffusion method with E coli may be a little highly detected for the residual fluoroquinolones.

Mass Production of Resting Egg of Korean Rotifer, Brachionus plicatilis (L and S-type) (한국산 Rotifer, Brachionus plicatilis (L과 S-type)의 내구란 대량생산)

  • Mass production of resting egg of the rotifer, Brachionus plicatilis was carried out in 1 $m^3$ tank for L-type rotifer and in 1 $m^3$ and 4 $m^3$ tank for S-type rotifer. L-type rotifer was fed on concentrated Chlorella + baker's yeast and only baker's yeast for 15 days. S-type rotifer was fed on concentrated Chlorella in 1 $m^3$ tank for 7 days and frozen Chlorella + baker's yeast in 4 $m^3$ for $6\~8$ days. Total number of resting egg and number of resting egg from $10^8$ rotifers and 1 g dry weight diet for L-type rotifer fed on Chlorella + baker's yeast were $54.5{\times}10^6$ eggs, $30.5{\times}10^6$ eggs and $100{\times}10^3$ eggs, respectively. These were higher than those of rotifer fed on baker's yeast only. Total number of resting egg and number of resting egg from $10^8$ rotifers and 1 g dry weight diet for S-type rotifer fed on concentrated Chlorella in 1 $m^3$ tank were $50\~104{\times}10^6$ eggs, $47\~82{\times}10^6$ eggs and $136\~260{\times}10^3$ eggs, respectively. In 4 $m^3$ tank with frozen Chlorella + baker's yeast, these were $149\~567{\times}10^6$ eggs, $36\~123{\times}10^6$ eggs and $131\~338{\times}10^3$ eggs, respectively. This result suggests that S-type rotifer are better than L-type rotifer for the mass production of resting egg of the Korean rotifer.

The Effect of Leukemia Inhibitory Factor on Embryos to the Blastocyst Formation (Leukemia Inhibitory Factor가 배의 배포형성에 미치는 영향)

  • Objective: To determine the effects of leukemia inhibitory factor (LIF) on embryonal development in in vitro culture. Methods: This is designed in vitro model using eggs from mouse. The eggs from mouse were assigned 29 for control group, 53 for 20 ng/ml of LIF, 88 for 40 ng/ml of LIF, 68 for 80 ng/ml of LIF respectively for in vitro fertilization. And 26 fertilized eggs at 2 cell stage from mouse also were assigned. The mouse embryos of all groups were cultured in medium supplemented with LIF in different concentrations, whereas the eggs in control group was cultured in medium without supplement of LIF. Results: At 72 hours culture of eggs from in vitro fertilization, there was a slight increas in rate of embryonal development to morula in both LIF-20 and LIF-40 as results of 64.15% and 75% respectively, while 42.65% in inferior rate of LIF-80, compare with 51.72% in control group. But the difference between these each groups were not significant in statistically ($p{\le}0.05$). And after 96 hours culture of eggs, the rates blastocyst formation was significantly higher in both LIF-20 and LIF-40 as 56.6% and 63.63% than those in control and LIF-80 as 44.83% and 35.29% respectively. On culturing eggs from in vivo fertilization, the rates of blastocyst formation was significantly not only higher as 85% and 81.81% respectively in medium supplemented with LIF-40 and LIF-80 than 42.3% in LIF-20 but also embryonal cell viability were remakedly improved at 96 hours after culture. Conclusion: The LIF in low dose is embryotrophic, but LIF in high dose is embryotoxic on eggs from in vitro fertilization. Whereas on culturing eggs from in vivo fertilization, LIF is more beneficial with dose dependent in high concentration.

  • Environmental Specimen Banks (ESBs) have been established widely in the world since 1979 for monitoring long-term bio-accumulation of environmental pollutants. The ESBs perform the retrospective analysis of representative biological samples collected regularly and store them in cryogenic condition. In Korea, National Environmental Specimen Bank (NESB) was established in 2009. Since then, NESB had prepared the standard operating procedures (SOPs) for the seven kinds of specimens (Red Pine (Pinus densiflora), Common Carp (Cyprinus carpio), and etc.) for monitoring the effect of environmental pollution on the terrestrial ecosystem and river ecosystem. In 2012, NESB added Black-tailed Gull (Larus crassirostris)'s eggs to the list of the environmental specimen for monitoring marine environmental pollution. In this study, we sampled the eggs of the Black-tailed Gull on two islands (Baekryeongdo of the West Sea and Hongdo of the South Sea). Especially, we selected eggs which remained in the early stages of embryo development for certifying the consistent and stable monitoring for environmental pollutants in egg contents. However, it was not considered to classify an order of eggs per clutch in this study. It is known that the concentration variations of pollutants exist among eggs in one clutch. Therefore, it is needed to consider the positions of eggs in the laying sequences to meet the objective of bioaccumulation monitoring. We collected 30 eggs in each site but the amount may be insufficient to achieve storage target (over 2,000 g). Therefore, we need to consider an optimal sampling size.

Distribution of Protease Inhibitors from Fish Eggs as Seafood Processing Byproducts (어류 알의 Protease Inhibitor 활성 분포)

  • To identify and examine the distribution of proteolytic inhibitory activity in crude extracts from fish eggs, and to determine the applicability of these protease inhibitors as anti-degradation agents in surimi-based products and fish meat, we compared the inhibitory activities of various extracts from fish eggs to those of commercial proteases, such as trypsin and papain. We used the optimal conditions for the screening of trypsin activity: 30 ug/uL of 0.1% trypsin and 0.6 mM Na-benzoyl-L-arginine-p-nitroanilide (BAPNA) with a pH of 8.0 at $40^{\circ}C$ for 60 min. The activities of papain and four commercial proteases were investigated after mixing with 100 ug/uL enzymes and 0.3% casein with a pH of 8.0 at $40^{\circ}C$ for 60 min. We performed a screening assay to detect the inhibitory activity (%) of crude extracts from eight species of fish eggs against the target proteases trypsin and papain. The assay revealed a wide distribution of trypsin and papain inhibitors in fish eggs. The specific inhibitory activities (11.6.28.6 U/mg) of crude extracts from fish eggs against trypsin and BAPNA substrate were higher than that (0.64 U/mg) of egg whites, used as a commercial inhibitor. The inhibitory activities of crude extracts from fish eggs against trypsin, and of egg whites against casein substrate (1.94.4.51 U/mg), were higher than those of papain (0.24.1.57 U/mg) and commercial protease (0.04.0.32 U/mg). The extracts from fish eggs were rich in protease inhibitors that exhibited strong inhibitory activity against trypsin, a serine protease, and papain, a cysteine protease.

Effect of Age of Laying Hens and Grade of Egg Shell Abnormality on Internal Egg Quality (산란계 주령과 계란 난각 형태이상 정도가 내부품질에 미치는 영향)

  • This study was carried out to investigate the effects of age of laying hens and grade of shell abnormality on internal egg quality. At each week, 1,000 eggs were randomly selected from 21,000 eggs laid by Hy-Line hens (100,000 birds) aged from 56 to 62 wk. The samples were divided into normal and externally abnormal eggs by an official egg grader. Then, egg quality including egg weight, egg white height, Haugh unit and yolk color was evaluated from each normal and abnormal eggs. The production rate of externally normal and abnormal eggs was 81.22 and 18.78%, respectively. No difference found in the production rate by the increase of hen's age, but Haugh unit of eggs from hens of 60-wk-old was higher (p<0.05) than that of 56-wk-old. When the same age groups were compared, egg white height and Haugh unit were lower in eggs which were externally abnormal. When the grade of abnormality increased, egg white height and Haugh unit were decreased further (p<0.05). Among the externally abnormal eggs, malformation showed the lowest Haugh unit. In conclusion, the shell abnormality of eggs laid by hens aged 56 to 62 wk was closely related with internal egg quality. Therefore, the grading external egg quality in the present egg grading system is directly related with the internal egg freshness.

Analysis of lipid composition and fatty acids in poultry eggs -cage system, open barn system's hen egg, moscovy duck's egg mallard's egg- (난류(卵類)의 지질성분 및 지방산 조성 분석 - 달걀, 기러기알, 청둥오리알 -)

  • The lipid and fatty acid composition of various poultry eggs were examined after extracting yolk oils from the eggs of caged hen, open barn-reared hen, mallard, and muscovy duck. Total lipid content in 100 g of each edible portion was the highest in muscovy duck egg followed by open barn-reared hen, mallard, caged hen, and natural hen eggs. The triglyceride contents in the egg were in the order of caged hen, open barn-reared hen, natural hen, mallard, and muscovy duck. The cholesterol contents in total lipid were the highest in caged hen and muscovy duck eggs(4.5%), and the lowest in mallard(3.3%). Caged hen eggs had higher neutral lipid ratio among total lipid, but had lower ratio of glycolipid and phospholipid compared with those of the open barn-reared hens. For fatty acid composition, linoleic acid was the highest in the muscovy and linolenic acid was the highest in open barn-reared hen eggs. The contents of arachidonic acid and Eicosapentaenoic acid(EPA) were the highest in muscovy eggs. On the other hand, the content of Docosahexaenoic acid(DHA) was the highest in muscovy duck eggs.
