Acute/Subacute Toxicity of Nano Calcium (나노 칼슘의 급성/아급성 안전성 평가)

  • The objective of this study was to obtain data on the safety-in-use of nano calcium as a dietary supplement by assessing its acute and subacute oral toxicities in female and male Sprague-Dawley rats. A single oral dose of 5,000 mg/kg of nano calcium did not result in mortality or significant changes in the general behavior and gross appearance of the internal organs of rats. For subacute toxicity study, nano calcium was administered orally at a dose of 1,000 mg/kg daily for 14 days. There were no significant differences in organ weights between control and treated groups of both sexes. Hematological analysis and blood chemistry revealed no toxic effects of nano calcium. Pathologically, neither gross abnormalities nor histopathological changes were observed. These results show that nano calcium possesses very low toxicity as indicated in a rat model.

The Daily Dose and Uses of Pinellia Tuber, Vinegar and Eggshell of Goju-tang in Treatise on Cold Damage Diseases (《상한론(傷寒論)》고주탕(苦酒湯)의 반하(半夏)와 고주(苦酒), 계자각(鷄子殼) 용량과 용도)

  • Objectives : The purpose of this study was to clarify the daily dose and uses of Goju-tang in Treatise on Cold Damage Diseases. Methods : We compared major editions of Treatise on Cold Damage Diseases with the original text of Supplement to the Essential Prescriptions Worth a Thousand Gold and chose the most appropriate text. If none of the existing texts seemed reasonable, We compared texts item by item. When there is nothing adequate even item by item, it was newly modified. These results were compared with currently distributed Pinellia Tuber and standards of eggs. Results : Goju-tang consisted of Goju(vinegar), Pinellia Tuber and an eggshell. Generally, a daily dose was extracted at once and taken 3 times a day, but in Goju-tang, 4 pieces of Pinellia Tuber and one-third Sheong of Goju(solvent) were used, which were one third of average volume. Pinellia Tuber was used cut in small pieces. And it was boiled gradually by low heat. Goju was mainly to reduce the pungency of Pinellia Tuber by cutting raphides of calcium oxalate. The Pot preparing medicines was eggshell which was chosen by its small size, because the volume of Pinellia Tuber and Goju was one-third of average volume. Since an egg shell is consisted of protein, it didn't react with vinegar(Goju). Conclusions : The 4 pieces of Pinellia Tuber and one-third Sheong of Goju(Vinegar) in the Goju-tang were one-third of average dose, and the eggshell was Pot preparing medicines.

Predicting ESP and HNT effects on the mechanical properties of eco-friendly composites subjected to micro-indentation test

  • The main goal of the present study was to assess the effects of eggshell powder (ESP) and halloysite nanotubes (HNTs) on the mechanical properties of abaca fiber (AF)-reinforced natural composites. For this purpose, a limited number of indentation tests were first performed on the AF/polypropylene (PP) composites for different HNT and ESP loadings (0 wt.% ~ 6 wt.%), load amplitudes (150, 200, and 250 N), and two types of indenters (Vickers or conical). The Young's modulus, hardness and plasticity index of each specimen were calculated using the indentation test results and Oliver-Pharr method. The accuracy of the experimental results was confirmed by comparing the values of the Young's modulus obtained from the indentation test with the results of the conventional tensile test. Then, a feed-forward shallow artificial neural network (ANN) with high efficiency was trained based on the obtained experimental data. The trained ANN could properly predict the variations of the mentioned mechanical properties of AF/PP composites incorporated with different HNT and ESP loadings. Furthermore, the trained ANN demonstrated that HNTs increase the elastic modulus and hardness of the composite, while the incorporation of ESP reduces these properties. For instance, the Young's modulus of composites incorporated with 3 wt.% of ESP decreased by 30.7% compared with the pure composite, while increasing the weight fraction of ESP up to 6% decreased the Young's modulus by 34.8%. Moreover, the trained ANN indicated that HNTs have a more significant effect on reducing the plasticity index than ESP.

Microbial Hygienic Status of Poultry Meats and Eggs Collected at the Public Markets in Seoul and Kyung-gi Regions in 1996 (1996년도 서울${\cdot}$경기지역에서의 시판계육과 계란에 대한 미생물학적 위생실태)

  • To determine the actual hygienic status of domestic chicken meats sold in public markets (conventional markets and department stores), microbial contamination levels (Total cells, Coliforms and Staphylococcal cells) and zoonotic pathogens (Salmonella species, Campylobacter species, Listeria species, and Staphylococcus aureus) isolation tests were conducted. Chicken meats and eggs tested were collected from the conventional markets (Si-Jang) and department-stores located in Seoul and Kyung-gi regions in 1996. In total cells and coliforms contamination tests, chicken meats sold in department stores were much lesser contamination status than those of Si-Jang, but staphylococcal cells level was much more higher than that of conventional markets. Salmonella isolation frequency was investigated as $68.8\%$, but Campylobacter jejuni and Listeria monocytogenes isolation frequency were appeared both $64.0\%\;and\;63.3\%$. In case of eggs sold in public markets, one of S. gallinarum strain $(0.7\%)$ was isolated only on the egg-shell part among the four-hundred and fourty-six. In comparison with foreign imported chicken meats, there were no big differences in microbial contamination status. On the other hand, both Salmonella and L. monocytogenes were isolated only in the chicken wings from Korea and China, but not from U.S.A. This data suggest that more hygienic control system in order to produce the safe and hygienic chicken meats and eggs is need in our country as soon as possible.

Effects of Supplementary Multiple Probiotics or Single Probiotics on the Performance, Intestinal Microflora, Immune Response of Laying Hens and Broilers (혼합 또는 단일 생균제가 산란계와 육계의 생산성, 소장내 미생물 균총 및 면역 체계에 미치는 영향)

  • This study was conducted to investigate the effects of dietary supplementation of multiple probiotics on the performance, small intestinal microflora and immune response in laying hens and broilers. In Exp.1, a total of 800, 82 wk old Hy-line Brown$^{(R)}$ laying hens were assigned to one of the following five dietary treatment; Control, Antibiotics (avilamycin 6 ppm), Probiotics; PB-M (Micro-ferm$^{(R)}$ 0.2%), PB-L (Lacto-sacc$^{(R)}$ 0.1%), PB-Y (Y University probiotics 0.2%). Each treatment was replicated eight times with 20 birds in each replicate and two birds were housed in each cage. Twenty birds units were arranged according to completely randomized block design. Feeding trial lasted 6 wk under 16 h lighting regimen. The Exp. 2, was conducted with a total of 1,000 broilers chicks (Ross$^{(R)}$). They were divided into five treatments, same as those of Exp. 1. Birds were fed starter (0~3 wk) and grower (4~5 wk) diets. Each treatment was replicated four times with 50 birds per pen comprising of deep litter. In Exp. 1, egg production parameters, such as hen-day and hen-house egg production, egg weight, broken and soft shell egg production, feed intake and feed conversion were not significantly different among treatments. However, strength and thickness of eggshell were significantly (P<0.05) different. Among the probiotics, PB-Y showed the highest strength and thickness of eggshell. Eggshell color, egg yolk color and Haugh unit were not significantly influenced. In Exp. 2, overall weight gain (0~5 wk) and mortality were not significantly different among treatments. However, weight gain of birds from PB-Y treatment during starter (0~3 wk) was significantly lower than the birds from Control and Antibiotic treatment. During the whole period (0~5 wk), birds from Antibiotics treatment had higher feed intake and Production Index (PI) and lower feed conversion than birds from Control treatment. Probiotics treatments were not significantly different from the Control on feed intake and feed conversion. In Exp.1, there were significant (P<0.05) differences in leukocytes parameters, such as white blood cell (WBC), hetrophil (HE), lymphocytes (LY), monocyte (MO), eosinophil (EO) and stress index (SI; HE/LY) in the blood of layers. Birds from Antibiotics and probiotics treatments tended to increase these parameters. In Exp. 2, however, only SI was significantly (P<0.05) decreased in Antibiotics treatments. Concentration of serum immunoglobulin (IgG) were higher (P<0.05) in PB-M and PB-Y treatments when compared with Control treatment in Exp. 1. The population of E. coli significantly (P<0.05) decreased in birds from Antibiotics, PB-L and PB-Y treatments when compared with birds from Control treatment in Exp. 1. Metalbolizability of crude fat decreased significantly (P<0.05) in birds from probiotic treatments in Exp. 2. It was concluded that the response of probiotics on the productivity of layers and broilers were different. Probiotics increased strength and thickness of eggshell in layers, and decreased feed conversion and increased PI in broilers. Leukocytes and IgG tended to increase by supplementation of antibiotics and probiotics in layers. Intestinal E. coli tended to decrease in layers. Digestibility of crude fat of diet decreased in probiotics treatments broilers. Parameters of blood and microbial were more sensitive in layers than broilers.

Comparison of F1 and F2 hybrid on authorized silkworm variety (누에 장려품종간 교잡누에의 실용형질 비교)

  • This study was conducted to consider practical use of F2 hybrid and intervariety crossing for the improvement of silkworm eggs export and silkworm eggs manufacture. Now Kumokjam and Baegokjam come into wide use to sericulture farmers but comparison test of F1, F2 hybrid and intervariety have not been done. We weighed against the productivity among economical character. While larval period of F2 among them is most long time, pupation percentage and cocoon yield per 10,000 3rd molted larvae are lower than intervariety. With comparing F2 and intervariety, single cocoon weight of F2 is lower 9 ~ 21% than intervariety, and cocoon shell weight of F2 was lower 10 ~ 18% than intervariety. By number of cocoons per liter, F2 hybrid was lower and more irregular size than intervariety. In coparison with number of egg laid, F2 hybrid Baegokjam was 655 ea (14%) and Kumokjam 625 ea (40%) more than F1 hybrid. Also, F2 hybrid for filament grade went down than F1 hybrid. The advantage of egg production in F2 hybrid can reduce costs because there is no necessity for sex discrimination and germination control. But besides increase of number of egg laid, the productivity including economical characters was greatly lowered. Except for being a lot of laying eggs, cocoon productivity of the F2 was greatly reduced by degeneration of practical traits. Therefore the further studies are needed in order to increase utilization value of the F2 hybrid silkworm.

Dietary Effect of Agaricus bisporus Feeds on Characteristics of Eggs (계란의 품질특성에 미치는 양송이 사료의 식이효과)

  • Dietary effects of Agaricus bisporus powder (ABP) on the quality characteristic of eggs from hens (Hy-Line) were investigated. Experimental groups (100 hens/group) were divided into normal control group (Control), 0.3% ABP supplemented diet group (M-0.3) and 0.6% ABP supplemented diet group (M-0.6), then fed for 12 weeks. There were no significant differences in shell thickness, while Haugh unit in M-0.3 and M-0.6 groups were 18.3$\sim$27.6% higher than that of control groups. Color $L^*$ and $b^*$ values of egg albumen and yolk in the M-03 and M-0.6 groups were lower whereas $a^*$ values were higher than those of NC group. In the yolk of M-03 and M-0.6 groups compared with control groups, the levels of total phospholipids and HDL-cholesterol were 4.5$\sim$13.9% and 86.1$\sim$87.0% higher, respectively, while the levels of total cholesterol, triglyceride and LDL-cholesterol were 29.4$\sim$31.9%, 51.9$\sim$52.4% and 49.1$\sim$53.5% lower than those of control groups, respectively. There were no significant differences in fatty acids. In the result of sensory evaluation, gory odor of raw eggs in the M-03 and M-0.6 groups was slightly lower compared with control groups. These results suggest that the feed containing mushroom of Agaricus bisporus can be used to reduce the egg yolk cholesterol content and gory odor from raw egg, and also to increase the phospholipids as a functional feed. Further studies on the mechanism are required.

Intestinal Permeability of Oyster Shell Calcium with Different Particle Sizes (패각 칼슘 입자 크기에 따른 흡수율)

  • In this study, we examined the ionization rate and permeability of nanocalcium prepared from oyster shells with various particle sizes. Four particle sizes of the calcium samples were prepared by centrifugation according to their density disparity in alcoholic solution: NC (normal calcium), C-1 (supernatant of 1,000 rpm), C-2 (supernatant of 2,000 rpm), and C-3 (supernatant of 3,000 rpm). Particle sizes of NC, C-1, C-2, and C-3 were $2,280.3{\pm}64.3nm$, $521.3{\pm}83.3nm$, $313.9{\pm}29.5nm$, and $280.0{\pm}3.4nm$, respectively. C-3 showed a slight increase in ionization rate compared with the other calcium samples, but their differences were not significant. Dialysis membrane-employed analysis showed that nanocalcium permeability increased as its particle size smaller; 32% of C-3 nanocalcium was transported to the outside of the membrane, whereas C-1 showed a 25% transport rate. We determined the permeability of the nanocalciums by using rat intestinal sacs, in order to provide different intestinal environments depending on pH level. Nanocalcium generally showed a higher permeability at pH 7, which represents an ileum environments compared to the duodenum and jejunum environments at pH 4.2 and pH 6.2, respectively. However, C-3 calcium showed the highest permeability, followed by C-2, C-1 and NS calciums. This result shows that the size of calcium positively affected its permeability in the intestinal sac. Taken together, nano-sized calcium derived from discarded oyster shell shows improved permeability in intestinal environments.

Malacological Studies on Parafossarulus manchouricus(Gastropoda: Prosobranchia) in Korea (한국산(韓國産) 왜우렁(Parafossarulus manchouricus)의 패류학적(貝類學的) 연구(硏究))

  • Five different populations of Parafossarulus manchouricus (Chongpyung, Chinju and Kunsan, Korea; and Japan and Taiwan), a population of Bitbynia (Gabbia) misella (Gongju, Korea) and two different populations of Bithynta tentaculata (Michigan, U.S.A. and Bodensee, Germany) were compared in regard to eff-laying characteristics, morphology, chromosome cytology, natural infections of parasites and ecology of habitats. A satisfactory culture method was devised for laboratory rearing of the snails. Tropical fish food (Terra SML) and powdered green leaves (Ceralife) were used as the main food sources for the snails. Benthic diatoms such as Navicula and Gomphonema from the periphyton were also essential for satisfactory growth, especially for the baby snails. The aquaria were stabilized with small stones from a local stream. Young P. manchouricus snails grew to adult size in about 54 days after hatching. They laid eggs 150-156 days after hatching. The whole cycle (birth to egg-laying) took approximately 5 months. The three species of bithyniid snails are iteroparous and lay eggs once a year. There were no major morphological differences in the shells of genera or subgenera studied here. They did exhibit the following rather minor differences. The shell of Parafossarulus has spirally raised ridges, and its apex is usually eroded; the other two genera lack these characteristics. The shell of B. (Gabbia) misella is small, nor exceeding 7.5 mm in length, while the shells of the other two species are larger, being more than 10 mm in length. Scanning electron microscopy (SEM) of the protoconch of P. manchouricus reveals nearly smooth sculpture with small, low, spiral wrinkles. This sculpture is quite different from that of the Hydrobiidae, a family to which the bithyniids are frequently assigned. Scanning electron microscopy of the radulae of the three bithyniid species showed that their radular morphologies are very similar, but there are some small differences, which may be species-specific. There were some statistical differences in shell heights between the Korean and the other populations of P. manchouricus, and between this species and the other two bithyniids as well. The shell differences between the several populations of Korean P. manchouricus may be related to environment. Edtails of the chromosome cycle of these bithyniid snails are similar to those reported for other snails. No specific differences were observed in the chromosome cycle between the various species and populations of snails employed in this study. Reporred for the first time in molluscs are two darkly stained "nucleolar organizers" during pachyterne stages of meiosis. Two different chromosome numbers were observed in the three bithyniid species: n=17 in B. tentaculata and P. manchouricus, and n=18 in B. (G.) misella. no sex chromosomes or supernumerary chromosomes were seen. There were no morphological differences in karyotypes of three Korean strains of P. manchouricus. The infection rates of cercariae of Clonorchis sinensis in Chinju and Kunsan strains of P. manchouricus were 0.14% and 1.25%, respectively. However, Clonorchis cercariae were found in Chongpyung strain of P. manchouriceu and Gongju strain of B. (G.) misella. The habitats of P. manchouricus around Jinyang Lake were relatively clean without any heavy pollution of aquatic microorganisms and organic materials during the period of this study. The levels of dissolved oxygen (D.O.) and biochemical oxygen demand (B.O.D.) of the water specimens sampled from the study areas ranged from 6.0 to 9.6 ppm and from 0.4 to 1.6 ppm, respectively. Eight metalic constituents from the water samples were also assayed, and all metalic ions detercted were remarkably low below the legal criteria. However, calcium ion in the water samples from the habitats of P. manchouricus was considerably higher than others.

Effects of Dietary Silicate Minerals on Ammonia Emission from Excreta and Performance of Laying Hens (규산염계 광물의 급여가 산란계분의 암모니아 발생과 생산성에 미치는 효과)

  • This study was conducted to investigate the effects of dietary silicate minerals on ammonia emission from the excreta and performance in laying hens. A total of one thousand and twenty 49 week-old Lohmann brown layers were divided into three treatment groups consisting two replicates of 170 birds each. The experimental diets were containing three kinds of silicate minerals (Sand, Na-bentonite and SY feldspar) added at the level of 2% by top-dressing. Ammonia emission from the excreta was significantly decreased (p<0.05) by the supplementation of Na-bentonite or SY feldspar. N concentration of the excreta was significantly higher (p<0.05) for the hens fed Na-bentonite or SY feldspar compared to those fed sand. However, no significant differences were observed in feed intake and laying performance with over 80% laying rate observed for all the treatments. Egg shell strength was diminished when SY feldspar was added to the diet. As a whole, the results obtained from the experiment indicated that supplementation of Na-bentonite or SY feldspar added at 2% to the laying hen diet should not affect laying performance but help to reduce ammonia from the excreta at the same time.
