Changes in Biochemical Components of the Ovary and the Trunk Tissues Including the Digestive Organ Associated with Gonadal Development of the Female Octopus ocellatus (주꾸미 (Octopus ocellatus) 의 생식소 발달에 따른 난소와 소화기관을 포함한 몸통부위 조직의 생화학적 성분변화)

  • Gonadosomatic index, condition index and reproductive cycle with the gonadal development of the female Octopus ocellatus were investigated by histological observations and morphometric data, from January to December, 2000. And changes in biochemical components of the ovary and the trunk tissues including the digestive organ associated with gonadal development were studied by biochemical analysis from January to October, 2001. The specimens were collected at the coastal waters of Buan, Jeollabuk-do, Korea, from January 2000 to October 2001. O. ocellatus is a dioecious organism. The gonad of O. ocellatus locates medially in posterior region of the body. Morphology of the ovary shows round and oval in shape, the average diameter and external colour of ripe ovary was 32 mm and semitransparent light brown in colour. As the ovary was getting mature, transparent elongated eggs covered with chorion were present in the ovarian cavity. Monthly changes in the gonadosomatic index (GSI) showed a similar pattern with those of the condition index. The GSI and condition index began to increase in March and reached the maximum in April. And then, their values decreased from May and reached the minimum in September. Reproductive cycle of O. ocellatus can be categorized into five successive stages: early developing stage (September to December), late developing stage (November to March), ripe stage (March to May), partially spawned stage (April to June), and degenerative/resting stage (June to October). Follicle cells attached to an oocyte were involved in vitellogenesis in the cytoplasm of the vitellogeneic oocyte and formation of chorion (secondary egg membrane) of the ovarian eggs. Spawning occurred between April and June. The spawning period was once a year and the peak took place between May and June. This species belongs to semelparity. According to changes in biochemical contents of the ovary and the digestive organ, monthly variations of moisture, total protein, total lipid and glycogen contents (%) in the ovary showed a negative correlationship with those of the trunk tissues including the digestive organ. Accordingly, it is assumed that the ovary only may be received nutrient supply (total lipid content) for gonadal development from the trunk tissues including the digestive organ (r = -0.55, p < 0.05).

  • The experiment was conducted to understand the effect of HCG upon ovulation induction and egg maturation in the sevenband grouper, Epinephelus septemfasciatus, as a part of the research for its resources management and seed production. Three different size groups of sevenband groupers showing 300~400g, 1,500~2,000g and 2,500~4,000g, were collected from June to August, 1996. They were treated two or three times with HCG (500~1,000 IU/kg). Gonadal development of the sevenband groupers were examined using the paraffin section method and then stained with haematoxylin and eosin. When smaller groups of sevenband grouper having 300~400g, 1,500~2,00g and 2,500~3,200g in weight were treated two or three time with HCG 9500~1,000 IU/g), their gonads contained oogonia, oocytes with the size of 20~30${\mu}{\textrm}{m}$, and stroma tissues. However, in case of two HCG treatments with a dosage of 1,000 IU/kg on sevenband groupers of 3,400~4,000g, both healthy eggs of $800\mu$m in diameter and regressing eggs were ovulated. A sevenband grouper of 4,000g obvulated 4,252 eggs in total. Of these, healthy and regressing eggs turned out to be 20.8% and 79.2%, respectively. In addition, previtellogenic oocytes of around $50\mu$m, oocytes of 20~$30\mu$m, and ovulation trace were also observed in the gonad.

Effects of young persimmon fruit powder on rice cookie quality (어린 감 과실 분말 첨가가 쌀쿠키의 품질특성에 미치는 영향)

  • This study was conducted to develop rice cookies added with young persimmon fruits. Effects of varying amounts (0-12%) of the fruit powder from a young astringent persimmon fruit (picked at July) on the quality characteristics of rice cookies were studied. Ingredients [rice (Oryza sativa subsp. japonica 'Ilpum') flours, persimmon (Diospyros kaki Thunb. 'Cheongdobansi') powder, sugar, butter, salt, baking powder, egg] were mixed, cut (thickness 3 mm, diameter 35 mm), baked at $170-180^{\circ}C$ for 9 min, cooled, and packaged in polyethylene/nylon bags. The loss rate and spread factor after baking of cookies increased and then decreased with an increase in the amount of fruit powder added. The moisture content and color values ($L^*$ and $a^*$) of cookies decreased with an increase in the amount of the persimmon powder added. Phenolic compounds content and DPPH radicals scavenging activity increased with an increase in persimmon powder content; in particular, the DPPH activity of the cookies sharply increased after the addition of 3% persimmon powder. These results suggest that the addition of the young persimmon fruit powder affected the quality characteristics of rice cookies and this fruit powder (approximately 3%) can be utilized as an additive during rice cookie processing.

Studies on Ecology and Injury Characteristics of Japanese Suniperus Bark Borer, Semanotus bifasciatus Mostschulsky (측백하늘소의 생태 및 가해특성에 관한 연구)

  • This study was conducted to investigate the bionomics, host range and damage aspect of Japanese juniperus bark borer, Semanotus bifasciatus M., which shoves the severe damage for the Juniperace in Chonnam province, recently. Host plants were Juniperus virginiana, J. chinensis var. kaizuka, J. chinensis, Biata orientalis var. nepalensis, Chamaccyparis ostus and Thujopsis dolabrata. Out of those, J. chinensis var. kaizuka is newly investigated in this study. Damage rate in each area was $16.5\%$ in Kwangju, $4.25\%$ in Damyang ana $6.5\%$ in Hwasoon. Adults appear once a year, with a peak at late March to early April. Adults emergence during the day showed the peak at 13 to 15 hours and were influenced in the maximum temperature in a day. Attack direction of larva after invasion in the stem was $62\%$ toward base, $22\%$ toward tip and $16\%$ toward horizon, and the damage in each DBH (Diameter of breast height) was the greatest at $30\~40mm$. At period of each stage, egg was $15.8\~19.7$ days, larva was $l12\~126$ days and pupa was $15\~21$ days. Longevity of adult was 19 days for female and 16 days for male.

The Egg Morphology and Larval Development of the Pipefish, Syngnathus schelegeli Kaup (실고기, Syngnathus schlegeli 난(卵)의 형태(形態) 및 산출(産出) 자치어(仔稚魚)의 형태(形態) 발달(發達))

  • The pipefish Syngnathus schelegeli was reared in the laboratory from May to June 1991 and observed the morphology of eggs and larvae squeezed from the parent fish(male). The diameter of inseminated eggs ranged from 0.72 to 1.01 mm (n=50), and yolk in yellow color were found in the eggs. The newly beared larvae were 10.9 mm in average standard length and had 59~60 myomeres. In 6 days after bearing, the post larvae attained 13.8 mm in average standard length and the low jaw was developed. The larvae of 14.1 mm in average standard length (8 days after bearing) had 40~42 fin rays in dorsal fin, 9 in caudal fin and 13~14 in pectoral fin. The juvenile of 14.7 mm in average standard length (10 days after bearing) had the well elongated snout along with the opercular and caudal fin similar to adult stage's.

On the Maturity and Spawning of the Greenling, Hexagrammos grammus(Temminck et Schlegel) (노래미, Hexagrammos agrammus(Temminck et Schlegel)의 성성숙과 산란)

  • Sexual maturation and spawning of the spottybelly greenling, Hexagrammos agrammus were histologically studied under photomicroscopy by considering changes in gonadosomatic index, hepatosomatic index and fatness, egg diameter composition, first sexual maturity and fecundity. The fish samples were collected monthly at the coastal area of Tongbaeksom, Pusan, Korea from July 1991 to July 1992. The gonadosomatic index(GSI) increased in September and reached the maximum value(female 4.31, male 1.61) in November when the gonad was mature and ripe. The values were decreased suddenly during the spawning season from December, and declined from January to August. The annual variations of hepatosomatic index(HSI) appeared to be correlated with those of GSI in female but were not significantly correlated in male. HSI in female began to increase in autumn with the increase of GSI, and reached the maximum in winter when the ovary was mature. Percentages of first sexual maturity in female and male fish were 50 % in 11.0~11.9 cm and 100 % in 13.0~13.9 cm groups. Both sexes participated in reproduction from one year old. H. agrammus was considered as a polycyclic species and spawns 3 times or more in the spawning season. Number of total and mature eggs in the absolute fecundity were proportional to standard length and body weight, respectively. Number of total and mature eggs in relative fecundity were also proportional to standard length, but rather decrease with increasing of body weight. Fatness coefficients reached the maximum value(female 15.32, male 15.14) in September(growing stage), and the values were sharply decreased after spawning. Thereafter, fatness values were gradually increased, therefore, the monthly changes in fatness coefficient closely correlated with the reproductive cycle. Sex ratios of female and male sexes of this species are showed 54.18 %, 45.82 %, respectively.

Maturity and Spawning of Spinyhead Sculpin, Dasycottus setiger (Bean) in the East Sea, Korea (동해안 고무꺽정이, Dasycottus setiger (Bean)의 성숙과 산란)

  • Maturity and spawning of spinyhead sculpin, Dasycottus setiger were investigated using samples collected by the eastern sea danish seine and gillnet in the East Sea of Korea from January to December 2004. We analyzed monthly changes in maturity stages, gonadosomatic index (GSI), egg diameter, fecundity, total length at 50% group maturity, and sex ratio. GSI began to increase in August, reached a maximum between September and November, and decreased in December. The spawning period was August to December, and the main spawning period was September to November. The fecundity ranged from 6,210 eggs at 22.3 cm (TL) to 15,898 eggs at 32.2 cm (TL). The relationship between total length (TL) and fecundity (F) was $F=25.475TL^{1.818}$ ($R^2=0.761$), and F increased with TL. The TL at 50% group maturity was estimated to be 23.4 cm. The sex ratio was 1 : 1.3 (female : male), indicating the male population was seem to be slightly predominant.

Spawning Character and Early Life History of the Endangered Korean Dwarf Loach, Kichulchoia brevifasciata (Teleostei: Cobitidae) (멸종위기종 좀수수치 Kichulchoia brevifasciata의 산란기 특징 및 초기생활사)

  • Eggs development and early life history of the endangered Korean dwarf loach, Kichulchoia brevifasciata (Teleostei: Cobitidae) was investigated to provide basic information regarding biological characteristics and restoration. Adult fish specimens were sampled using a spoon net at Geurnsan-myeon, Goheung-gun, Jeollanam-do, Korea from June to July 2011. Since, spawning characteristics were analyzed, and females were induced to spawn by injecting Ovaprim (0.5 mL/kg) and their eggs were artificially fertilized with sperms by the dry method in the laboratory. Total length of mature female were 46~76 mm with GSI $9.6{\pm}3.77%$, and total length of mature male was 42~52 mm with GSI $3.5{\pm}1.04%$. Sex ratio (♂/♀) was 0.10, and there were no secondary sexual characteristics. The number of mature eggs was averaged $60{\pm}28.7$ per female. The lemon yellow eggs were slightly adhesive $1.46{\pm}0.07mm$ in diameter. The embryo hatched approximately 66 h after fertilization at $25^{\circ}C$, and the hatched larvae were averaged $5.5{\pm}0.07mm$ in total length (TL). At 6 days after hatching, the larvae averaged $9.0{\pm}0.29mm$(TL) and their yolk sac was completely absorbed. At 17 days after hatching, they entered the juvenile stage and reached $12.6{\pm}0.24mm$ (TL). At 80 days after hatching, the band patterns and external form of the juveniles were similar to those of adults, and they averaged $33.0{\pm}2.19mm$(TL).

Structural Changes of Zona Pellucida Surface of Immature, In vivo and In vitro Matured Canine Oocytes Using Scanning Electron Microscopy

  • Zona pellucida (ZP), a primarily representative coat of mammalian egg and embryo, has an extremely heterogeneous morphology during different developmental stages. The objective of the present study was to compare the morphological changes of the ZP surface of immature, in vitro and in vivo matured canine oocytes by using scanning electron microscopy (SEM). Canine ovaries were collected from local veterinary hospitals to recover immature oocytes. The ovaries were sliced and the released cumulus oocyte complexes (COCs) were washed with TL-HEPES. The selected COCs were randomly divided into two groups, first group was processed immediately at immature state and the second group was processed 72 h after in vitro maturation, and compared with in vivo derived oocytes. Oocytes were fixed, critical point dried and examined under SEM. The diameters of oocyte and outer holes of the ZP were measured on a total of 249 oocytes; the results were analyzed using One-way ANOVA. Our results showed that, the diameter of immature oocytes significantly differed (p < 0.05) from that of in vivo matured oocytes ($79.60{\pm}0.77{\mu}m$ vs. $101.46{\pm}1.07{\mu}m$, respectively). Similarly, a significant difference (p < 0.05) in the diameters between those of in vitro and in vivo matured oocytes were found ($79.51{\pm}2.36{\mu}m$ vs. $101.46{\pm}1.07{\mu}m$, respectively). Moreover, the diameters of the outer holes of the ZP were significantly (p < 0.05) larger in in vivo matured ($1.48{\pm}0.42{\mu}m$) than in vitro matured for 72 and immature oocytes ($1.10{\pm}0.16$ and $0.43{\pm}0.12{\mu}m$, respectively). Taken together, these data indicates that the ZP surface is related to oocyte maturity in canine.

A study on change of reproductive biology and fishing business of snailfish, Liparis tanakae in Korea (우리나라에 분포하는 꼼치(Liparis tanakae)의 생식생태와 어업경영에 관한 연구)

  • This study analyzed the reproductive biology, fishing characteristics and changes in fishing business of Liparis tanakae, snailfish collected from September to March. It was the period when they were mainly caught from 2018 to 2020. The average length was generally small in September and October and was large in January and February. The average body weight was generally around 1,500 g and the average body weight in autumn was lower and in winter was higher. The sex ratio of male and female was 0.40:0.60 (��2 test, p < 0.05). The spawning period was estimated from October to February and the main spawning period was from December to February through the GSI. The egg diameter of matured staged female L. tanakae was 0.11-1.48 mm, which was the main spawning period and the relationship between body weight and fecundity was F = 1849TL0.1093 (r2 = 0.2401). The monthly catch of L. tanakae was high from November to February, the time of migrating to the coastal area. Coastal gillnet fishery showed the highest percentage of all fisheries catching Liparis spp. Liparis spp. were caught at a high rate in winter in Chungnam, Jeonbuk, Jeonnam and Gyeongnam region, and revenue and cost was increased since 2017. Assuming a situation where there is no catch of Liparis spp., the fishing profit that can be obtained was the highest in Gyeongnam region and the dependence on fishing of Liparis spp. by coastal gillnet fishery was high.
