The Studies on Pregnancy after Transfer to Recipient with Blastocyst Derived In Vitro in Korean Cattle (한우 유래의 체외수정란의 이식후 임신에 관한 연구)

  • These studies were carried out to establish an effective in vitro embryo transfer methods by analyzing several factors. The base media were TCM-199 solution for in vitro maturation(IVM) of bovine follicular oocytes and Fer-TALP solution for in vitro fertilization(IVF) and CRlaa medium for in vitro culture(IVC). IVC used the fertilized oocytes of 24-hr culture (day 1)after IVF. Embryos were cultured in drop-culture that contained 25 embryos per 10${mu}ell$. Blastocysts cultured for 7 to 9 in vitro were transferred to recipients. The results obtained were as follows: 1. The pregnancy rate according to different region of embryo transfer were 33.8%, 48.1%, 45.0% and 35.3% respectively. 2. The pregnancy rate according to the parity of recipient when embryos were transferred to nulliparous (42.9%) was higher than that of 1∼3nd parlous(36.9%), however there were not show significant difference each other. 3. According to the stage of blastocyst, the pregnancy rate when middle blastocysts (MB) (45.5%) were transferred to recipients were higher than that of late blastocysts (LB) (41.0%). 4. When IVF-derived blastocysts cultured for 7 to 9 day were transferred to recipients, the pregnancy rate was higher 7 day of blastocyst than that of 8 day or 9 day of blastocyst. The results of embryo transfer according to the regions, the parity of recipient and the development stage showed that blastocyst formed for 7day transferred to nulliparou were higher pregnancy rate than others.

Isoflavone Content and its Relationship with Other Seed Quality Traits of Soybean Cultivars Collected in South Korea

  • The 117 soybean cultivars were collected from nine provinces in Korea, and various seed quality traits along with isoflavone contents were evaluated to elucidate their relationship. The 100-seed weight of the black soybean (31.2 g) was significantly higher (p<0.05) than yellow soybeans (28.6 g). The composition of genistein, daidzein, and glycitein accounted for 75.8, 22.8, and 1.4 % of total isoflavone in yellow soybean cultivars, while their compositions in black soybeans were 58.5, 39.7, and 1.8%, respectively. The mean contents of total isoflavone in yellow and black soybean were $l,561.6{\mu}g\;g^{-1}\;and\;l,018.3{\mu}g\;g^{-1}$. The isofalvone content showed significant variation among cultivars when classified by the seed size. In the yellow soybeans, total isoflavone content was higher in small size soybean cultivars $(1,776.0{\mu}g\;g^{-1})$ and medium size soybean cultivars $(1,714.3{\mu}g\;g^{-1})$ compared to large size ones $(1,518.5{\mu}g\;g^{-1})$. Genistein content was proved as the major factor determining the relationship between isoflavone content and 100-seed weights (r =-0.206*). Daidzein and glycitein, however, showed no significant relationship with the 100-seed weights. Isoflavone content was not significantly correlated with color parameters L (lightness) and a (redness) values, but color parameter b (yellowness) was positively correlated with glycitein (r=0.264*) in the yellow soybeans, while its negative correlation between daidzein (r=-0.245*) and total isoflavone (r=-0.256*) were observed in black soybeans. However, these findings suggested that the seed color value may not serve as an effective parameter for estimating the isoflavone intensity of the soybeans. Variation of protein and lipid contents between yellow soybeans (n=58) and black soybeans (n=59) was relatively stable, however, protein and lipid contents have no significant relationship with isoflavone content.

Production of Bioactive Compounds from Fungi Grown on Ginseng-Steaming Effluent (인삼 유출액에서 생육한 곰팡이로부터 생리 활성 물질의 생산)

  • We described production of bioactive compounds from fungi grown on Korean ginseng-steaming effluents (GSE) for develop high-value added nutraceuticals from Korean GSE. Hansenula anomala KCCM 11473, which grew well in Korean GSE had high RNA content, and its optimal autolysis conditions were established to produce 5'-ribonucleotides (13.9~28.5 mg/g of biomass) at $55^{\circ}C$ and pH 5.0 for 24 h. 5'-Phosphodiesterase and adenyl deaminase were not effective in increasing the yield of 5'-ribinucleatides, but the yield of IMP increased significantly only after the addition of 1.0% adenyl deaminase. Saccharomyces cerevisiae showed the highest growth in the GSE medium. 267.1 mg of S. cerevisiae biomass was produced from 1 g of GSE solid and medicinal ginsenoside-$Rg_3$ contents was determined with 0.033 mg. Mucor miehei KCTC 6011 produced approximately 120 mg of chitosan per g-dry mycelium in 84 h at $25^{\circ}C$ when grown in the GSE (pH 8.0) supplemented with 0.5% yeast extract and 0.002% $CuSO_4$. Chitosan produced by M. miehei KCTC 6011 have deacetylated approximately 56% and its viscosity and molecular weight of the chitosan were 80 cps and $1.07\times10^3$ kDa, respectively. The chitosan at 1.5 mg/ml inhibited 73.9% of the mycelium growth of Rhizotonia solani in 60 h.

  • Pulsed electric fields(PEF) technology is one of the latest nonthermal methods of food processing for obtaining safe and minimally processed foods. This technology can be effectively explored for obtaining safe food with minimum effect on nutritional, flavor, rheological and sensory qualities of food products. The process involves the application of high voltage(typically 20 ${\sim}$ 80 kv/cm) to foods placed between two electrodes. The mode of inactivation of microorganism; by PEP processing has been postulated in term; of electric breakdown and electroporation. The extent of destruction of microorganisms in PEF processing depends mainly on the electric field strength of the pulses and treatment time. For each cell types, a specific critical electric field strength and specific critical treatment time are required depending on the cell characteristics and the type and strength of the medium where they have been present. The effect also depends on the types of microorganisms and their phase of growth. A careful combination of processing parameters has to be selected for effective processing. The potential applications of PEF technology are numerous ranging from biotechnology to food preservation. With respect to food processing, it has already been established that, the technology is non-thermal in nature, economical and energy efficient, besides providing minimally processed foods. This article gives a brief overview of this technology for food processing applications.

  • The quality of image from the system that creates medical images by using X-ray depends on the various different reasons such as the X-ray generator, the subject and the image transmission medium. In other words, thereare various factors existing that can influence on the quality of image from the moment when the X-ray is generated and until the final image is created. Therefore, the operator who creates images at the clinical site should make continuous evaluation and observation from the final image. There are various methods of evaluating the medical images, but it is assumed that the MTF measurement method can be suitable for measuring actual or effective resolution. So in this study, the MTF measurement method by using X-ray film has been avoided and the MTF features according to the deterioration of the X-ray system have been measured by using the software (the program used Borland C++ builder software and LEAD tools software) that can measure the MTF of the digital medical images. As the result of this measurement, it has been found out through the MTF graph that the resolution and sharpness from the old x-ray generator with a many years of using and many numbers of times of using were deteriorated for the quality of image comparing to those from the new system. Also a simple and easy measurement method for the MTF from the digital medical images can be obtained in this study.

Polycyclic Aromatic Hydrocarbon (PAH) Binding to Dissolved Humic Substances (HS): Size Exclusion Effect

  • Binding mechanisms of polycyclic aromatic hydrocarbons (PAHs) with a purified Aldrich humic acid (PAHA) and its ultrafiltration (UF) size fractions were investigated. Organic carbon normalized binding coefficient ($K_oc$) values were estimated by both a conventional Stern-Volmer fluorescence quenching technique and a modified fluorescence quenching method. Pyrene $K_oc$ values depended on PAHA concentration as well as freely dissolved pyrene concentration. Such nonlinear sorption-type behaviors suggested the existence of specific interactions. Smaller molecular size PAH (naphthalene) exhibited higher $K_oc$ value with medium-size PAHA UF fractions whereas larger size PAH (pyrene) had higher extent of binding with larger PAHA UF fractions. The inconsistent observation for naphthalene versus pyrene was well explained by size exclusion effect, one of the previously suggested specific mechanisms for PAH binding. In general, the extent of pyrene binding increased with lower pH likely due to the neutralization of acidic functional groups in HS and the subsequent increase in hydrophobic HS region. However, pyrene $K_oc$ results with a large UF fraction (>100K Da) corroborated the existence of the size exclusion effect as demonstrated by an increase in $K_oc$ values at a certain higher pH range. The size exclusion effect appears to be effective only for the specific conditions (HS size or pH) that render HS hole st겨ctures to fit a target PAH.

Effects of Different Seeding Rates on Disease Incidences of Wheat Sharp Eyespot and Selection of Fungicides (밀 잎집눈무늬병의 발생에 파종량이 미치는 영향과 방제 약제 선발)

  • This study was conducted to examine the effects of the seeding rate on the disease incidence of sharp eyespot(Rhizoctonia cerealis) on three different varieties and to select effective chemicals to control the disease. When the seeds were sown twice as many as the recommendation, the disease incidence increased by approximately 13%. However, the susceptible variety 'Jopummil' alone showed the significantly enhanced disease incidence at a two-fold seeding rate. Two chemicals such as Hexaconazole EC and Tebuconazole EC highly inhibited the fungal growth on agar medium. However, two strobilurin fungicides such as Pyraclostrobin EC and Trifloxystrobin SC were relatively weak. The fungicides tested displayed the similar in vivo antifungal activities as in vitro activities. Hexaconazole EC and Tebuconazole EC showed the strongest both protective and curative activities and the protective activities of the chemicals were generally higher than the curative activities. Hexaconazole EC and Tebuconazole EC controlled the disease by 64% and 73%, respectively, and the two chemicals reduced the disease by 45% and 39%, respectively, when they were applied one day after pathogen inoculation. These results indicate that both Hexaconazole EC and Tebuconazole EC could be used to control sharp eyespot on wheat.

Static Bending Strength Performances of Hybrid Laminated Woods Composed of Wood-Wood Based Boards (목재와 목질보드 복합적층재의 휨강도성능)

  • To study an effective use of woods, three-ply hybrid laminated woods instead of crosslaminated woods which are composed of spruce in the face and three kinds of wood-based boards (MDF, PB, OSB) in the core were manufactured, and the effect of constitution elements for the core laminae on bending strength performances was investigated. Bending modulus of elasticity (MOE) of hybrid laminated woods had the highest values for the hybrid laminated wood types arranging OSB laminae in the core, and had the lowest values for those arranging MDF laminae in the core. These values were higher than those of various cross-laminated woods. The estimated bending MOEs of the hybrid laminated woods which were composed of perpendicular-direction lamina of spruce in the faces were similar to their measured values, regardless of wood-based boards in the core. However, those of the hybrid laminated woods which were composed of parallel-direction lamina of spruce in the faces had much higher values than those of their measured values, and it was necessary to revise the measured values. Bending modulus of rupture (MOR) of the hybird laminated woods had the highest value for those arranging OSB laminae in the core, and had the lowest values for those arranging PB laminae in the core unlike the bending MOE. By hybrid laminating, the anisotropy of bending strength performances was markedly decreased, and the differences of strength performances among wood-based boards were also considerably decreased.

Influence of Ammonium Phosphate on Mycelial Morphology during Submerged Cultivation of Ganoderma lucidum (영지의 액체배양에 있어서 균사체 형태에 미치는 Ammonium Phosphate의 영향)

  • The mycelial morphology during submerged cultivation of Ganoderma ludium using by air-lift fermenter system were analyzed by image processing system and the characterization of mycelial morphology were investigated. In submerged culture using medium with different ammonium phosphate concentrations, the various morphological forms of G. lucidum mycelium were observed. The filamentous forms such as non-branched long filamentous mycelium, non-branched short mycelium, branched long filamentous mycelium, branched short mycelium, entangled mycelium and clump were observed, and also, and also, the pelleted forms such as smooth pellet, rough pellet and hollow rough pellet were observed. The mycelial morphology was changed from the filamentous to the pelleted forms by addition of ammonium phosphate. The fractal dimensions of pelleted and filamentous forms were 1.05 and 1.3, respectively, while the fractal dimension of mixtures of pelleted and filamentous forms was 1.16. Therefore, the fractal dimension was found to be more effective index for the detection of the mycelial morphology and morphological change during batch cultivation. The circularity was also found to be useful for evaluating the surface growth of pelleted mycelium.
