Production Enhancement of Benzophenanthridine alkaloids in the Suspension Cultures of California poppy using Cyclodextrin (양귀비 세포 현탁배양계에서 Cyclodextrin을 이용한 Benzophenanthridine alkaloids의 생산성 증대)

  • In this research, an extractive production system for alkaloids, where production and some degree of separation occur simultaneously, was developed in a way that the fast removal of alkaloid produced from the suspension cultures was done by capturing alkaloid with cyclodextrins. The alkaloid production was substantially enhanced up to 40 fold when the solid cultures of E. califonica cells treated with ${\beta}$-cyclodextrin compared to the control. The enhancement of alkaloid production was also observed in the suspension cultures. Interestingly, the production pattern seemed to change when the cultures were treated with ${\beta}$-cyclodextrin so that the major part of the alkaloids in the treated cultures was present in the medium, while the non-treated cultures produced the alkaloids intracellularly. ${\beta}$-cyclodextrin was the most effective one in terms of the alkaloid production among the cyclodextrilns(${\alpha}$-cylodextrin, ${\beta}$-cyclodextrin and ${\gamma}$-cyclodextrin) tested in the suspension cultures. ${\beta}$-cyclodextrin showed no adverse effect on the cell growth. The most effective concentration of ${\beta}$-cyclodextrin was observed around 1.5% (w/v) in the suspension cultures. The formation of the inclusion complex of the alkaloids with ${\beta}$-cyclodextrin in the suspension cultures was confirmed by detecting the shift of UV absorbance from 274 nm to 282 nm with a UV spectrophotometer.

Factors Required to Sustain Pastoral Farming Systems and Forage Supply In Winter-Cold Zones in Korea (한국의 동계한냉지역에 있어서 초지개발과 조사료 공급의 활성화에 필요한 요인)

  • The area of grassland established and the arable land in forage production have been decreased recently in Korea. As a result, forages available from the grassland and the arable land provided only 40% of the total roughage consumed by cattle in 1991, compared with 54% in 1989. The decreasing trend of the forage production has been impinged on by a number of legislative. socio-economic. technical and other constraints negatively affecting grassland and forage development. Consequently. effective systems and strategies are quite necessary to have sustainable grassland farming in Korea. In spring and autumn, temperatures are too low for subtropical forage crops, and the summer monsoon climate is another serious limitation to the use of cool-season species. Therefore it is an imperative that not only the research-based grassland development but also the forage crop production should be strongly supported by the government authorities to overcome such climatic limitations. Private forest land holdings are of relatively small units in Korea. Accordingly. it is necessary and important to enlarge the forest land holdings per farm to develop as economic units of grassland. For this the government should introduce new policy measures such as for example: long-term leases of Sorest land and the idle arable land of absentee owners ; rational rental system of national and public forest lands; integrating livestock and forest production. All the laws and regulations standing in the way of the grassland development in forest lands should be modified and revised to open the way for much easier development of grassland. It is also proposed that a high level of financial incentives -hould be provided for structural improvements for the grassland development. They may be: mechanization facilitation, construction of new roads relating to grassland, and land exchange and consolidation. And it is basically necessary that financial enumeration or profits must be garanteed in order to motivate farmers to keep up farming continuously. For more efficient grassland development in mountainous areas, reorganization of Alpine Experiment Station and NAB1 Namwon Branch Station may be needed. Research should be strengthened for completion of pasture mixtures, development of maximum forage production methods with a view to saving labor and reducing production costs, introduction of grazing and forage conservation techniques. and utilization of rice straw as more palatable and nutritious forage source. In order to have more efficient and effective transfer of advanced forage production technologies to livestock farmers the user-clients. it is also essential that special training measures should be given to livestock farmers through making greater use of specialized research and extension workers.

  • The practical studies on the method of particle contamination control for yield enhancement in the cleanroom were carried out. The method of the contamination control was proposed, which are composed of data collection, data analysis, improvement action, verification, and implement control. The partition check method for data collection and data analysis was used in the cellular phone module production lines. And this method was evaluated by the variation of yield loss between before and after improvement action. In case that the partition check method was applied, the critical process step was selected and yield loss reduction through improvement actions was observed. From these results, it is concluded that the partition check method is effective solution for particle contamination control in the cleanroom production lines.

  • In order to evaluate the antiallergic effects of CSYJT, studies were done. We measured the cytotoxic activity for cytokines transcript expression, production of IL-4, IgE, $IFN-{\gamma}$, proliferation of B cell in HRF plus anti-CD40mAb plus rIL-4 stimulated murine splenic B cells. and cytokines transcript expression of IgE in Mast cell The results were obtained as follows: 1. CSYJT decreased the expression of IL-4 in mast cell significantly. 2. CSYJT decreased the production of IL-4 significantly. 3. CSYJT decreased the expression of IgE in mast cell significantly. 4. CSYJT decreased the production of IgE significantly. 5. CSYJT increased the appearance of $IFN-{\gamma}$. The facts above prove that CSYJT is effective against the allergy. Thus, I think that we should study on this continuously.

  • Nowadays, the modeling of systems have been enabled by various models and methodologies that are used for different purpose and perspectives upon the systems. The integrated modeling methodology that integrates these models and methodologies has become challenging issues in system integration such as CIM and PDM. The primary objective of this paper is to propose the comprehensive integrated modeling methodology that integrates the Extended IDEF0 function model, OOIDEF0 information model, navigation model, interface model, and organization model for an effective analysis and design of the CIMS for ship production. This methodology can be used for the integrated system, and cover the systems development life cycle in an integrated fashion. In order to prove the consistency and efficiency of the proposed methodology, the CIMS for ship production is modeled.

  • The effectiveness of resting cells of a photosynthetic bacterium, Rhodospirillum rubrum N-1, was investigated on the production of extracellular ${\delta}-aminolevulinic$ acid(ALA). The ALA generating system required 3hr-incubation in the presence of 10mg of resting cells per ml to obtain the maximal yield of extracellular ALA. and also, under this condition the effect of ALA inducers, i.e., 30mM levulinic acid (LA) and L-glutamic acid($C_5$ pathway precursor) was relatively higher than that of produced extracellular ALA($83{\mu}M$). The volume of system and proper cell density appeared to be important factors for the effective production of extracellular ALA.

  • Solid-state fermentation of lovastatin by Aspergillus terreus was investigated using commercially available 1.2 L polypropylene bottle designed for mushroom cultivation. Moist solid raw materials such as com, rice, and soy bean were tested and com was found to be most suitable for an economical production of lovastatin. 50% or higher water addition prior to the sterilization of com was effective for the maximal lovastatin production. About 0.5% (w/w) lovastatin content in dried cells and corn mass was obtained after 20 days of solid-state fermentation at 30$^{\circ}C$. For safety concerns, aflatoxin Bl and citrinin levels after fermentation were assayed but they were not detected. Lovastatin containing cells and corn residue after fermentation were autoclaved, dried, crushed, and fed to chicken for a period of 3 weeks. Approximately 20% reduction of blood cholesterol level of chicken was observed.

  • Anaerobic hydrogen production was executed using mixed anaerobic sludge. pH control (pH $5.5^{\sim}6.2)$ and heat treatment of anaerobic sluge at $92^{\circ}C$ was not effective for repressing the methane formation. The addition of $nitrate(500^{\sim}2,000\;mg/L\;KNO_3)$ made it possible to repress the methane formation, which resulted in the improvement of hydrogen production. By using SEM(Scanning Electron Microscope), less methane forming microbial with spagetti shape was observed when nitrate was supplied to anaerobic reactor.

  • Temperature and pH conditions were studied for an effective biosurfactant production by Pseudomonas aeruginosa YPJ-80. Efficient methods of biosurfactant separation were also investigated. pH-uncontrolled experiments at 35$^{\circ}C$ and an initial pH of 8 resulted in the best cell growth (3.6 g/L) and biosurfactant production (0.073 g biosurfactant/g cell). Biosurfactant separation was most efficient using solvent extraction with chloroform/methanol (2:1 vol%) followed by acidification using 1N HCl.

  • Optimal culture conditions for efficient in vitro propagation and polysaccharide production of Orostachys japonicus were established. The highest growth yield was achieved in 1/2 MS medium, while the lowest growth yield was obtained in 4 MS medium. The patterns of polysaccharide formation were a little similar in all cases, but on MB5 medium, the po]ysaccharide contents of plant were higher than others. Among the modified nitrate levels, effective growth level were obtained in 1/4 N and 1/2 N. High contents of polysaccharide were obtained in 4 N. Different concentration of potassium and calcium did not improve the growth and polysaccharide production. The micropropagated shoots were successfully acclimatized artificial soils.
