Phyllosticta musarum Infection-Induced Defences Suppress Anthracnose Disease Caused by Colletotrichum musae in Banana Fruits cv 'Embul'

  • Anthracnose development by Colletotrichum musae was observed to be significantly less in the fruits of the banana cultivar 'Embul' (Mysore, AAB) infected with Phyllosticta musarum than in fruits without such infections. Anthracnose disease originates from quiescent C. musae infections in the immature fruit. P. musarum incites minute, scattered spots, referred to as freckles, in the superficial tissues of immature banana peel which do not expand during maturation or ripening. P. musarum does not appear to have a direct suppressive effect on C. musae as conidia of C. musae germinate on both freckled and non-freckled fruit forming quiescent infections. Our investigations have shown that P. musarum infection induced several defence responses in fruit including the accumulation of five phytoalexins, upregulation of chitinase and ${\beta}$-1,3-glucanase, phenylalanine ammonia lyase (PAL) activity and cell wall lignification. $^1H$ and $^{13}C$ NMR spectral data of one purified phytoalexin compared closely with 4'-hydroxyanigorufone. Some of the P. musarum-induced defences that retained during ripening, restrict C. musae development at the ripe stage. This paper examines the potential of P. musarum-induced defences, in the control of anthracnose, the most destructive postharvest disease in banana.

Hydrolysis Mechanism of Phenyl-N-benzoylchlorothioformimidate Derivatives (Phenyl-N-benzoylchlorothioformimidate 誘導體의 加水分解 反應메카니즘)

  • The rate constants of hydrolysis of phenyl-N-benzoylchlorothioformimidates were determined by UV spectrophotometry in 30% (v/v) aqueous dioxane at $25^{\circ}C$. On the basis of rate equation, general base catalysis, solvent effect, substituent effect, thermodynamic parameters, frontier orbital interaction and hydrolysis product analysis, it may be concluded that the hydrolysis of phenyl-N-benzoylchlorothioformimidates proceeds through $S_N1$ mechanism via azocarbocation intermidiate below pH 10.0, while above pH 10.00 the hydrolysis proceeds through nucleophilic addition-elimination ($Ad_{N-E}$) mechanism. In the range of pH from 10.0 to 11.0 these two reaction occur competitively.

Morphology of the Conducting Poly-N-vinylcarbazole-coated Silica Gel Nanocomposites

  • We report the effect of surface morphology on the conductivity of the poly-N-vinylcarbazole (PVK)/silica gel ($SiO_2$) nanocomposites as a function of $SiO_2$ weight percentage (%).The polymerization of PVK was initiated by a free-radical polymerization. The surface morphology of the prepared composite shows the incorporation of $SiO_2$ in the prepared PVK-$SiO_2$ (PS) nanocomposites. The conductivity increased from $9.2{\times}10^{-5}S\;cm^{-1}$ to $9.6{\times}10^{-4}S\;cm^{-1}$ with the increase in the percentage of silica gel from 5 to 30%. The nanocomposites show a percolation behavior having a threshold value between 15 and 20%.

Poduction of Organic Compost to Exclusive use in a Ginseng (인삼 전용 유기질 퇴비제조 및 시용효과)

  • In order to the development of a new organic compost to exclusive use in a ginseng, we conducted the study of the preparation of organic fertilizer and the application effect of organic fertilizer in cultivation of ginseng. The new organic compost was composed of dryed broad leaves of acorn 40%, puffed husks of rice 40% and organic material mixture 20%. The chemical properties of the organic compost shown that the values of the total nitrogen, available phosphate, pottasium, calcium, magnesium, organic matter and C/N ratio were $17g\;kg^{-1}$, $8g\;kg^{-1}$, $11g\;kg^{-1}$, $14g\;kg^{-1}$, $4g\;kg^{-1}$, $794g\;kg^{-1}$, and 26.7, respectivly. The application effect of organic compost in the cultivation of ginseng shown that survival ratio of above ground plant of ginseng was shown the high value than control, and the ratio of root rot and red colored root were decreased. The optimal amount of organic fertilizer for ginseng seedling was $24M/T\;ha^{-1}$. The chemical properties of the ferilized soil were analyzed.

Development of Bacteria for Removal of the Nitrogen in Wastewater (하ㆍ폐수 고도처리를 위한 다기능의 질소원 분해능 균주의 분리)

  • Ninety strains of photosynthetic bacteria were isolated from a local stream at Kyonggi-do, Korea and were further screened. Using these isolated strains, experiments were performed under various light and oxygen conditions in order to select strains with high nitrogen $(NH_3-N,\; NO_3^--N)$ removal efficiencies. Results showed that all the strains screened removed $NH_3-N$, the light had no effect on nitrogen removal, and the nitrogen removal rate was higher aerobically than anaerobically. The removal of $NO_3^--N$ was showed up to 35.3% in some specific strains. Results of batch experiments using Rhodocyclus gelatinosus, an isolated strain with a superior removal rate of $NH_3--N$ and $NH_3-N$, under the anaerobic condition, showed that the removal rate of organics and $NH_3-N$ was the highest (98.2 and 89.0%, respectively) at the CODcr (mg/L)/biomass (mg/L) ratio of 0.2, and the $NH_3-N$ concentration did not increase with the decreasing $NH_3-N$ concentration. Experimental results from various C/N ratios confirmed that the effective removal rate (75.8%) of $NH_3-N$ occurred even at the low (5:1) C/N ratio as well as high ratios, and the simulataneous removal of $NO_3^--N$ (96.0%).

  • Rate constants for the hydrolysis of para-substituted phenyl N-(p-chlorobenzoyl)chloroformimidate (I) derivatives in 1 : 4 dioxane-water at $25^{\circ}C$ have been determined. Rate data, substituent effect $(\rho\>{\rho}^+)$, product analysis and MO calculation indicate that the uncatalyzed reaction proceeds through an $S_N1$ mechanism involving the formation of azocarbonium ion (II) below pH 3.0, and the base-catalyzed reaction proceeds through an $S_N2$ mechanism via transition state (III) above pH 4.0. The relative stability of four peri planar conformational isomers were (E-ap) > (Z-ap) > (E-sp) > (Z-ap), respectively, and the most stable stereo structures shows that the Y-substituted phenyl group $(C_6H_4-Y)$ occupy vertical $(90^{\circ})$ position on the plane of the benzimidochloroformyl group in (E-ap) conformer. The nucleophilic substitution of water molecule occurs by sigma attack to the activatived azomethine carbon atom of (I) derivatives.

Application Effects of Biochar Derived from Pruned Stems of Pear Tree on Growth of Crops and Soil Physico-chemical Properties (배 전정지 바이오차 시용이 작물 생육 및 토양이화학성에 미치는 영향)

  • This study was conducted to develop the manufacturing method of biochar using pruned stems of pear tree and its application effect on the crop growth and soil physico-chemical properties. In this study, biochar derived from pruned stems of pear tree at heating temperature of $300^{\circ}C$, $500^{\circ}C$ and $700^{\circ}C$ in heating times of 2, 3 and 4 hours, were tested in the changes of their chemical properties during biochar processing. The pH, Exch. K, Exch. Mg and cation exchange capacity (CEC) increased as the pyrolysis temperature increased during the production of biochar, and the change of these properties rapidly occurred at $500^{\circ}C$. However, as the pyrolysis temperature increased, ash content increased and total carbon (T-C), yield decreased. And the change of the properties in response to the heating time was not shown. It was thought that it would be desirable to set the production conditions of biochar at $500^{\circ}C$ for 2 hours in consideration of the change of chemical properties and the ash content and yield. And also, were conducted the experiments to establish manufacturing method of farm-made biochar using drum biochar manufacturing machine and investigate the application effects of biochar on the cultivation of chinese cabbage and tomato. Application of biochar derived from pruned stems of pear tree could enhance pH, organic matter (OM), total carbon (T-C) of soil. On the other hand, soil electrical conductivity (EC), NO3-N were lowered compared to the control which has no application. The bulk density, porosity and aggregate formation of soil were improved by biochar application. The fresh matter yields of chinese cabbage and tomato were significantly increased in proportion to the application rate of biochar. This study demonstrated the effect of the biochar derived from agricultural byproduct to be as a low cost potential soil ameliorant by physico-chemical properties in eco-friendly greenhouse cultivation.

  • The aim of the present study was to evaluate the variation in ultimate pH of commercial populations of pure-breed (Landrace, Duroc and Yorkshire) pig's longissimus muscles and their effect on objective meat quality traits and sensory characteristics. Fifty boars were sampled from 184 pigs, which were reared at three breeding farms and slaughtered at a commercial abattoir. The selection was determined based on ultimate pH, and animals were segregated into three groups: low pH (pH $\leq$ 5.5, n=13), medium pH (pH 5.5 to 5.6, n=18) and high pH (pH $\geq$ 5.6, n=16). The breeds had no significant effects; however, pigs with a higher ultimate pH had significantly (p<0.05) higher intramuscular fat content, lower level of polyunsaturated fatty acids, lower level of lipid oxidation and higher eating quality compared to those with lower ultimate pH. As the ultimate pH increased, the relative proportion of C14:0, C16:0 and C18:1 increased while C18:2n6 and C20:4n6 decreased. The present study demonstrates that the economic value of pigs can be characterized by the ultimate pH and/or intramuscular fat content. However, these results do not necessarily indicate that a high ultimate pH directly corresponds to high intramuscular fat content and vice versa.

Analgesic Effects of Transcranial Direct Current Stimulation on Central Neuropathic Pain in Spinal Cord Contusive Rat Model

  • The aim is to investigate the analgesic effect of transcranial direct current stimulation(tDCS) on central neuropathic pain(CNP) in spinal cord contusive rat model. Twenty Sprague-Dawley rats($250{\pm}50$ g, male) were used. Thoracic spinal cord(T10) was contused using New York University(NYU) spinal cord impactor. The animals were randomly assigned to two groups; GroupI: Non-treatment after SCI induction(n=10), GroupII: application of tDCS(0.1 mA, 20 min/time, 2 times/day, 5 days/6week) after SCI induction(n=10). Assess the effect of tDCS using the Basso Beattie Bresnahan(BBB) locomotor rating scales, Touch $test^{TM}$ sensory evaluator(TTSE), Plantar test$^{\circledR}$after contusion at the $2^{nd}$, $3^{rd}$, $4^{th}$, $5^{th}$, $6^{th}$ week and the immunohistochemistric response of c-fos in the thalamus, cerebral cortex after contusion at the $3^{rd}$, $6^{th}$ week after SCI. The scores of BBB scales were significantly different from $3^{rd}$week. TTSE were different significantly over time, but there were no differences at each evaluation times on between-measure time effects. Plantar test were different significantly over time and there were difference at the $4^{th}$, $6^{th}$ week after SCI on between-measure time effects. Also, immunohistochemistric response of c-fos was reduced significantly from $3^{rd}$, $6^{th}$ week after SCI in tDCS group compared with control group in thalamus and cortex. These results identified that tDCS of non-invasive therapeutic method may have beneficial analgesic effect on CNP after SCI with behavioral test and immunohistochemical test.

Effect of Oyster Shell Meal on Improving Soil Microbiological Activity (굴패화석 비료 시용이 토양의 생물학적 활성에 미치는 영향)

  • The effect of oyster shell meal, which is made of a simple crushing and alkaline calcium materials, on soil microbial properties, microbial biomass C, N and P contents, and enzyme activities were evaluated in silt loam soil. The oyster shell meal fertilizer was added at the rates of 0, 4, 8, 12 and $16Mg\;ha^{-1}$. Microbial biomass C, N and P contents were significantly increased with increasing application of oyster shell meal. Soil enzyme activities, such as urease, ${\beta}$-glucosidase and alkaline phosphomonesterase were increased significantly by shell meal application, due to increased soil pH towards neutral range and increased nutrient availability in soil. In particular, the increased microbial biomass P content and phosphomonoesterase activities were strongly correlated with available P content in soil. Conclusively, oyster shell meal fertilizer could be a good supplement to improve soil microbial activities.
