Inhibitory Effects of Various Edible Plants and Flavonoids from the Leaves of Cedrela sinensis on Human Immunodeficiency Virus Type 1 Protease

  • The inhibitory effect of extracts from 15 edible plants on the protease of human immunodeficiency virus (HIV) type 1 was investigated. Protease activity was determined by incubating the extracts in a reaction mixture containing protease and substrate His-Lys-Ala-Arg-Val-Leu-(p-NO$_2$-Phe)-Glu-Ala-Nle-Ser-NH$_2$ to inhibit proteolytic cleavage. Of various plants tested, the leaves of Cedrela sinensis inhibited the HIV-1 protease by 42% at a concentration of 100$\mu\textrm{g}$/ml. A major flavonoid isolated from the leaves of C. sinensis, quercetin 3-O-$\alpha$-L-rhamnoside showed inhibitory activity of 19% at a concentration of 100$\mu$M.

On the Accumulation of Radioactive Materials in Marine Organisms Along the Coast of Korea 2. Strontium-90 Activities in Several Edible Marine Algae

  • Continued to the previous paper, the present investigation deals with stronitium-90 activities and calcium contents among 54 samples of edible marine algae collected along the coast of Korea during September, 1973 and April, 1974. The calcium contents are variable 2.0-17.8%, and 6.38% on an average. Most of the members investigated contain 2-4% in green and red algae, and 8-10% in brown algae. Strontium-90 activities are 0.32-0.37 pCi/l in sea-water, and 0.80-28.66 pCi$\^$90/Sr/g Ca in edible seaweeds. Among the algal phyla, they are 12.49 in green, 3.34 in brown, and 9.39 pCi$\^$90/Sr/g Ca in red algae, while they are 6.25 pCi$\^$90/Sr/g Ca on an average. In a single species collected at the same season, the highest activities appear mostly from the eastern coast, and from the western and southern coasts, in turn. The green algae, Capsosiphon- Enteromorpha complex show about 3 times higher activities, 28.66 pCi$\^$90/Sr /g Ca compared with the other members showing rather higher activities, and would be an indicator plant of strontium-90 activities of the marine algae along the coast of Korea.

Effects of Pinching times on Root Yield and Quality of Codonopsis lanceolata (S.et Z.) Trautv. (더덕순 채취가 수량 및 품질에 미치는 영향)

  • To produce edible Codonopsis shoots in spring, the effect of shoot pinching times on root yield and quality of Codonopsis was evaluated during 3 years from 1992 to 1994. Shoot growth decreased by increasing picking times and one time pinching of Codonopsis shoot was optimal for seed production. In case of 3 times shoots harvest per year. yield of edible Codonopsis shoots amounted to 29.5MT/ha for 3 years. Increase of shoot picking times exerted harmful effect on root growth and decreased marketable value of harvested shoots. It was recommended from the results. One-time pinching of shoot a year for the production of marketable shoots and roots after 3-year cultivation.

  • Continued to the previous papers, the present investigation is carried out to clarify cesium-137 activities and potassium contents among 54 samples of edible marine algae collected along the coast of Korea during September, 1973 and April, 1974. The potassium contents are variable, 1.5-29.6%, and 11.36% on an average, while most of the members contain them about 10-16% generally. Among the algal phyla, they are 13.49% in green, 10.82% in brown and 16.46% in red algae, respectively. Cesium-137 activities are variable, 0.19-8.13 pCi$\^$137/ Cs/g K, and 2.35 pCi$\^$137/Cs/g K on an average in the samples investigated. Among the algal phyla they are 4.35 in green, 2.17 in brown and 0.89 pCi $\^$137/Cs/g K in red algae, respectively. Green algae contain especially higher activities compared with the other two groups. The brown alga, Myelophycus caespitosus shows the highest activities, 8.13 pCi$\^$137/Cs/g K, among the members investigated. Considering the concentration factor, this plant would be an indicator plant of cesium-137 among the marine algae along the coast of Korea.

Antimicrobial Activity of an Edible Wild Plant, Apiifolia Virgin's Bower (Clematis apiifolia DC)

  • An edible wild perennial plant with extremely potent antimicrobial activity was found and identified as apiifolia Virgin's Bower (Clematis apiifolia DC) which is easily found around wet wildernesses. Fresh fruit extract of C. apiifolia exhibited minimum inhibitory concentrations (MIC) in the vicinity of 0.1% against various yeasts and of less than or equal to 0.4% for non-lactic acid bacteria. MICs against lactic acid bacteria were about 2.0%. The antimicrobial activity of C. apiifolia fruit was even more potent than that of garlic which has been known for its potent antimicrobial activity. The principal antimicrobial compound of fruit extract of C. apiifolia was isolated and identified by high performance liquid chromatography and gas chromatography as protoanemonin (a gamma lactone of 4-hydroxy-2,4-pentadienoic acid). The antimicrobial activity of C. apiifolia was stable at high temperatures, and the activity was maintained after heating at $121^{\circ}C$ for 10 min. The antimicrobial compound of C. apiifolia was supposed to inhibit microorganisms by reacting with sulfhydryl groups of cellular proteins.

  • Moisture and quantum were measured to investigated out the growth characteristics of Adenocaulon himalaicum which is one of the important plants at the forest plants. The herbal medicines of Adenocaulon himalaicum known as one of the main mountain edible herbs have great value of resources according to the report that dopamine causing schizophrenia is greatly reduced with it. The moisture is a primary factor for the growth of Adenocaulon himalaicum. In this study we showed that the site with little change of temperature and under $200\;{\mu}mol\;s^{-1}m^{-2}$ quantum is suitable. Also, the maintenance of relative humidity over $50\%$ is a very important factor in the proliferation of shady spot plants.

Analysis of Melatonin Content from Domestic Edible Plants (국내산 식용식물체의 멜라토닌 함량 분석)

  • Melatonin, which is a hormone secreted from pineal gland of brain and known to prevent oxidative damages of various tissues, was analyzed in 26 domestic edible plants. For the preparation of melatonin fraction, 50% ethanol extract prepared from lyophilized plant powder was filtered and applied on TLC plate. Melatonin position on TLC developed with acetone was identified by fluorescence light and extracted with methanol. This methanolic fraction was injected into HPLC comprising ODS-A column, fluorescence detector, and mobile phase consisting of a mixture (30 : 70, v/v) of 70% ammonium acetate and methanol at a flow rate of 1.0 mL/min. Melatonin was identified at the retention time of 17 min. Results revealed that celery, leek, broccoli, and cauliflower had higher melatonin contents than others.

Synergistic Growth Inhibition of Herbal Plant Extract Combinations against Candida albicans

  • Many skin diseases are caused by microbial infections. Representative pathogenic fungus and bacterium that cause skin diseases are Candida albicans and Staphylococcus aureus, respectively. Malassezia pachydermatis is a fungus that causes animal skin diseases. In this study, we propose a method for removing pathogenic microorganisms from the skin using relatively safe edible herbal extracts. Herbal extracts were screened for skin health through the removal of pathogenic microorganisms, and combinations for effective utilization of the screened extracts were identified. In this study, among methanol extracts of 240 edible plants, C. albicans, S. aureus, and M. pachydermatis were killed by extracts of 10 plants: Acori Gramineri Rhizoma, Angelicae Tenuissimae Radix, Cinnamomi Cortex, Cinnamomi Ramulus, Impatientis Semen, Magnoliae Cortex, Moutan Cortex Radicis, Phellodendri Cortex, Scutellariae Radix, and Syzygii Flos. By evaluating the synergistic antifungal activities against C. albicans using all 45 possible combinations of these 10 extracts, five new synergistic antifungal combinations, Acori Gramineri Rhizoma with Magnoliae Cortex extracts, Acori Gramineri Rhizoma with Phellodendri Cortex extracts, Angelicae Tenuissimae Radix with Magnoliae Cortex extracts, Magnoliae Cortex with Phellodendri Cortex extracts, and Phellodendri Cortex with Syzygii Flos extracts, were identified. By utilizing the selected extracts and five combinations with synergistic antifungal effects, this work provides materials and methods to develop new and safe methods for treating candidiasis using natural products.

Anti-oxidative Activities of Commercial Edible Plant Extracts Distributed in Korea (국내 유통 중인 식용식물 추출물의 항산화효과)

  • Many plant extracts are known to have antioxidative effects. However, their activities can be reduced or disappeared during mass production process. The purpose of this study is to compare antioxidative effects of edible plant extracts distributed in Korea. forty three kinds of edible plant extracts commercially available in Korea were selected and investigated for their total phenolics contents and antioxidative potentials(DPPH radical and superoxide anion radical scavenging activities). In contents of total phenolics, the commercial plant extracts from Artemisia annua(whole plant), Ilex paraguariensis(leaf, Silybum marianum(fruit and leaf, Ulmus pumila(bark), Coliolus versicolor(fruit), and Curcuma longa(root and stem) contained over 70 mg/g of powder, DPPH radical scavenging activities($SC_{50}$, 50% scavenging concentration) of A. annua, I. paraguariensis, Pinus densiflora(leaf),S. marianum, U. pumila, and C. longa were $53.96{\pm}0.81\;ppm,\;24.61{\pm}2.12\;ppm,\;35.96{\pm}1.11\;ppm,\;57.46{\pm}2.13\;ppm,\;55.25{\pm}1.65\;ppm\;and\;12.99{\pm}1.67ppm$, respectively, while that of positive control(vitamin C) was $3.86{\pm}0.81\;ppm$. $SC_{50}$ values against superoxide anion radical of A. annua, Cinnamomum zeylanicum(bark), I. paraguariensis, Rubus coreanus(fruit and leaf), Morus alba(leaf), P. densiflora, S. marianum, U. pumila, C. versicolor, C. longa, Perilla frutescens var. acuta(leaf), and H. sabdariffa(leaf and newer) were $53.21{\pm}1.83ppm,\;50.12{\pm}2.12ppm,\;5.59{\pm}0.84ppm,\;41.60{\pm}8.93ppm,\;20.19{\pm}0.97ppm,\;15.19{\pm}1.66ppm,\;21.20{\pm}1.88ppm,\;15.71{\pm}0.91ppm,\;55.48{\pm}2.42ppm,\;52.12{\pm}2.44ppm,\;23.80{\pm}1.98ppm\;and\;11.14{\pm}0.51ppm$, respectively($SC_{50}$ value of vitamin C: $9.61{\pm}0.93ppm$). In particular, both 1 paraguariensis and P. densiflora had high content of phenolics as well as high scavenging activities of DPPH radical and superoxide anion radical. Consequently, above two commercial extracts may be useful as a source of antioxidative nutraceutics.

Expression of Helicobacter pylori urease in plants to use as an edible vaccine

  • Helicobacter pylori is the etiologic agent of human gastritis and peptic ulceration and produces urease as the major protein component on its surface. H. pylori urease is known to serve as a potent immunogen as well as major virulence factor. In order to express the recombinant urease in tobacco plants, a DNA fragment containing the minimal H. pylori urease gene cluster was subcloned into a plant expression vector. The recombinant vector was transformed to tobacco plants. The integration of the recombinant plasmids into tobacco chromosomal genome was verified by genomic PCR. Expression to mRNA was confirmed by Northern blot analysis, and expression to recombinant urease protein was observed by Western blot analysis. These results showed that the recombinant urease can be produced in tobacco plants and will be tested for immune response to use as an edible vaccine.
