Bad Subjects and the Transnational Minjung: The Poetry of Jason Koo and Ed Bok Lee

  • In light of Korean inclusion of its diaspora as part of the nation, a "creolized" approach that brings together constructions of the bad subject of Asian American studies with conceptions of the Korean minjung grounds an analysis of two poets as they might be considered from a bi-national, Korean and U.S. American, perspective. The poets Ed Bok Lee and Jason Koo show different ways of being the bad subject. Lee is clearly a bad American subject, resisting American white racial hegemony, and his poetry often addresses a kind of American minjung multiculturalism, as is shown in poems from his first two books Real Karaoke People and Whorled. He challenges some aspects of contemporary Korea, and might be a kind of Korean bad subject in those challenges. Koo, on the other hand, resists the call to bad subjectivity, so that his poetry may not fit the preferred paradigm of Asian American studies, as he recognizes. As he resists that paradigm, he also gives little attention to his Korean heritage, so his not-bad American subjectivity becomes bad Korea subjectivity. He recovers some measure of badness in the final poem of Man on Extremely Small Island when he connects briefly to his Korean heritage and his Asian American present. The creolized juxtaposition of the bad subject with the minjung suggests the use of these poems in considering both American and Korean society.

  • We propose a new sampling design for the 2001 Health Index Survey at Seoul. In this stratified two-stage sampling design, the ED(enumeration district) of 2000 Population and Housing Census is used as primary sampling unit and the Gu is used as stratification variable in order to obtain the sub-domain estimate for 25 Gu's as well as population estimate for Seoul. The sample ED's are systematically selected after the Ed's are ordered by location and property to obtain a representative sample. And also, the imputation methods for item nonresponses are suggested.

  • Active fraction, $P_2$, was Isolated from natural anti-cancer drug, $CP_2$, by HPLC. We confirmed that $P_2$ includes most of the Isoguanosine and minor components, Berberine and other protoberberine alkaloids, by $^1H-NMR$ and $^{13}C-NMR$ and measured tile cytotoxicity of $P_2$ against various tumor cell-lines.' $P_2$ was very effective to all tumor cell-lines, especially to human colon cancer SNU-C2A$(ED_{50};\;24{\mu}g)$ and liver cancer HEP-3B$(ED_{50};\;27{\mu}g)$.

Identification and Characterization of Thermoplasma acidophilum 2-Keto-3-Deoxy-D-Gluconate Kinase: A New Class of Sugar Kinases

  • The thermoacidophilic archaeon Thermoplasma acidophilum has long been known to utilize D-glucose via the non-phosphorylated Entner-Doudoroff (nED) pathway. We now report the identification of a gene encoding 2-keto-3-deoxy-D-gluconate (KDG) kinase. The discovery of this gene implies the presence of a glycolysis pathway, other than the nED pathway. It was found that Ta0122 in the T. acidophilum genome corresponded to KDG kinase. This enzyme shares no similarity with known KDG kinases, and belongs to a novel class of sugar kinases. Of the five sugars tested only KDG was utilized as a substrate.

Clinical Impact of a Quality Improvement Program Including Dedicated Emergency Radiology Personnel on Emergency Surgical Management: A Propensity Score-Matching Study

  • Objective: To investigate the clinical impact of a quality improvement program including dedicated emergency radiology personnel (QIP-DERP) on the management of emergency surgical patients in the emergency department (ED). Materials and Methods: This retrospective study identified all adult patients (n = 3667) who underwent preoperative body CT, for which written radiology reports were generated, and who subsequently underwent non-elective surgery between 2007 and 2018 in the ED of a single urban academic tertiary medical institution. The study cohort was divided into periods before and after the initiation of QIP-DERP. We matched the control group patients (i.e., before QIP-DERP) to the QIP-DERP group patients using propensity score (PS), with a 1:2 matching ratio for the main analysis and a 1:1 ratio for sub-analyses separately for daytime (8:00 AM to 5:00 PM on weekdays) and after-hours. The primary outcome was timing of emergency surgery (TES), which was defined as the time from ED arrival to surgical intervention. The secondary outcomes included ED length of stay (LOS) and intensive care unit (ICU) admission rate. Results: According to the PS-matched analysis, compared with the control group, QIP-DERP significantly decreased the median TES from 16.7 hours (interquartile range, 9.4-27.5 hours) to 11.6 hours (6.6-21.9 hours) (p < 0.001) and the ICU admission rate from 33.3% (205/616) to 23.9% (295/1232) (p < 0.001). During after-hours, the QIP-DERP significantly reduced median TES from 19.9 hours (12.5-30.1 hours) to 9.6 hours (5.7-19.1 hours) (p < 0.001), median ED LOS from 9.1 hours (5.6-16.5 hours) to 6.7 hours (4.9-11.3 hours) (p < 0.001), and ICU admission rate from 35.5% (108/304) to 22.0% (67/304) (p < 0.001). Conclusion: QIP-DERP implementation improved the quality of emergency surgical management in the ED by reducing TES, ED LOS, and ICU admission rate, particularly during after-hours.

수소 버블을 이용한 전기화학적 그래핀 박리법

  • 그래핀은 전기적, 광학적, 역학적, 열적 특성 등이 아주 좋은 소재이다. Thermal Chemical Vapor Deposition (T-CVD) 장비를 이용 저 진공, 고온에서 CH4과 H2를 가스를 사용하여 그래핀을 합성을 하였다. 그래핀은 탄소만으로 이루어진 2차원 층상구조를 가지고 있다. 촉매 금속 위에서 합성이 이루어지기에 합성된 그래핀을 바로 다른 응용하기에는 어려움이 따른다. 따라서 촉매 금속에서 그래핀을 분리하여 원하는 곳으로 옮기는 과정이 필요한데, 이를 전사공정이라 한다. 최근 전기분해를 이용하여 발생되는 수소 버블을 사용하여 그래핀을 촉매로 사용되는 금속으로부터 분리해내는 Electrochemical Delamination(ED) 전사방식이 소개가 되었다. 이러한 전사 방식의 장점은 촉매기판을 제거하지 않음으로써, 다시 재활용이 가능하고, 공정에 필요한 시간이 짧다. 또한 표면에서 직접적으로 분리하는 방식이기에 촉매 금속의 양면을 사용이 가능하다. 이러한 ED방식의 장점이 있기에 공정의 최적 조건을 잡기 위하여 변수들을 바꾸어가면서 실험을 하였다. 전사된 그래핀은 표면을 광학현미경으로 확인하였고, 라만 분광기를 사용하여 라만 스펙트럼과 기본적인 전기특성을 확인하여 특성을 평가하면서, 기존의 전사방식을 사용한 그래핀 샘플과 비교분석 하였다.

Purification and Characterization of Glycerate Kinase From the Thermoacidophilic Archaeon Thermoplasma acidophilum: An Enzyme Belonging to the Second Glycerate Kinase Family

  • Thermoplasma acidophilum is a thermoacidophilic archaeon that grows optimally at $59^{\circ}C$ and pH 2. Along with another thermoacidophilic archaeon, Sulfolobus solfataricus, it is known to metabolize glucose by the non-phosphorylated Entner-Doudoroff (nED) pathway. In the course of these studies, the specific activities of glyceraldehyde dehydrogenase and glycerate kinase, two enzymes that are involved in the downstream part of the nED pathway, were found to be much higher in T. acidophilum than in S. solfataricus. To characterize glycerate kinase, the enzyme was purified to homogeneity from T. acidophilum cell extracts. The N-terminal sequence of the purified enzyme was in exact agreement with that of Ta0453m in the genome database, with the removal of the initiator methionine. Furthermore, the enzyme was a monomer with a molecular weight of 49kDa and followed Michaelis-Menten kinetics with $K_m$ values of 0.56 and 0.32mM for DL-glycerate and ATP, respectively. The enzyme also exhibited excellent thermal stability at $70^{\circ}C$. Of the seven sugars and four phosphate donors tested, only DL-glycerate and ATP were utilized by glycerate kinase as substrates. In addition, a coupled enzyme assay indicated that 2-phosphoglycerate was produced as a product. When divalent metal ions, such as $Mn^{2+},\;CO^{2+},\;Ni^{2+},\;Zn^{2+},\;Ca^{2+},\;and\;Sr^{2+}$, were substituted for $Mg^{2+}$ the enzyme activities were less than 10% of that obtained in the presence of $Mg^{2+}$. The amino acid sequence of T. acidophilum glycerate kinase showed no similarity with E. coli glycerate kinases, which belong to the first glycerate kinase family. This is the first report on the biochemical characterization of an enzyme which belongs to a member of the second glycerate kinase family.

Analysis of Impact Acoustic Property of Apple Using Piezo-Polymer Film Sensor (고분자 압전 박막 센서를 이용한 사과의 충격 음파 특성 분석)

  • In this study, the PVDF (polyvinylidene fluoride) piero-film sensor was applied to measure the internal quality of apple. The developed sensor detected the response signal through apple after mechanical impact on the surface of apple. The acoustical parameters at time domain such as rise time (RT), ring down count (RC), energy (EN), event duration (ED) and peak amplitude (PA) and acoustical parameter at frequency domain such as spectral density (SE) were analyzed. The size of waveform decreased as storage time of apple increased. The frequency at maximum magnitude was shifted to lower frequency band according to the storage time. The acoustical parameters showed strong relationship with storage time. The multiple linear regression equation was developed to estimate storage time of apple using the acoustical parameters at time domain and its coefficient of determination was 0.97. The internal quality of apple according to storage time is predictable using developed PVDF sensor and acoustical parameters defined in this study.

Development of Homogeneous Enzyme Immunoassay for the Organophosphorus Insecticide Fenthion

  • A rapid, convenient homogeneous competitive enzyme immunoassay for estimating the amount of fenthion is described. The assay utilizes glucose-6-phosphate dehydrogenase-hapten conjugates that are inhibited in solution by antibodies obtained from bovine serum albumin-hapten conjugates. In order to investigate the effects of bridging group recognition on the sensitivity of dose response characteristics, the bridging groups of varying alkyl chain length were attached at the phosphate position of fenthion. Among the antibodies used, the one obtained from the use of hapten (fenthion analog) with the same bridging group structure that was used in preparing the enzyme-fenthion conjugates showed maximum inhibition (up to 51.8%) in the absence of fenthion. In the presence of fenthion, the activity of the enzyme-hapten conjugate is regained in an amount proportional to the fenthion concentration. Under the optimized condition, the $ED_{50}$ value for fenthion was $0.809{\mu}g/ml$. The assay developed in this study is a rapid effective screening method for fenthion prior to precise analysis.

Synthesis and In Vitro Cytotoxicity of Cinnamaldehydes to Hyman Solid Tumor Cells

  • Cinnamaldehydes and related compounds were synthesized from various cinnamic acids based on the $2^{I}$-hydroxycinnamaidehyde isolated from the bark of Cinnamomum cassia Blume. The cytotoxicity to human solid tumor cells such as A549, SK-OV-3, SK-MEL-2, XF498 and HCT15 were measured. Cinnamic acid, cinnamates and cinnamyl alcohols did not show any cytotoxicity against the human tumor cells. Cinnamaldehydes and realted compounds were resistant to A549 cell line up to 15 .mu.g/ml. In contrast, HCT15 and SK-MEL-2 cells were much sensitive to these cinnamaidehyde analogues which showed $ED{50} values 0.63-8.1{\mu}g/ml.$Cytotoxicity of the saturated aldehydes was much weak compared to their unsaturated aldehydes. From these studies, it was found that the key functional group of the cinnamaldehyde-related compounds in the antitumor activity is the propenal group.p.
