Dental Pulp Stem Cell: A review of factors that influence the therapeutic potential of stem cell isolates

  • Undifferentiated stem cells are being studied to obtain information on the therapeutic potential of isolates that are produced. Dental Pulp Stem Ccell (DPSC) may provide an abundant supply of highly proliferative, multipotent Mesenchymal Stem Cells (MSC), which are now known to be capable of regenerating a variety of human tissues including bone and other dental structures. Many factors influence DPSC quality and quantity, including the specific methods used to isolate, collect, concentrate, and store these isolates once they are removed. Ancillary factors, such as the choice of media, the selection of early versus late passage cells, and cryopreservation techniques may also influence the differentiation potential and proliferative capacity of DPSC isolates. This literature review concludes that due to the delicate nature of DPSC, more research is needed for dental researchers and clinicians to more fully explore the feasibility and potential for isolating and culturing DPSCs extracted from adult human teeth in order to provide more accurate and informed advice for this newly developing field of regenerative medicine.

EFFECT OF ORCHARDGRASS GROWTH STAGE ON POOL SIZE AND KINETICS OF DIGESTA PARTICLES IN THE RUMEN OF SHEEP

  • The differences in rumen particle pool size, passage rate and rumen degradability in sheep receiving three varieties of orchardgrass hay harvested at pre-heading (H1), early-bloom (H2) and late-bloom (H3) were investigated using four ruminal-cannulated wethers (68 kg) fed 1,300 g of the hay once a day. Representative samples of whole rumen contents were collected at different times after feeding and the quantities of rumen particle pools [large particle pool (LPP), retained on a $1,180{\mu}m$ sieve; small particle pool (SPP), retained on a 47 but passed a $1,180{\mu}m$ sieve; and soluble fraction (SOL), passed a $47{\mu}m$ sieve (SOL)] were determined by a wet-sieving technique. The fullowing results were obtained: 1) The dry weight of whole rumen contents were significantly lower (p < 0.05) for HI than for H2 or H3. The reduction rate of whole rumen contents was slightly but significantly greater for HI that, the other hay varieties. 2) The LPP disappearance rates were 26.2, 25.3 and 21.7 g DM/h for H1, H2 and H3, respectively, and no statistical differences were found among the hay varieties. Appreciable changes were not observed with SPP and SOL throughout measurements for all hay varieties; however the SPP was markedly greater (p <0.05) for H2 and ill than for HI, while SOL did not differ among hay varieties. 3) The SPP passage rate (g DM/h) and effective rumen degradability (%) for HI, H2 and ill were, respectively, 9.7, 56.6; 16.9, 42.3; and 18.0, 28.9. The ruminal tum-over rate for SPP appeared to be higher for HI than for the other hay varieties.

  • A Literature study was done for identifying the effects of Bloodletting on C.V.A. The major results of the study were as follows. 1. The frequency of points of Bloodletting on C.V.A. were in order Twelve well point, Ship son, Gold SalivaJade Fluid, Paekoe, Chungchung, Sugu, Sosang, Taechu, Wijung, Kwanchung, etc. 2. The frequency of meridians of Bloodletting on C.V.A. were in order Extra-point, Tongmaek-kyong, Su-gworum-Shimpo-Kyong, Susoyang-Samcho-Kyong, Sutaeum-Pye-Kyong, Choktaeyang-Panggwang-Kyong. ete. 3. The frequency of the site of points of Bloodletting on C.V.A. were in order four extremities, face, neck and head, etc. 4. The effects of Bloodletting on C.V.A. is clear away heat and alleviate pain, therapy for waking up a patient from unconsciousness, dredge the meridian passage, expel wind-evil and promote blood circulation, emergency treatment for collapse, etc, 5. The effects of Bloodletting on the early stage of C.V.A. were wake up the patient from unconsciousness by clearing away the heat and The effects of Bloodletting on sequence of C.V.A. were dredge the meridian passage, 6. The frequency of points and meridians of Bloodletting on Hemiplegia were in order Twelve well point, Kyonjong, Extra-point, Chok soyang-Tam-Kyong, etc. 7. The frequency of points and meridians of Bloodletting on Aphasia were in order Gold Saliva Jade Fluid, Amun, Extra-point, Tongmaek-Kyong, etc. 8. 강릉지역 2009~2018년 해풍 발생 특성 (Occurrence Characteristics of Sea Breeze in the Gangneung Region for 2009~2018)

  • The Gangneung region has the complicated geographical characteristics being adjacent to East Sea and Taeback mountains, and thus sea breeze could play an important role in local weather in various aspects. This study aims to understand overall characteristics of sea breeze largely based on long-term (2009~2018) ground-based observation data. We also propose a selection criteria of sea breeze occurrence day; 1) daily precipitation is less than 10 mm, 2) surface wind direction is 0~110° (northerly to easterly) for more than 3 hours during the daytime, 3) wind direction is 110~360° for more than 3 hours during the nighttime, and 4) land and sea temperature difference is positive during the daytime, 5) sea and land sea-level pressure difference is more than 0.5 hPa. As a result, a total of 595 days was selected for the past 10 years. The occurrence of sea breeze is the highest in late Spring to early Summer (May to June). The passage time of sea breeze at the inland station (1.6 km farther inland) is one hour later than the coastal station. On the typical sea breeze event of April 12, 2019, the passage speed and duration of sea breeze was 15 km hr-1 and about 9 hours, respectively, with its depth of about 500 m and its head swelling. The current results emphasize the critical role of sea breeze in forecasting surface temperature and wind, and contribute to relieve heat wave especially in summer in the Yeongdong region.

도시 지하차도 침수 분석을 통한 강제배제시설 평가 (Evaluation of Drain Pump System by Inundation Analysis in Urban Underground Passage)

  • 도시유역의 강우 유출은 대부분 불투수지역인 도시유역의 유출 특성상 빠른 도달시간을 갖는다. 일반적으로 도시유역에서의 강우 유출은 배수펌프장으로 집수되어 하천으로 배제된다. 그러나 설계강우를 초과하는 호우가 발생하였을 때에는 배수펌프장의 용량을 초과하여 전량 하천으로 배제하기가 어려우며 따라서, 도시유역에서의 내수침수위험성이 증가하게 된다. 현행의 배수펌프장에서의 펌프 운영 기준은 배수펌프장의 집수정 또는 유수지의 수위에 따라서 가동되도록 구성되어있다. 그러나 최근 빈번히 발생하는 국지적 집중호우는 배수펌프장의 설계용량을 초과하는 경우가 대부분이며 배수펌프장이 제어할 수 없는 강우 유출은 도시유역에서의 내수침수를 발생시킨다. 본 연구에서는 도시 지하차도에서의 실제 내수침수 사례에 대한 모의를 통하여 당시 침수 발생의 요인을 분석하고 펌프 운영에 대한 변화를 통하여 배수펌프장 강제배제시설에 대하여 평가하였다.

견인강선을 이용한 경골극 견열 골절의 관절경적 치료 (Arthroscopic Treatment of Fractures of the Intercondylar Eminence of the Tibia Using Pull-Out Wire)

  • Recently, a variety of arthroscopic techniques have been reported for the treatment of the displaced tibial eminence fracture. The purpose of this study was to describe details of arthroscopic technique using pull-out wire and to evaluate the results. Eleven patients with irreducible type II and type III tibial eminence fractures underwent the arthroscopic reduction and internal fixation using double strand pull-out wiring. The anterior cruciate ligament tibial drill guide was utilized for the reduction of fracture and passage of the guide pins. The tibial eminence fracture was firmly fixed with double strand 26-gauge pull-out wire(0.45mm diameter). Fracture union was achieved at 7.2 weeks (range, six to eight weeks) after operation. All cases were united at the last follow-up. Subjectively, nine patients had no pain and no restriction of daily activities. Two patients with combined injuries had limitation of knee motion(10 to 130 degrees, respectively) and one patient showed mild anterior laxity. Early rehabilitation was enabled without loss of reduction and breakage of pull-out wire. The arthroscopic reduction and internal fixation using pull-out wire showed good results including early rehabilitation, early fracture union, minimal morbidity, and no requirement of the second operation for hard ware removal.

The importance of post-thaw subculture for standardizing cellular activity of fresh or cryopreserved mouse embryonic stem cells

  • Objective: Remarkable difference in cellular activity was found between early and late subpassaged embryonic stem cell (ESCs) lines, which can be created by subtle changes in cell manipulation protocol. This study subsequently examined whether post-thaw subculture of early subpassaged ESC lines could further affect the activity of the ESCs. Methods: Fresh (as a control treatment) or cryopreserved F1 hybrid (B6CBAF1) early ESC lines (C57BL/6xCBA) of the 4 (P4) or the 19 passage (P19) were subcultured once, twice or six times under the same condition. The post-thaw survival of the ESCs was monitored after the post-treatment subculture and the ability of cell proliferation, reactive oxygen species (ROS) generation, apoptosis and mitochondrial ATP synthesis was subsequently examined. Results: Regardless of the subculture number, P19 ESCs showed better (p<0.05) doubling time and less ATP production than P4 ESCs and such difference was not influenced by fresh or cryopreservation. The difference between P4 and P19 ESC lines became decreased as the post-treatment subculture was increased and the six times subculture eliminated such difference. Similarly, transient but prominent difference in ROS production and apoptotic cell number was detected between P4 and P19 ESCs only at the 1st subculture after treatment, but no statistical differences between two ESC lines was detected in other observations. Conclusion: The results of this study suggest that post-thaw subculture of ESCs under the same environment is recommended for standardizing their cellular activity. The activity of cell proliferation ability and ATP synthesis can be used as parameters for quality control of ESCs.

낙동강 하구역의 동물플랑크톤 군집 분포특성 (Distributional characteristics of mesozooplankton community in Nakdong river estuary)

  • 2015년 평수기인 5월과 태풍 "두쥐안"이 통과한 직후인 10월 초에 낙동강 하구역에서 동물플랑크톤 군집과 환경특성을 조사하였다. 10월에 출현한 동물플랑크톤 군집의 우점종은 따개비 유생, 유공충류와 야광충이었고, 환경특성은 낙동강 하굿둑 수문개방에 의한 담수유입, 난류수 유입과 태풍통과에 의한 수괴혼합에 의해 영향을 받았다. 반면 평수기인 5월에는 모든 정점에서 저염분 해수가 강하게 분포하여 담수 기인 지각류(Evadne nordmanni, Podon polyphemoides)가 우점하여 분포하였다. 모든 시기에 울타리 섬을 기준으로 내측에서 표층염분이 가장 낮았고, 외측으로 가면서 증가하였다. 10월의 표층과 저층 평균수온이 동일하여, 수괴혼합이 활발함을 지시했고, 부유물질 평균농도는 5월보다 10월에 높았다. 5월의 영양염 농도가 엽록소-a 농도와 함께 10월에 비해 높았다. 그러나 동물플랑크톤 총 평균 개체수는 엽록소-a 농도와 관련된 시기적 차이가 없었으며, 집괴분석결과 동물플랑크톤 분포특성은 모든 시기에 염분분포와 관련되어 세 그룹(울타리 섬 내측, 중간측, 외측)으로 구분되었다. 본 연구결과는 조사시기동안 낙동강 하구역에서 관찰된 동물플랑크톤 군집의 시기적 분포특성이 상향적 조절(영양염-엽록소-a 농도-동물플랑크톤)보다 수문개방에 의한 담수유입, 난류수 유입정도 그리고 태풍통과 기인 수괴혼합과 관련있음을 지시하였다.

과잉치 치수유래 줄기세포의 장기 배양 특성 (Characteristics of Supernumerary Tooth-derived Pulp Cells during Long-term Culture)

  • 이 연구는 과잉치 치수로부터 얻은 세포를 더 이상 증식하지 못한다고 판단될 때까지 계대배양을 시행하면서 초기, 중기, 후기 계대의 특성을 비교 분석하였다. 2명의 건강한 6세 남아로부터 3개의 과잉치를 발거하여 치수조직에서 줄기세포를 얻었다. 이를 SNT1(supernumerary tooth 1), SNT2라고 하고, 다른 아이로부터 얻은 과잉치는 SNT3로 명명하였다. SNT1과 SNT2는 똑 같은 시간으로 계대배양 되었고 SNT3은 조금 빠르게 계대배양 되었다. 전체 계대배양의 평균 기간은 $3.6{\pm}1.1$일이었다. 총 $23.3{\pm}0.6$계대까지 배양되었으며, 83일이 소요되었다. 이를 계대를 기준으로 3등분하여 세 군으로 나누었다. I군에서 II군까지 계대배양에 소요된 시간 증가율은 11.90%였으나, II군에서 III군 사이의 증가율은 28.62%로 2.4배가 증가하였다. 22계대까지의 배양기간에 대한 소요 시간을 회귀분석한 결과 y = 0.1169x + 2.25 ($r^2=0.4778$)과 y = 0.1169x + 2.0 ($r^2=0.6444$)으로 추론되었다. 과잉치 치수유래 줄기세포의 정상 배양은 20계대까지 가능할 것으로 판단되었으며, 후기로 넘어가면 계대배양 시간이 2.4배 증가였다. 후기 계대에서는 세포내 부유물이 증가하였고, 형태도 선형으로 변형되는 양상이 관찰되었다.

도갑사대웅보전(道岬寺大雄寶殿)의 조영(造營)에 관한 연구(硏究) -전통중층목조건축의 주칸설정과 상층체감기법을 중심으로- (A Study on the Construction of Main Sanctuary of Dogapsa Temple -Focused on Establishing Bay of columns and Setback Technique in Upper Stories of Traditional Architecture with Multi Roof-)

  • The results of examining the architectural features and changes of the main building of Dogapsa temple which is multi roofs wooden structure are as follows. The passage comparing 'Dogapsa' of <>, account of Lee Ha Gon's trip as the literature showing the appearance of Dogapsa temple in the early 18th century to Borimsa through verse of 'Dutacho' was noticeable. Dogapsa temple at Yeongam was distant over 100 ri from Borimsa temple at Jangheung and it was considered that there were many temples at Mt. Wolchul, Yeongam and there were also many temples to be comparable with it. But, Dogapsa temple was compared to Borimsa temple because verses 'many-storied building is high and immense' of 'Dogapsa' at <> and 'Dogapsa is lower than Borimsa at Jangheung' at 'Dutacho' of <> were interpreted as the existence of multi roofs Buddhist temple which had something in common with Dogapsa and Borimsa and was comparable to them. According to existing materials, it was assumed that the main building of Dogapsa was burnt through the Japanese invasion of Korea in the 52nd year of the sexagenary cycle (Eulmyooebyeon, 1555) and Jungyujaeran (1597), but record of major history of the main buildings of Dogapsa and Borimsa indicated that multi roofs wooden structure of the two temples were built at the same period. Since multi roofs wooden structure of main building of Borimsa was rebuilt in 1692, these buildings existed from the early 18th century to middle 18th century.
