Antioxidative Effect of Ethanol Fraction for Several Korean Medicinal Plant Hot Water Extracts (몇 가지 생약재의 열수 추출물에 대한 Ethanol 분획물의 항산화 효과)

  • The hot water extracts of Paeonia japonica, Paeonia moutan, Eucommia ulmoides, Bupleurum falcatum, Cornus officinalis were freeze dried and dissolved with 70% ethanol. These extracts were fractionated with ethanol soluble fraction(ESF) and ethanol insoluble fraction(EIF). To compare oxidative stability effect of these extracts with that of BHA, linoleic acid with 3,000ppm extracts was incubated at 35$^{\circ}C$. Induction period of BHA was 9 days but that of Paeonia japonica ESF and Scutellaria baicalensis ESF was 12, 9 days respectively. And the higher antioxidant activity was observed for ESF than EIF of these extracts.

The Effect of Equatorial Spread F on Relative Orbit Determination of GRACE Using Differenced GPS Observations (DGPS기반 GRACE의 상대궤도결정과 Equatorial Spread F의 영향)

  • In this paper, relative orbit of Low Earth Orbit satellites is determined using only GPS measurements and the effects of Equatorial Spread-F (ESF), that is one of biggest ionospheric irregularities, are investigated. First, relative orbit determiation process is constructed based on doubly differenced GPS observations. In order to see orbit determination performance, relative orbit of two GRACE satellites is estimated for one month in 2004 when no ESF is observed. The root mean square of the achieved baselines compared with that from K-Band Ranger sensor is about 2~3 mm and average of 95% of ambiguities are resolved. Based on this performance, the relative orbit is estimated for two weeks of two difference years, 2003 when there are lots of ESF occurred, and 2004 when only few ESF occurred. For 2003, the averaged baseline error over two weeks is about 15 mm. That is about 4 times larger than the case of 2004 (3.6 mm). Ionospheric status achieved from K-Band Ranging sensor also shows that more Equatorial Spread-F occurred at 2003 than 2004. Investigation on raw observations and screening process revealed that the ionospheric irregualarities caused by Equatorial Spread-F gave significant effects on GPS signal like signal loss or enhancement ionospheric error, From this study, relative orbit determination using GPS observations should consider the effect of Equatorial Spread-F and adjust orbit determination strategy, especially at the time of solar maximum.

Anti-obesity Effect of Ethanol Extracts from Silkworm (Bombyx mori) Pupae Powder Fermented with Cordyceps militaris in the Primary Adipocytes and High Fat Diet-induced Obesity Model Mice (일차 지방세포와 고지방식이로 유발한 비만모델동물에서 Cordyceps militaris로 발효시킨 누에 (Bombyx mori) 가루 에탄올 추출물의 항비만 효과)

  • Silkworm (Bombyx mori) pupae have been widely worked in wound dressing, hepatoprotective activity, antigenotoxicity, control of glucose level and anti-cancer activity. To investigate the anti-obesity activity of ethanol extract of silkworm pupae powder fermented with Cordyceps militaris (ESfC), the free glycerol release and cAMP concentration as well as fat accumulation were measured in the primary adipocytes of SD (Sprague Dawley) rats and high fat diet (HFD)-treated C57BL/6 mice treated with 12 weeks. Firstly, the presence of the cordycepin with lipid lowering effect was confirmed in ESfC using HPLC analysis. The level of free glycerol and cAMP concentration was significantly increased in the primary adipocytes treated with high dose of ESfC ($400{\mu}g/ml$) although these levels were consistently maintained in other dose ESfC treated groups. In HFD-induced obesity model, the increased fat weight and size of adipocytes in HFD+Vehicle treated group was recovered in HFD+ESfC treated group. Also, the liver weight and the number of lipid droplets were higher in HFD+Vehicle treated group than No treated group. But, this level was significantly decreased in HFD+ESfC treated group compared with HFD+Vehicle treated group. Furthermore, a similar recovery was detected on the phosphorylation of periliphin and HSL, and ATGL expression. Overall, the results of the present study provide some scientific evidences that ESfC can stimulate lipolysis in primary adipocytes and prevent fat accumulation in HFD-treated obesity model, and therefore have the potential for use as anti-obesity agents to treat obese patient.

The Effects of Gilts Housed Either in Group with the Electronic Sow Feeding System or Conventional Stall

  • This experiment was conducted to assess the welfare and productivity of gestating gilts in groups with the electronic sow feeding (ESF) system compared to conventional stalls. A total of 83 gilts ($Yorkshire{\times}Landrace$) were housed into individual stalls to be artificially inseminated. Gilts confirmed pregnant were introduced to their treatment, conventional stalls (ST) or groups with the ESF system. All gilts were taken to the farrowing crates one week prior to their expected farrowing date. In the gestation period, there were no significant differences between gilts allocated to ST and ESF on growth performance. However, backfat thickness gain (p = 0.08) and body condition score (BCS) at 110 days of gestation (p = 0.10) tended to be higher in ESF gilts than ST. Likewise, gilts housed in group showed significantly higher estimated body muscle contents at 110 days of gestation (p = 0.02) and body muscle change during gestation (p = 0.01). There was a trend for a shorter parturition time in ESF gilts (p = 0.07). In the lactation period, group housed gilts showed a tendency to increased BCS changes (p = 0.06). Reproductive performance did not differ with the exception of piglet mortality (ST = 0.2 no. of piglets vs ESF = 0.4 no. of piglets; p = 0.01). In blood profiles, ST gilts showed a higher cortisol level at 110 days of gestation (p = 0.01). Weaning to estrus interval was shorter in gilts housed in ESF than ST (p = 0.01). In locomotory behaviors, ESF gilts recorded a tendency to elevate locomotion score at 36, 70, and 110 days of gestation (p = 0.07, p = 0.06, and p = 0.06, respectively). Similarly, ESF gilts showed significantly higher incidence of scratches at 36, 70, and 110 days of gestation (p = 0.01). Moreover, farrowing rates were higher in stall treatment (97.6%) compare to group housing treatment (95.2%). In conclusion, while group housed gilts with ESF system positively affected welfare status in combination with less physiologically stressful environments and activity, it negatively effects piglet mortality, farrowing rates and injuries of gilts.

Characteristics of Eggshell Powder as Carriers of Probiotics (생균제의 부형제(운반체)로서의 난각분말의 특성)

  • Eggshell (ES) is a by-product of table eggs with high content of calcium carbonate which can be used as a calcium source in feed. In this study, we have first illuminated the potential application of ES as a novel carrier for probiotics. The carriers used in the study include a SBM (Soybean meal), ESL (Eggshell powder with large particles), ESF (Eggshell powder with fine particles), and the complex carriers (SBM+ESL, SBM+ESF). The structure of carriers absorbed by L. plantarum was confirmed by SEM image. Among these carriers, the complex carrier SBM+ESF showed the highest viability of L. plantarum with pH 7~8 during four weeks storage at room temperature. The SBM+ESF was further tested as a carrier for various probiotic strains at $4^{\circ}C$ or $30^{\circ}C$. All the probiotic strains showed high viability at $4^{\circ}C$ storage. However, a significant reduction of Lactobacillus cells was observed at $30^{\circ}C$ storage. B. lichenifomis maintained high viability whereas B. subtilis, B. amyloliquefaciens, and S. cerevisiae showed the reduction of $2{\log}_{10}$ (CFU/g). These results suggest that if the ESF as a calcium source in feed was mixed with SBM, it can be used as an effective complex carrier for improving the viability of some probiotics including B. licheniformis.

Effect of Individual, Group or ESF Housing in Pregnancy and Individual or Group Housing in Lactation on the Performance of Sows and Their Piglets

  • To evaluate different housing systems, 80 gilts were randomly allocated at puberty to four treatments: i) sow stall in gestation followed by farrowing crate (SC), ii) group housing with individual feeding in gestation followed by farrowing crate (GC), iii) ESF (Electronic Sow Feeding) system in gestation followed by farrowing crate (EC), and iv) ESF system followed by group farrowing pen (EG). The results showed that stalled sows had a longer interval between puberty and second estrus (p<0.001). The sows kept in the ESF system gained more body weight (p<0.01) and backfat (p<0.05) prior to service, and more backfat during gestation (p<0.05), but also had greater backfat losses in the subsequent lactation (p<0.01). Sows changing from loose housing to confinement at farrowing had longer gestation length (p<0.001). Total litter size did not differ significantly between gestation treatments, but the number of stillborn piglets was significantly higher in the SC treatment (p<0.01). After weaning, SC sows had the longest interval for rebreeding (p<0.001). Some EG sows came into heat before weaning, giving this treatment the shortest interval. These results indicate that gestation confinement in sow stalls had several detrimental effects on sow performance relative to group housing.

Molecular Cloning and Functional Expression of esf Gene Encoding Enantioselective Lipase from Serratia marcescens ES-2 for Kinetic Resolution of Optically Active (S)-Flurbiprofen

  • An enantioselective lipase gene (esf) for the kinetic resolution of optically active (S)-flurbiprofen was cloned from the new strain Serratia marcescens ES-2. The esf gene was composed of a 1,845-bp open reading frame encoding 614 amino acid residues with a calculated molecular mass of 64,978 Da. The lipase expressed in E. coli was purified by a three-step procedure, and it showed preferential substrate specificity toward the medium-chain-length fatty acids. The esf gene encoding the enantioselective lipase was reintroduced into the parent strain S. marcescens ES-2 for secretory overexpression. The transformant S. marcescens BESF secreted up to 217kU/ml of the enantioselective lipase, about 54-fold more than the parent strain, after supplementing 3.0% Triton X-207. The kinetic resolution of (S)-flurbiprofen was carried out even at an extremely high (R,S)-flurbiprofen ethyl ester [(R,S)-FEE] concentration of 500 mM, 130 kU of the S. marcescens ES-2 lipase per mmol of (R,S)-FEE, and 1,000 mM of succinyl ${\beta}-cyclodextrin$ as the dispenser at $37^{\circ}C$ for 12h, achieving the high enantiomeric excess and conversion yield of 98% and 48%, respectively.

Radical Scavenging and Antioxidant Activities of Fermented Laminaria japonica Extracts

  • Radical scavenging and antioxidant activities of Laminaria japonica and fermented its extracts were evaluated. Freeze-dried L. japonica was fermented by Aspergillus oryzae and extracted with distilled water. The extract solution was mixed with ethanol and centrifuged. The supernatant was ethanol soluble fraction, non-polysaccharide fraction (ESF), and residue was ethanol insoluble precipitation, polysaccharide fraction (EIP). ESF was subjected to sequential fractionation with dichloromethane, ethyl acetate, butanol and water. To determine the radical scavenging and antioxidant activities of these, DPPH radical, hydroxyl radical, superoxide anion radical scavenging activities and linoleic acid oxidation were tested. Among the extracts, ESF of fermented L. japonica showed the highest radical scavenging activity. The ESF showed DPPH radical scavenging activity of 64.33% at concentration of $50{\mu}g/mL$. It was higher than 57.70% of vit. C. Ethyl acetate and butanol fraction had high value of radical scavenging and antioxidant activities, especially butanol fraction of fermented L. japonica was 79.48 % of hydroxyl radical scavenging activity at concentration of $50{\mu}g/mL$. The fermented L japonica had radical scavenging and antioxidant activities higher than L. japonica. These results suggest that fermented L japonica is healthy food having radical scavenging and antioxidant activities.

POD-based representation of the alongwind Equivalent Static Force for long-span bridges

  • This paper develops and discusses a method by which it is possible to evaluate the Equivalent Static Force (ESF) of wind in the case of long-span bridges. Attention is focused on the alongwind direction. The study herein carried out deals with the classical problems of determining the maximum effects due to the alongwind action and the corresponding ESFs. The mean value of the maximum alongwind displacement of the deck is firstly obtained both by the spectral analysis and the Gust Response Factor (GRF) technique. Successively, in order to derive the other wind-induced effects acting on the deck, the Gust Effect Factor (GEF) technique is extended to long-span bridges. By adopting the GRF technique, it is possible to define the ESF that applied on the structure produces the maximum alongwind displacement. Nevertheless the application of the ESF so obtained does not furnish the correct maximum values of other wind-induced effects acting on the deck such as bending moments or shears. Based on this observation, a new technique is proposed which allows to define an ESF able to simultaneously reproduce the maximum alongwind effects of the bridge deck. The proposed technique is based on the GEF and the POD techniques and represents a valid instrument of research for the understanding of the wind excitation mechanism.
