Calcium Ionophore를 이용한 소 배반포로 부터의 배아주 유사세포의 효과적인 분리 (Efficient Establishment of Presumptive Embryonic Stem Cells from Bovine Blastocysts by Exposure to Calcium Ionophore)

  • 소 배반포로부터 배아주 (embryonic stem, ES) 유사세포를 분리하기 위해서는 영양외배엽 (trophectoderm, TE) 세포를 제거하는 것이 효과적이다. 따라서 본 실험은 효과적으로 TE를 제거하기 위한 calcium ionophore A23187 (CIPA) 처리조건을 확립하고, 분리해낸 ES 유사세포의 in vitro 다능성 (pluripotency)을 검증하고자 수행하였다. CIPA 농도 및 처리시간을 달리 하였을 때 50 $\mu$M에서 25분간 처리한 군이 colony 형성율 (51%)및 10 passage 까지의 배양성적 (4.8%)에서 가장 좋은 결과를 나타내었다. 또한 CIPA를 처리하지 않은 군과의 비교에서도 약 5배의 높은 결과를 보임으로서 본 실험에서 확립된 CIPA 처리조건은 가시적인 toxicity 없이 ES 유사세포의 확립에 이용될 수 있음을 시사하였다. 확립된 ES 유사세포는 heterogeneous한 alkaline phosphatase (AP) 활성을 보여 소 ES 유사세포에 대한 타 보고들과 유사한 결과를 보였다. In vitro 부양배양 (suspension culture)에서는 embryoid body로 분화가 가능하였으며, 약 70% 정도의 euploidism을 보였다. 따라서 본 실험에서 확립된 CIPA의 처리조건이 소 배반포로부터 ES 유사세포를 확립하는데 효과적으로 이용될 수 있음을 확인할 수 있었다.

BCF1 생쥐 배반포기 유래 배아간세포 작성에 관한 연구 (Maintenance and Differentiation of Pluripotential Embryonic Cell Lines from Mouse Blastocysts)

  • 생쥐 배반포기 내부세포괴를 체외에서 분리 배양하여 분화가 억제된 내부세포괴 유래 증식세포를 미분화 상태에서 무한히 증식할 수 있는 전능성을 지닌 배아간세포(embryonic stem cell : ES cell)로 확립하고자 본 연구를 실시하였다. BCF1 생쥐 배반포를 10% FCS, 0.1mM nonessential amino acid, 0.1mM sodium pyruvate, 0.1mM 2-mercaptoethanol과 1,000U/ml LIF(세포분확억제인자)가 첨가된 DMEM 기초배양액에 mitomycin-C를 처리한 STO 단층배양세포에서 배양하여 분화가 억제된 내부세포괴 유래의 배아간세포를 분리하였다. 배반포를 STO 단층배양세포에서 4일간 배양하여 내부세포괴세포를 신선한 STO 단층배양세포에서 약 5일 간격으로 반복하여 계대배양을 실시하였다. 5차 계대배양후 뚜렷한 분화 양상없이 배양된 미분화 세포군에 대한 alkaline phosphatase (AP)염색과 체외분화능 검색을 실시한 결과 적색의 미분화 AP 양성반응이 확인되었으며 체외에서 배분화 형성이 유도됨에 따라 배양된 배아간세포주의 다능성 배아간세포 특성을 확인할 수 있었다.

Factors Affecting Primary Culture of Nuclear Transfer Blastocysts for Isolation of Embryonic Stem Cells in Miniature Pigs

  • Pluripotent embryonic stem (ES) cells isolated from inner cell mass (ICM) of blastocyst-stage embryos are capable of differentiating into various cell lineages and demonstrate germ-line transmission in experimentally produced chimeras. These cells have a great potential as tools for transgenic animal production, screening of newly-developed drugs, and cell therapy. Miniature pigs, selectively bred pigs for small size, offer several advantages over large breed pigs in biomedical research including human disease model and xenotransplantation. In the present study, factors affecting primary culture of somatic cell nuclear transfer blastocysts from miniature pigs for isolation of ES cells were investigated. Formation of primary colonies occurred only on STO cells in human ES medium. In contrast, no ICM outgrowth was observed on mouse embryonic fibroblasts (MEF) in porcine ES medium. Plating intact blastocysts and isolated ICM resulted in comparable attachment on feeder layer and primary colony formation. After subculture of ES-like colonies, two putative ES cell lines were isolated. Colonies of putative ES cells morphologically resembled murine ES cells. These cells were maintained in culture up to three passages, but lost by spontaneous differentiation. The present study demonstrates factors involved in the early stage of nuclear transfer ES cell isolation in miniature pigs. However, long-term maintenance and characterization of nuclear transfer ES cells in miniature pigs are remained to be done in further studies.

  • Human embryonic stem (ES) cell lines derived from the inner cell mass of human blastocysts have potential to differentiate into any cell types. We have established in vitro neural differentiation of human ES cells. After the formation of embroid bodies (EBs), the differentiating EBs formed neural tube-like rosettes in the presence of basic fibroblast growth factor (bFGF). The rosettes were selectively isolated by the treatment of dispase and cultured in a medium for human neural precursors in the presence of bFGF. (omitted)

Expression of HERV-HX2 in Cancer Cells and Human Embryonic Stem Cells

  • The endogenous retrovirus-like elements (HERVs) found on several human chromosomes are somehow involved in gene regulation, especially during the transcription level. HERV-H, located on chromosome Xp22, may regulate gastrin-releasing peptide receptor (GRPR) in connection with diverse diseases. By suppression subtractive hybridization screen on SV40-immortalized lung fibroblast (WI-38 VA-13), we discovered that expression of HERV-HX2, a clustered HERV-H sequence on chromosome X, was upregulated in immortalized lung cells, compared to that of normal cells. Expression of HERV-HX2 was then analyzed in various cell lines, including normal somatic cells, cancer cells, SV40-immortalized cells, and undifferentiated and differentiated human embryonic stem cells. Expression of HERV-HX2 was specifically upregulated in continuously-dividing cells, such as cancer cells and SV40-immortalized cells. Especially, HERV-HX2 in HeLa cells was highly upregulated during the S phase of the cell cycle. Similar results were obtained in hES cells, in which undifferentiated cells expressed more HERV-HX2 mRNA than differentiated hES cells, including neural precursor and endothelial progenitor cells. Taken together, our results suggest that HERV-HX2 is upregulated in cancer cells and undifferentiated hES cells, whereas downregulated as differentiation progress. Therefore, we assume that HERV-HX2 may playa role on proliferation of cancer cells as well as differentiation of hES cells in the transcriptional level.

체외수정 유래 소 배반포로부터 유사 배아 줄기 세포의 확립 및 유지 (Establishment and Maintenance of Embryonic Stem-like Cell Lines from In Vitro Produced Bovine Blastocysts)

  • 본 연구는 소 배반포의 내부 세포괴로부터 다능성(pluripotency)을 지닌 배아 줄기 세포(embryonic stem cell) 또는 그 유사 세포를 분리 및 배양함으로써 줄기 세포 관련 분야의 기반 기술을 확립하고자 하였다. 소 체외수정란을 $10{\sim}12$일간 체외배양하여 생산된 부화 배반포를 세포분열이 불활성화된 생쥐 태아 섬유아 세포(mouse embryonic fibroblast, MEF) 위에서 배양하여 콜로니 형성을 유도하였으며, 이들로부터 내부 세포괴 유래의 형태를 지닌 것만을 광학현미경 하에서 물리적으로 분리하여 약 $5{\sim}7$일 간격으로 계대배양을 실시하였다. 이러한 방법을 통하여 배아 줄기 유사 세포의 특성을 40계대 이상 유지하는 2개의 세포주를 확립하였다. 각각의 세포주들은 높은 alkaline phosphatase(AP) 활성을 지니고 있었으며, 형광 면역 염색법과 PCR 기법을 사용하여 Oct-4, Nanog, STAT3, SSEA3 및 SSEA4의 발현을 관찰할 수 있었다. 이러한 결과를 종합하여 볼 때, 본 연구에서는 소 배반포로부터 배아 줄기 세포주를 확립하는 제반 기술이 확립되었다고 판단되며, 향후 관련 분야 연구에 활용될 수 있을 것으로 기대된다.

Comparison of Three Different Culture Systems for Establishment and Long-Term Culture of Embryonic Stem-like Cells from In Vitro-Produced Bovine Embryos

  • Although embryonic stem cells (ESCs) or ES-like cells are reported from many mammalian species other than the mouse, the culture system for murine ESCs may not be suitable to the other species. Previously many other research groups have modified either human or mouse ESC culture systems for bovine ESC culture. In this study, we compared three different culture mediums consisting of DMEM, ${\alpha}$-MEM or KnockOut$^{TM}$-DMEM (KO), which are modified from human or mouse ESC culture system, for the generation of bovine ESCs. In this study, some pre-requisite events which are important for establishment and long-term propagation of ESCs such as inner cell mass (ICM) attachment on feeder cells, primary colony formation and sustainability after passaging. Once the ICM clumps attached on feeder cells, this was designated as passage 0. In regards to the rate of ICM attachment, ${\alpha}$-MEM was superior to the other systems. For primary colony formation, there was no difference between DMEM and ${\alpha}$-MEM whereas KO showed lower formation rate than the other groups. For passaging, the colonies were split into 2~4 pieces and passed every 5~6 days. From passage 1 to passage 3, DMEM system seemed to be appropriate for maintaining putative bovine ESCs. On the other hand, ${\alpha}$-MEM tended to be more suitable after passage 6. Although ${\alpha}$-MEM support to maintain a ES-like cell progenies to passage 15, all three culture systems which are modified from human or mouse ESC culture media failed to retain the propagation and long-term culture of putative bovine ESCs. Our findings imply that more optimized alternative culture system is required for establishing bovine ESC lines.
