Insecticide Susceptibility in the Different Larva of Tobacco Cutworm, Spodoptera litural Fabricius (Lepidoptera: Noctuidae) Collected in the Soybean Fields of Milyang, Korea (밀양산 콩포장 담배거세미나방 유충의 약제에 대한 감수성)

  • The susceptibility of the different larval stages of Spodoptera litura to nine insecticides was evaluated using the perilla leaf-dipping method. Median lethal concentration ($LC_{50}$) was increased with larval development in the range of 0.5 ppm to 5.6 ppm, 9.9 ppm to 27.9 ppm, 9.6 ppm to 125.1 ppm and 24.3 ppm to 546.6 ppm in the 1st, 2nd, 3rd and 4th instar, respectively. The tolerance ratio (TR), which is the TR of 90 percent lethal concentration (LC/ sub 90/) to the recommended concentration, was 0.04 to 0.8 in the 1 st, 0.2 to 7.5 in the 2nd, 0.7 to 115.3 in the 3rd and 1.2 to 485.4 in the 4th instars. Lower D$LC_{50}$ and DTR, which is the difference between the $LC_{50}$ and the TR of 4th and other instars, respectively, were observed in chlorfenapyr, chlorpyrifos and EPN while higher ones were lufenuron, chlorfluazuron and teflubenzuron. These results mean that insecticides with lower D$LC_{50}$ and DTR are effective in controlling larva of S. litura collected in Milyang, Korea.

Enzyme Activities of a Honeybee(Apis mellifera L.) Associated with the Degradation of Some Insecticides (서양종(西洋種)꿀벌의 살충제분해효소에 관(關)한 연구(硏究))

  • In order to determine the approptiate usage of insecticides to honeybee(Apis mellifera L.), median effective dose to seven insecticides were studied. $LC_(50)$ value of DDT was the highest as being 58 ppm, and that of EPN was the lowest as being 1.61ppm. Various detoxifying enzymes from the midget cf adult worker bee, including microsomal oxidases, glutathione Stransferases, esterases, and DDT-dehydrochlorinase were assayed. Effects of various insecticides on microsomal enzyme activities were as follows: Aldrin epoxidase activity was inhibited by malathione and permethrin treatment. N-demethylase activity was induced by diazinon and EPN treatment and O-demethlase activity was induced by diazinon treatment. Of the glutathione S-transferases, aryltransferase(DCNB conjugation) activity was significantly induced by diazinon, and moderately induced by permethrin. Of the esterases, ${\alpha}-NA$ esterase activity was moderately inhibited by malatjione and permethrin. Acetylcholinesterase activity was not affected by the sublethal exposure of honeybee to the insecticides. Sublethal exposure of honeybee to the insecticides had no effect on DDT-dehydrochlorinase activity, except carbaryl and permethrin were significantly induced.

  • Four technical grade organophosphate pesticides (Fenitrothion, Fenthion, Dizninon and EPN) were investigated for their effects on the murine immune function. Among the immunotoxicological assay parameters of NIEHS, humoral immune parameter and pathotoxicological indicators were examined in this study. Subchronic exposure of rodents to these pesticides resulted in marked suppression of humoral immune function and moderate histological changes of lymphoid organ any significant alterations of clinical status.

  • Eight cattle and ten sheep were administrated various degree of PMA-B and EPN, and S-GOT, S-GPT and serum cholinesterase activity were tested. Serum cholinesterase activity showed no typical tendency, however, S-GOT/S-GPT ratios were decreased by the administration of PMA-B. It was suggested some degree of liver damage by the administration of the chemicals was recognized.

  • We applied the simulation method using Petri Nets to a FMS model. Generally ordinary Petri Net would be short of describing a real FMS operations. Hence we adopted the extended Petri-Mets(EPN) and timed places in order to have performance evaluation. Our simulator use data based modeled of Petri Nets in simulation. We can enhance the overall production rate of the system with the obtained results over a number of simulations.

Fate of Some Pesticides during Brining and Cooking of Chinese Cabbage and Spinach

  • Chinese cabbage and spinach applied with eight pesticides at two different rates were subjected to brining, heat-cooking, and blanching to determine residue or transfer ratios of those pesticides. Residue ratio in discarded inedible portion varied significantly, 0-94%, depending on pesticides applied, vegetable type, and cooking processes. Average reduction ratios of residues during cooking process were 78, 46, 23, 12, 10, 9, 8, and 2% in dichlorvos, diazinon, chlorpyrifos, endosulfan, EPN, cypermethrin, deltamethrin, and fenvalerate, respectively.

  • Organophosphorus insecticide residues were investigated in six kinds of fruits and five kinds of vegetables.The materials used in this experiment were grape, musk melon, apple, peach, plum, apricot, lettuce, green pepper, cucumber, pumpkin and tomato which were collected from June to september 1986 in Seoul. Residual pesticides investigated were Diazinon, Parathion, MEP (Fenitrothion), Malathion, EPN, MPP (Fenthion), PAP (Phenthoate) and Dimethoate and all samples were analysed by gas chromatographic technique with NPD (Nitrogen Phosphorus Detector). No sample was found to approach proposed national maximum residue limits in Korea.

  • We have developed the bio-electrode measuring the variance of the amount of acetylcholine affected by residual pesticide. The working electrode of the biosensor was made by combination of cobalt phthalocyanine and carbon organic compounds. The biosensors were constructed by screen-printing method. The principle of working electrode is similar to thiocholine sensor. We have fabricated the biosensor using standard screen printing method. Generally, the biosensor made by printing method formed thick film biosensor. When the electrodes were made by electrochemical cells, the generation of current by the addition of enzyme substrate was inhibited by standard solutions of organo-phosphate pesticides. The detection limit of sensor is about 0.5 $\mu{g}/L$ for carbofuran. We could improve the responsibility of the sensor by controlling the cobalt phthalocyanine and thiocholine concentration ratio. Also we have tested the EPN and Chlorpyrifos pesticides and found that the biosensor is applicable to fast determination of residual pesticides.

Purification of Paclitaxel and Its Derivatives by Supercritical Fluid Chromatography(SFC) (초임계 유체 크로마토그래피(SFC) 방법에 의한 Paclitaxel 및 그 유도체의 분리 정제)

  • Studies were carried out to elucidate the effects of pressure, temperature and mobile phase composition on supercritical $CO_2$ chromatographic separations of paclitaxel, baccatin III, 10-deacetyl baccatin III, 7-epi-10-deacetyltaxol, cephalomanine, and 10-deacetyltaxol. High resolutions of paclitaxel, 10-deacetylbaccatin III, 10-deacetyltaxol were observed with optimized pressure, temperature, and mobile phase composition. The highest resolution between paclitaxel and 10-deacetylbaccain III was observed at 275 kg/$\textrm{cm}^2$, $40^{\circ}C$ with the mobile phase composition of gradient mixture of 3.9-3.6 mL/min $CO_2$, 0.1-0.4 mL/min methanol for 20 min. Resolutions of baccatin III, capalomannine, and 7-epi-10-deacetyltaxol were found to be low in this study. On-line coupled SFE/SFC process was applied to isolate paclitaxel from yew tree powder. As a consequence, paclitaxel with a purity of 95% was obtained with a recovery yield of 38%.

Studies on Pretreatment for Analysis of Pesticides by Using HPLC and GC (HPLC 및 GC에 의한 농약분석에서 전처리에 대한 연구)

  • For determination of separated pesticides by using GC and HPLC, liquid-liquid extraction(LLE) and solid phase extraction(SPE) have been carried out to separate and concentrate the organophophorous pesticides such as Diazinon, Fenitrothion, Phosmet, Phosalon and EPN in environmental water samples. ln determination of pesticides by HPLC/UV, SPE has resulted in higher recovery and more precision than LLE, while in determination of pesticides by GC/FPD, vice versa. HPLC/UV after the pretreatment process of sample by solid phase extraction (SPE-HPLC/UV) has suggested the possibility of determination of pesticides ppb level. ln comparison of detection limit, both SPE-HPLC/UV and LLE-GC/FPD are reasonably suitable for analysis of residue pesticides. ln the respect of the rapidity and the solvent required, SPE-HPLC/UV method has proven to be superior to LLE-GC/FPD.
