An Evaluation of EOCS Regarding Safety Management Effects on Buried Gas Pipelines and Convenience of the Excavators and the Operators of Gas Companies

  • We introduced the Excavation One Call System (EOCS) as a pilot system, in Seoul, Korea. The system utilizes the phone and internet to transfer information about digging underground and buried gas pipelines, although currently written forms are used in accordance with the City Gas Business Law. After one year, we evaluated the business model by surveying the excavators and the operators of the gas companies. This paper shows that the EOCS was more effective in preventing the buried gas pipelines from being damaged than the existing method that has to use due form. It also shows that the EOCS was more convenient and cost efficient than the present policies in place. We come to the conclusion that the EOCS should be extended nationwide and gradually include other subsurface facilities.

SENSITIVE DETERMINATION OF ELEVEN PHENOLIC ENDOCRINE-DISRUPTING CHEMICALS IN HUMAN URINE USING GAS CHROMATOGRAPHY/MASS SPECTROMETRY -SELECTED ION MONITORING

  • I improved an analytical method for determining trace amounts of eleven phenolic endocrine-disrupting chemicals (11 phenolic EOCs) in human urine. The 11 phenolic EOCs were subjected to hydrolysis and then to solid phase extraction with a XAD-4 column. Alkylphenols, chlorophenols, and bisphenol A in XAD-4 column were eluted with acetonitrile, and the eluate was concentrated under a nitrogen stream, and then tert-butyldimethylsilylation. Separation and determination were done by gas chromatography, using mass spectrometry operating in the selective ion monitoring mode for quantitation. For tert-butyldimethylsily (TBDMS) derivatization the recoveries were $91.2{\sim}125.9%$, the limits of quantitation (LOQ) for the 11 phenolic EOCs in the nanogram-per-milliliter range ($0.025{\sim}1.000\;ng/mL$) were thus achieved by using 1 mL of urine, and the SIM responses were linear with the correlation coefficient varying by $0.9300{\sim}0.9943$. Based on the results for urine samples from unexposed individuals, 4-tert-octylphenol and pentachlorophenol were detected in hydrolysed urine sample. Other alkylphenols, chlorophenols and bisphenol A were not detected.

  • 정부가 도입하려 하는 원콜시스템(EOCS: Excavation One Call System)은 원콜센터에 전화 한통화를 하면 현장에서 만나 도시가스 매설배관을 지면에 표시해 주기 때문에 신청서 작성이 필요없고 곧바로 업무처리가 가능하다. 이 시스템 도입은 관계부처 협의를 거쳐 법제처 심사를 앞두고 있으며 올 하반기쯤 시행될 예정이다.

Melt Rheology of Ethylene 1-Octene Copolymer Blends Synthesized by Ziegler-Natta and Metallocene Catalysts

  • The melt rheology of four binary blends of ethylene 1-octene copolymers (EOCs) which consist of one component by Ziegler-Natta and another by metallocene catalysts, was studied to elucidate miscibility in the melt by using torsion rheometer at 200$\^{C}$ and different shear rates. The four blend systems, designated into the FA+FM, SF+FM, RF+EN, and RF+PL blend, are divided and interpreted based on the melt index (MI), the density and the comonomer contents. The melt viscosity such asη', η", and η$\^$*/ is weight average value if the comonomer contents are similar, otherwise they show different manner. The experimental resole are analyzed based on the Cole-Cole plot of logη' uersus log η", the logarithmic plots of the dynamic storage modulus (G') versus the dynamic loss modulus (G") for various blend compositions, and the melt viscosity of 11', n", and f" as a function of blend compositions. As a cerise-quence, the FA+FM blend is miscible, but the SF+FM, RF+EN, and RF+PL blends are not in the melt. Thus miscibility of the blends studied in this communication is suggested to strongly influence by the comonomer contents rather than the density or the MI.

Methylation Profile of BRCA1, RASSF1A and ER in Vietnamese Women with Ovarian Cancer

  • DNA methylation is considered a promising biomarkers for diagnosis of cancer in general and of ovarian cancer in particular. In our study, we validated the accuracy of methylation specific polymerase chain reaction (MSP) to analyze the methylation pattern of BRCA1, RASSF1A and ER in 59 and 10 Vietnamese patients with epithelial ovarian cancer (EOC) and benign ovarian tumors, respectively. We found methylation of BRCA1, RASSF1A and ER in 11/59 (18.6%), 40/59 (67.8%) and 15/59 (25.4%) of EOC cases, while methylation of BRCA1 was only detected in 2/10 (20%) benign ovarian patients. Forty five out of the 59 EOCs (78%) demonstrated methylation at one or more genes. The methylation frequency of RASSF1A was significantly associated with EOC (p<0.0005). No significant association was observed between methylation status of these genes and the clinical and pathological parameters of tumors collected from Vietnamese women suffering from ovarian cancer.

Investigating Preoperative Hematologic Markers for Prediction of Ovarian Cancer Surgical Outcome

  • Purpose: The current study aimed at assessing the association between neutrophil-lymphocyte ratio (NLR) and platelet lymphocyte ratio (PLR) for the prognosis of the surgical outcome of epithelial ovarian cancer (EOC). Materials and Methods: EOC patient medical records of surgical operations between January, 2005 and December, 2015 were reviewed and their data of clinicopathological complete blood counts (CBCs) and surgical outcomes were collected. To assess their effects on surgical outcomes, PLR and NLR optimal predictive values were determined and then compared with each other. Results: A statistically significant relation was found between surgical outcomes and NLR and PLR (p<0.001 and p<0.001), for which new cutoff points were gained (PLR: 192,3,293; NLR: 3). The sensitivity and specificity were 0.74 and 0.67, respectively for PLR and 0.74 and 0.58, for NLR. Conclusions: NLR and PLR seem to be useful methods for the prediction of surgical outcomes in patients with EOCs. Increased NLR and PLR proved to be beneficial for poor surgical outcomes. Moreover, PLR increase showed further help in the predicting outcome of EOC suboptimal debulking.

Overexpression of Semaphorin4D Indicates Poor Prognosis and Prompts Monocyte Differentiation toward M2 Macrophages in Epithelial Ovarian Cancer

  • Previously, we demonstrated overexpression of semaphorin4D (SEMA4D, CD100) to be closely related to tumor angiogenesis in epithelial ovarian cancers (EOCs). However, the function and expression of SEMA4D in the EOC microenvironment has yet to be clarified in detail. In this study, we confirmed that overexpression of SEMA4D in primary tumors and ascites was related to low differentiation, platinum resistance and a refractory status (P<0.05), while high M2 macrophage count and percentage were evident in EOC patients with advanced FIGO stage and platinum resistance (P<0.05), using immunohistochemistry, enzyme-linked immunosorbent assay (ELISA), and fluorescence-activated cell sorting (FACS), respectively. The data showed correlations of SEMA4D expression and M2 macrophage counts in primary tumors and M2 macrophage percentage in ascites (r=0.281 and 0.355, each P<0.05). In the Cox proportional hazard mode, SEMA4D expression was an independent indicator of overall survival (OS) and progression-free survival (PFS) for EOC patients. Furthermore, higher expression of SEMA4D in ovarian cancer cell lines (SKOV3, A2780, and SW626) and their supernatants were found than that in a human primary cultured ovarian cell and its supernatant by reversed transcript PCR (RT-PCR), Western blotting and ELISA, respectively. Interestingly, peripheral blood monocytes (MOs) tended towards the M2-polarized macrophage phenotype ($CD163^{high}$) in vitro after human recombined soluble SEMA4D protein stimulation. These findings suggest that SEMA4D might possibly serve as a reliable tool for early and accurate prediction of EOC poor prognosis and could playan important role in promoting tumor dissemination and metastasis in the EOC microenvironment. Thus SEMA4D and its role in macrophage polarization in EOC warrants further study.

  • Effect of poly(propylene-co-octene) as a compatibilizer in toughened polypropylene/ poly(ethylene-co-octene) (EOC) was investigated. The EOCs used were metallocene catalyzed commercial linear low density polyethylene and they are elastomeric materials. The poly(propylene-co-octene) was synthesized by metallocene catalyst in our laboratory to be used as a compatibilizer in PP/EOC blends. PP/EOC blends without compatibilizer shows very low mechanical properties in specimens with weldlines while incorporation of a compatibilizer significantly increases the mechanical properties of specimens with weldlines. However, compatibilized PP/EOC blends does not show increased impact property in a weldline free specimen and it is attributed to low molecular weight of the poly(propylene-co-octene) synthesized in present study. It is expected that the poly(propylene-co-octene) having increased molecular weight provides very good performance as an effective compatibilizer in toughened polypropylene/EOC blends.

  • Established in the 1960s, high pressure underground pipelines in Ulsan and Yeosu industrial estate are underground as toxic gas as well as combustible gas that is heavier than city gas and low combustion range. Especially, industrial pipelines occupy more than 20 years old pipes. In this way, the industrial estate pipeline was installed before the introduction of the supervision of construction, However, unlike the city gas pipeline, the pipeline is managed without any legal obligation. In this study, the safety management status of high pressure underground pipelines and urban gas underground pipelines in the industrial estate is analyzed and comparison of laws, extent of damage impact, using the pipe inspection model for pipe inspection of high pressure piping system with the existing piping system. it is intended to cuntribute to improving the safety of industrial estate are underground pipeline.

XIAP Associated Factor 1 (XAF1) Represses Expression of X-linked Inhibitor of Apoptosis Protein (XIAP) and Regulates Invasion, Cell Cycle, Apoptosis, and Cisplatin Sensitivity of Ovarian Carcinoma Cells

  • Background: X-linked inhibitor of apoptosis protein (XIAP) associated factor 1 (XAF1) exhibits aberrantly low or absent expression in various human malignancies, closely associated with anti-apoptosis and overgrowth of cancer cells. However, limited attention has been directed towards the contribution of XAF1 to invasion, apoptosis, and cisplatin (DDP)-resistance of epithelial ovarian cancer (EOC) cells. This study aimed to evaluate the potential effects of XAF1 on invasion, cell cycle, apoptosis, and cisplatin-resistance by overexpressing XAF1 in SKOV-3 and SKOV-3/DDP cells. Methods and Results: The pEGFP-C1-XAF1 plasmid was transfected into SKOV-3 and SKOV-3/DDP cells, and the expression of XAF1 at both mRNA and protein levels was analyzed by reverse transcription-PCR and Western blotting. Overexpression of XAF1 suppressed XIAP expression in both SKOV-3 and SKOV-3/DDP cells. Transwell invasion assays demonstrated that XAF1 exerted a strong anti-invasive effect in XAF1-overexpressing cells. Moreover, flow cytometry analysis revealed that XAF1 overexpression arrested the cell cycle at G0/G1 phase, and cell apoptosis analysis showed that overexpression of XAF1 enhanced apoptosis of SKOV-3 and SKOV-3/DDP cells apparently by activating caspase-9 and caspase-3. Furthermore, MTT assay confirmed a dose-dependent inhibitory effect of cisplatin in the tested tumor cells, and overexpression of XAF1 increased the sensitivity of SKOV-3 and SKOV-3/DDP cells to cisplatin-mediated antiproliferative effects. Conclusions: In summary, our data indicated that overexpression of XAF1 could suppress XIAP expression, inhibit invasion, arrest cell cycle, promote apoptosis, and confer cisplatin-sensitivity in SKOV-3 and SKOV-3/DDP cells. Therefore, XAF1 may be further assessed as a potential target for the treatment of both cisplatin-resistant and non-resistant EOCs.
