Development of Luteinizing Hormone Releasing Hormone (LHRH) Delivery Systems for Vaginal Mucosal Route

  • The objective of this study was to find a rational dosage form for vaginal mucosal delivery of LHRH. Vaginal absorption of LHRH was estimated by measuring its ovulation inducting effect in rat and in vitro vaginal membrane permeation study in rabbit. THe effects of different hydrogel bases, such as Polycarbophil and Pemulen compared with solutions on vaginal membrane permeation of LHRH were investigated. Sodium laurate, disodium ethylenediamine brane permeaiton of LHRH were investigated. Sodium ethylenediamine tetraacetate (EDTA) and sodium tauro-24, 25-dihydrofusidate (STDHF), which are effective peptidase inhibitors were chosen as additives to a LHRH hydrogel delivery system and LHRH solutions. A Polycarbophil compared with a solution formulation 3.4 times increase in LHRH vaginal membrane permeability compared with a solution formulation. Vaginal membrane permeability from the Polycarbophil was greater than that from Pemulen hydrogels. This may be due to the larger bioadhesive values. LHRH solution with EDTA(2%), STDHF(1%) and sodlaurate(0.5%) showed 4.1 times, 4.8 times and 6.0 times of ovulation inducing activity compared with control. These results suggest that enzyme inhibition effect of EDTA, STDHF and sod, laurate may be result in substantial enhancement of vaginal absorption. By administraiton of Polycarbophil hydrogels containing LHRH the ovulation inducing activity was 3.3 times greater than the solutions. This result indicates the bioadhesive hydrogels as well as peptidase in hibition significantly improved absorption of LHRH. By coadministration with these inhibitors the ovulation inducing activity of Polycarbophi hydrogel containing LHRH was comparable with subcutaneous administration in ovulation inducing activity.

Screening for Antioxidant Effects of Aerial Part Extracts Obtained from Sixteen Compositae Species (국화과 식물 16종 지상부 추출물의 항산화효과 탐색)

  • The attempts to develop natural antioxidants have been made with aerial part of 16 Compositae species by analyzing their phenolic compound contents, scavenging activities on 1,l-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis-(3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) radicals, ferrous ion chelating effects and inhibition effects on peroxidation of linoleic acids. Total polyphenol and flavonoid contents were highest in Matricaria recutica. Scavenging activity on DPPH and ABTS radicals were highest with extracts of Echinacea angustifolia and Serratula coronata var. insularis f. insularis, respectively. Aerial palt extracts of all species showed lower DPPH scavenging activity than ascorbic acid and 2,6-Di-tert-butyl-4-methylphenol (BHT). But Serratula coronata var. insularis f. insularis demonstrated higher ABTS scavenging activity than ascorbic acid and BHT. In Hieracium pilosella, Echinacea angustifolia, Matricaria reculica extracts showed higher ABTS scavenging activity than BHT. Ferrous ion chelating effects was highest with Matricaria recutica extract, but the effects were much lower than ethylenediaminetetraacetic acid (EDTA). The inhibition activity in lipid peroxidation of linoleic acids was highest in Eupatorium japonicum with 90.06% inhibition 4 days after reaction and 40.52% after 24 days. This demonstrated higher inhibition activity and longer lasting than BHT. Aerial part of Matricaria recutica for extraction source, rather than flower, has higher potential for antiox.idant material. In conclusion, development of natural antioxidants in Compositae is possible by studying antioxidant activity of each species.

Effects of Phytic Acid on Viability of Vibrio vulnificus and on Septicemia-Induced Mice (피틴산이 비브리오균의 생존과 마우스의 패혈증에 미치는 영향)

  • EDTA is known to have bacteriocidal effect on Vibrio vulnificus, pathogen of septicemia by osmotic shock in seafoods. Attempts were made to elucidate the bacteriocidal effect of phytic acid (PA) as a substitute for EDTA against V. vulnificus and its inhibition effect on the septicemia, which induces liver damage of the mice by the pathogen. Viable cells of V. vulnificus with the initial titre of $1.7{\times}10^6$ c.f.u. $ml^{-1}$ decreased by 90.6% after 1 min and 99.6% after 5 min in distilled water. The titre decreased by 65.9% and 94.5% in 2 mM solution of $Mg^{2+}$. In 0.1 mM solution of PA, the rate of decrease in titre was 97.4% after 1 min of incubation and 99.8% after 5 min, compared to 95.7% and 99.8% in 0.1 mM solution of EDTA. The bacteriocidal effect of PA solution at a concentration of 1 mM was marked: the rate of decrease in titre was 99.9% after 1 min. In relation to the bacteriocidal effect, PA was evaluated as a potential therapeutic agent for V. vulnificus septicemia in mouse. When the survival periods of mice were investigated by PA and EDTA treatment after the pathogen injection, the group of mice which infected by a low concentration of the strain survived longer than that inoculated at high concentration; also, the ratio of survival was 1.3 times higher in PA than in EDTA, showing that the fatal rate depended on the inoculation concentration. Although survival periods of mice induced with liver damage by carbon tetrachloride and then inoculated with the strain showed a similar trend, the fatal rate of mice was 2 times faster than those inoculated with only pathogen into normal liver, These results indicate that the infection by V. vulnificus was more fatal to those with liver disease. Also, symptoms of hemorrhage and inflammation on the mice with induced liver damage were reduced in case there was phytic acid treatment at each concentration.

Characterization of Alternaria alternata ${\alpha}-Amylase$ (Alternaria alternata ${\alpha}-Amylase$의 특성에 관한 연구)

  • The ${\alpha}-amylase$ of Alternaria alternata was purified through ammonium sulfate precipitation, dialysis and Sephadex G-100 column chromatography. One single band was obtained in SDS-polyacrylamide gel electrophoresis. The optimum pH for enzyme activity was 5.0 and the enzyme activity was maintained at $3.6{\sim}7.0$pH range. The optimum temperature for ${\alpha}-amylase$ activity was $40^{\circ}C$ and 71% of the activity was still maintained until 30 min after heating at $80^{\circ}C$. The ${\alpha}-amylase$ was slightly activated by $Mn^{2+},\;Zn^{2+}\;and\;Sn^{2+}$, but inhibited by $Ba^{2+},\;Pb^{2+},\;Co^{2+}\;and\;Ag^{1+}$. The $Hg^{2+}\;and\;Ag^{2+}$ slightly inhibited the activity of the enzyme at concentrations of $10^{-3}\;and\;10^{-4}M$. The Michaelis constant $(K_m)$ to soluble starch was $6.50{\times}10^{-2}M$ and inhibition constant $(K_i)$ by the 1mM EDTA was $8.0{\times}10^{-2}M$. The inhibition of this enzyme by EDTA was competitive one.

Inhibition of Glutamate-Induced Change in Mitochondrial Membrane Permeability in PC12 cells by 1-Methylated β-carbolines

  • 1-Methylated $\beta$-carbolines (harmaline and harmalol) and antioxidants (N-acetylcysteine and ascorbate) reduced the loss of cell viability in differentiated PC 12 cells treated with 5 mM glutamate. $\beta$-Carbolines prevented the glutamate-induced decrease in mitochondrial membrane potential, cytochrome c release and caspase-3 activation in PC 12 cells. $\beta$-Carbolines reduced the formation of reactive oxygen species and depletion of glutathione due to glutamate in PC12 cells. $\beta$-Carbolines revealed a scavenging action on hydrogen peroxide and reduced the iron and EDTA-mediated degradation of 2-deoxy-D-ribose. The results suggest that I-methylated $\beta$-carbolines attenuate the cytotoxic effect of glutamate on PC12 cells by reducing the alteration of mitochondrial membrane permeability that seems to be mediated by oxidative stress.

Partial Purification and Characterization of an Extracellular Protease from Xenorhabdus nematophilus a Symbiotic Bacterium Isolated from an Entomopathogenic Nematode, Steinernema glaseri

  • Entomopathogenic nematodes are used for insect control. Herein, an extracellular protease was partially purified from a culture supernatant of Xenorhabdus nematophilus, a symbiotic bacterium of an entomopathogenic nematode, Steinernema glaseri: using precipitation with $80\%$ v/v isopropyl alcohol followed by gel permeation chromatography with a packed Sephacryl S-300 HR media. The partially purified protease exhibited maximal activity at pH 7 in the presence of 1 mM $CaCl_2$. The protease was identified as a metallo-protease based on the inhibition of its activity by the metal chelating agent, EDTA.

Characterization of a Fibrinolytic Serine Protease from a Wild Mushroom, Lepista nuda

  • Fibrinolytic enzyme was purified from the fruiting bodies of Lepista nuda, using DEAE-Cellulose chromatography, Phenyl Sepharose chromatography, and Mono-S column chromatography. The substance has a molecular weight of 30006.62 Da as measured by MALD-TOF mass spectrometry. The N-terminal amino acid sequence of the enzyme was Tyr-Pro-Ser-Pro-Ser-His-Gln-Thr-Ala-Val-Asn-Ala-Ile-Ile-X. The activity of the enzyme was inhibited by PMSF, indicating that the enzyme is a serine protease. No inhibition was found with E-64, pepstatin, and EDTA. It has broad substrate specificity for synthetic peptides. The enzyme was stable up to $30^{\circ}C$. The enzyme hydrolyzes both Aa and y chains of human fibrinogen but did not show any reactivity for $B{\beta}$ chain of human fibrinogen.

  • Metalloenzyme was purified from the fruiting bodies of Tricholoma sejunctum. MALDI-TOF and ICP/MS analyses revealed that the enzyme had a molecular weight of 18788.25 and includes $Zn^{2+}$ ion. The N-terminal amino acid sequence of the enzyme was Ala-Thr-Tyr-Lys-Ile-X-Ser-Ala-Thr-His-Gln-X-X-Leu-Val. The activity of the enzyme was inhibited by EDTA and 1,10-phenanthroline, indicating that the enzyme was a metalloprotease. No inhibition was found with E-64 and pepstatin. It has broad substrate specificity for synthetic peptides. The enzyme was stable up to $40^{\circ}C$. The activity of the enzyme was increased by $Zn^{2+}$ and $Co^{2+}$, while it was totally inhibited by $Hg^{2+}$. The enzyme hydrolyzes $A{\alpha}$ subunit of human fibrinogen but did not show any reactivity for $B{\beta}$ and ${\gamma}$ form of human fibrinogen.

Effects of Calcium Antagonists on Superoxide Generation, NADPH Oxidase Activity and Phagocytic Activity in Activated Neutrophils (칼슘 길항제가 활성화된 호중구에서의 $O_{\overline{2}}$의 생성, NADPH oxidase활성도 및 탐식작용에 미치는 영향)

  • NADPH oxidase dependent superoxide generation and phagocytosis in neutrophils stimulated with opsonized zymosan or heat aggregated IgG were coincided with the process of calcium uptake. The responses in activated neutrophils were enhanced with increasing concentrations of extracellular calcium and these effects were significantly inhibited by calcium chelators, EGTA and EDTA. The superoxide generation in activated neutrophils was reduced by dantrolene and chlorpromazine. Calcium antagonists, bepredil, diltiazem, verapamil, nifedipine and nimodipine effectively inhibited the calcium uptake, superoxide generation and phagocytosis in activated neutrophils, and NADPH oxidase activity was also inhibited. The results suggest that calcium antagonists may inhibit the superoxide generation and phagocytosis in activted neurtophils by the inhibition of calcium influx and by the action on intracellular redistribution of calcium and NADPH oxidase system.

  • Polyphenol oxidase in Prunus salicina(Red) was extracted, some properties and its partially purification were investigated as follows; Polyphenol oxidase was purified about 15 folds after ammonium sulfate saturation and about 64 folds after Sephadex G-100 column chromatography. Polyphenol oxidase showed optimum pH for activity at 6.5 and optimum temperature at at $35^{\circ}C$ and high affinity to catechol in o-diphenol compounds. Thermal stability were about 85% and 75% of initial polyphenol oxidase activity remained after heating at $50^{\circ}C$ for 5 minutes and 30 minutes respectively. The Michaelis constant of the enzyme was 2.58mM. L-cysteine, glutathione, ascorbic acid and potassium cyanide appeared to be most effective inhibitors. EDTA showed a slight inhibition.
