Development of Refolding Process to Obtain Active Recombinant Human Bone Morphogenetic Protein-2 and its Osteogenic Efficacy on Oral Stem Cells

  • BMP-2 is a well-known TGF-beta related growth factor, having a significant role in bone and cartilage formation. It has been employed to promote bone formation in some clinical trials, and to differentiate mesenchymal stem cells into osteoblasts. However, it is difficult to obtain this protein in its soluble and active form. hBMP-2 is expressed as an inclusion body in the bacterial system. To continuously supply hBMP-2 for research, we optimized the refolding of recombinant hBMP-2 expressed in E. coli, and established an efficient method by using detergent and alkali. Using a heparin column, the recombinant hBMP-2 was purified with the correct refolding. Although combinatorial refolding remarkably enhanced the solubility of the inclusion body, a higher yield of active dimer form of hBMP-2 was obtained from one-step refolding with detergent. The refolded recombinant hBMP-2 induced alkaline phosphatase activity in mouse myoblasts, at $ED_{50}$ of 300-480ng/ml. Furthermore, the expressions of osteogenic markers were upregulated in hPDLSCs and hDPSCs. Therefore, using the process described in this study, the refolded hBMP-2 might be cost-effectively useful for various differentiation experiments in a laboratory.

Antihypertensive Effects of Amlodipine Besylate and Its New Salts (암로디핀의 베실레이트염과 신규 염들의 항고혈압작용 비교평가)

  • The vascular relaxant and antihypertensive effects of newly developed salts of amlodipine-maleate and camsylate-were evaluated on isolated aorta from rats and in spontaneously hypertensive rats, and compared with those of amlodipine besylate, a standard drug. Amlodipine besylate concentration-dependently inhibited $Ca^{2+}$-induced contraction in depolarised rat aorta($IC_{50}$/: 4.17 nM), with a very slow onset of action. Amlodipine maleate and amlodipine camsylate also showed vascular relaxant effect with a pattern and a potency similar to those of amlodipine besylate($IC_{50}$/: 3.62 and 3.28 nM, respectively). Amloclipine besylate produced a dose-dependent and long-lasting(>10∼24h) antihypertensive effect with a slow onset of action (ED$_{20}$: 2.31 mg/kg) in spontaneously hypertensive rats. Amlodipine maleate and amlodipine camsylate also exerted antihypertensive effects with a pattern and a potency similar to those of amlodipine besylate(ED$_{20}$: 2.09 and 2.21 mg/kg, respectively). These results suggest that amlodipine maleate and amlodipine camsylate are not statistically differ with amlodipine besylate in relaxant effect of $Ca^{2+}$-induced contraction in depolarised rat aorta and in antihypertensive effect in spontaneously antihypertensive rats.

Endophytic Diaporthe sp. ED2 Produces a Novel Anti-Candidal Ketone Derivative

  • This study aimed to examine the anti-candidal efficacy of a novel ketone derivative isolated from Diaporthe sp. ED2, an endophytic fungus residing in medicinal herb Orthosiphon stamieus Benth. The ethyl acetate extract of the fungal culture was separated by open column and reverse phase high-performance liquid chromatography (HPLC). The eluent at retention time 5.64 min in the HPLC system was the only compound that exhibited anti-candidal activity on Kirby-Bauer assay. The structure of the compound was also elucidated by nuclear magnetic resonance and spectroscopy techniques. The purified anti-candidal compound was obtained as a colorless solid and characterized as 3-hydroxy-5-methoxyhex-5-ene-2,4-dione. On broth microdilution assay, the compound also exhibited fungicidal activity on a clinical strain of Candida albicans at a minimal inhibitory concentration of $3.1{\mu}g/ml$. The killing kinetic analysis also revealed that the compound was fungicidal against C. albicans in a concentration- and time-dependent manner. The compound was heat-stable up to $70^{\circ}C$, but its anti-candidal activity was affected at pH 2.

수소 버블을 이용한 전기화학적 그래핀 박리법

  • 그래핀은 전기적, 광학적, 역학적, 열적 특성 등이 아주 좋은 소재이다. Thermal Chemical Vapor Deposition (T-CVD) 장비를 이용 저 진공, 고온에서 CH4과 H2를 가스를 사용하여 그래핀을 합성을 하였다. 그래핀은 탄소만으로 이루어진 2차원 층상구조를 가지고 있다. 촉매 금속 위에서 합성이 이루어지기에 합성된 그래핀을 바로 다른 응용하기에는 어려움이 따른다. 따라서 촉매 금속에서 그래핀을 분리하여 원하는 곳으로 옮기는 과정이 필요한데, 이를 전사공정이라 한다. 최근 전기분해를 이용하여 발생되는 수소 버블을 사용하여 그래핀을 촉매로 사용되는 금속으로부터 분리해내는 Electrochemical Delamination(ED) 전사방식이 소개가 되었다. 이러한 전사 방식의 장점은 촉매기판을 제거하지 않음으로써, 다시 재활용이 가능하고, 공정에 필요한 시간이 짧다. 또한 표면에서 직접적으로 분리하는 방식이기에 촉매 금속의 양면을 사용이 가능하다. 이러한 ED방식의 장점이 있기에 공정의 최적 조건을 잡기 위하여 변수들을 바꾸어가면서 실험을 하였다. 전사된 그래핀은 표면을 광학현미경으로 확인하였고, 라만 분광기를 사용하여 라만 스펙트럼과 기본적인 전기특성을 확인하여 특성을 평가하면서, 기존의 전사방식을 사용한 그래핀 샘플과 비교분석 하였다.

  • A series of nurmerical analysse for the earhtquake of Iran railway tunnles under construction by NATM(New Astrian Tunnelling Method) were careid out throuth a pseudo-dynamic analyses techique used in a FFM computer program, DWTAP(Daewoo Tunnel Analysis Program), and the results are described in the paper. The analyses were performErl for two case;one is for the primary supports and the other is for the rompletEd permanent roncrete lining. The horizontal and verical groW1d accelerations for the design were estimatEd as 0.34 g and 0.23 g, respectively based on the historical reismic rerords in the proj3et area and the empirical equations. The results show that the turmel would be safe W1der the anticipitOO earthquake motion with the permanent roncrete lining, but some minor cracks rnigt be developErl in the primary shotcrete lining without any significant structural damages.

The Physicochemical Stabilities and Biological Activities of Pigment Extracts from Pseudoalteromonas sp. Ju11-1 and Pseudoalteromonas sp. Ju14 (Pseudoalteromonas sp. Ju11-1과 Pseudoalteromonas sp. Ju14의 색소 추출물의 물리화학적 안정성과 기능성)

  • We investigated the physicochemical stabilities and biological activities of ethanol- extracted pigment from marine bacteria Pseudoalteromonas sp. Ju11-1 and Pseudoalteromonas sp. Ju14. The bacterial pigment of strain Ju11-1 was very stable at pH 5.0 below $25^{\circ}C$. The stability of the pigment showed higher stability in the presence of metal ions such as $Cu^{2+}$ and $Mg^{2+}$. The pigment has activity of free-radical scavenging ($IC_{50}$ $95.2{\mu}g$/ml) and the protective antioxidant effect ($ED_{50}$ $82.3{\mu}g$/ml) against DNA damage in human lymphocytes. The bacterial pigment of strain Ju14 was very stable at pH range between 4.0 and 8.0 below $40^{\circ}C$. In the presence of light, the pigment was also very stable, showing more than 90 percent of remaining absorbance during 14 days at $25^{\circ}C$. The stability of the pigment, when metal ions were present, showed higher stability in all examined metal ions except for $Fe^{2+}$, $Al^{3+}$, and $Cu^{2+}$, especially in the presence of $Na^+$. The pigment has activity of freeradical scavenging ($IC_{50}$ $208.6{\mu}g$/ml) and the protective antioxidant effect ($ED_{50}$ $ 96.4{\mu}g$/m) against DNA damage in human lymphocytes. The result indicates that the bacterial pigments from marine bacteria, Pseudoalteromonas sp. Ju11-1 and Pseudoalteromonas sp. Ju14 showed higher physicochemical stability and significant effects for reduction in oxidative DNA damage. Therefore, the results suggest that these bacterial pigments could be used as a natural colorant having the advantages of antioxidant.

  • This paper presents the investigations and the reduction methods for EMI in the Elevator(E/L) system. In the large capacity E/L. Power factor controlled IGBT converter is used, so that the conducted EMI level is mostly determined by a high frequency input current. We suggest the low cost EMI countermearsure for E/L system using by a LCL filter ed a low cost noise filter.

Pharmacological properties of the reversible inhibitor of the gastric $H^+/K^+$ ATPase, AU-164

  • AU-164 was synthesized as a reversible gastric $H^+/K^+$ ATPase inhibitor, and its effects were tested in various systems. AU-164 inhibited rabbit gastric $H^+/K^+$ ATPase with an $IC_{50}$/ of 9 $\mu$M. On the other hand, AU-164 was a weak inhibitor for dog kidney $Na^+/K^+$ ATPasc, indicating the selectivity for gastric $H^+/K^+$ ATPase. The reversible property of the AU-164-induced inhibition of $H^+/K^+$ ATPase was confirmed by filtering the inhibition mixture through Sephadex G-25M column. In vivo basal acid secretion was also inhibited by AU-164 under the pylorus ligation of Sprague-Dawley rats. In addition, AU-164 protected dose dependently gastric lesion induced by ethanol in rats. The $ED_{50}$ value of 62 mg/kg p.o was estimated. These results suggest that AU-164 is a potent, selective and reversible gastric $H^+/K^+$ ATPase inhibitor, and that AU-164 has a potential use for the clinical therapeutics of peptic ulcer disease.

Toxic Evaluation and Chromatographic Analysis of Cucurbitacin D and F from Sorbaria sorbifolia (쉬땅나무(Sorbaria sorbifolia) 성분으로서 cucurbitacin D, F의 독성평가 및 정량)

  • Cucurbitacin D and F, the protostane type triterpenoid of S. sorbifolia, were isolated with chromatograpic method and used as the standard substances for quantitative analysis. The compounds were identified with $^1H$-NMR, FAB-MS and UV spectrophotometer. They were separated on YMC-Pack ODS-AQ(303)[$250{\times}4.6mm$ I.D., $S-5{\mu}m$, 120A] column by HPLC. Cucurbitacin F was detected at 10.73mg/kg in cortex of S. sorbifolia, but cucurbitacin D was not. The compounds were shown to exihibit significant cytotoxicity($ED_{50}$<$0.1{\mu}g/mL$) against several tumor cell lines and acute toxicity(cucurbitacin D: 4.7mg/kg/day, cucurbitacin F: 2.5mg/kg/day) against BDF-1 mouse.

Effect of different harvesting times on the nutritive value and fermentation characteristics of late and early-maturing forage oats by rumen microbes

  • Late-maturing Dark Horse, and early-maturing High Speed oat varieties were seeded on March 3, 2016 and harvested on three periods: May 31, June 10, and June 20 coded as early, mid, and late-harvest, respectively. Dried and ground samples were subjected to chemical analysis to determine nutritional values such as crude protein (CP), neutral detergent fiber (NDF), acid detergent fiber (ADF), ether extract (EE), organic matter (OM), and total digestible nutrient (TDN). Effective degradability (ED) of nutrients and fermentation characteristics including volatile fatty acid (VFA) composition, pH, gas production, and ammonia-N concentration were evaluated through an in vitro digestion method. Varieties of oat hays showed significant difference in terms of nutritional value, ED, and fermentation characteristics. Dark Horse showed higher CP and OM, and lower EE contents than High Speed. Dark Horse also showed higher EDDM (dry matter), NDF, ADF, and OM than High Speed, and although High Speed showed higher pH and ammonia-N, it had lower gas and total VFA production than Dark Horse. However, in terms of harvest period, significant difference was only observed in Dark Horse where early-harvest increased the CP, and late-harvest increased the NDF and OM contents. In addition, early-harvest of Dark Horse increased the EDDM and EDNDF of the forage. Therefore, early-harvest of late-maturing Dark Horse would give better nutrient efficiency than High Speed. Allowing Dark Horse to advance in maturity would decrease its nutrient productivity and efficiency.
