니오븀의 전자빔 용접성 (Electron beam weldability of Niobium)

  • Electron beam (EB) weldability of pure grade Nb sheet was studied. One of Nb sheets was as-annealed and the other was cold rolled. Microstructures, Vickers hardness, and transverse weld tensile test were carried out for the base metal, the heat affected zone (HAZ) and weld metal. In the case of the EB welds made using the annealed Nb sheeet, fine equiaxed grains and coarse grains were dominant at the base metal and the HAZ, respectively, and columnar grains were observed at the weld metal. For the EB welds made using the cold rolled Nb sheet, elongated grains in the rolling direction at the base metal, and the microstructures of the weld metal and the HAZ are similar to those of the EB welds made using the annealed Nb sheet, respectively. For both annealed and cold rolled Nb sheet, the width of the HAZs are unusually wide in spite of using high density heat source, i.e. electron beam, and the grain sizes of both HAZs are similar. When tensile test was carried out using the transverse weld specimens, the failure occurred at the HAZ for both EB welds made using Nb sheets annealed and cold rolled, respectively and the tensile strengths of both specimens were 161MPa. Vickers hardness of EB welds made using annealed Nb was 56-57 Hv at both base metal and weld metal, 52-53 Hv at the HAZ. On the other hand, Vickers hardness of EB welds made using cold rolled Nb was 97-99 Hv at the base metal, but the hardness values of weld metal were similar to those obtained at the weld metal of annealed Nb.

  • A business process specification (BPS) plays the role of a contracted business scenario in the execution of the B2B business process instances. XML-based specification languages, such as BPSS, WS-BPEL, etc., are usually adopted for the specifications. However, composing complex XML-based specifications are not easy for humans. As an alternative, graphical modeling languages such as UML and BPMN can be employed. This paper presents an UML-based modeling method for graphically specifying ebXML BPSS instances (namely, business process specifications). UML diagrams, directly matching with ebXML BPSS, can be reused for understanding and generating ebXML BPSS instances. We discuss the value of tile method by developing a business reference library in the area of supply chain management.

  • Plasma resistant nanocrystalline $Y_2O_3$ films were deposited on alumina substrates through the electron-beam PVD technique. Increasing substrate temperature to $600^{\circ}C$ resulted in the textured microstructures with significantly enhanced adhesion force of the coating to the substrate. During the exposure to fluorine plasma, erosion rate of the coated specimen was higher than that of a sintered yttria specimen, but significantly lower than that of a single crystalline alumina. Considering the adhesion and erosion behaviors observed in the coated specimen prepared at $600^{\circ}C$, the deposition technique appears effective in reducing contamination particles generated from the ceramic parts in the plasma environment.

화살나무 및 느릅나무 추출물이 면역계세포의 활성에 미치는 영향 (Effect of Euonymus alatus and Ulmus clavidiana var japonica on the immune system)

  • We have previously shown that crude water extract of Euonymus alatus (EA) had strong prophylactic effect against chemically induced-and tumor cell implanted-cancer, and that the mechanisms responsible for its antitumor effects were due to nonspecific enhancement of the NK cell activities and the cell mediated immunity. However, it was unknown that any components of crude extract did work so, since it consisted of several components. In this paper, we fractionated the crude watar EA-extract into several fraction such as hexane-, ethylether-, ethyl acetate-, n-butanol- and water soluble-fraction, and screened the immune regulating activities of each fraction by the evaluation of lymphokine production and activated lymphocyte proliferation. As a result of the component fraction of EA-extract, it was found that n-butanol fraction was a potent immunostimulator, and the remained water soluble fraction also contained some stimulator, But, other fraction did not showed any remarkable effect. It is therefore suggested that EA-glycosides in n-butanol fraction may be new one of the potent biological response modifiers. The present study was also undertaken in an efforts to investigate the effects of elm-bark(EB, Ulmus clavidiana var japonica), which has been used for curing ulcer and inflammation as a folk medicine without any kind of experimental evidence to support this, on the cellular- and humoral-immune responses, lymphocyte function and NK cell activities in mice. Regardless of time and duration of EB-treatment, Arthus reaction and antibody response to SRBC were not modified by EB, but delayed hypersensitivity to SRBC was significantly enhanced only when EB was treated prior to SRBC-sensitization. EB slightly inhibited the proliferation responses of splenocytes to PHA-stimulation, but it significantly augmented the responses of these cells to S aureus Cowan 1 and Con A-activation, and these effects were manifested only when EB was added at culture initiation. EB did not influence Ig secretion of spleen cells but it significantly augmented the Con A-induced 1L 2 and MIF production of splenocytes. NK cell activities of splenocytes were markedly riled when effector cells were pretreated with EB and this augmentation was dine to the increase of binding affinity of effector cells to target cells and the target cell lytic activities of effector cells. These results led to the conclusion that EB triggers increase of cellular immune responses, such as delayed hypersensitivitiy, lymphokine production and NK cell activities. Also these results suggested that EB contains potent immune stimulants, which may provide the rational basis for their therapeutic use as one of the new biological response modifiers.

탄저병균에 대하여 길항작용을 보이는 Burkholderia cepacia EB215로부터 분리한 Pyrrolnitrin의 항균활성 (In vivo Antifungal Activity of Pyrrolnitrin Isolated from Burkholderia capacia EB215 with Antagonistic Activity Towards Colletotrichum Species)

  • 오이 뿌리조직으로부터 분리한 식물내생세균 EB215균은 탄저병균인 Colletotrichum species에 대하여 강한 항균활성을 보였다. 이 균은 생리 생화학적 특성과 Biolog 실험 및 16S rDNA 유전자 서열에 의해 Burkholderia cepacia로 동정되었다. 이 균의 항균물질 생산을 위한 최적배지는 nutrient 액체(NB) 배지로 그리고 배양기간은 3일로 결정되었다. B. cepacia EB215 균주의 NB 배양체로부터 원심분리, n-hexane 분획, silica gel 컬럼, preparative TLC 및 in vitro 생물검정 등을 통하여 한 개의 항균물질을 분리하였다. 이 물질은 질량분석과 핵자기공명 분석을 통하여 pyrrolnitrin으로 동정되었다. Pyrrolnitrin은 고추 탄저병(Colletotrichum coccodes), 오이 탄저병(Colletotrichum orbiculare), 벼 도열병(Magnaporthe grisea), 벼 잎집무늬마름병(Corticium sasaki) 등의 4가지 식물병에는 $11.1\;{\mu}g/ml$ 낮은 농도에서도 90% 이상의 높은 방제활성을 보였다. 그리고 또한 토마토 잿빛곰팡이병(Botrytis cinerea)과 밀 붉은녹병(Puccinia recondita)에 대해서는 $33.3\;{\mu}g/ml$ 이상의 농도에서 90% 이상의 방제활성 보였다. 하지만 Phytophthora infestans에 의한 토마토 역병에 대해서는 전혀 항균활성이 없었다. 앞으로 B. cepacia EB215균을 이용한 미생물살균제 개발에 대하 연구를 진행할 예정이다.

Construction of Shuttle Promoter-probe and Expression Vectors for Escherichia coli and Bacillus subtilis, and Expression of B. thuringiensis subsp. kurstaki HD-73 Crystal Protein Gene in the Two Species

  • A shuttle promoter-probe vector, pEB203, was derived from pBR322, pPL703 and pUB110. Using the vector, a useful DNA fragment, 319 bp EcoRI fragment, having strong promoter activity has been cloned from Bacillus subtills chromosomal DNA. Selection was based on chloramphenicol resistance which is dependent upon the introduction of DNA fragments allowing expression of a chloramphenicol acetyl transferase gene. The nucleotide sequence of the 319 bp fragment has been determined and the putative -35 and -10 region, ribosome binding site, and ATG initiation codon were observed. This promoter was named EB promoter and the resultant plasmid which can be used as an expression vector was named pEBP313. The crystal protein gene from B. thuringiensis subsp. kurstaki HD-73 was cloned downstream from the EB promoter without its own promoter. When the resultant plasmid, pBT313, was introduced into Escherichia coli and B. subtilis, efficient synthesis of crystal protein was observed in both cells, and the cp gene expression in B. subtilis begins early in the vegetative phase. The cell extracts from both clones were toxic to Hyphantria cunea larvae.

전자선을 이용해 가교된 SPEEK 기본 물질로 하는 이온 교환막의 특성 분석 (The Characterization of Crosslinked SPEEK Based Ion Exchange Membranes Prepared by EB Irradiation Method)

  • Crosslinked SPEEK/PVDF membrane were prepared by EB radiation method with various contents of PVDF. The prepared membranes were subjected to a comparative study of proton exchange membranes for fuel cell appreciations. The crosslinked SPEEK/PVDF membranes were characterized by using DMA, DSC and SAXS. The DMA data indicate that the ionic modulus values and cluster $T_g$ decrease with increasing PVDF content. Thus, it was suggested that the number of clustering in the crosslinked membranes can be reduced with increasing PVDF content. The DSC results were shown that the degree of crystalline of the membrane increased with increasing PVDF content. The morphology of the crosslinkied membranes was shown that with increasing PVDF content, the number of crystalline domain of the SPEEK/PVDF membranes increased but ionic aggregation of the membranes decreased. The water uptake behavior, ionic exchange capacity (IEC) and proton conductivity were decreased with increasing PVDF content. The overall findings suggest that the crosslinked membranes offer the possibility for improving the performance of PEMFC, provided that the membranes have thermal and hydration stability.

Electron Beam 조사가 고추 및 생강 분말의 미생물학적 및 관능적 품질에 미치는 영향 (Effects of Electron Beam Irradiation on Microbiological and Organoleptic Qualities of Powdered Red Pepper and Ginger)

  • 향신료의 살균기법을 연구할 목적으로 전자선(electron beam) 이용하여 고추 및 생강 분말의 미생물학적 및 관능적 품질에 미치는 영향을 감마선과 비교 평가하였다. 사용된 시료의 총세균은 $10^5{\sim}10^6\;CFU/g$, 효모 및 곰팡이는 $0{\sim}10^3$의 수준이었고, 대장균군은 $0{\sim}10^2\;CFU/g$ 정도로 혼입되어 있었다. 전자선 조사(5 kGy)는 일반세균을 $2{\sim}3\;log$ cycles 정도 감소시킬 수 있었고, 대장균군은 2.5 kGy 조사에서도 검출되지 않았다. 초기 미생물 농도를 90% 사멸시키는데 필요한 조사선량$(D_{10}\;value)$을 계산하여 보았을 때, 고추분발의 경우 전자선 $1.50{\sim}1.54\;kGy$, 감마선 $1.68{\sim}1.80\;kGy$ 범위였고, 생강분말에서는 전자선 $1.30{\sim}2.27\;kGy$, 감마선 $1.45{\sim}2.77\;kGy$로 시료의 미생물 농도와 에너지의 종류에 따라 다소 상이한 방사선 감수성을 나타내었다. 살균처리 후 실온에서 4개월 간 저장된 시료의 미생물 농도는 처리구간에 큰 차이 없이 증식현상을 거의 나타나지 않았다. 시료의 기계적 색도(Hunter's color)는 방사선 조사에 의한 영향은 거의 없었고, 4개월 저장에 따른 영향은 크게 나타났다 향신료의 색깔 및 냄새에 대한 관능검사에서 살균선량의 전자선 조사는 고추 및 생강 분말의 관능적 품질에 대하여 안정한 것으로 나타났다.
