Rheological Properties of White Pan Bread Dough Prepared with Angelica gigas nakai Powder (당귀 분말을 첨가한 식빵 반죽의 물리적 특성)

  • The effects of Angelica gigas nakai powder(AP) addition on bread dough were investigated by preparing dough with 0-10%(w/v) powder. Dough raising power, gluten levels, rapid visco properties, and falling number, were investigated. The rheological properties of dough as measured by mixography, farinography, alveography, color assessment, and with scanning electron microscopy, were examined. Increase in AP concentration resulted in a linear decrease in gluten content. Dough raising power and extensibility were decreased by water absorption rate, and resistance increased. Dough stability and rose when AP was present at 1-2%(w/v). Water absorption, dough stability, and dough valorimeter values also rose when AP was present at 1-2%(w/v), but AP induced weakness in the dough, as revealed through farinography, and also resulted in a lowering of initial pasting temperature and the temperature at peak viscosity. A decrease in viscosity at the peak point, and(as revealed by RVA), a decrease in extensibility, an increased resistance to extension, and a rise in the energy required for extension, were also seen when AP powder was added, as was an increase in the R/E ratio. Overall, the addition of AP to dough to levels of 1% or 2%(both w/v) is thought to be useful in the preparation of a functional white pan bread, and results in quality improvements.

A study on the practicability of genotyping and cytotoxicity of Actinobacillus actinomycetemcomitans isolated from periodontal patients (Actinobacillys actinomycetemcomitans의 유전자형의 유용성과 세포독성에 관한 연구)

  • The purpose of this study was to evaluate the genotypic characterization of Actinobacillus actinomycetemcomitans (A. actinomycetemcomitans) using arbitrarily primed polymerase chain reaction (AP-PCR), to investigate the cytotoxicity of both clinical isolates and standard strains of A. actinomycetemcomitans for the human Jurkat T cells, and to measure the osteoclastogenic cytokines released by Jurkat cells infected with these bacterial strains. The random sequence primer 15 and 16 could distinguish different AP-PCR profiles between clinical isolates of A. actinomycetemcomitans. A. actinomycetemcomitans significantly suppressed Jurkat cell viability in time dependent fashion and the results of DNA fragmentation assay indicated that this bacterial species induced apoptosis in Jurkat cells undergoing apoptosis released the osteoclastogenic cytokine, IL-1$\beta$, IL-6, TNF-$\alpha$. These data support the hypothesis that induction of apoptosis is at least one essential step in A. actinomycetemcomitans induced local immunosuppressive pathway, and that A. actinomycetemcomitans can modulate the immunomodulatory cell population in the periodontal tissue by inducing T cell death through apoptosis, and that apoptosis of local resident T cells may play an active role in bone resorption by releasing osteoclastogenic cytokines, e.g. IL-1$\beta$, IL-6, TNF-$\alpha$.

JS-III-49, a hydroquinone derivative, exerts anti-inflammatory activity by targeting Akt and p38

  • Since previous studies have reported that hydroquinone (HQ) exerted immunosuppressive and anti-inflammatory activity, various HQ derivatives have been synthesized and their biological activities investigated. In this study, we explored the anti-inflammatory activity of JS-III-49, a novel HQ derivative, in macrophage-mediated inflammatory responses. JS-III-49 suppressed the production of the inflammatory mediators nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) and down-regulated the mRNA expression of the inflammatory enzymes cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) as well as the expression of the pro-inflammatory cytokines interleukin-6 (IL-6) and IL-$1{\beta}$ without cytotoxicity in LPS-stimulated RAW264.7 cells. JS-III-49 inhibited nuclear translocation of the $NF-{\kappa}B$ transcription factors p65 and p50 by directly targeting Akt, an upstream kinase of the $NF-{\kappa}B$ pathway, in LPS-stimulated RAW264.7 cells. However, JS-III-49 did not directly inhibit the kinase activities of Src and Syk, which are upstream kinases of Akt, in LPS-stimulated RAW264.7 cells. Moreover, JS-III-49 suppressed the nuclear translocation of c-Fos, one of the components of AP-1, by specifically targeting p38, an upstream mitogen-activated protein kinase (MAPK) in the AP-1 pathway in LPS-stimulated RAW264.7 cells. These results suggest that JS-III-49 plays an anti-inflammatory role in LPS-stimulated macrophages by targeting Akt and p38 in the $NF-{\kappa}B$ and AP-1 pathways, respectively.

Factors Influencing Preferential Utilization of RNA Polymerase Containing Sigma-38 in Stationary-Phase Gene Expression in Escherichia coli

  • In order to understand the molecular basis of selective expression of stationary-phase genes by RNA polymerase containing$\sigma$$\^$38/ (E$\sigma$$\^$38/) in Escherichia coli, we examined transcription from the stationary-phase promoters, katEP, bo1AP, hdeABP, csgBAP, and mcbP, in vivo and in vitro. Although these pro-moters are preferentially recognized in vivo by E$\sigma$$\^$38/, they are transcribed in vitro by both E$\sigma$$\^$38/ and E$\sigma$$\^$70/ containing the major exponential $\sigma$, $\sigma$$\^$70/. In the presence of high concentrations of glutamate salts, how-ever, oldy E$\sigma$$\^$38/ was able to efficiently transcribe from these promoters, which supports the concept that the promoter selectivity of $\sigma$$\^$38/-containing RNA polymerase is observed only under specific reaction con-ditions. The examination of 6S RNA, which is encoded by the ssr1 gene in vivo, showed that it reduced E$\sigma$$\^$70/ activity during the stationary phase, but this reduction of activity did not result in the elevation of E$\sigma$$\^$38/ activity. Thus, the preferential expression of stationary-phase genes by E$\sigma$$\^$38/ is unlikely the con-sequence of selective inhibition of E$\sigma$$\^$70/ by 6S RNA.

COMPARISON OF LINEAR POLYMERIZATION SHRINKAGE IN COMPOSITES AND COMPOMER POLYMERIZED BY PLASMA ARC OR CONVENTIONAL VISIBLE LIGHT CURING (리노미터를 이용한 할로겐 가시광선 광조사기와 플라즈마 아크 광조사기의 복합레진 및 컴포머의 광중합 양상 비교)

  • The purpose of this study was to evaluate the effectiveness of plasma arc curing (PAC) unit for composite and compomer curing. To compare its effectiveness with conventional quartz tungsten halogen (QTH) light curing unit, the polymerization shrinkage rates and amounts of three composites (Z100, Z250, Synergy Duo Shade) and one compomer, that had been light cured by PAC unit or QTH unit, was compared using a custome made linometer. The measurement of polymerization shrinkage was peformed after polymerization with either QTH unit or PAC unit. In case of curing with the PAC unit, the composite was light cured with Apollo 95E for 6s, the power density of which was recorded as 1350 mW/$\textrm{cm}^2$ by Coltolux Light Meter. For light curing with QTH unit, the composite was light cured for 30s with the XL2500, the power density of which was recorded as 800 mW/$\textrm{cm}^2$ by Coltolux Light Meter. The amount of linear polymerization shrinkage was recorded in the computer every 0.5s for 60s. Ten measurements were made for each material. The amount of linear polymerization shrinkage for each material in 10s and 60s which were cured with PAC or QTH unit were compared with t test. The amount of polymerization shrinkage in the tested materials were compared with 1way ANOVA with Duncan's multiple range test. As for the amounts of polymerization shrinkage in 60s, there was no difference between PAC unit and QTH unit in Z250 and Synergy Duo Shade. In Z100 and Dyract AP, it was lower when it was cured with PAC unit than when it was cured with QTH unit (p<0.05). As for the amounts of polymerization shrinkage in 10s, there was no difference between PAC unit and QTH unit in Z100 and Dyract AP. The amounts of polymerization shrinkage was significantly higher when it was cured with PAC unit in Z250 and Synergy Duo Shade (p<0.05). The amounts of polymerization shrinkage in the tested materials when they were cured with QTH unit were Z250 (6.6um) < Z100 (9.3um), Dyract AP (9.7um) < Synergy Duo Shade (11.2um) (p<0.05). The amount of polymerization shrinkage when the materials were cured with PAC unit were Dyract AP (5.6um) < Z100 (8.1um), Z250(7.0um) < Synergy Duo Shade (11.2um) (p<0.05).

  • In this work $Cd_{4}GeSe_{6}$ and $Cd_{4}GeSe_{6}$ : $Co^{2+}$ single crystals were grown by the chemical transport reaction method and the structure of $Cd_{4}GeSe_{6}$ and $Cd_{4}GeSe_{6}$ : $Co^{2+}$ single crystals were monoclinic structure. The temperature dependence of optical energy 9ap was fitted well to Varshni equation. Also, the entropy, enthalpy and heat capacity were deduced from the temperature dependence of optical energy gap.

  • Cervical cancer is one of the leading causes of female death from cancer worldwide with about 500,000 deaths per year. A strong association between certain human papilloma viruses (HPV types 16 and 18) and cervical cancer has been well known. An extract of natural products, named as Somatid, has been used to investigate whether this agent has the ability of inhibiting the oncogenes E6 and E7 of HPV type 16. This Somatid inhibited the proliferation of human cervical cancer cell lines (C-33A, SiHa, CaSki) and HaCaT keratinocytes in a dose response manner, In vitro binding assay and ELISA showed that Somatid inhibited the in vitro biding of E6 and E6AP which are essential for the binding and degradation of the tumor suppressor p53. In addition, Somatid inhibited the in vitro binding of E7 and Rb which is essential tumor suppressor for the control of cell cycle. The levels of mRNA for E6 and E7 were also decreased by Somatid. Our data suggested that Somatid inhibited the oncogenecity of E6 and E7 of HPV 16 type, thus can be used as a putative anti-HPV agent for the treatment of cervical carcinomas caused by HPV.

An extract of Saururus chinensis, named as PE-46, has been used to investigate whether this agent has the ability of inhibiting the oncogenes E6 and E7 of HPV type 16. PE-46 inhibited the proliferation of human cervical cancer cell lines in a dose response manner. PE-46 also inhibited the in vitro binding of E6 and E6AP which are essential for the binding and degradation of the tumor suppressor p53. In addition, PE-46 inhibited the in vitro binding of E7 and Rb which is essential tumor suppressor for the control of cell cycle. The levels of mRNA for E6 and E7 were also decreased by PE-46. SiHa cells treated with PE-46 induced G0/G1 arrest, resulting in inhibition of growth. Our study showed that the PE-46 can inhibit the cervical carcinomas via both inhibition of bindings between oncogenes and tumor suppressors, and inhibition of G1longrightarrowS transition. PE-46 inhibited the oncogenecity of E6 and E7 of HPV 16 type, thus could be used as a putative modulating agent for the treatment of cervical carcinomas caused by HPV.

Evaluation of Effects on Tritium Measurement According to HTO Type Sample Preparation Conditions (HTO 형태 시료 조제 조건에 따른 삼중수소 계측에 미치는 영향 평가)

  • In this study, for the measurement of 3H(tritium) radioactivity concentration, a study was conducted on whether the type of cocktail and the material of the vial had an effect on the measurement before liquid scintillation counter measurement on HTO-type samples that had undergone physical and chemical pretreatment. As a result of the study, the efficiency according to the type of cocktail was higher in Ultima Gold LLT than Ultima Flo-AP cocktail with polyethylene (1.49%), glass (5.10%), and teflon (6.58%), respectively. Regarding the effect according to the type of vial, the efficiency and SQP(E) of both Ultima Gold LLT and Ultima Flo-AP showed the highest values in the order of teflon, polyethylene, and glass.
