감자로부터 Eukaryotic Translation Initiation Factor 5A (elF-5A) 유전자의 동정 및 발현 분석 (Isolation and Characterization of Eukaryotic Translation Initiation Factor 5A (eIF-5A) from Potato)

  • 감자 (Solanum tuberosum L. cv. Irish Cobbler)의 괴경형성과정 (tuberization) 동안에 발현하는 유전자들의 발현양상을 조사하고자 differential display법을 실시하였다. Differential display를 이용하여 분리된 eIF5A DNA단편을 probe로 사용하여 감자의 cDNA library screening을 통하여 eIF5A full-length cDNA를 감자에서 처음으로 분리하였다. 감자의 eIF5A, clone은 토마토의 eIF5A cDNA 염기서열과 94.8%. 아미노산 서열에서는 97.5%로 매우 높은 유사성을 나타내었다. 감자의 eIF5A 유전자는 길이가 716 bp로 하나의 단백질 code영역 (ORF)을 포함하고 있었다. 이 영역은 분자량 17.4 kD, pI 5.5로 추정되는 160개의 아미노산으로 구성된 eIF5A단백질을 code하고 있었다. eIF5A 단백질들에서 12개의 아미노산 서열 (STSKTGKHGHAK)은 효모에서 사람에 이르기까지 완벽하게 보존되어 있는 것으로 알려져 있는데, 감자에서도 또한 잘 보존되어 있었다. 이 영역은 eIF5A 단백질의 활성을 나타내는 데 있어서 필수적인 hypusine을 생성하는 전사 후 수식 부위가 들어 있는 아주 중요한 곳이다. 감자에서 eIF5A 유전자의 발현양상을 조사한 결과 감자의 전조직에서 발현을 보였는데, 성숙잎이나 괴경보다는 세포분열 및 물질축적이 활발히 일어나고 있는 꽃기관들 (stamen, ovary, petal. sepal), 과실 (fruit)과 stolen 등의 조직들에서 비교적 활발히 발현되고 있었다.

  • Structure-activity relationship studies of allylamine type of antimycotics were carried out to evaluate the effect of naphthyl and methyl portion of naftifine. Compounds with 3,4-difluorophenyl (2a-5a), 4-hydroxyphenyl (2b-5b), 3-nitro­phenyl (2c-5c), 4-chlorobenzothiazoly (2d-5d) and 5-methylfurfural (2e-5e) instead of naphthyl group, and with hydrogen (3a-3e), methyl (4a-4e) and ethyl (5a-5e) in the place of methyl in naftifine were synthesized and tested for their in vitro anti-fungal activities against five different fungi. Fourteen compounds (3a, 4a, 5a, 3b, 4b, 5b, 3c, 4c, 5c, 3d, 4d, 3e, 4e and 5e) showed significant anti-fungal activities against T. mentagrophytes. (E)-N-(3-Phenyl-2-propenyl)-3,4-difluoro-ben­zenemethaneamine(3a), (E)_N_(3_phenyl_2_propenyl)_4_hydroxy_benzenemethaneamine(3b) and (E)-N-ethyl-N-(3-phenyl­2-propenyl)-3-nitro-benzenemethaneamine (5c) displayed moderate anti-fungal activities against all five different fungi.

단백질합성인자 eIF5B의 저 발현 효모벡터의 제조 및 특성 (Construction and Characterization of Vector Expressing Low Level of Translation Factor eIF5B)

  • eIF5B는 단백질합성의 개시 인자로서 Met-$tRNA^{Met}$을 AUG 개시코돈에 전달하고, 리보솜의 두 소단위체 결합을 유도한다. 이 인자의 기능을 연구할 목적으로 eIF5B를 코딩하는 FUN12 유전자의 5'말단 부위를 삭제하는 연구를 수행하였다. 프로모터의 대부분을 삭제한 FUN12를 함유한 pRS효모벡터를 FUN12가 삭제되어 천천히 자라는 돌연변이주 ($fun12{\Delta}g$)에 전달하였을 때 그 표현형을 부분적으로 상보하였다. 위와 같이 제조된 벡터에서 N-말단이 상실된 eIF5B 단백질이 발현되었고, 그 양이 정상 균주에서 발현되는 eIF5B 양의 약 5%에 불과하였다. 이와 같이 부분적으로 성장을 상보한 균주에서 발현된 적은 양의 단백질합성개시 인자 eIF5B는 직접적으로 그 성장을 제한하는 요소로 작용한다. 이러한 균주에서 성장의 제한인자인 eIF5B는 in vitro 에서도 역시 전체 단백질 합성의 활성을 조절하였다.

분벽성 옥수수에 대한 Esterase 동위효소의 유전분석 (Genetic Analysis of Esterase Isozymes in Tillering Maize)

  • 분벽성 옥수수에 대한 esterase 동위효소의 유전양상을 알아보고자 acrylamide gel 6.5%를 사용하여 분벽성인 IK 계통과 대조구로 무벽성인 A-type 계통을 교배하여 얻은 $F_1$$F_2$ 종자를 28$^{\circ}C$ 암조건하에서 발아시킨 유조직 중심주를 공시하여 전기영동한 결과는 다음과 같다. 1. 본 실험에 공시된 중심주조직은 음극효소를 포함하여 총 13개의 동위효소가 확인되었으며 이동거리에 따라 유전분이 양상이 5개군으로 구분되었다. 2. 원점으로부터 0.5cm에 존재하는 E$_{0.3}$, E$_{0.4}$, E$_{0.5}$ 동위효소는 2개의 대립인자에 의해 지배되었고, E$_{0.3}$+E$_{0.4}$ 변이체는 공우성 대립인자에 의해 지배되었다. 3. 밴드의 이동거리가 1.0cm에서 1.8cm에 존재하는 E$_{1.0}$ , E$_{1.2}$ , E$_{1.5}$ , E$_{1.8}$ 의 4개의 동위효소는 2개의 대립인자에 의해 지배되었고 존재하지 않는 비활성 밴드가 우성으로 작용하였다. 한편 변이체로 나타난 E$_{1.2}$ +E$_{1.5}$ 의 동위효소는 공우성 대립인자로 작용하였다. 4. 밴드의 이동거리가 2.8cm에서 3.5cm에 존재하는 E$_{2.8}$ , E$_{3.0}$ , E$_{3.5}$ 인 세 개의 밴드는 이들 중에서 두 개씩 나타나는 밴드가 세 개 모두 나타나는 개체수보다 현저히 많이 나타났다. ($\chi$$^2$=0.327$^{**}$ ). 5. 밴드의 이동거리가 3.8cm 이상에서 나타난 동위효소는 활성이 매우 낮았고 이들 효소는 모두가 두 개의 대립인자에 의해 지배되었다. 따라서 분벽성 옥수수 중심주 조직에 대한 esterase 동위효소는 멘델식 유전법칙에 의해 지배된다고 생각된다.각된다.각된다.

  • 7'-Aldehyde-nucleoside 유사체(2a, 2c)을 6-N-benzoyl-2',3'-O-isopropylideneadenosiner과 uridine으로부터 합성하였다. 2와 Wittig reagent인 ethoxycarbonylmethylene을 축합시켜 ethoxycarbonyl group을 갖는 탄소수를 증가시킨 공액 이중 결합을 지닌 nucleosi de 유사체, ethyl-1',5',6',7',8'-pentadeoxy-1'-(adenin-9-yl)-$\beta$-D-ribo-nnona-5'(E).7'(E)-dienofuranuronate(4b), ethyl-1',5',6',7',8'-pentadeoxy-1'-(uracyl-1-yl)-$\beta$-D-ribo-nona-5'(E),7'(E)-dienofuranuronate(4c)을 얻었다. 또한 2와 CBr4, Ph3P을 반응시켜 공액 이중 결합을 지닌 9-[8',8'-dibromo-5',6',7',8'-tetradeoxy-$\beta$-D-ribo-octa-5'(E),7'(E)-diene]nucleosides (6b,6c) 유사체를 각각 합성하였다.

GIOVE-B 위성 E5a Secondary Code 결정 및 신호처리 결과 (Secondary Code Determination and Signal Processing Results of GIOVE-B E5a)

  • GIOVE-B is the second experimental Galileo satellite for the Galileo In-Orbit Validation, which was launched by a Soyuz/Fregat rocket departing from the Baikonur cosmodrome in Kazakhstan on 27 April and now operated successfully. This paper presents the results obtained from processing of the E5a signal transmitted from the GIOVE-B. The acquisition and tracking of the data and pilot channels are performed by the E5a software receiver implemented by ETRI. Moreover, the paper suggests the GIOVE-B E5a-I/E5a-Q secondary code, which is determined by analyzing the correlation output of the primary correlator using the primary code.

Interaction of Escherichia coli K1 and K5 with Acanthamoeba casfellanii Trophozoites and Cysts

  • The existence of symbiotic relationships between Acanthamoeba and a variety of bacteria is well-documented. However, the ability of Acanthamoeba interacting with host bacterial pathogens has gained particular attention. Here, to understand the interactions of Escherichia coli K1 and E. coli K5 strains with Acanthamoeba castellanii trophozoites and cysts, association assay, invasion assay, survival assay, and the measurement of bacterial numbers from cysts were performed, and nonpathogenic E. coli K12 was also applied. The association ratio of E. coli K1 with A. castellanii was 4.3 cfu per amoeba for 1 hr but E. coli K5 with A. castellanii was 1 cfu per amoeba for 1 hr. By invasion and survival assays, E. coli K5 was recovered less than E. coli K1 but still alive inside A. castellanii. E. coli K1 and K5 survived and multiplied intracellularly in A. castellanii. The survival assay was performed under a favourable condition for 22 hr and 43 hr with the encystment of A. castellanii. Under the favourable condition for the transformation of trophozoites into cysts, E. coli K5 multiplied significantly. Moreover, the pathogenic potential of E. coli K1 from A. castellanii cysts exhibited no changes as compared with E. coli K1 from A. castellanii trophozoites. E. coli K5 was multiplied in A. castellanii trophozoites and survived in A. castellanii cysts. Therefore, this study suggests that E. coli K5 can use A. castellanii as a reservoir host or a vector for the bacterial transmission.

일부(一部) 무의촌지역(無醫村地域)과 병원(病院)에서 분리(分離)한 E.coli의 항생제(抗生劑) 감수성(感受性)에 대(對)하여 (Studies on the Antibiotic Sensitivity of Escherichia coli Isolated from a Doctorless Area and Seoul National University Hospital)

  • 40 strains of E. coli isolated from residents of a doctorless area in Korea in 1976 and 40 strains of E. coli isolated from patients of Seoul National University Hospital from 1975 to 1976 were examined for susceptibilities to 14 antimicrobial agents by the agar dilution method. The susceptibilities of the two groups to each antimicrobial agent were compared and correlations in the antimicrobial susceptibility of the 80 strains of E. coli among the 14 antimicrobial agents were also analyzed. The results were obtained as follow: 1. With Tetracycline, Oxytetracycline, Doxycycline and Ampicillin, the mean MIC's of E. coli isolated from patients of Seoul National University Hospital were 8.6 to 14 times higher than. those of E. coli isolated from residents of a doctorless area. 2. With Streptomycin, Minocycline and Carbenicillin, the mean MIC's o{ E. coli isolated from patients of Seoul National University Hospital were 4.1 to 5.6 times higher than those of E. coil isolated from residents of a doctorless area. 3. With Kanamycin, Penicillin and Cotrimoxazole, the mean MIC's of E. coli isolated from patients of Seoul National University Hospital were 2.6 to 3.7 times higher than those of E. coli isolated from residents of a doctorless area. 4. There were no significant differences in susceptibility to Erythromycin respectively between E. coli isolated from patients of Seoul National University coli isolated from residents of a doctorless area. 5. E. coli isolated from patients of Seoul National University Hospital were resistant to Erythromycin(100%), Streptomycin(75%), Tetracycline(72.5%), Oxytetracycline(72.5%), Doxycycline(72.5%), Minocycline(67.5%), Penicillin(82.5%), Ampicillin(60%) and Carbenicillin(65%) respectively and were sensitive to Gentamicin(97.5%), Cephalexin(92.5%) and Kanamycin(72.5%) respectively. 6. E. coli isolated from residents of a doctorless, area were resistant to Erythromycin(100%), Streptomycin(40%) and Penicillin(50%) respectively and were sensitive to Gentamicin(100%), Kanamycin(92.5%), Tetracycline(87.5%), Oxytetracycline(87.5%), Doxycycline(87.5%), Minocycline(87.5%), Ampicillin(95%), Carbenicillin(92.5%) and Cephalexin(97.5%) respectively. 7. There were high correlations among the suscebtibilities of the 80 strains of E. coli to Tetracycline analogues(Tetracycline, Oxytetracycline, Doxycycline and Minocycline) and among susceptibilities of the 80 strains of E. coli to Penicillin analogues(Penicillin, Ampicillin and Carbenicillin). 8. There were relatively high correlations between the susceptibilities of the 80 strains of E. coli to Penicillin analogues and those to Tetracycline analogues, between the susceptibilities to Penicillin analogues and those to Streptomycin and between the susceptibilities to Tetracycline analogues and those to Streptomycin.
