Kisspeptin-10 Enhanced Egg Production in Quails Associated with the Increase of Triglyceride Synthesis in Liver

  • Our previous results showed that kisspeptin-10 (Kp-10) injections via intraperitoneal (i.p.) once daily for three weeks notably promoted the egg laying rate in quails. In order to investigate the mechanism behind the effects of Kp-10 on enhancing the egg laying rate in birds, this study focused on the alternations of lipids synthesis in liver after Kp-10 injections. 75 female quails (22 d of age) were allocated to three groups randomly, and subjected to 0 (control, Con), 10 nmol (low dosage, L) and 100 nmol (high dosage, H) Kp-10 injections via i.p. once daily for three weeks, respectively. At d 52, quails were sacrificed and sampled for further analyses. Serum $E_2$ concentration was increased by Kp-10 injections, and reached statistical significance in H group. Serum triglyceride (TG) concentrations were increased by 46.7% in L group and 36.8% in H group, respectively, but did not reach statistical significance, and TG contents in liver were significantly elevated by Kp-10 injections in a dose-dependent manner. Serum total cholesterol (Tch) concentrations significantly decreased in H group, while in H group the hepatic Tch content was markedly increased. The level of non-esterified fatty acid (NEFA), apolipoprotein A1 and B (apoA1 and apoB) were not altered by Kp-10 injections. The genes expression of sterol regulatory element binding protein-1 (SREBP-1), fatty acid synthetase (FAS), apolipoprotein VLDL-II (apoVLDL-II), cholesterol $7{\alpha}$-hydroxylase (CYP7A1) and vitellogenin II (VTG-II) were significantly up-regulated by high but not low dosage of Kp-10 injection compared to the control group. However, the expression of SREBP-2, acetyl-CoA carboxylase ($ACC_{\alpha}$), malic enzyme (ME), stearoyl-CoA (${\Delta}9$) desaturase 1 (SCD1), apolipoprotein A1 (apoA1), fatty acid binding protein 2 (FABP2), 3-hydroxyl-3-methyl glutaryl-coenzyme A reductases (HMGCR), estrogen receptor ${\alpha}$, ${\beta}$($ER{\alpha}$ and ${\beta}$) mRNA were not affected by Kp-10 treatment. In line with hepatic mRNA abundance, hepatic SREBP1 protein content was significantly higher in H group. Although the mRNA expression was not altered, the content of $ER{\alpha}$ protein in liver was also significantly increased in H group. However, SREBP-2 protein content in liver was not changed by Kp-10 treatment. In conclusion, exogenous Kp-10 consecutive injections during juvenile stage significantly advanced the tempo of egg laying in quails, which was associated with the significant elevation in hepatic lipids synthesis and transport.

Identification of CM1 as a Pathogenic Factor in Inflammatory Diseases and Cancer

  • Background: CM1 (centrocyte/-blast marker 1) was defined by a mAb against concavabalin-A (ConA) activated PBMC. It is expressed in germinal center of human tonsil and on the surface of activated PBMC as well as cancer cells. Recently, increased productions of pro-inflammatory mediators were detected from activated PBMC by CM1 ligation. Methods: However, there is a limitation to explain the exact role of CM1 on inflammation and its related mechanisms, since the identity of CM1 is still not clarified. In our previous study, we have already confirmed that soluble form of CM1 was produced by Raji. Therefore, we performed Q-TOF analysis after immunoprecipitation of concentrated Raji culture supernatant using anti-CM1 mAbs. Results: As a result, we found that CM1 is identical to enolase-1(ENO1), a glycolytic enzyme, and we confirmed that results by silencing ENO1 using siRNA. It was also confirmed through competition assay between anti-CM1 and anti-ENO1 mAbs. Finally, we investigated the possible role of CM1 in inflammatory response and cancer. The ligation of CM1 on Raji cells with anti-CM1 mAbs induces the extensive production of prostaglandin $E_2(PGE_2)$. In addition, the increased activity of matrix metalloproteinase (MMP)-2/9 was shown in NCI-N87, stomach cancer cell line by CM1 stimulation. Conclusion: CM1 is identical to ENO1 and it might be an important role in the regulation of inflammatory responses.

Ficin 처리 우육의 소화에 관한 연구 (Studies on the Digestion of Beef by Ficin Treatment)

  • 전보에서 무화과 latex에서 ficin을 분리 정제한 바 있다. 본 연구에 있어서 조 ficin을 우육에 처리하여 그의 연육제로서의 단백질 분해효과를 살피기 위하여 ficin을 농도별로 우육에 작용했을 때 생기는 유리아미노산 및 질소화합물의 함량을 측정 하였으며 이들을 숙성 및 조리시의 함량과 비교검토하였으며 그 결과는 다음과 같다. 유리 아미노산의 함량은 $1^{\circ}C$$8^{\circ}C$에서 숙성시 처리온도 및 시간에 따라 증가하였고 생우육에 ficin 처리시는 더욱 증가하였으며 증가율 또한 현저하게 높았다. 조리후 ficin 처리의 경우는 "삶기", "찌기", "굽기"의 경우중 찌기가 유리아미노산이 가장 많이 증가하였으며 또한 아미노산 증가량은 숙성중($1^{\circ}C$, 3일) 생육에 ficin 처리 (0.1% 8시간) 조리후 ficin 처리(0.1% 2시간)을 비교하였을 때 각각 13%, 292%, 137%로서 ficn 처리시의 두 group이 숙성중인 것보다 월등히 증가하였다. 총질소, 비단백태질소 및 암모니아태질소의 함량 변화는 $1^{\circ}C$$8^{\circ}C$에서 숙성시 처리온도와 시간에 따라 증가하였으며 생우육에 ficin 처리시는 더욱 증가하였으며 그 증가율이 현저하게 높았다. 조리후 ficin의 처리 경우에는 "삶기", "찌기", "굽기"의 경우에서 "찌기"가 암모니아태질소를 제외하고는 그 수치가 가장 높았다. 또한 숙성중($1^{\circ}C$, 3일)인 생육과 ficin 처리시의 생육의 총질소에 대한 비단백태질소의 증가율을 비교하였을 때 ficin 처리한 생육이 숙성중인 생육보다 크게 증가하였다.

Conjugated Linoleic Acid가 대장암 세포인 HT-29의 증식에 미치는 영향 (Effect of Conjugated Linoleic Acid on the Proliferation of the Human Colon Cancer Cell Line, HT-29)

  • Conjugated linoleic acid(CLA) is a group of positional and geometric isomers of linoleic acid(LA) and exhibits anticarcinogenic activity in multiple experimental animal models. Cis-9,trns-11(c9t11) and trans-10,cis-12(t10c12) CLA are the principal isomers found in foods. The present study was performed to determine whether CLA and the two isomers inhibits HT-29 cell proliferation and to assess whether such an effect was related to changes in secretion of eicosanoids. Cells were incubated in serum-free medium with various concentrations(0 to 20$\mu$M) of CLA or LA. CLA inhibited cell proliferation in a dose-dependent manner, with maximal inhibition(70 $\pm$ 1%) observed at 20$\mu$M concentration after 96 hours. However, LA had no effect at the same concentration range. To compare the ability of c9f11 and t10c12 to inhibit cell proliferation, cells were incubated with increasing concentrations(0 to 4$\mu$M) of these isomers. T10c12 inhibited cell proliferation in a dose-dependent manner. A 66 $\pm$ 2% decrease in cell number was observed within 96 hours after addition of 4$\mu$M t10c12. By contrast, c9t11 had no effect. The concentrations of CLA and the two isomers in the plasma membrane were increased when they were added to the incubation medium. However, they did not alter the levels of arachidonic acid in plasma membrane. To assess whether the proliferation inhibiting effect of CLA was related to changes in eicosanoid production, prostaglandin E$_2$(PGE$_2$) and leukotriene B$_4$(LTB$_4$) concentrations in conditioned media were estimated by a competitive enzyme immunoassay. Both CLA and t10c12 increased the production of materials reactive to PGE$_2$ and LTB$_4$ antibodies in a dose-dependent manner. By contrast, c9t11 had no effect. These results indicate that inhibition of HT-29 cell proliferation by CLA is attributed to the effect of the t10v12 isomer. The materials reactive to PGE$_2$ and LTB$_4$ antibodies may inhibit growth stimulatory effect of arachidonic acid-derived eicosanoids on HT-29 cell proliferation.

백색목재부후균중 Biopulping에 이용가능한 선택적 리그닌분해균의 스크리닝 (Screening of White Rot Fungi with Selective Delignification Capacity for Biopulping)

  • 선택적 리그닌 분해능을 가지고 생물펄프공정에 사용가능한 백색부후균을 얻기 위하여 94종류의 목재부후균을 검정하였고 선발된 7개종에 대하여 활엽수와 침엽수의 부후능력을 측정하였다. 우선 백색부후균은 셀룰로오즈 분해효소, 페놀산화효소, laccase, peroxidase 등의 효소활성을 간단한 방법으로 검정하여 선발하였는데, Bavendamm test에서 양성을 나타내는 대부분의 균들은 syringaldazine을 사용한 laccase test에서도 강한 반응을 나타낸 반면, 음성반응을 나타낸 대부분의 균들은 laccase와 peroxidase test에서도 음성반응을 나타내었다. 선택적 리그닌 분해능력을 지닌 부후균을 선발하기 위하여 부후균을 은사시나무와 일본잎갈나무(낙엽송) 목재블럭에 접종하여 12주간 배양한 후에 부후된 목재의 중량감소율, 리그닌 량의 감소, 형태적 변화들을 화학분석과 주사전자현미경을 통하여 분석하는 목재부후 실험을 실시하였다. 이 실험에서 사용한 거의 모든 균주는 목재블럭의 중량감소율이 일본잎갈나무 보다 은사시나무에서 2배이상 높게 나타났으며 균을 접종하지 않은 목재블럭에서는 중량감소가 전혀 나타나지 않았다. Ceriporiopsis subvermispora와 Phanerochaete chrysosporium이 다른 균주에 비해서 침엽수와 활엽수의 리그닌을 모두 잘 분해시키는 것으로 나타났으나 분해 능력은 Ceriporiopsis subvermispora가 더욱 우수하였다. Bjerkandera adusta와 미동정된 2균주는 은사시나무에서만 상대적으로 높은 리그닌 분해능력을 나타내었다. B. adusta는 모든 세포벽 성분을 동시에 분해시켜서 2차세포벽을 얇게 만들었으나 다른 균주들은 선택적 리그닌 분해력을 나타내어 두 세포의 세포벽 사이에 위치하는 중벽에 존재하는 리그닌을 분해시켜서 세포를 분리시키는 것이 관찰되었다.

Chito-oligosaccharides as an Alternative to Antimicrobials in Improving Performance, Digestibility and Microbial Ecology of the Gut in Weanling Pigs

  • A total of 126 crossbred weanling pigs (average body weight of $6.3{\pm}0.3$ kg) were used to investigate the effect of chito-oligosaccharide (COS) on growth performance, nutrient digestibility, pH of gastro-intestinal tract (GI), intestinal and fecal microflora of young piglets. Pigs were allocated to three dietary treatments based on body weight and gender in a single factorial arrangement. Treatments were control (No COS), T1 (0.2% COS during starter (6-13 kg) and 0.1% COS during grower (13-30 kg) phases, and T2 (0.4% COS during starter (6-13 kg) and 0.3% COS during grower (13-30 kg) phases, respectively. Each treatment had 3 replicates and 14 pigs were raised in each pen. COS is a low molecular weight water-soluble chitosan that can be obtained from chitin of the crab shell after deacetylation with concentrated sodium hydroxide at high temperature and then further decomposition by chitosanase enzyme in the presence of ascorbic acid. For the starter and grower periods, there were no significant differences (p>0.05) in average daily gain (ADG) and feed to gain ratio among treatments. However, during the overall period (6-30 kg), T2 showed better (p<0.05) feed to gain ratio than other treatments. A digestibility study was conducted at the end of grower phase which showed improvement (p<0.05) in DM and crude fat digestibility in T2 over the control. At 25 kg body weight, 6 pigs per treatment (2 per replicate) were sacrificed to determine the effect of diets on pH and microbial count at different sections of the GI tract. The pH of the cecal contents in pigs fed 0.1% COS was higher (p<0.05) than in the other treatments. Total anaerobic bacterial number increased from cecum to rectum in all treatments. The weekly total bacterial counts showed higher (p<0.05) in feces of pigs fed COS than that of untreated pigs at the $8^{th}$ week. The number of fecal E. coli in untreated pigs at $4^{th}$ wk was 7.35 log CFU/g compared to 6.71 and 6.54 log CFU/g in 0.1 and 0.3% COS-treated pigs, respectively. Similarly, at $8^{th}$ wk, fecal clostridium spp. were lower in pigs fed 0.3% COS (5.43 log CFU/g) than in untreated pigs (6.26 log CFU/g). In conclusion, these results indicated that chito-oligosaccharide could improve feed efficiency in young pigs and inhibited the growth of harmful bacteria.

미생물을 이용한 저식염 멸치젓의 속성발효에 관한 연구 1. 젓갈에서 분리한 단백질분해균 및 단백질분해효소의 생화학적 특성 (Studies on the Processing of Rapid Fermented Anchovy Prepared with Low Salt Contents by Adapted Microorganism 1. Biochemical Characterization of Proteolytic Bacteria and their Extracellular Pretense Isolated from Fermented Fish Paste)

  • 산업적으로 속성젓갈을 대량 생산하기 위하여, 우선 시중 젓갈에서 단백질분해력이 강한 균주 4 종을 분리하여 속성 분해정도와 분해 생성물의 풍미를 비교 검토한 결과 B. subtilis p-4 및 B. licheniformis p-5 균이 가장 양호하였으며, 이들 균주와 그 pretense의 특성은 다음과 같다. pH 7.0, $40^{\circ}C$ 식염$1\%$ 농도에서 균체 및 조효소 활성이 가장 양호하였으며, 이때의 비증식속도는 p-4 균 및 p-5 균이 각각 0.48/hr, 0.49/hr이었고, 최대 효소활성 농도는 p-4 는 30시간 후 335n mole-Tyr/min.ml, p-5는 28시간 후 300n mo1e-Tyr/min.ml이였다. 그리고 sephadex G-100 겔 여과에 의한 효소 정제도는 조효소에 비해 비활성이 p-4 및 p-5 균 protease의 경우 각각 25.4배, 8.6배씩 증가하였으며, pH 7.0, $50^{\circ}C$에서 활성이 가장 높았다. 또한 겔여과에 의한 분자량은 p-4가 18,000, p-5 protease가 30,000이었고, 저해제의 효과에서는 두 Protease 모두 $Ni^{2+},\;Cu^{2+}$ 이온과 EDTA, o-phenanthroline에 의해 강하게 활성이 소실되는 것으로 보아 metal chelator sensitive neutral protease에 속하는 것으로 추정되었다.

동물모델을 이용한 짚신나물 물 추출물의 위염 예방효과 및 유전독성 평가 (Preventive Effect of the Water Extract of Agrimonia pilosa Ledeb and Micronucleus Assay-Based Evaluation of Genotoxicity in Gastritis Animal Models)

  • Background: Agrimonia pilosa Ledeb has been used as a traditional medicine for the treatment of hematuresis and uterine bleeding in Korea. It has been reported to have anti-obesity, anti-diabetes and anti-inflammaotry effect by regulating the inflammatory signaling pathway. However, the preventive effect of Agrimonia pilosa Ledeb on gastritis has not been elucidated. Thus, in the present study, we evaluated the effects of the water extract of Agrimonia pilosa Ledeb (APW) using HCl/EtOH-induced gastritis rat models. Method and Results: Gastritis was induced in rats by HCl/EtOH administration. The rats in each group were orally administered with two doses of APW (100 and 500 mg/kg). Omeprazole was used as a positive control drug. An enzyme-linked immunosorbent assay (ELISA) was used to measure the prostaglandin $E_2$ ($PGE_2$) levels in stomach. The treatment with 500 mg/kg APW reduced the gastric ulcer area. The APW treatment prevented a decreased in $PGE_2$ concentration induced by HCl/EtOH in rats. In the micronucleus test, the ratio of micronucleated polychromatic erythrocytes to polychromatic erythrocytes showed no significant change in the APW-treated group compared with the control group. Conclusions: These results indicate that APW could be used to prevent the gastritis caused by the HCl/EtOH-induced damage to stomach lining. In addition, the APW treatment showed no significant change in results of the micronucleus test. However, further experiments are required to determine how APW influenced the secretion of mucus and gastric acid using the chromosome aberration test and bacterial reverse mutation assay.

Environment-wide association study of elevated liver enzymes: results from the Korean National Environmental Health Survey 2018-2022

  • Background: Environmental exposure is characterized by low concentration, chronic, and complex exposure. Traditional epidemiological studies show limitations in reflecting these characteristics since they usually focus on a single or very limited number of exposure factors at a time. In this study, we adopted the methodology of environment-wide association study (EWAS) to figure out the association of human liver function with various environmentally hazardous substances. Methods: We analyzed 2,961 participants from the Korean National Environmental Health Survey Cycle 4 (2018-2020). Using generalized linear model (GLM) analysis, we analyzed the association of 72 variables with 3 liver function indices (aspartate aminotransferase [AST], alanine aminotransferase [ALT], and gamma glutamyl transferase [GGT]). Finally, we visualized our results with Manhattan plot. Results: In GLM analysis, perfluorooctanesulfonate were positively associated with ALT (odds ratio [OR]: 2.2; 95% confidence interval [CI]: 1.39-3.46; padjusted = 0.0147) and perfluorodecanoic acid showed positive association with GGT (OR: 2.73; 95% CI: 1.36-5.5; padjusted = 0.0256). Plasma mercury showed positive association with GGT (OR: 1.45; 95% CI: 1.14-1.84; padjusted = 0.0315). Using a plastic container while keeping food in the refrigerator was associated with elevated GGT compared to using a glass container (OR: 1.51; 95% CI: 1.16-1.95; padjusted = 0.0153). 2-ethyl-5-oxohexyl phthalate, showed a negative trend with all 3 indices, with AST (OR: 0.54; 95% CI: 0.39-0.73; padjusted = 0.00357), ALT (OR: 0.5; 95% CI: 0.34-0.75; padjusted = 0.036), GGT (OR: 0.55; 95% CI: 0.4-0.76; padjusted = 0.00697). Bisphenol S and frequent use of sunblock cream showed negative association with ALT (OR: 0.77; 95% CI: 0.66-0.89), and GGT (OR: 0.25; 95% CI: 0.11-0.55), respectively. Conclusions: We conducted an exploratory study on environmental exposure and human liver function. By using EWAS methodology, we identified 7 factors that could have potential association with liver function.

젓갈등속(等屬)의 정미성분(呈味成分)에 관(關)한 미생물학적(微生物學的) 및 효소학적(酵素學的) 연구(硏究) (Microbiological and Enzymological Studies on the Flavor Components of Sea Food Pickles)

  • 우리나라에서 알려진 젓갈 30여종(餘種)에서 그중(中) 대표적(代表的)인 것으로 알려진 조기젓. 조개젓(고식(高食) 염성(鹽性)인 20%내외(內外))과 굴젓, 오징어젓(저식염성(低食鹽性)인 10%내외(內外))의 4종(種)을 시료(試料)로하여 일반성분(一般成分) 분석(分析), microflora, 주요발효미생물(主要醱酵微生物)의 동정(同定) 및 효소(酵素)의 특성(特性)을 조사(調査)하는 동시(同時)에 숙성(熟成)에 관여(關與)하는 미생물(微生物)의 효소작용(酵素作用)이 정미성(呈味性) 5’-mononucleotide 및 amino 산(酸)의 생성(生成)에 미치는 영향(影響)을 조사(調査)하여 다음과 같은 결과(結果)를 얻었다. (1) 4종(種) 젓갈에 대(對)한 microflara를 조사(調査)한 결과(結果)는 다음과 같다. a) 젓갈담금 $1{\sim}2$개월(個月)의것은 생균수(生菌數) total counts가 $10^7$이었으며 담금후(後) 6개월(個月)의것은 $10^4$이였다. b) 젓 갈 담금 $1{\sim}2$개월(個月)의것은 Micrococcus 속(屬)이 $10{\sim}20%$, Brevibacterium 속(屬)이 $10{\sim}20%$, Sarcina 속(屬)이 $0{\sim}30%$, Leuconostoc 속(屬) $20{\sim}30%$, Bacillus 속(屬)이 30%내외(內外) Pseudomonas 속(屬)이 $0{\sim}10%$ Flavobacterium 속(屬)이 $0{\sim}10%$ Yeast가 $0{\sim}20%$인 분포상(分布相)을 나타냈다. c) 젓갈숙성(熟成)에 관여(關與)하는 미생물(微生物)로서 초기이후(初期以後)에는 주(主)로 내염성(耐鹽性) Bacillus subtilis, Leuconostoc mesenteroides, Pediococcus halophilus, Sarcina litoralis 등(等)임을 동정(同定)하였고 이들의 효소활성(酵素活性)이 젓갈숙성(熟成)을 지배(支配)하는 요소(要素)임을 알 수 있었다. d) 숙성(熟成)에 관여(關與)하는 세균중(細菌中)에 Sarcina litoralis 8­14 및 8-16. 양균주(兩菌株)는 영양요구성(榮養要求性)이 현저(顯著)하여 전자(前者)는 purine, pyrimidine base 및 cystine 후자(後者)는 purine, pyrimidine base 및 glutamic acid가 공존(共存)하여야 생육(生育)이 가능(可能)하였다. (2) 젓갈원료(原料) 및 젓갈에서 분리(分離)한 관여미생물(關與微生物)에 대(對)하여 효소특성(酵素特性)을 검토(檢討)한 결과(結果)는 다음과 같다. a) 젓갈원료중(原料中)에 protease는 소량(少量) 존재(存在)하나 식염농도(食鹽濃度) 7%에서 protease 활성(活性)이 현저(顯著)하게 조해(阻害)를 받아 그 효소활성(酵素活性)의 $30{\sim}60%$나 감소(減少)하였다. b) 내염성세균(耐鹽性細菌)인 Bacillus subtilis 7-6, 11-1, 3-6 9-4 등(等) 4 균주(菌株)는 complete media에서 생성(生成)한 protease 활성(活性)은 식염농도(食鹽濃度) 7%에서 약간 조해(阻害)를 받아 활성(活性)이 $10{\sim}30%$가 감소(減少)하였고 Sarcina litoralis 8-14 및 8­16 양균주(兩菌株)는 동배지(同培地)에서 생성(生成)한 protease 활성(活性)은 식염농도(食鹽濃度) 7%에서 그 발효활성(醱酵活性)의 $10{\sim}20%$가 감소(減少)하였다. c) 젓갈 원료중(原料中)의 단백질(蛋白質)은 자가효소(自家酵素)에 의한것보다 주(主)로 젓갈숙성중(熟成中) 관여미생물(關與微生物)의 protease에 의(依)한 가수분해(加水分解)로 유리(遊離) amino 산(酸)이 생성(生成)됨을 알 수 있었다. d) 젓갈원료(原料) 및 젓갈의 RNA-depolymerase는 젓갈중(中) RNA를 nucleoside 및 유리인산(遊離燐酸)까지 분해(分解)하므로 정미성(呈味性) 5'-mononucleotide로서 축적(蓄積)되기 어렵다 e) 조개젓에서 분리(分離)한 Bacillus subtilis 3-6 균주(菌株)가 생성(生成)한 효소(酵素)는 RNA를 분해(分解)하여 5’-mononucleotide로 축적(蓄積)하므로 이 균주(菌株)가 생성(生成)한 RNA 분해효소(分解酵素)는 5’-phosphodiesterase임을 밝혔다. f) Bacillus subtilis 3-6 strain의 효소생성배지중(酵素生成培地中)에 corn steep liquor 0.5% 농도(濃度)로 첨가(添加)한 구(區)와 조개젓 0.5% 농도(濃度)로 첨가(添加)한 구(區)에서 5’-phosphodiesterase 생성증가(生成增加)를 보았으므로 corn steep liquor 및 조개젓 성분(成分)이 각각(各各) 5’-phosphodiesterase 생성증가제(生成增加劑)가 됨을 밝혔으며 또한 이 효소(酵素)는 10%의 식염농도(食鹽濃度)에서 활성조해(活性阻害)를 받아 $10{\sim}30%$에 해당(該當)되는 활성(活性)이 감소(減少)되었고 식염농도(食鹽濃度) 20%에서 이 효소활성(酵素活性)의 $40{\sim}60%$가 감소(減少)함을 나타냈다. 그리고 이 균주(菌株)의 5’­phosphodiesterase에 의(依)하여 젓갈중(中) 5’-mononucleotides가 생성(生成)되는 것이라고 생각된다. (3) 젓갈의 유리(遊離) amino 산(酸)을 Autoanalyzer로 정량(定量)한 결과(結果)는 다음과 같다. a) 조개젓은 산성(酸性) amino 산(酸)인 glutamic acid, asp artic acid 함량(含量)이 다른 젓갈보다 $2{\sim}10$배(倍) 정도(程度)로 현저하게 많아서 조개젓의 구수한 맛이 발현(發現)되는 것이라고 생각(生覺)된다. b) 조기젓에는 basic amino acid인 arginine, histidine함량(含量)이 다른 젓갈보다 특이(特異)하게 많았다. c) 오징어젓은 함류황(含硫黃) amino산(酸)인 cystine함량(含量)이 다른 젓갈의 $17{\sim}130$배(倍), methionine함량(含量)이 $7{\sim}17$배정도(倍程度)로 현저하게 많았다. d) 굴젓에는 필수(必須) amino 산(酸)인 lysine, threonine iso leucine, leucine 함량(含量)이 다른 젓갈보다 현저하게 많을 뿐아니라 또한 감미성(甘味性) amino 산(酸)인 alanine 함량(含量)이 다른 젓갈들보다 4배(倍), glycine 함량(含量)이 $3{\sim}14$배정도(倍程度)로 함유(含有)하고 있어 이것이 굴젓 특유(特有)의 단맛을 발현(發現)하는 것이라 생각(生覺)된다. (4) 4종(種) 젓갈에 대(對)한 5'-mononucleotide를 ion exchange column chromatography로 측정(測定)한 결과(結果)는 다음과 같다. a) 조개젓에는 5'-adenylic acid, 3’-adenylic acid 가 많이 들어있고 5'-inosinic acid가 미량(微量)들어 있다. b) 굴젓에는 5'-adenylic acid와 3’-adenylic acid가 다른 mononucleotides보다 현저하게 많았다. c) 오징어젓에는 5'-adenylic 및 3'-adenylic acid만이 함유(含有)되어 있는데 이것은 패류(貝類)의 젓갈들과 같 이 무척추연체동물(無脊椎軟體動物)이며 이것들은 adenylic deaminase가 결여(缺如)되어 있어 mononucleotide중(中) adenylic acid의 함량(含量)이 높아 adenylic acid-type 임을 알았다. d) 조기젓 중(中)에는 5'-inosinic acid 함량(含量)이 다른젓갈보다 현저하게 많았으므로 이것은 다른 척추동물(脊椎動物)의 어육(魚肉)이나 수육(獸肉)과 같이 adenylic deaminase가 있어 5'-inosinic acid가 많은 inosinic acid-type임을 알았다. (5) 정미성(呈味性) amino 산(酸) 및 5'-mononucleotide의 조성(組成)과 함량(含量)으로 맛의 조화성(調和性)을 비교(比較)하여 보면 다음과 같다. a) 조기젓은 glutamic acid와 aspartic acid가 풍부(豊富)하고 소량(小量)의 5'-inosinic acid가 공존(共存)된 상태(狀態)이므로 정미성분조성(呈味成分組成)과 함량(含量)이 잘 조화(調和)되어 구수한 맛의 상승작용(相乘作用)이 적당(適當)하게 발현(發現)됨을 구명(究明)하였다. b) 조개젓의 맛은 정미성(呈味性) 5’-mononucleotides에서 유래(由來)되는것 보다는 현저하게 많이 들어있는 glutamic acid 및 aspartic acid에 기인됨을 알았다.
